1.Optimization of liquid culture conditions of dihydropyrimidinase producing strain Pseudomonas putida 9801

Purpose The aim is to optimize the liquid culture conditions of dihydropyrimidinase producing strain Pseudomonas putida 9801. Methods Plackett-Burman design and spherical symmetric desig n were used.Results Optimum conditions for dihydropyrimidinas e formation of Pseudomonas putida 9801 were defined:yeast extract 2.39%, Glu cose 1.81%,Uracil 0.06%,K2HPO4*3H2O 0.2%, MgCl2*6H2O 0.05%and NaCl 0 .3%,when the strain was cultured at 32℃ for 10 h,about 3.02 units/ml of hydanto inase was obtained. This value was quite consistent with the theory value(2.91 u nits/ml).Conclusion The liquid culture conditions of dihydrop yrimidinase producing strain were optimized.
2.Investigation in the burden of dementia patients caregivers in Gushan town,Fuzhou city

Objective To study the burden of dementia patients' caregivers and related influencing elements.Methods A survey was carried out in 152 dementia patients' caregivers after studying the prevalence of dementia in Gushan town using self-designed questionnaires.Results Among the 152 earegivers,65.13% received primary school education or below.And those taking care of disabled dementia patients,taking care of patients with higher ADL scores,attending on without professional helper,having insufficient knowledge,reluctant to give care,and spending much time on it got higher burden score.Conclusions Among caregivers of dementia patients,females are more than males;sons,daughters and spouse are the primary caregivers.Their education level is low,and the caregivers don't have enough knowledge on dementia care and can't give enough service to the senile dementia patients.
3.Investigation and Analysis of Patients with Lung Infection After Tracheal Intubation General Anesthesia

OBJECTIVE To know the risk factors about lung infection in patients after tracheal intubation general anesthesia and take control measures.METHODS All the patients with tracheal intubation general anesthesia from Jan to Dec in 2007 were investigated retrospectively.RESULTS Among 2914 operation cases,471 cases were of tracheal intubation general anesthesia and 95 cases were infected(20.17%).The infection rate was 5.38%(24 hours).Sputum culture: G-bacteria accounted for 57.65%,G+ bacteria 28.83%,fungi(13.52%).CONCLUSIONS The infection rate increase with longer intubation time.Mechanical ventilation is a risk factor of lung infection.The measures to control infection include shortening intubation hours,reducing mechanical ventilation and strictly sterilization and isolation.
4.Clinical observation of Pentoxifylline combined with mechanical ventilation in acute respiratory distress syndrome

Objective To explorer the clinical effects of Pentoxifylline(PTX group) with mechanical ventilation in patients with acute respiratory distress syndrome(ARDS). Methods 64 patients with ARDS were randomized into a control group(n = 32 ,ARDS conventional treatment) and a PTX group(n = 32 ,PTX 50ml iv bid in addition to the conventional treatment). The patients were closely monitored with their curative effect, blood gas indexes and serum levels of TNF-α and IL-8 after the treatment. Results The PaO_2 and PaO_2/FiO_2 got significantly higher in the PTX group than in the control group on 3d and 7d (P < 0.05). The serum levels of TNF-α and IL-8 and the mortality of ARDS were significantly decreased in the PTX group after the combined with Pentoxifylline (P < 0.05 or P <0.01). Conclusion The treatment combined with the Pentoxifylline can effectively improve the Oxygenation and prognosis of ARDS patients by inhibiting the inflammatory responses.
5.Development of polyvinyl alcohol-collagen composite and its biocompatibility as tissue substitute

BACKGROUND: Polyvinyl alcohol (PVA) displays limitation to cell adsorbability. Can collagen improve the adsorbability of PVA to cells?OBJECTIVE: To develop a novel type composite of PVA and collagen, and explore the feasibility to serve as soft tissue substitute.DESIGN: Single sample observation.SETTING: Guangzhou Red Cross Hospital, Jinan University Medical College, Guangzhou Institute of Traumatic Surgery.MATERIALS: Fifteen New Zealand rabbits of 2.0-3.0 kg, either male or female, were provided by Medical Experimental Animal Center of Guangdong Province. The experiment was carried out in the Laboratory of Guangzhou Institute of Traumatic Surgery, and the experimental procedure was accorded with the animal ethical standards. Bovine typeI collagen was purchased from Guangzhou Trauer Biotechnology Co., Ltd. and PVA-124 from Guangzhou Chemical Reagent and Instrumentation Co., Ltd.METHODS: The experiment was performed in Guangzhou Red Cross Hospital, Jinan University Medical College between July 2003 and December 2006. ①Preparation of PVA-collagen material: 5 g/L bovine type I collagen was mixed with 5% PVA-124 at a ratio of 1 : 1. The mixture was freeze-dried at vacuum until becoming gelatinous. The internal structure was observed under the use of scanning electron microscope. ②Cytotoxicity test: PVA-collagen composite was cut into pieces of 10 mm×5 mm× 1 mm, put into 48-well culture plate after sterilized by Y ray, cultured with 1×104 3T3 cells in each well. Cell growth was observed under scanning electron microscope and laser scanning confocal microscope. ③Embedding test in vivo: Two longitudinal incisions were cut at the two sides of spine. The subcutaneous tissue was separated bluntly to form subcutaneous lacuna. Four pieces of PVA-collagen material were implanted in the lacuna and fixed. Nine specimens and the surrounding tissues were harvested from three rabbits each at one, four, eight and sixteen weeks postoperatively for pathological observation.MAIN OUTCOME MEASURES: The internal structure of gel film under scanning electron microscope, cytotoxicity test and embedding test in vivo results.RESULTS: ①Internal structure of PVA-collagen material:PVA-collagen material showed white gel shape after freeze-drying at vacuum. Penetrating three-dimensional pores were observed in the surface and inner section under scanning electron microscope. ②Cytotoxicity test results showed that 3T3 cells grew normally on the PVA-collagen material. ? Embedding test in vivo results suggested that one week after PVA-collagen implantation, foreign body reaction occurred, and the interface between material and tissue was clear. Four weeks later, only rare lymphocyte infiltration was observed, and a great amount of fibroblast hyperplasia formed collagen fibrils and false simple cuboidal epithelium coating material. In 8 weeks, no lymphocyte infiltration, neutrophilic granulocyte infiltration or foreign body giant cell were found; dense capsule wall and capsule coating material generated from a great amount of fibroblasts were observed. In 16 weeks, extending collagen fibrils were found arranged regularly with shrank nucleus, showing long ovoid or long fusiform in shape; no new formation small vessels, lymphocyte, neutrophilic granulocyte infiltration or foreign body giant cell infiltration were observed. The capsule wall was stable and thinned. CONCLUSION: PVA-collagen composite has good cell compatibility and tissue compatibility but no toxic or adverse effect. It can serve as in vivo implant.
6.Heat-clearing and Toxicity-removing Therapy for the Treatment of Acute Coronary Syndrome: An Observation of 55 Cases

[Objective] To observe the preventive effect and mechanism of heat-clearing and toxicity-removing therapy for the treatment of acute coronary syndrome (ACS), and to explore the relationship of ACS with inflammation. [Methods] Fifty five ACS patients were randomized into 2 groups:41 patients in group A were treated with Sanhuang Tablets (mainly composed of Radix et Rhizoma Rhei, Radix Scutellariae and Rhizoma Coptidis) p.o., 4 tablets one time, twice a day, and a routine treatment for anticoagulation, anti-platelet aggregation and anti-myocardial ischemia; 14 patients in group B were given routine treatment for anticoagulation, anti-platelet aggregation and anti-myocardial ischemia. The treatment course lasted 2 weeks. After treatment, improvement in symptoms such as chest pain, chest distension, short breath, palpitation, fever, spontaneous sweating, insomnia, lassitude, aversion to cold and cold limbs was compared between group A and group B by scoring method. Meanwhile, inflammatory indicators in the two groups were also observed before and after treatment. 8.Clinical value of combined detection of of SCC and CEA in the diagnosis of lung cancer

Objective To explore the clinical value of combined detection of SCC and CEA in the diagnosis of lung cancer.Methods 382 cases from January 2015 to April 2016 in the respiratory department of our hospital were selected in this survey.Among which 172 cases with lung cancer,110cases with benign lung diseases and 100 healthy person.The CEA and SCC content in serum of all these cases were determined by chemical chemiluminescence,and the diagnostic efficacy of lung cancer by CEA or SCC alone and CEA and SCC combined examination was compared.Results In lung cancer group,the serum levels of CEA and SCC content were significantly higher than those of the normal control group and lung benign disease group(P<0.05),and benign lung disease group were significantly higher than that of the normal control group(P<0.05).and compared with squamous cell carcinoma and small cell lung cancer,CEA has the highest content in adenocarcinoma(P<0.05).Meanwhile,the content of SCC in squamous cell carcinoma was obviously higher than that of small cell lung cancer and adenocarcinoma(P<0.05),The sensitivity of CEA was significantly higher than that of SCC(P<0.05),and the sensitivity and negative predictive value in combined detection group was significantly higher than that of CEA or SCC(P<0.05).Conclusion SCC and CEA could be used to identify different types of lung cancer,and the combination of the two methods could improve the positive diagnosis rate,facilitate the early detection of lung cancer,early treatment,has the promotion value.
9.Study on clinical characteristic and outcomes of primary lung cancer combined with venous thromboembolism

Objective To observe the clinical characteristic and prognosis of primary lung cancer patients with venous thromboembolism (VTE).Methods 589 primary lung cancer patients were selected and divided into VTE group(n =49) and non VTE group(n =540).49 cases with VTE were divided into pulmonary thromboembolism (PTE) group(n =15),including single PTE and PTE combined with deep venous thrombosis(DVT) and DVT group (n =34).Single factor and multiple logistic regression analysis were performed to determine the factors influencing primary lung cancer patients with VTE.Clinical manifestation,time of onset and prognosis of patients with VTE were analyzed.Results 49 patients with VTE included 10 patients(20.4%) with single PTE,34 patients(69.4%) with single DVT and 5 PTE patients combined with DVT(10.2%).D-dimer(OR =1.560,95% CI =1.018 ~ 2.392,x2 =4.161,P =0.041),interleukin-1 (IL-1,OR =1.846,95% CI =1.054-3.234,x2 =4.594,P =0.033),tumor necrosis factor (TNF OR =1.486,95% CI =1.014-2.178,x2 =4.126,P =0.042),adenocarcinoma (OR =2.854,95%CI=1.217-6.695,x2 =5.812,P=0.016) and phase Ⅲ-Ⅳ(OR =2.198,95%CI=1.122-4.305,x2 =5.272,P =0.022) were the factors influencing primary lung cancer patients with VTE.Chest tightness,coughing,accelerated heart rate,swelling and pain in lower limb were common clinical manifestations of primary lung cancer patients with VTE.Most patients with VTE occurred within 3 months after a diagnosis of primary lung cancer.There was no significant difference in the time of onset between PTE group and DVT group(P >0.05).As of July 2014,31 cases (63.2%) died,12 cases (24.5 %) survived,and 6 cases (12.2%) lost in 49 patients with VTE.The median survival time of 49 patients with VTE was 9.5 months.The median survival time of PTE group was 5.8 months,while DVT group was 15.2 months,but no significant difference between them (P > 0.05).Conclusion Increased D-dimer,increased IL-1,increased TNF,adenocarcinoma and phase Ⅲ-Ⅳ could increase the risk of primary lung cancer patients with VTE.There were little typical.clinical symptoms in most patients with VTE,which occurred with in 3 months after a diagnosis of primary lung cancer.They had high mortality and needed to take early diagnosis and treatment through auxiliary examination.

Objective To establish the quality standard for Chanhou Zhuyu Capsules.Methods Leonurus Japonicus and Ligusticum chuanxiong in Chanhou Zhuyu Capsules were identified by TLC,and the content of stachydrine hydrochloride was determined by TLC Scanning.Results The quality identification by TLC was specific.Stachydrine hydrochloride had a good linearity in the range of 4.0~12.0 ?g,r=0.999 7,and the average recovery was 99.87 %,RSD=1.67 %.Conclusion The methods of identification and content determination are simple,reliable and reproducible.They can be used effectively for the quality control of Chanhou Zhuyu Capsules.
