1.Investigation and Analysis of Patients with Lung Infection After Tracheal Intubation General Anesthesia

OBJECTIVE To know the risk factors about lung infection in patients after tracheal intubation general anesthesia and take control measures.METHODS All the patients with tracheal intubation general anesthesia from Jan to Dec in 2007 were investigated retrospectively.RESULTS Among 2914 operation cases,471 cases were of tracheal intubation general anesthesia and 95 cases were infected(20.17%).The infection rate was 5.38%(24 hours).Sputum culture: G-bacteria accounted for 57.65%,G+ bacteria 28.83%,fungi(13.52%).CONCLUSIONS The infection rate increase with longer intubation time.Mechanical ventilation is a risk factor of lung infection.The measures to control infection include shortening intubation hours,reducing mechanical ventilation and strictly sterilization and isolation.
2.Primary observation on bone mesenchymal stem cells accelerating bone-tendon junction healing

[Objective]To evaluate tho effects of Bone Mesenchymal Stem Cells(BMSCs) on healing of bone-tendon junction in a patellar tendon complex.[Method]The BMSCs of rabbits were harvested by bone marrow aspiration. Adherent cells were selected as BMSCs after the whole marrow was cultured.Standard partial patellectomy was conducted in 36 18-week-old rabbits divided into 2 groups. The experimental group was planted into by the composite of BMSCs and Pluronic F-127 while the control group was only operated without BMSCs planting into.The patellar-patellar tendon complexes were harvested at weeks 6,12,18 postoperatively for radiographic, gross and histological evaluations.[Result]Radiographic measurements showed significantly more newly formed bone at the patellar tendon-patellar healing junction in the experimental group compared with that of the controls at weeks 6,12,18(P
3.Characteristics of EDC/NHS-crosslinked collagen type Ⅱmatrix

[Objective]Porous collagen type Ⅱ matrices crosslinked by 1-ethyl-3-(3-dimethyl aminopropyl)carbodⅡmide(EDC) and N-hydroxysucinimide(NHS) were developed as scaffolds for cartilage tissue engineering.[Method]Collagen type Ⅱ matrix was crosslinked by EDC/NHS,and then freeze-dried to achieve collagen scaffolds.The porous characteristics of the prepared scaffolds were tested by using diffenent methods,including laser scanning confocal microscopy (LSCM),scanning electron microscopy (SEM) and mechanical testing machine.The chondrocytes labeled by GFP were planted in crosslinked collagen type Ⅱ,and observed by LSCM.[Result](1) The mechanical properties of cross-linked matrices increased significantly,reaching 2.18?0.47MPa.(2)The degradation of cross-linked matrices decreased by 8.28% after 4 hours.The pore size was about 90?m,and their average porosity and water capacity reached 93.39% and 97.78%,respectively.3.Chondrocytes were in good condition in the crosslinked collagen.[Conclusion]The present work indicates that EDC/NHS-crosslinked collagen type Ⅱ could keep the properties and biocompatibility of collagen,besides,the mechanical strength increased and the degradation was decreased.It will be suitable for cartilage tissue engineering purposes.

5.Long-term effect of maternal hyperglycemia and postnatal overfeeding on offsprings of rats

Objective To establish the animal model of gestational diabetes mellitus (GDM to observe the long-term effect of maternal hyperglycemia and postnatal overfeeding on the metabolism of offsprings. Methods Streptozotocin (STZ, 50 mg/kg) was administered to Wistar rats intraperitoneally on the 5th day of gestation to induce animal GDM model. Offsprings were allocated into 3 groups: Group CP (n=8) were exposed to normal maternal hyperglycemic environment, group DP-1 (n=8) exposed to intrauterine hyperglycemia and every 8 pups breast-fed by one GMD mother and group DP-2 exposed to intrauterine hyperglycemia and every 4 pups breast-fed by one GDM mother (n=8). Weight gain between 3-10 weeks were recorded, OGTT at 10 weeks and hyperinsulinemia-euglycemia clamp technique was performed, and plasma insulin level was measured by ELISA at 11 weeks. Results Offsprings in DP-1 and DP-2 group had greater weight gain than group CP at 3-10 weeks after birth [(6.68±1.70)% and (6.03±1.85)% vs (4. 74±0.93)%, P< 0. 05], but no difference was found among the three groups in OGTT results (P> 0. 05). The baseline plasma insulin level was lower in the DP-1 group than in the DP-2 and CP group [(0. 76± 0. 37)μg/L vs (1.13±0. 25)μg/L and (1.06±0. 14)μg/L](P<0. 05). GIR60-120 was higher in DP-1 Offsprings of Wistar rats, exposed to intrauterine hyperglycemia, experience excessive weight gain at 3-10 weeks after birth. Overfeeding in early life expedites the decrease of insulin sensitivity in these offspings.
6.Intrauterine hyperglycemia and leptin resistance of offsprings

Objective To study the effect of intrauterine hyperglycemia on leptin level and offspring development in rats.Methods Female and male adult Wistar rats were mated,streptozotocin (STZ,50 mg/kg) was administered intraperitoneally on 5th day of gestation to induce diabetic model,diabetic pups (DP) were exposed to intrauterine hyperglycemia;control pups(CP) were exposed to controls,which was injected with citrate buffer,8 pups were choosed from each group.Weight gain between 3-10 weeks were recorded.Plasma leptin was detected by enzyme-linked immunosorbent assay (ELISA) when the rats were 11 weeks old,and the expression of leptin receptor in hypothalamus was measured at protein level by histomorphology and mRNA level measured by realtime PCR[expressed with integral absorbance(IA)]in 11weeks in order to discuss the relation of leptin and offspring development.Results The fasting blood glucose level was significantly higher in diabetic mother compared with the controls[(28.3±5.1) mmol/Lvs.(6.3±1.4) mmol/L,P<0.05].However,there was no difference between the fasting blood glucose level in DP group and CP group[(5.1±0.8) mmol/L vs.(5.3± 0.6) mmol/L,P > 0.05].The growth rate between 3-10 weeks was significantly higher in DP group 649.7% than CP group 479.2%,P <0.05.The base insulin level was lower in DP group[(0.76±0.37) μg/L vs.(1.06±0.14)μg/L,P < 0.05];while there was no difference in plasma leptin and the expression of ieptin receptor in hypothalamns[(113±37) μg/L vs.(128±40) μg/L,P >0.05].The growth rate was not associated with plasma leptin in DP group (r=-0.501,P=0.311) but associated in CP group (r=-0.553,P=0.001).The protein level of DP group (4125±414) did not significantly differ from that of CP group (4244±511).The median of mRNA of leptin receptor in hypothalamus in DP group did not altered significantly compared with that of CP group(1.25 vs 1.80,P>0.05).Conclusions Intrauterine hyperglycemia accelerated growth rate of offsprings between 3 and 10 weeks,however,plasma leptin was not discreased,which indicated leptin resistance.Intrauterine hyperglycemia did not influence the expression of leptin receptor in hypopthalamus in offsprings,this suggested the leptin resistance may be not caused by the quantity of leptin receptors.
7.Construction and primary application of Sh-2K~d-HBc tetramer

Objective:To prepare and test tetrameric sH-2K~d-HBc complex for the further measurement of the specific CTL response.Methods:PE labled streptavidin with 4 biotinylated binding sites can bind to 4 biotinylated monomer to form the corresponding tetramer.Mice were immunized via different methods of genetic immunization by use of the construted pcDNA3-C plasmid to get the specific CTLs.Then our prepared tetramer was applied to stain the specific CTLs by the analysis of flow cytometry.Results:We applied our prepared tetramer to stain the cells from the experimental groups and control group.The results showed the tetramer was able to discriminate the frequencies of specific CTL induced by the three immunol methods(0.24%,0.26%,0.36% vs 0.07%,P≤0.05).This demonstrated that the prepared tetramer could bind its targets specifically and efficiently.The three immunol methods induced different levels of immune responses.Compared with the traditional muscle injection,gene gun induced weaker humoral immune response and stronger cellular immune response,and hydrodynamic injection induced the strongest humoral and cellular immune responses.Conclusion:Have successfully constructed the sH-2K~d-HBc tetramer.The techniques and methods can be used for preparation of tetramers of other types of MHCⅠ molecules.
8.Clinical value of combined detection of of SCC and CEA in the diagnosis of lung cancer

Objective To explore the clinical value of combined detection of SCC and CEA in the diagnosis of lung cancer.Methods 382 cases from January 2015 to April 2016 in the respiratory department of our hospital were selected in this survey.Among which 172 cases with lung cancer,110cases with benign lung diseases and 100 healthy person.The CEA and SCC content in serum of all these cases were determined by chemical chemiluminescence,and the diagnostic efficacy of lung cancer by CEA or SCC alone and CEA and SCC combined examination was compared.Results In lung cancer group,the serum levels of CEA and SCC content were significantly higher than those of the normal control group and lung benign disease group(P<0.05),and benign lung disease group were significantly higher than that of the normal control group(P<0.05).and compared with squamous cell carcinoma and small cell lung cancer,CEA has the highest content in adenocarcinoma(P<0.05).Meanwhile,the content of SCC in squamous cell carcinoma was obviously higher than that of small cell lung cancer and adenocarcinoma(P<0.05),The sensitivity of CEA was significantly higher than that of SCC(P<0.05),and the sensitivity and negative predictive value in combined detection group was significantly higher than that of CEA or SCC(P<0.05).Conclusion SCC and CEA could be used to identify different types of lung cancer,and the combination of the two methods could improve the positive diagnosis rate,facilitate the early detection of lung cancer,early treatment,has the promotion value.

This study investigated the expression profiles of IL-10 gene in three human hepatoma cell lines including Huh7, HepG2, and HepG2 transfected with a plasmid containing hepatitis B virus (HBV) named HepG2.2.15. RT-PCR analysis demonstrated that IL-10 message RNA was absent in HepG2 and Huh7 cells, whereas it was present in HepG2.2.15 cells, which was consistent with ELISA result. Furthermore, except for lamivudine other antiviral treatments did not significantly decrease the HBV DNA level in HepG2.2.15 cells, while they had different effects on the expression of IL-10 protein, although stimulation by LPS had no significant effect. In addition, except for poly(I:C), the other treatments decreased the expression of IL-10 protein to different degrees, but had no significant effects on the expression of NF-κB and MyD88. Meanwhile, all treatments we used had effect on the expression of STAT1. In conclusion, IL-10 was expressed in HepG2.2.15 cells and STAT1 pathway might be involved in the regulation of IL-10 expression in HepG2.2.15 cells, but it was not the sole pathway, the exact mechanism warrants further study.
10.Finite Element Analysis on Different Internal Fixations for Treating Proximal Humeral Fractures in Children

Objective To compare biomechanical characteristics of external fixator, Kirschner’s wire, elastic stable intramedullary nailing (ESIN) for fixing proximal humeral fractures in children by finite element method.Methods The CT scanning data from the healthy humerus of an 8-year-old patient with proximal humeralfractures were collected, and the image data were imported in Mimics 21. 0 to establish the rough humeralmodel, which was imported in Geomagic 2013 to construct the three-dimensional (3D) model of cancellous and cortical bones of the humerus. After the model was assembled with 3 fixators ( external fixator, Kirschner’swire, ESIN), it was imported in ANSYS 2019 to simulate the upper limb under quiet, abduction, adduction, flexion, extension, external rotation, internal rotation working conditions. The maximum displacement of the distal humerus, the maximum stress of the fixture, and the maximum displacement of the distal fracture surface were analyzed. Results The minimum values of the maximum displacement of the distal humerus in models fixed by external fixator, Kirschner’s wire, ESIN appeared under extension (2. 406 mm), external rotation (0. 203 mm), external rotation (0. 185 mm) working conditions, respectively. Conclusions External fixator is the most unstable fixation of proximal humeral fractures in children, and the biomechanical performance of ESIN is better than that of external fixator and Kirschner’s wire fixation
