1.Clinical observation of polysomnographic in patients with different subtypes of Parkinson's disease

Objective To evaluate the changes parameters of polysomnographic ( PSG) with different subtypes of primary Parkinson's disease ( PD ) and analyze the clinical characteristics of sleep disorders in PD patients. Methods Ninety patients with primary PD [ tremor-predominant PD group ( n = 40 ) , postural instability gait disorder ( PIGD )-predominant PD group ( n = 50 ) ] and 50 healthy controls were detected by full night PSG monitoring. And the results were compared. Results Compared with tremor-predominant PD group, the total sleep time, bed time, sleep wake-up times, sleep efficiency and rapid eye movement ( REM ) sleep time of PIGD-predominant PD group were significantly lower (all P<0. 05), however, the sleep latency, S1 (min), S1 (%), S2 (min), S2 (%), S3+S4(min), S3+S4 (%) had no statistical significance (all P>0. 05). Compared with the controls, the total sleep time and bed time of tremor-predominant PD group were significantly lower (all P<0. 05), however, the sleep wake-up times, sleep efficiency, REM sleep time, sleep latency, S1 ( min ) , S1 (%) , S2 (min), S2 (%), S3 + S4 (min), S3 + S4 (%) had no statistical significance (all P>0. 05). Combined with video surveillance video observation,the abnormality of REM sleep behavior disorder ( RBD) was 70% with PIGD-predominant PD group, in which the abnormality of tremor-predominant PD group was 2. 5%. Conclusions The PIGD-predominant PD patients are more likely to appear sleep disorders and RBD than tremor-predominant PD patients. It may be related to clinical heterogeneity of different PD subtypes.

Objective:To investigate the expression of matrix metalloproteinase-9(MMP-9) and its correlation with cancer cell proliferation and metastasis in oral squamous cell carcinoma (OSCC). Methods: Immunohistochemical SP method was used to detect the expression of MMP-9 and Ki-67 in 40 cases of OSCC.Results:The expression of MMP-9 was closely correlated with lymph nodes metastasis and TNM stage of OSCC(P0.05). 3.Effect of Clopidogrel on Plasma Endothelin and Calcitonin Gene Related Peptide in Patients with Unstable Angina Pectoris

Objective To investigate the effect of Clopidogrel on the plasma endothelin(ET)and calcitonin gene related peptide(CGRP)in patients with unstable angina pectoris(UAP).Methods Patients with UAP were divided into two groups,the regular treated group(n =50)was treated with Isoket and the Dansben group(n =50)was treated with combined Isoket and Clopidogrel.The plasma ET and CGRP were measured before and after treatment by radioimmunoreactive assay.Data were compared between the two groups and also compared with those measured in 20healthy subjects for control.Results ET level was significantly higher and CGRP level was lower in UAP patients than that in healthy subjects(P < 0.05).After Clopidogrel treatment,ET was significantly decreased(P < 0.01)and CGRP increased(P <0.05)in combined treatment group compared with patients received lsoket only.Conclusion Clopidogrel was an effective drug for UAP treatment,which could efficiently regulate the ET and CGRP metabolism.
4.Bacterial Biofilm Formation on Indwelling Double J Stent Observed by Scanning Electron Microscopy

OBJECTIVE To observe the bacterial biofilm formation on indwelling double J stent in the human body.METHODS A prospective study of 42 cases with double J stent indwelling in the human body after operation was conducted.The stent was divided into 4 segments then detected microbiologicaly in a separate manner and observed with scanning electron microscope for the presence of bacterial biofilm.RESULTS From 42 cases with douple J stent,only one case was without any bacteria in four segments.20 cases(47.6%) had the same bacteria in four segments.The bacterial positive rate of each segment was pelvic segment(81.0%),upper ureter segment(64.3%),lower ureter segment(71.4%) and bladder segment(83.3%);totally 18 kinds and 126 bacterial strains were flund.the most popular pathogens were Escherichia coli(23.0%),Pseudomonas aeruginosa(15.9%),corynebacterium(15.1%) and Staphylococcus epidermidis(13.5%).On the surface of unused double J stent,there was uneven but clean,without any attachment and encrustation,but on the surface of double J stent indwelling in the human body,the inflammatory affachment and encrustation were observed as well as bacterial biofilm,encapsulated in the abundant fibrous membrane.CONCLUSIONS Our study found that bacterial biofilm can be encrustated on the surface of double J stent indwelling in the human body,it may be one of the reasons for catheter-associated urinary tract infection.
5.Analysis of a survey results on brucellosis in high-risk areas of Urumqi

Objective The main purpose is to investigate the status of brucellosis infection in high-risk areas of Urumqi and population characteristics,and to provide a basis for prevention of brucellosis.Methods In Urumqi City,Dabancheng District,Gaoxin District,Midong,Zone and Urumqi County were selected as survey sites;according to pastoral,agro-pastoral and agricultural,all towns in each survey site were classified into three categories;150 to 200 residents in each township were selected as subjects.Basic demographic information was collected,and blood samples were collected for serological detection.rose bengal plate agglutination test (RBPT) was used for preliminary screening,RBPT positive persons were further confirmed by standard tube agglutination test (SAT).Different regions,gender,age,occupation,exposure difference and brucellosis infection rates were studied.x2 test was used to compare rates.Results The infection rate was 5.42% (29/535).Furthermore,the infection rate ofthe Dabancheng District [10.34% (12/116)] was higher than those of Gaoxin District [2.94% (3/102)] and Urumqi County [2.58% (4/155),x2 =4.643,7.199,all P < 0.05],the differences were statistically significant (x2 =9.327,P <0.05).Besides,infection rate of the average exposure occupation [8.33% (24/288)] was higher than those of low exposure occupation [0(0/61)] and high exposure occupation [2.72% (5/184),x2 =5.459,6.140,all P < 0.05].The differences were statistically significant (x2 =10.846,P < 0.05).The differences of infection rates in ages and nations showed no statistical significance (x2 =2.396,4.639,all P > 0.05).Conclusions High risk areas of brucellosis are still exist in Urumqi.Health education should be strengthened in the future,in order to enhance residents' seff-protection awareness in exposed population.
6.Effect of Prophylactic Use of Vitamin B6 with Chlorpheniramine on the Adverse Reactions in Fundus Fluores-cein Angiography

OBJECTIVE:To explore the effect of prophylactic use of vitamin B6 with chlorpheniramine on the adverse reactions in fundus fluorescein angiography (FFA). METHODS:326 patients with FFA were randomly divided into observation group and control group. Observation group was orally given Vitamin B6 tablet 10 mg 30 min before angiography+Chlorpheniramine maleate tablet 4 mg;control group was orally given Metoclopramide tablet 10 mg+Chlorpheniramine maleate tablet 4 mg. The occurrence time and incidence of adverse reactions in 2 groups were observed and correlation analysis was conducted. RESULTS:There was no significant difference in the occurrence time of adverse reactions between 2 groups(P>0.05);the incidence of adverse reactions in observation group was significantly lower than control group,the difference was statistically significant(P<0.05). The correlation analysis of adverse reactions in control group showed the occurrence of adverse reactions had no correlation with age and gender of patients(P>0.05). CONCLUSIONS:Prophylactic use of vitamin B6 with chlorphenir arnine can reduce the occurrence of adverse reactions in FFA.
7.Clinical Observation of Fuyankang Combined with Azithromycin in the Treatment of Pelvic Inflammatory Disease

OBJECTIVE:To observe the clinical efficacy and safety of Fuyankang combined with azithromycin in the treat-ment of pelvic inflammatory disease. METHODS:90 patients with pelvic inflammatory disease were selected and randomly divided into observation group and control group,with 45 cases in each group. Control group was given Lactose acid azithromycin for injec-tion 500 mg,ivgtt,qd,and then Azithromycin capsule 250 mg,po,qd;observation group was additionally given Fuyankang Tab-let 1.56 g,tid,on the basis of control group. A treatment course of 2 groups lasted for 7 d,and both received 3 courses of treat-ment. Clinical efficacies of 2 groups were observed as well as the level of CRP,IL-2 and life quality score before and after treat-ment,and the occurrence of ADR was compared between 2 groups. RESULTS:Total effective rate of observation group was 97.78%,which was significantly higher than that of control group (86.67%),with statistical significance (P<0.05). The level of CRP,IL-2 and life quality score of 2 groups were improved significantly after treatment,and the observation group was more better than the control group,with statistical significance (P<0.05). No obvious ADR was found in 2 groups during treatment. CONCLUSIONS:Fuyankang combined with Azithromycin is effective for pelvic inflammatory disease,and can improve the quali-ty of life,reduce inflammation cytokines infiltration and the generation of inflammatory medium with good safety.
8.Effect of thrombin on malignant biological behavior of esophageal cancer cell line Eca109

Objective To study the effect of thrombin on proliferation and invasion of esophageal cancer cell line Eca109, and to explore its possible mechanism. Methods The proliferation and invasion of Eca 109 cells treated with thrombin were detected by MTT and Transwell assay, respectively. The activity of matrix metalloproteinase 2 (MMP-2) and MMP-9 in the supernatant of Eca109 cells was detected by gelatin zymography. Reverse transcription polymerase chain reaction (PCR) and immunocytochemistry were used to study the mRNA expression of protease-activated receptor 1 (PAR-1), the important receptor of thrombin, and subcellular localization of PAR-1 protein in Eca109 cells, respectively. Results Thrombin could promote Eca109 cells proliferation in a dose-dependent manner. Cell proliferative rates of 0.5 U/ml and 1.0 U/ml thrombin were 34.38 % and 57.19 %, respectively (P< 0.05). Compared to that of control group, the number of Eca109 cells incubated with 1.0 U/ml thrombin invading through the basement membrane of Transwell was increased (303.33 ±6.66 vs. 116.33 ±11.51, P< 0.05). When treated with various concentrations of thrombin for 24 h, the activities of MMP-2 and MMP-9, especially MMP-9, in the supernatant of Eca109 cells were increased in a dose-dependent manner. Eca109 cells expressed PAR-1 mRNA, and PAR-1 protein was mainly located on the cellular membrane. Conclusion Thrombin increases proliferation and invasion of esophageal cancer Eca109 cells and enhances the activities of MMP-2 and MMP-9 in cells supernatant, which might be induced by activation of PAR-1 located on cellular membrane.

Objective To investigate the gene carrying rate,gene type and composition ratio of thalassemia among pre-pregnant population in Chongqing area.Methods A total of 1054 people were enrolled in the hospital from April 2014 to March 2016 for thalassemia screening.The content of screening included mean corpuscular volume (MCV),mean corpuscular hemoglobin (MCH) and hemoglobin electrophoresis.Thalassemia gene was examined in people with any abnormal term of screening result.Results In 10854 cases,1117 cases showed positive in thalassemia primary screening,and the positive rate was 10.29%.458 cases were tested positive of thalassemia gene,the carrying rate of thalassemia was 4.21%.In which,253 cases of pure a-thalassemia were tested.The carrying rate of α-thalassemia was 2.33%.The most common kind in α-thalassemia was--SEA whose constituent ratio were 52.17 %.197 cases of pure β-thalassemia were tested,the carrying rate of β-thalassemia was 1.81%.The most common kind in β-thalassemia was CD17 (A→T),whose constituent ratio were 31.47 %.11 cases were diagnosed with αβ-thalassemia.Conclusion Chongqing is high-prevalence area of thalassemia.It is important to conduct thalassemia genetic screen before pregnancy which plays a vital role in improving population quality and achieving prepotency.
10.Expression of miR-134 in Methamphetamine-induced Neuronal Injury and Its Effect on Evoked Action Potential

[Objective]To investigate the expression change of miR-134 in methamphetamine(MA)-induced neuronal injury in PC12 cells and its effect on neuronal excitability and understand the pathogenesis of methamphetamine-induced neuronal injury.[Methods]PC12 cells in the logarithmic phase were divided into control group and MA group. The MA group was treated with 800μmol/L MA to establish the model of neuronal injury. The cellular injury was observed under microscope. The neuronal apoptosis was detected by Hoechst3342/PI double staining,and miR-134 expression was measured by using real-time quantitative PCR (Real time-PCR). Furthermore,we constructed miR-134 interference vector and observed its effect on evoked action potential.[Results]The cultured PC12 cells were damaged under the 800 μmol/LMA treatment ,and neurites became shorter ,the apoptotic cells were evidence. Real time-PCR showed that miR-134 expression was increased after MA treatment. Electrophysiological data showed that the evoked action potential increased after miR-134 interference.[Conclusions]High concentration of MA can induce neuronal damage and apoptosis and also increase miR-134 expression. While silence miR-134 expression can increase neuronal excitability.Our study provides an experimental basis for elucidating the possible mechanism of MA-induced neuronal injury and the role of miR-134 in neurotoxicity and neuronal excitability.
