Objective To evaluate the application value of MiniFilerTM kit in LCN-STR genotyping in forensic science.Methods Compared the testing results of MiniFilerTM and IdentifilerTM kits in 49 general quantity biological samples and 39 LCN-biological samples,including blood stains,sperm stains,bones,and so on.Meanwhile the sensitivity of these two kits were also compared.Results Full concordance between IdentifilerTM and MiniFilerTM kits was observed in all of 49 general quantity biological samples.For 39 LCN-biological samples,only 22 samples could be genotyped in partial loci and 17 samples negative with the IdentifilerTM kit,while 30 samples could be genotyped in all loci,5 samples in partial loci and 4 samples negative with the MiniFilerTM kit.Therefore,there was a higher success rate for LCN-biological samples typing with MiniFilerTM kit than with IdentifilerTM kit.In addition,the sensitivity of the MiniFilerTMkit was also a little higer than the IdentifilerTMkit.Conclusion The results demonstrated that the MiniFilerTM kit can markedly improve the typing success rate of LCN-biological samples,and is suitable for analyzing difficult biological samples in forensic practice.

Background and purpose:Multiple microRNAs (miRNAs) are abnormally expressed in breast cancer and play an important role in the regulation of breast cancer. miRNAs may be a new target for the treatment of breast cancer. This study aimed to investigate the expression of miR-199a-3p in breast cancer and the effect of miR-199a-3p on proliferation and apoptosis of breast cancer.Methods:Real-time PCR was used to test the expression of miR-199a-3p in breast cancer tissues, normal breast tissues, breast cancer cells and normal breast cells. Overexpression (or silencing the expression) of miR-199a-3p was conducted by transfecting MDA-MB-231 with miR-199a-3p mimics (or inhibitors). The proliferation of MDA-MB-231 was detected by MTT method. The apoptosis of MDA-MB-231 was investigated by Hoechst staining and caspase-3 activity assay kit.Results:Compared to corresponding non-tumor breast tissues (or normal breast cell HBL-100), lower levels of miR-199a-3p were expressed in breast cancer tissues or breast cancer cells. Overexpression of miR-199a-3p induced by miR-199a-3p mimic inhibited the proliferation and promoted the apoptosis of MDA-MB-231, while silencing the expression of miR-199a-3p induced by miR-199a-3p in-hibitor increased the proliferation and suppressed the apoptosis of MDA-MB-231.Conclusion:The expression of miR-199a-3p is lower in breast cancer, which shows its tumor suppression effect by regulating the proliferation and apoptosis of breast cancer cells.

Objective To investigate the impact of oral contrast agent for assisting in outlining the small bowel on pelvic volumetric modulated arc therapy (VMAT) dose in patients with cervical cancer.Methods Nine cervical cancer patients for postoperative radiotherapy underwent CT scans,and the target volumes and organs at risk including the small bowel were contoured.The VMAT plan was designed for each case.Then another plan was generated by re-calculating the radiation dose after changing the electron density of the small bowel.The first plan (plan A) was the conventional VMAT plan,and the second one (plan B) specified the electron density of the small bowel.Paired t-test was used to compare the dose distribution between the two plans.Results The Dg8,D5o,conformity index,and homogeneity index of plans A and B were 4 989.1 vs.5 000.1 cGy (P =0.026),5 208.6 vs.5 191.6 cGy (P =0.005),0.766 vs.0.765 (P =0.920),and 0.081 vs.0.074(P =0.055),respectively.The volumes of the small bowel receiving at least 30 Gy for plans A and B were 309.3 vs.314.3 cm3(P =0.207),while bladder V45 of the two plans was 52.4％ vs.51.1％ (P =0.168).To achieve the same prescribed dose,plan A and plan B needed 893.3 MU and 865.8 MU (P =0.093).Conclusions The contrast agent filling the small bowel does not lead to a significant increase in the pelvic VMAT dose in patients with cervical cancer after surgery.

Objective To compare the effect of silica-extraction method and Silico membrane based method in DNA purification from bones and teeth.Methods DNA samples were purified respectively with the silica-extraction method and MinElute PCR Purification kit from 6 bones and 8 teeth,then tested STR types by GlobalFiler? kits. And evaluated the two methods with the success rate and the peak height. Results Both of the two purification methods can successfully obtain the STR markers of the 14 samples. And there was no statistical difference between the two methods in the average peak height from bones and teeth. Conclusion The Silico membrane based method which have more advantages in operation is an efficient method to purify DNA from bones and teeth, and there is no significant difference compared with the silica-extraction method. But the cost is higher. It can be selectively used in forensic practice.

Clinical features and related information on diagnosis and treatment of 45 cases of Castleman's disease (CD) were retrospectively analyzed.Based on the clinical classification, localized CD (LCD) was found in 26 cases, multicentric CD (MCD) was found in 19 cases.Most cases of LCD presented the symptoms of compression, while MCD had complicated and non-specific clinical manifestations, making the early diagnosis more difficult.All 26 cases with LCD underwent surgery, among which only 2 cases relapsed.Sixteen out of 19 patients with MCD were treated with glucocorticoids or combined chemotherapy, and 14 cases achieved complete or partial remission.The results show that patients with CD have variant manifestation and the diagnosis depend on CT scan or histopathology examination.Most LCD can be cured by complete surgical resection, and MCD can achieve remission by the treatment with glucocorticoids or combined chemotherapy.

Objective The aim of this study was to investigate mutations of 41 STR loci. Methods 4546 bloodstain samples were typed from 1932 father–mother–child trios by using AGCU_21+1, AGCU_EX22 and GlobalFiler_ExpressTM amplification Kit. Calculate the mutation rates of STR loci. Results 154 mutations were identified at 32 of the 41 loci. The average mutation rate was 1.0×10-3per locus(95%CI: 0.8~1.1×10-3), and the mutations of SE33 was highest. 152(98.7%) mutation events were one-step mutation, 2(1.3%) events were two-steps. The mutation events occurred in 150 father–mother–child triplets. The mutations in 146(97.3%) triplets occurred at single locus, 8 mutations were observed at two loci in 4(2.7%) triplets simultaneously. 104 paternal and 22 maternal mutations could be determined under 79212 paternal and maternal allelictransfers. The ratio of paternal versus maternal mutations was 4.7:1, and 28 unassigned mutations were observed. Conclusion STR mutation are common in paternity testing, and we should pay more attention to it.

Objective To demonstrate the incidence and relative factors of hypothyroidism after partial thyroidectomy (PTC). Methods The records of all euthyroidsm patients who underwent hemithyroidectomy from 1988 to 2000 were reviewed to determine the incidence of postoperative hypothyroidism and the predisposing factors. All the patients age, gender, serum TSH TGA and TPO levels,and the weight of resected thyroid tissue were evaluated. Hypothyroidsm patients were evaluated for the symptoms , timing of diagnosis , and thyroxine therapy. Results Hypothyroidism was diagnosed in 41(3.4%) of 1 210 patients ,inclnding subclinical hypothyroidsm in 28 and overt in 13.The mean postoperative serum TSH level was (9.22?3.36)mU/L. The mean preoperative serum TSH level was (3.14?1.05)mU/L in hypothyroidsm patients but in euthyroid patients was (1.07?0.72)mU/L(P

This article describes the design and development of an ultrasonic power meter which is consist of an electronic balance, a practice target, an acoustic enclosures and a blocking. The electronic balance mounted on the blocking is linked with the practice target by connecting rod. By adjusting the blocking makes the practice target suspended above ultrasound probe, and then the ultrasonic power can be measured. After initial tests, the ultrasonic power meter performanced with good stability and high precision.

Objective To analyze the sexuality situation and the influencing factors of 1 732 cases of migrants in Chongqing , and provide the scientific basis for improving the sexual health of migrants and the service level of the relevant departments .Meth‐ods The multi stage random cluster sampling method was used to select the subjects .A total of 1 732 married and unmarried mi‐grants who had sexuality were included into the structured questionnaire survey .The SAS9 .3 software was use to statistical analy‐sis .The χ2 test and Ordinal regression analysis and Logistic regression analysis were use to univariate and multivariate analysis .Re‐sults The communication frequency of sexuality of male migrants was higher than the females ,OR=1 .53(95% CI:1 .24-1 .89);young people had the high communication frequency of sexuality ;the frequency of low education groups lower than higher .The fre‐quency of subjects worked in Hong Kong ,Taiwan ,overseas‐funded enterprises are higher than other workplace .The sexual life fre‐quency of male migrants is higher than the female ,OR=1 .84(95% CI:1 .40-2 .42);the sexual life frequency of low age group was higher than high age group ,the frequency of sexual life in factory workers was lower than the Hong Kong ,Taiwan ,overseas‐funded enterprises workers .Compared with the subjects who communicate the sexuality issues often ,the frequency of sexual depression and suppression was higher in the migrants who communicate general and rarely ,the OR was 2 .75 (95% CI:1 .71 -4 .41) and 2 .60 (95% CI:1 .45-4 .66) .Conclusion Male ,younger ,highly educated migrants have a high sexual issues communication frequency . The age and the workplace was the most important factor of the frequency of sexual life .Ease the work pressure and increase the communication of sexual issues will help to relax the sexual depression and depression .
