Cyclins, CDKs and CKIs in cell cycle play important roles in maintaining cell cycle and regulation of the process cell cycle. Many endogenous and extraneous inhibitors have effects on cell cycle by modulating functions of cyclins, CDKs and CKIs. Several highlighted points in the researches of cell cycle and related proteins, inhibitors were reviewed, including the latest advances on their applications in cancer therapy.
2.High Performance Liquid Chromatography Coupled with Electrospray Tandem Mass Spectrometry for Analysis of Pacific Ciguatoxin in Fishes

A sensitive method was developed for the determination of pacific ciguatoxin (P-CTX1) residue in fishes by high performance liquid chromatography coupled with electrospray tandem mass spectrometry ( HPLC-MS/MS). Homogenized samples of fish was extracted with acetone,degreased by freezing under -20℃ and filtered with filter paper. Then the supernatant was cleaned up with PSA column. Identification was achieved by electrospray ionization (ESI) in positive mode using multiple reaction monitoring. The calibration curve of P-CTX1 showed good linearity in the range of 0.5 -20μg/L with correlation coefficient of 0.9998. The limit of quantitation (LOQ) was 0. 1 mg/L( S/N = 10). The recoveries of P-CTX1 were between 82. 3% and 87. 2% (n =7) at spiked levels of 1.0 -20.0 ng/kg. The relative standard derivations( RSD) of the method were less than 7.8%.

Two sample pretreatment methods of pesticide residues in Panax notoginseng of Chinese traditional medicine were developed. For Method I, the residues were extracted from homogenized tissue with n-hexane-dichloromethane (6:4) by means of ultrasonication, the crude extract was purified by an Envi-carb/NH2 solid-phase extraction (SPE) column. For Method II, matrix solid-phase dispersion (MSPD) technique was used for extracting and cleaning up. The eluates were concentrated by rotary evaporation, and then were redissolved in dichloromethane prior to GC-MS determination. The determination was performed in selected ion monitoring (SIM) mode with the external calibration for quantitative analysis. Under the optimal conditions, the results indicated that the methods are easier and faster, the recoveries of method I for the spiked standards at concentration of 0.01, 0.5, and 2.0 mg x kg(-1) were 81.90%-102.10% with the relative standard deviations (RSDs) of 3.60%-7.10%. The recoveries of method II were 96.26%-104.20% with the RSDs of 3.52%-7.94%. The detection limits (S/N) for residues of pesticides were in the range of 0.48-1.34 ng x g(-1). The results indicated that these multiresidue analysis methods can meet the requirements for determination of residue pesticides and can be appropriate for trace analysis of residue pesticides in Panax notoginseng.

5.Effect of ischemic preconditioning on myocardial Bcl-2 expression and mitochondrial structure during heart valve replacement surgery

AIM: To investigate the effect of ischemic preconditioning (IP) on myocardial Bcl-2 expression and mitochondrial structure during heart valve replacement surgery under cardiopulmonary bypass. METHODS: Fifty-four patients were prospectively randomized to receive or not ischemic preconditioning (IP) before cold cardioplegic arrest. Ischemic preconditioning in the IP patients (n=22) was induced by a single 2-min ischemia followed by 3-min reperfusion just before aortic clamping and cold crystalloid cardioplegia for myocardial protection. The control group (n=32) received no ischemic preconditioning before cold cardioplegic arrest. The levels of ejection fraction (EF), fractional shortening(FS) and stroke volume (SV) in both groups were measured and compared. troponin T (c-TnT) level, Bcl-2 protein expression and microscopic changes of myocardial mitochondrial structure were recorded for each group before and after surgery. RESULTS: The level of EF, FS and SV in IP group was higher than those in control group (P<0.05). No significant difference in preoperative c-TnT levels between two groups was observed. The level of c-TnT in IP group was lower than that in control group and with a declining trend over time of 6 h, 24 h, 48 h, 72 h and 5 d after surgery, respectively. The preoperative positive unit of Bcl-2 expression between two groups showed no statistical difference (P> 0.05). Postoperatively, the positive unit of Bcl-2 expression in IP group was 19.85±5.88, significantly increased as compared to the preoperative value (P<0.05). In control group, the positive unit of Bcl-2 expression was 14.17±3.39, showed no statistically significant difference to the preoperative value (P>0.05). Postoperative Bcl-2 expression between two groups showed a significant difference (P<0.05). In the control group, microscopic observation revealed swollen mitochondrion, with a hardly visible or disrupted membrane for some mitochondrion;mitochondrial crista were obviously dissolved and loose with a large number of vacuoles formation. However in IP group, myocardial mitochondrion appeared with intact membrane, concentrated mitochondrial cristae with high electron density and no vacuoles formation was observed. CONCLUSION: IP may up-regulate the expression of myocardial anti-apoptotic protein Bcl-2 to protect the mitochondrion, thus protecting cardiocytes and cardiac functions.
6.Development of a Gene Chip for the Detection of Common Pathogens Causing Urogenital Sexually Transmitted Infections

Objective To develop a gene chip for the detection of common pathogens causing urogenital sexually transmitted infections. Methods The target pathogens were divided into three groups: viruses, bacteria, and lower eukaryotes. Three pairs of universal primers were designed and applied to amplify the target genes of these different pathogens in one PCR reaction system. The gene chips were then prepared via immobilization of the specific probes onto specially treated glass slides. Finally, the labeled amplicons were hybridized with the gene chips, scanned and analyzed using computer software. Results Amplicons were detected by agarose gel electrophoresis. The fluorescence signals for specific pathogens could be recognized in the gene chips, and were identical with the positions of the specific probes. Conclusions Gene chip is a specific, sensitive and rapid method for simultaneous detection of multiple sexually transmitted infections.
7.Diterpenoid alkaloids from Aconitum handelianum.

OBJECTIVETo investigate the alkaloids from Aconitum handelianum.
METHODThe column chromatographic methods were employed for the isolation and purification of the chemical constituents. The structures were elucidated by spectroscopic methods.
RESULTEight diterpenoid alkaloids were isolated and identified as acoforine (1), acoforestinine (2), 14-O-acetylsachaconitine (3), vilmorrianine C (4), vilmorrianine D (5), talatizamine (6), chasmanine (7) and yunaconitine (8).
CONCLUSIONAll compounds were isolated from this plant for the first time.

Cemento-ossifying fibroma is a rare benign tumor from periodontium, which usually occurs in mandible body and mandible ramus. It consists of collagen fibrils, fibroblast, and cementoblast. This article reported a case of giant cemento-ossifying fibroma and discussed the clinical features and treatment.

9.Application of nursing intervention based on acceptance and commitment therapy in patients with inflammatory bowel disease

Objective:To explore the application effect of nursing intervention based on acceptance and commitment therapy (ACT) in patients with inflammatory bowel disease (IBD) .Methods:Using the convenient sampling method, a total of 71 IBD patients who were hospitalized in Department of Gastroenterology in the Affiliated Tumor Hospital of Harbin Medical University from June 2018 to October 2019 were selected and they were randomly divided into the experimental group (36 cases) and the control group (35 cases) . The control group received routine care, while the experimental group implemented ACT-based nursing intervention on this basis. The self-efficacy, psychological flexibility and quality of life scores of patients of the two groups before and after intervention were compared.Results:Finally, 33 patients in the experimental group and 34 patients in the control group completed the study. After the intervention, Acceptance and Action Questionnaire-2nd Edition score of the experimental group was lower than that of the control group, and the difference was statistically significant ( P<0.01) . After the intervention, scores of Inflammatory Bowel Disease Self-efficacy Scale and Inflammatory Bowel Disease Questionnaire of the experimental group were higher than those of the control group, and the differences were statistically significant ( P<0.05) . Conclusions:Nursing intervention based on acceptance and commitment therapy can improve psychological flexibility and self-efficacy of IBD patients and improve their quality of life.
10.Optimization of Primary Processing Technology of Gentiana rigescens by Box-Behnken Design-response Surface Method

OBJECTIVE:To optimize the primary processing technology of Gentiana rigesce ns. METHODS :HPLC method was adopted for content determination of loganic acid ,swertiamarin and gentiopicroside in G. rigescens ,and overall desirability value (OD value ) of the contents of above 3 components was taken as index to carry out single factor test on blanching temperature,blanching time and drying temperature. Based on that ,Box-Behnken design-response surface methodology was used to optimize primary processing technology of G. rigescens . Validation test was also performed. The samples prepared by optimized technology were compared with those dried in the shade. RESULTS :The optimal primary processing technology of G. rigescens included blanching time of 5 min,blanching temperature of 40 ℃ and drying temperature of 60 ℃. Validation test showed that the average OD value of the 3 components was 0.565 2,with a deviation of 0.94% from the predicted value (0.570 6). Compared with samples dried in the shade ,OD value of 3 components in samples prepared by optimized technology were increased significantly , indicating the quality of the samples prepared by the optimized technology was better. CONCLUSIONS :The optimal technology is stable and feasible ,and can be used for the primary processing of G. rigescens .
