1.Study of primary cells of breast cancer with special immune response induced by CIK co-cultured with self-dendritic cells

Objective To investigate effect to kill primary breast cancer cells by dendritic cell (DC)with self-freeze thawing antigens of primary breast cancer cells co-cultured with cytokine induced killer cell (CIK). Methods Self-neoplasm antigen by primary cells of breast cancer in log phase growth was prepared and DC and CIK cells was cultured from peripheral blood. The CIK of co-cultured DC loaded self-neoplasm antigen was compared to PBMC, CIK cells with self-neoplasm antigen and the single CIK cells. Results Killing efficiency of PBMC, CIK, Ag-CIK and Ag-DC-CIK were (34.35±3.28) %, (45.91±2.78) %, (50.88±3.22) %, (62.10±5.94) %. There were significant difference between Ag-DC-CIK group and CIK-Ag and CIK group to primary cells of breast cancer (P <0.01). Conclusion The CIK induced from cocultured DC loaded with self-neoplasm antigen show a special killing activity to self-primary tumor cells.
2.How to break through the spell of intensive care unit infection

Nosocomial infection is one of the most common diseases in the intensive care unit (ICU), which has received much concern due to its multi-type infection, more severe infection, difficulty in treatment and high mortality. However, the drug resistance of bacteria is increasingly serious with the wide use of the antibiotics which is no longer an ideal strategy. Instead, the control and prevention of infection from their sources is the key to reduce ICU infection. Especially for some particular nosocomial infections, such as hospital acquired pneumonia (HAP) and ventilator associated pneumonia(VAP), comprehensive measures are needed to take to break the spell of ICU infection.
3.The expression and significance of Toll-like receptors-4 in peripheral blood and placental tissuein gestational diabetes mellitus patients

Objective To investigate the expression and significance of Toll-like receptors-4 in peripheral blood and placental tissue in gestational diabetes mellitus (GDM) patients.Methods From February 2013 to February 2015.a total of 30 cases of gestational diabetes mellitus patients(GDM group) and 30 cases of normal pregnant people(health group)were selected as research objects.Peripheral blood and placental tissue of two groups were collected.The expression of TLR4 mRNA in peripheral blood mononuclear cells was detected by RT-PCR,the expression of TLR4 protein in placenta tissue was detected by immunohistochemistry.Results The expression of TLR4 mRNA in peripheral blood mononuclear cells of the observation group was significantly higher than that of the health group[(0.63±0.12) vs.(0.32±0.07)],the difference was statistically significant(t=12.223,P<0.05).The expression of TLR4 in villous trophoblast cells,decidual cells and amniotic epithelial cells in GDM group was significantly higher than that in the health group(Z=2.325,2.374,2.162,P<0.05).Conclusion The expression of TLR4 in peripheral blood mononuclear cells,villous trophoblast cells and decidual cells in gestational diabetes mellitus patients significantly enhanced,suggesting that TLR4 might be related to the e gestational diabetes mellitus.
4.The changes of D-D, t-PA and PAI in acute renal failure patients during blood purification and its clinical significance

Objective To study the dynamic changes and clinical significance of D-D, t-PA and PAI in patients with acute renal failure during the process of different methods of blood purification. Methods Thirty-seven ARF patients were divided into three groups: HD group, HDF group and HF group. Plasma D-D level, t-PA and PAI activity were determined 1 hour, 4 hours before and after treatment. Normal control group consisted of fourteen healthy people. Objective To explore the psychometric properties of the Chinese version of integrated visual and auditory continuous performance test (IVA-CPT) ,and to assess the diagnosis value of the Chinese version of IVA-CPT in two samples of children with attention deficit hyperactivity disorder (ADHD) in clinic and school.Methods 112 participants were divided into 4 groups:clinic group, clinic control group, school group and control group from school.The participants were measured by IVA-CPT and the results were compared.Results ①The integrated quotients of IVA-CPT had concordance with the dimensions of the DSM-ⅣV and ASRS.Each integrated quotients of IVA-CPT and each dimension of the DSM-ⅣV and ASRS were negatively correlated(school: r=-0.277, P<0.05;clinic: r=-0.423, P<0.05).The score of HI dimension was negatively correlated with integrated attention quotient and integrated inhibition quotient of IVA-CPT (r=-0.480, P<0.01;r=-0.452, P<0.01).② The results of the IVA-CPT and the comprehensive assessment had consistency for diagnosis of children with ADHD from the total subjects,the clinic ,and the school.And there were no difference between the sensitivity, the specificity, the coincidence rate, the missed diagnosis rate and the misdiagnosis rate of IVA-CPT when used to diagnose children with ADHD in clinic and school(x2=3.396, P>0.05).③The IVA-CPT evaluation system had good stability according to the retest of a random sample of 25 subjects(P<0.01).The pretest and posttest results of each full quotients of IVA-CPT were significantly correlated (P<0.01).Conclusion The IVA-CPT not only can be used as auxiliary diagnostic tools to diagnose children with ADHD in clinic,it can be used as auxiliary diagnostic tools to diagnose children with ADHD in the community.
6.Clinical diagnosis and surgical treatment of cervical spondylosis with proximal upper extremity amyotrophy

Objective To summarize the clinical features and diagnostic flow of cervical spondylosis with proximal upper extremity amyotrophy;and further analyze the clinical effect of cervical anterior decompression and fusion on cervical spondylosis with proximal upper extremity amyotrophy.Methods Twenty-two cases of cervical spondylosis with proximal upper extremity amyotrophy were analyzed retrospectively from June 2006 to December 2013.Seventeen males and 5 females with an average age of (55.73 ± 8.64) years (38 to 68 years) were included.The mean preoperative course of disease was (19.2 ± 21.86) months (1-72 months).Clinical symptoms,imaging findings and electrophysiological findings were analyzed.The muscular strength recovery of atrophic muscles was evaluated by Manual Muscle Testing (MMT).The clinical improvement rate was evaluated by the Japanese Orthopedic Association (JOA) score,and the clinical satisfaction was assessed at followed up.Results The muscles involved in patients of cervical spondylosis with proximal upper extremity amyotrophy are mainly the deltoid muscle,biceps and scapula levator muscle.Most cases of imaging findings showed multi-segmental degeneration,of which C4,5,C5,6 segments were most common.Neuroelectrophysiological examination showed that affected muscles experienced obvious denervation and decreased action potential.The average follow-up time was (44.14 ± 20.51) months (14 to 102 months).At the last follow-up,the JOA score (16.29 ±0.59) in 17 cases was higher than preoperative (15.12 ± 0.93),the difference was statistically significant (F=51.814,P=0.000),and the average improvement rate was 73.3%.MMT assessment showed that 19 patients (86.4%) in this group had muscle strength recovery for more than 1 grade at the last follow-up.The average clinical satisfaction was 83.7%.Conclusion The clinical diagnosis of cervical spondylosis with proximal upper extremity amyotrophy requires a combination of clinical symptoms,imaging findings and neurophysiological examination results for comprehensive judgment.Anterior cervical decompression and fusion in the treatment of cervical spondylosis with proximal upper extremity amyotrophy patients can achieve good clinical results.
7.Determination of Residual Organic Solvents in Meropenem by Headspace Gas Chromatography

OBJECTIVE:To develop a headspace GC method for determination of residual organic solvents invovling methanol,alcohol,acetone,acetonitrile,dichloromethane and n-butanol in meropenem.METHODS:The residual organic solvents were separated on DB-624 capillary column using temperature programming.FID was used as detector with a temperature of 250 ℃ with nitrogen as carrier gas and water as solvent.The inlet temperature was 200 ℃.RESULTS:The linear range were 12~600 ?g?mL-1 (r=0.999 8) for methanol,20~1 000 ?g?mL-1 (r=0.999 9) for alcohol,20~1 000 ?g?mL-1 (r=0.999 9) for acetone,1.64~82 ?g?mL-1 (r=0.999 8) for acetonitrile,2.4~120 ?g?mL-1 (r=0.999 6) for dichloromethane and 20~1 000 ?g?mL-1 (r=0.999 9) for n-butanol with RSD of 1.0%,2.2%,0.9%,2.2%,1.6%,1.1%,respectively.The average recoveries were ranged from 98.8% to 99.6%.The LOD were about 0.06~0.43 ng.The residual levels of six organic solvents in 3 batches samples were all up to the standard stated in Chinese Pharmacopeia (2005 edition).CONCLUSION:The method is simple,accurate,sensitive and reproducible for determination of residual organic solvents in meropenem.
8.Effects of salmeterol fluticasone on the glucose metabolism and bone density of the elderly patients with chronic obstructive pulmonary disease combined with type 2 diabetes mellitus

ObjectiveTo observe the effects of salmeterol fluticasone on the glucose metabolism and bone density of the elderly patients with chronic obstructive pulmonary disease (COPD) combined with type 2 diabetes mellitus (T2DM).MethodsThirty-one patients with COPD combined with T2DM were divided into 2 groups by random digits table,14 cases in control group were given conventional therapy,17 cases in experimental group were given conventional therapy and fluticasone propionate 50/500 μg,twice a day,for 3 months.The fasting blood glucose,postprandial 2 hours glucose,glycosylated hemoglobin,plasma cortisol and bone density respectively before and after treatment were detected.ResultsIn experimental group,the fasting blood glucose,postprandial 2 hours glucose,glycosylated hemoglobin and bone density before treatment was (5.25 ± 0.21 ) mmol/L,(7.14 ± 0.33 ) mmol/L,(5.58 ± 0.26 )%,( 1.96 ± 0.11 ) g/cm2,and after treatment was(5.31 ± 0.27 ) mmol/L,(7.22 ± 0.29 ) mmol/L,(5.67 ± 0.23 )%,(2.03 ± 0.15 ) g/cm2,there was no significant difference (P > 0.05).In control groups,the fasting blood glucose,postprandial 2 hours glucose,glycosylated hemoglobin and bone density before treatment was (5.33 ± 0.35) mmol/L,( 7.26 ± 0.29 ) mmol/L,( 5.62 ± 0.19 )%,( 1.88 ± 0.20 ) g/cm2,and after treatment was ( 5.36 ± 0.31 ) mmol/L,(7.30 ± 0.35 ) mmol/L,( 5.69 ± 0.26 )%,( 1.98 ± 0.17 ) g/cm2,there was no significant difference (P > 0.05 ).There was no significant difference in plasma cortisol before and after treatment in two groups (P >0.05).ConclusionInhaling salmeterol fluticasone for elderly patients with COPD combined with T2DM is safe.
9.Discussion in measuring the curvature of cervical endplate based on reverse engineering software

Objective Method discussion of measuring curvature of cervical endplate based on reverse engineering software.Methods The CT scan images data (From C2 to C7) of 8 normal males were selected and inputted into Mimics 10.0 software for 3D reconstruction and separation (cervical spine is divided into C1-C7 isolated reconstructed 3D images).Another software NX Imageware 13.2 is used for further curvature measuring in lines named left,middle,right,anterior,middle,posterior which are derived from each superior and inferior endplate.In each line,15 different points which can perfectly show morphologic characters of the endplate are selected for statistic comparing between different points in the same endplate,different lines in the same endplate,different endplates in the same individual and the same endplate in different individuals.In addition,the correlation between the concavity and the width and depth of endplate is also analyzed.Results From inferior endplate of C2 to that of C6,except superior endplate of C7,there is significant difference between different points in the same line (P<0.05),while it is opposite in the compare between lines in the same endplate,different endplates in the same individual and the same endplate (except inferior endplate of C4) in different individuals (P>0.05).It showed significant correlation between the concavity and the width and depth of endplate.Conclusion Reverse engineering software is feasible to be used in evaluate morphologic characters of normal cervical endplate as a new method for measuring the detail parameters of them,it also explore the laws of the curvatures of cervical endplate initially and both of them will be of great use for cervical interbody cage design.
