1.Study on HPLC fingerprint chromatograms of Arisaematis Rhizoma.

The fingerprint chromatograms of Arisaematis Rhizoma were established by HPLC. The analysis was performed on a Lichrospher C18 (4.6 mm x 200 mm, 5 microm) column with acetonitrile-water (containing 0.1% acetic acid) as mobile phase at a flow rate of 1.0 mL x min(-1). The detection wavelength was set at 270 nm, and the column temperature was 30 degrees C. The similarities of the fingerprint chromatograms were calculated over 0.9 between 11 batches of Arisaematis Rhizoma samples by analyzing 14 common peaks with adenosine as reference substance. However, their fingerprint chromatograms were significantly different from those of Pinellia pedatisecta and P. ternate. Adenine, hypoxanthine, xanthine, uridine, guanosine, adenosine, schaftoside, and isoschaftoside were identified by comparing the retention times and their ultraviolet spectra. The method is repeatable, exclusive and can be used for identification and evaluation of Arisaematis Rhizoma.

2.Analysis of the current situation of work-family conflicts in Emergency nurses and its influencing factors

Objective? To explore the status quo of work-family conflicts among nurses in Emergency Departments and analyze its influencing factors, so as to provide basis for nursing managers to formulate intervention measures. Methods? A cluster sampling method was used, from June 2018 to December 2018, a total of 137 Emergency nurses from 5 Class Ⅲ Grade A hospitals in Jilin Province were selected as the research objects in this study. Nurses' General Information Questionnaire and Work-family Conflict Scale were used to investigate them. Results? The total score of Work-Family Conflict Scale of 137 emergency nurses was (55.67± 8.06). The scores of each dimension of work-family conflict were higher than that of family-work conflict, and the differences were statistically significant (P<0.05). Multivariate linear regression analysis showed that gender, marital status, fertility and frequency of night shift per month were the influencing factors of work-family conflict among emergency nurses (P< 0.05). Conclusions? The work-family conflict of emergency nurses is not optimistic. The work-family conflict is greater than the family-work conflict. Nursing managers should pay enough attention to this problem and pay more attention to the family situation of nurses in order to alleviate the work-family conflict of nurses.
3.The prevention effect of intermittent pneumatic compression pump on venous thrombosis in patients undergoing knee joint replacement

Objective To investigate the prevention effect of intermittent pneumatic compression pump on DVT in patients undergoing knee joint replacement.Methods A total of 318 patients with knee joint replacement in our hospital were selected as study objects,and 108 cases with routine nursing were as control group,and 210 cases with intermittent pneumatic compression were as observation group.The changes of D-D,PLT and Hb,and DVT of two groups were compared.Results PLT and D-D after 6 d of the operation in the observation group was lower than that in the control group,the difference was statistically significant(P <0.05).Venous velocity of lower limb after 6 d of operation in the observation group was quicker than that in the control group,the detection rate of DVT was lower than that in the control group,DVT diameter was less than that in the control group,the differences were statistically significant (P < 0.05).Conclusion Intermittent pneumatic compression pump can reduce PLT,D-D and the incidence of DVT after knee replacement.

5.Job engagement and its influencing factors of nurses working in emergency department

Objective? To describe the current situation of nurses' job engagement in emergency department and analyze its main influencing factors. Methods? A total of 146 emergency nurses from 6 Class 3 hospitals from June to September 2017 were selected by convenience sampling method. The nurses were investigated using general data questionnaires, Utrecht Work Engagement Scale (UWES), and Nursing Organization Atmosphere Scale. Results? A total of 146 questionnaires were distributed and 138 valid questionnaires were collected. The average score of UWES for the 138 emergency nurses was (70.34±13.72). The results of multiple linear regression analysis showed that nurses with or without children, education level, monthly income, physical condition, frequency of night shifts, and nursing organization atmosphere were the main factors affecting the job engagement of nurses in emergency department (P< 0.05). Conclusions? The current situation of emergency work nurses' job engagement is at a medium level. Managers should attach great importance to this issue and adopt more individualized intervention programs and management to increase the level of job engagement and work enthusiasm of nurses in emergency departments.

TotaLLy 150 patients who received chemotherapy after surgery for coLorectaL cancer in a CLassⅢ Grade A hospitaL in a city from January 2017 to February 2018 were seLected as convenient sampLing and investigated with the generaL information questionnaire, Chemotherapy Patient Comfort ScaLe and SociaL Support Rating ScaLe (SSRS). Univariate anaLysis, Pearson correLation anaLysis and muLtivariate Linear regression anaLysis were used to expLore the factors affecting the patients' comfort LeveLs. ResuLts? The comfort LeveL of these patients with coLorectaL cancer after surgery totaLed (72.36±9.34). Comfort LeveLs were positiveLy correLated with sociaL support (P< 0.05). MuLtivariate Linear regression anaLysis showed that educationaL background, enterostomy received or not, compLication, treatment protocoL and sociaL support were the main factors affecting the patients' comfort LeveL. ConcLusions? The comfort LeveL of patients receiving chemotherapy after surgery for coLorectaL cancer stands at a medium LeveL. MedicaL and nursing workers shouLd improve sociaL support for the patients and take more targeted interventions based on the patients' actuaL situation to improve their comfort LeveLs and quaLity of Life.
7.Study on the impact of Trp1707Ser mutation on the binding mechanism of rF VIII light chain with VWF.

OBJECTIVETo disclose the impact of Trp1707Ser mutation on the binding mechanism of rFVIII light chain (rFVIII LC) with VWF.
METHODSUsing long-chain PCR technique, we constructed rFVIII LC plasmids of both wild type and Trp1707Ser mutant type. BL21 competent cells were used for protein expression. Gradient renaturation was employed to refold protein. SDS-PAGE and Western blot were performed to identify the molecular weight of expressed protein. GST-Sefinose was used for protein purification and surface plasmon resonance (SPR) was employed to detect binding of B-domain-deleted rFVIII (BDD-rFVIII), wild and mutant rFVIII LC with VWF, respectively.
RESULTSThe results of SDS-PAGE and Western blot showed a molecular weight of 110×10(3) of expressed proteins, which were consistent with objective proteins. The expression quantity of wild type was higher than that of mutant type. A concentration-dependent combination of the 3 testing proteins with VWF was found. The KD value of BDDrFVIII (12.2) was lower than that of both rFVIII LCs (wild type 48.9 and mutant type 46.3), whereas there was no discrepancy between wild rFVIII LC and mutant rFVIII LC.
CONCLUSIONTrp1707Ser mutation didn't impact the binding of rFVIII LC expressed by BL21 competent cells with VWF. The heavy chain played a more important role in impacting the binding of FVIII with VWF.
