Objective To evaluate the impact of psychological and behavioral intervention on self-concept of school- age children with asthma. Methods The intervention group and the control group in-cluded 50 patients each. Those in the intervention group attended a 6-month psychological behavioral inter-vention in addition to routine follow-up visit and inhalation therapy, while the control group received the routine follow-up visit and inhalation therapy only. Piers-Harris children's self-concept scale (PHCSS) was administered to evaluate the self- concept of the sample repeatedly at 3 months and 6 months. Data about the incidence times of asthma, time of absent from school and hospitalization time were collected within 6-month intervention. Results After 6- month intervention ,the incidence times of asthma and time of ab-sent from school in the intervention group were less San those of the control group. No statistical difference was seen in hospitalization time between the two groups. The total self- concept level and the scores of most subscales, including intelligence and academic competence, physical appearance and attribute, anxiety and popularity in the intervention group were higher than those of the control group. Conclusions Psycholog-ical behavioral intervention was effective in reducing psychosomatic effect of asthma, improving self-con-cept of school- age children with asthma, and promoting the physical and mental health of them.

A method of qualitative and quantitative analysis on fluid catalytic cracking (FCC) full range gasoline by comprehensive two-dimensional gas chromatography coupled with time-of-flight mass spectrometry (GC×GC-TOF MS) was established. The results showed that paraffins, olefins, naphthenes and aromatics in FCC gasoline had regional and zonal distribution in two-dimensional contour plots. The distinctions of boiling points and polarity between different compounds were used to achieve the accurate separation and determination in GC×GC-TOF MS analysis, and consequently the co-current flow which was often present in the conventional GC analysis was greatly suppressed. The difference of ionizing efficiency between different compounds was modified by response factors, and moreover, a good quantitative dependency was found between the analysis results of GC and GC×GC-TOF MS on FCC gasoline. Due to the high separation resolution, GC×GC-TOF MS gave more accurate results about the group compositions of FCC gasoline. GC×GC-TOF MS provided an effective way with high precision for the characterization of FCC gasoline.

AIM: To establish human umbilical vein endothelial cells (HUVECs) to express green fluorescent protein (GFP), and to study the suppression of GFP by siRNA in HUVECs. METHODS: Using lipofectamine 2000 to transform plasmid pN_3-EGFP encoding GFP into HUVECs. The HUVEC containing pN_3-EGFP, named HUVEC-GFP, was screened and selected by antibiotic G418. Using in vitro transcription T7 kit, GFPsiRNA targeting GFP mRNA and control-siRNA used as control were synthesized. The siRNAs were transfected into HUVEC-GFP with oligofectamine. 48 h later, the expression levels of GFP protein and mRNA in HUVEC-GFP were determined. RESULTS: The HUVEC-GFP was screened to express GFP in the presence of G418. The agarose gel electrophoresis analysis showed that the siRNAs prepared were integrated. 48 h after transfection with siRNAs, compared to control group, the level of GFP fluorescence was obviously decreased in the HUVEC-GFP transfected with GFPsiRNA. The results of RT-PCR detection showed that GFP mRNA expression was obviously suppressed by GFPsiRNA at the rate of 40%, and no obvious suppression of GFP mRNA expression was found in the HUVEC-GFP transfected with control siRNA. CONCLUSION: The siRNA targeting GFP mRNA, synthesized in vitro, efficiently suppresses the GFP expression in HUVECs.

Objective:
To study infection of coxsackievirus A6 (CV-A6) in Mongolian gerbils.
Methods:
To screen the optimal ages of Mongolian gerbils, five groups with different ages were infected with 1×105 TCID50 dose of CV-A6 XS45 strain by intraperitoneal, and symptom scores of Mongolian gerbils were collected. Then to estimate the dose-effect, three doses of virus were injected to the Mongolian gerbils. Quantitative reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry(IHC) were used to determine virus load and tissues infection in muscle, brain and intestinal tract.
Results:
Mongolian gerbils infected with 1×105 TCID50 dose CV-A6 consistently exhibited clinical signs, and the morbidity (death) rates of five age groups were up to 100%. There was a positive correlation between the trend of symptom scores changes and ages. The morbidity (death) rates of three doses (1×103 TCID50, 1×104 TCID50, 1×105 TCID50) also were up to 100% in 28 days Mongolian gerbils. The correlation between the trend of symptom scores changes and doses were negative. Virus loads were detected in muscle, brain and intestinal tract of pathogenesis animal. The virus loads of muscle were higher than others. IHC results showed virus infection and cytopathic effects in three tissues.
Conclusions
Mongolian gerbils had high susceptibility to CV-A6, and were best for animal model of CV-A6 infection.
