Objective To prepare human Fab fragment antibodies against Interleukin-2 and identify their antigenic specificity and combining activities with antigens. Methods The specific Anti-interleukin-2 clones were screened from a natural human Fab fragment antibodies phage display library against the immobilized interleukin-2 antigens. Then the phagemid DNA from the specific clones was digested with Spe I and Nhe I to delete gⅢ (about 660bp). The digested 4.7kb DNA, which was purified after separation of bands from agarose gel using purification kit, was ligated with T4-DNA ligase and the ligated reaction mixture were transformed to the BL21 (DE3) pLysS. Positive clones on the LB agar plates were inoculated to liquid LB culture medium, and when the bacteria were grown to OD600≈0.5 at 37℃ with continuous shaking, IPTG was added to induce the expression of soluble Fab fragment antibodies at 30℃. The expressed products containing Fab fragment antibodies were determined by SDS-PAGE, Western blot and ELISA. Results The soluble products were identified as containing human Fab fragment antibodies against Interleukin-2 by Western blot and formed a Mr 47?103 band under non-reducing condition on SDS-PAGE. The band was then proved as anti-human Fab fragment antibodies by Western blotting. ELISA demonstrated that Fab fragments possessed good antigenic specificity as well as excellent combining activity with interleukin-2 antigens, and the fragments did not react with bovine serum albumin and IL-4 in ELISA. Conclusions The soluble human anti-interleukin-2 Fab fragment antibodies have been highly expressed and successfully identified, and an effective way has been searched out for constructing the engineering antibodies. All of the results may lay a potentially good foundation for engineering human Fab antibodies, and for the clinical application of the antibodies on the immunotherapy of tumor diseases.

Objective To investigate if diastolic function could be improved by cardiac sympathetic nerve blockade in patients with dilated cardiomyopathy (DCM). Methods Sixty consecutive cases of DCM were randomed into therapeutic group and control group. Patients in therapeutic group were administered thoracic epidural blockade (TEB) (T 1-5 ) with 0.5 % lidocaine intermittent injection every two hours for four weeks, in addition to some routine medicine, while patients in control group were only administered routine treatment. Then the parameters of left ventricular diastolic function were evaluated with Doppler echocardiography before and after four weeks. Results ①All the patients in TEB group felt better within five minutes after injection of lidocaine within local epidural cavity.Their symptoms were relieved more rapidly than those of the drug therapy alone. Some patients with NYHA class Ⅳ could lie down supine in a short time. Exercise tolerance increased in patients with NYHA class Ⅱ to Ⅲ. But symptoms and signs of patients in control group were relieved very slowly, some patients even got worsen. ② Thirteen of thirty in therapeutic group got improvement on diastolic function after treatment. One of thirty in this group deteriorated. In detail, six of fifteen patients with demonstrated improvement of left ventricular diastolic filling changed from having a restrictive filling pattern to having a pseudonormal left ventricular filling pattern. Four of nine patients with a pseudonormal filling pattern changed to have a delayed filling pattern.Three of six patients, left ventricular filling pattern moved from a pseudonormal left ventricular filling pattern to a normal filling pattern. Only one patient acquired a restrictive filling pattern from a pseudonormal filling pattern. In contrast, in control group one of thirty got improvement on diastolic function after treatment. Nine of thirty in this group deteriorated. Individual changes of left ventricular filling pattern showed that only one patient improved from a delayed filling pattern to normal filling pattern, whereas seven patients had a restrictive left ventricular filling pattern developed from a pseudonormal filling pattern and another two patients achieved a pseudonormal filling pattern from delayed filling pattern. Conclusions Diastolic function in patients with DCM was improved significantly by cardiac sympathetic nerve blockade. The approach has beneficial effects on the patients of DCM.

Objective:To investigate the expression of matrix metalloproteinase-9(MMP-9) and its correlation with cancer cell proliferation and metastasis in oral squamous cell carcinoma (OSCC). Methods: Immunohistochemical SP method was used to detect the expression of MMP-9 and Ki-67 in 40 cases of OSCC.Results:The expression of MMP-9 was closely correlated with lymph nodes metastasis and TNM stage of OSCC(P0.05). Objective To investigate cell injury and apoptosis of L-6TG cells induced by simulated ischemia-reperfusion,and to explore its regulating mechanism.Methods Cultured L-6TG cells were randomly divided into control group(C)and simulated ischemia-reperfusion group(IR).The L-6TG cells of C group were cultivated under normal condition in mimicking reperfusion solution,while the L-6TG cells of IR group were cultivated in mimicking ischemic solution for 4 hours,and then cultivated in mimicking reperfusion solution for 4 hours.Superoxide dismutase(SOD),xanthine oxidase(XOD)and free calcium in L-6TG cells were determined in each group;mitochondrial respiration was measured by MTT method.Apoptosis was identified by flow cytometer with propidium iodide(PI)staining method and agarose gel electrophoresis;the immunohistochemical method was used to determine the expression of bcl-2 and caspase-3,and their profiles were half-quantitatively analyzed by automatism image analytical system.Results Compared with the L-6TG cells of C group,after suffering from ischemia and reperfusion each for 4 hours,XOD,free calcium in L-6TG cells and apoptotic percentage were all increased significantly(287.88?8.47 vs 42?3.94 U/g prot,1.547?0.017 vs 7.23?0.28 lnMCF,7.23%?0.28% vs 0.69%?0.07%,P

Objective To evaluate the effectiveness of chronic diseases self-management model for the aged patients with type 2 diabetes mellitus in communities.Methods Patients with T2DM aged over 60 years and over in two communities in Baoding were selected:one as intervention community and the other as control community.The intervention community underwent the chronic diseases self management theory based education by expert,face to face consultation and patient communication,while the control community received general community management.The scores of self-efficacy were collected before the sessions and 3 months after the end of the sessions,and the effectiveness of the intervention was evaluated by the changes of scores,fasting blood-glucose and hemoglobin A1c (HbA1c) before and after the session.The pure increment level of scores of diabetes self-efficacy was higher in the intervention community than in the control community [(16.9 ± 5.3)scores vs.(0.4±1.3) scores,t=21.317,P＜0.01].Results All dimension values were also significantly higher in the intervention group than in control community (P＜ 0.01).The pure decrement level of fasting blood glucose (1.8 ± 1.7) ％ and HbA1c (0.7 ± 1.2) ％ after intervention was significantly higher in the intervention than in control community (t=6.096,P＜0.01 ;t=2.930,P＜0.05).Conclusions The chronic diseases self-management education may significantly improve patients' self-efficacy and decrease the level of blood-glucose and HbAlc.

Emphysematous cystitis is a rare inflammation of the bladder, which can be life-threatening in severe cases. A senior gentleman with emphysematous cystitis was recently admitted to our hospital. He got bilateral renal hydronephrosis and bilateral terminal pneumatosis of the ureter. The comorbidity was prostatic hyperplasia and diabetes. After indwelling catheter , glucose levels control and anti-infection treatment for 3 days, the repeated CT showed the gas inside the bladder the end of ureter decreased significantly. The blood routine, urine routine, renal function was better than before. The patient was instructed to continue anti-infection treatment. The above laboratory examination indicators returned to normal after a week's re-examination.
