1.IMMUNOHISTOCHEMICAL STUDY OF ENDOCRINE CELLS IN THE DIGESTIVE TRACTS OF CANIS LUPUS AND CANIS FAMILIARIS

Objective To clarify the morphological features,types,regional distributions and cell densities of gastrointestinal endocrine cells in various parts of the digestive tracts of Canis lupus and Canis familiaris. Methods Immunohistochemical techniques of streptavidin-peroxidase(SP) method was used. Results Seven kinds of gastrointestinal endocrine cells were found positive in the digestive tracts of Canis lupus and Canis familiaris.The endocrine cells were mainly located between the epithelia cells of gastric gland,intestinal villus and intestinal gland.There were also some endocrine cells distributed in the lamina propria and submucosa.The endocrine cells found in the digestive tract were in various shapes.Most of the endocrine cells had long cell processes run ning out to the lumen or the nearby cells.Among these endocrine cells,5-hydroxytryptamine(5-HT) positive cells had a widest distribution,followed by vasointestinal poplypetide(VIP) positive cells.The distributions of other cells were limited.From the comparison of the characters of endocrine cells of Canis lupus and Canis familiaris,most of the endocrine cells had similar morphological features and regional distributions except pancreatic polypeptide(PP) positive cells and substance P(SP) positive cells.There were differences in cell densities of some endocrine cells.Conclusion The results of Canis lupus and Canis familiaris reflect the similarity of mammals in the characteristics of the morphological features,types,regional distributions and cell densities of gastrointestinal endocrine cells,as well as the similarity in digestive physiology of canine animals.At the same time,the results of Canis lupus also exhibit characteristics of gastrointestinal endocrine cells of wild animals.
2.Hemangiopericytoma in nasal cavity: a case report.

We report a case of a 46 year old female patient with nasal hemangiopericytoma. She complained of left nasal congestion, pus snot for 10 years, sometimes with left nasal bleeding. Physical examination: in the left nasal tract saw red soft neoplasm, roughness surface, easy bleeding when touched. Sinus CT shows: bilateral maxillary sinus, ethmoid sinus, sphenoid sinus and the left posterior nasal cavity lesions, considering inflammation with the formation of polyps, tumor not excluded. The left nasal cavity neoplasm biopsy shows: hemangioma of left nasal cavity. After admission in general anesthesia, we do transnasal endoscopic sinus openning operation and the left nasal cavity neoplasm resection. Postoperative pathological examination shows: the left nasal cavity hemangiopericytoma. Immunohistochemical showed: Vimentin(+), Smooth muscle actin(+), Desmin(-), endothelial cells CD31(-) and CD34(-). No postoperative radiotherapy or chemotherapy, no tumor recurrence. After one year of follow-up, the contact was lost.

3.Loop-mediated isothermal amplification for detecting Streptococcus pneumoniae and its clinical appfication

Objective To establish loop-mediated isothermal amplification(LAMP)method for detecting Streptococcus pneumoniae in clinical samples.Methods Four Streptococcus pneumoniae-specific LAMP primers were designed according to the published sequence of strain R6(GenBank accession number AE008540).Genomie DNA in positive blood cultures was extracted by nanidine hydrochloride and benzene-methanol.Then lytA was amplified by LAMP at 63℃ for 45 minutes.We observed the turbidity in thereaction tube.For further confirmation.The amplified products were also detected using electrophoresis in 2% agarose gels,followed by ethidium bromide staining.Resuits 16 strains of Streptococcus pneumoniae were detected in 196 positive blood cuhure bottles by LAMP.Compared with traditional method.It8 sensitivity and specificity were both 100%and the detection could be finished in an hour.The assay had a minimum detection limit of 102 CFU/m1.Conclusions This IJAMP-based assay is simple,rapid.Sensitive and specific.It can be used to detect trept OCOCCUS pneumoniae in clinical samples.
4.Efficacy of neuronavigation-guided selective percutaneous radiofrequency aiming at the target point semilunar ganglion in treatment of trigeminal neuralgia

Objective To evaluate the efficacy of neuronavigation-guided selective percutaneous radiofrequency aiming at the target point semilunar ganglion in the treatment of trigeminal neuralgia.Methods One hundred and forty-seven patients with primary trigeminal neuralgia of both sexes,aged 32-99 yr,with VAS score ≥ 8,were randomly divided into 2 groups:C-arm group (group C,n =72) and neuronavigation group (group N,n =72).Hartel anterior puncture was used and the C-arm guided puncture was performed at the target point foramen ovale in group C.In group N,three-dimensional reconstruction was made after the skull MRI images were transmired to the navigational system of StealthStation,and then the puncture path and point were designed after the target in the trigeminal ganglion was determined.The successful puncture and puncture-and radiofrequency-related complications were also recorded.The VAS scores were recorded at 1 and 7 days and 1,6,12 and 24 months after operation and the analgesic efficacy was evaluated.The therapeutic effect was evaluated by using Barrow Neurological Institute scoring system at 1 and 24 months after operation.Results No puncture-related side effects,damage to the oculomotor nerve or tinnitus developed in group N.The success rate of puncture at first attempt and the effective analgesia rate at different time points after operation were significantly higher,and the treatment effect was better in group N than in group C (P < 0.05).There was no significant difference in the time for location of the nerve and incidence of facial numbness between the two groups (P > 0.05).Conclusion Neuronavigation-guided selective percutaneous radiofrequency aiming at the target point semilunar ganglion in the treatment of trigeminal neuralgia provides a better efficacy,and a lower recurrence rate and a higher probability of successful puncture,with fewer complications.
5.Inhibitory effect of nicotinylsalicylic acid on rabbit platelet aggregation induced by ADP,AA and Collagen

Aim To investigate the effect of nicotinyl salicylic acid(NSA) on rabbit platelet aggregation induced by Collagen,ADP and AA.Methods Platelet aggregation induced by collagen,adenosine diphosphate(ADP) or arachidonic acid(AA) was studied with turbidimetry in rabbtis blood,in vitro and vivo.Results NSA significantly inhibited the platelet aggregation induced by Collagen and ADP,in vitro and vivo.The inhibition by NSA was dose-dependent.NSA had no effect on the platelet aggregation induced by AA.Conclusion NSA can inhibit rabbit platelet aggregation induced by Collagen and ADP.
7.Effect of high ilioinguinal approach in the treatment of acetabular fractures involving quadrilateral plate

Objective To study effect of high ilioinguinal approach in treatment of acetabular fractures involving quadrilateral plate. Methods 28 cases of acetabular fractures were randomly divided into two groups according to admission time. 16 patients in the observation group were given high ilioinguinal approach operation. 12 patients in the control group were treated with traditional ilioinguinal approach op-eration. Operation condition, fracture reduction, the hip function recovery and postoperative complications of the two groups were compared. Results The amount of bleeding, the length of incision, operation time and operation conditions of the two groups had no significant differ-ence (P>0. 05). The satisfactory fracture reduction rate of observation group was higher than the control group (P<0. 05). Hip function score of observation group was significantly higher than that of the control group (P<0. 05). In observation group, the excellent rate of hip joint function was better than that of the control group (P<0. 05). The incidence rate of complications in the observation group and the con-trol group were 12. 50% and 16. 67% respectively, and there was no significant difference between the two groups (P>0. 05). Conclusion High ilioinguinal approach can improve the fracture reduction effect, effectively improve the hip joint function in patients, and it has higher security which is a kind of ideal treatment approach for acetabular fractures involving quadrilateral plate.
8.Influence of cervical curvature change on the nerve root and vertebral artery of patients with cervical spondylosis and clinical observation of manipulation treatment

OBJECTIVE: To observe the influence of the change of cervical curvature on the nerve root and the vertebral artery of the patients with cervical spondylosis and the curative effect of therapeutic manipulations. METHODS: Sixty cases of cervical spondylosis with the change of cervical curvature, 60 cases of cervical spondylosis without the change of cervical curvature and 60 normal people were chosen. They were clinically observed and the cervical spondylosis was treated by manipulations and the data was processed according to the principle of statistics. RESULTS: The change of cervical curvature affected the nerve root and the vertebral artery and therapeutic manipulations produced a satisfactory curative effect. CONCLUSION: Cervical spondylosis with change of cervical curvature has special characteristics in the etiology, course and state of the disease. The change of cervical curvature produces an effect on the nerve root and the vertebral artery. Selected therapeutic manipulations produce a better curative effect.
9.The application value of estimating 24 h urinary iodine excretion in the evaluation of individual iodine nutrition level

Objective:To explore the application value of creatinine correction method in estimating 24 h urinary iodine excretion of healthy adults.Methods:Healthy adults(half males and half females) aged 18 to 59 yesrs old in Tangshan City were selected as the research subjects. Random urine within 1 d, morning urine and 24 h urine samples of 5 d in a row of all subjects were collected to detect urinary iodine and creatinine concentrations. Twenty-four hours urinary iodine excretion was estimated by creatinine correction formula [24 h urinary iodine excretion estimated value(μg/d) = urinary iodine/creatinine concentration ratio (μg/g Cr) × 24 h urinary creatinine excretion (g Cr/d)]. The estimated value of 24 h urinary iodine excretion obtained from random urine within 1 d and continuous 5 d morning urine corrected by creatinine were studied by the coefficient of variation, and compared with the measured value of 24 h urinary iodine excretion.Results:The coefficient of variation of the estimated 24 h urinary iodine excretion of random urine within 1 d ranged from 8.8% - 20.2%, with an average value of 13.1%; the coefficient of variation of 24 h urinary iodine excretion estimated value from continuous 5 d morning urine and the 24 h urinary iodine excretion measured value ranged from 4.7% - 17.2% and 4.0% - 10.4%, and the average values were 10.1% and 6.4%, respectively. In the detection of random urine within 1 d, there was no significant difference between the 24 h urinary iodine excretion estimated value of the first, second, third, and forth time random urine and the 24 h urinary iodine excretion measured value ( P > 0.05). In the detection of continuous 5 d morning urine, there were no significant differences in the 24 h urinary iodine excretion estimated value of the first, second, third, forth and fifth day compared with the 24 h urinary iodine excretion measured value on the same day, respectively ( P > 0.05). Conclusions:When the daily creatinine excretion is accurately known, the estimated 24 h urine iodine excretion obtained by the creatinine correction method can better reflect the actual 24 h urinary iodine excretion. Further study on prediction criteria of creatinine excretion suitable for Chinese people will help to better detect the iodine intake of corresponding individuals.
10.Establishment of a real-time PCR assay for detection of the activity of HIV-1 reverse transcriptase

Objective To reconstruct the initiative procedure of HIV-1 reverse transcription in vitro and establish a methodology of assessing activity of HIV-1 reverse transcriptase (RT) with real-time PCR Methods The tRNALys-3 gene was amplified from genome in healthy individuals through polymerase chain reaction (PCR), and then T7 transcription promoter was added in 5'-terminal of the tRNALys-3. The tRNA[Lys-3 cRNA product was obtained by applying T7 RNA polymerase through a transcription reaction. The 5'-LTR-PBS DNA was also obtained by transcription reaction from the HIV-1 infectious clone and inserted into pGEM-T easy vectors. 5'-LTR-PBS cRNA was obtained by applying SP6 RNA polymerase whose combining site was located in pGEM-T easy vectors. Then the two RNA samples was catalyzed by two kinds of standard reverse transcriptases (SuperScript Ⅲ and HIV-1 standard reverse transcriptase, respectively) and the cDNA was synthesised. The relative activity of RT was determined with the real-time PCR. Results The tRNALys-3 primer and the SP6-5'-LTR-PBS RNA were procured accurately, whose length were 93bp and 872 bp, respectively. After the following serial dilution of Super Script Ⅲ and HIV-1 standard reverse transeriptase:1 : 10, 1: 100, 1:1 000, 1:10 000, each step of reverse transcription process worked successfully. Real-time PCR results showed that Ct values of the two groups were 13.9, 18. 3, 20. 9, 24. 9 and 20. 4, 25. 5, 28. 7, 32. 5 respectively. Conclusion A novel real-time PCR method is developed to assay the RT activity directly through reconstructing the initiation of HIV reproduction, which may be helpful for clinical management, screening of new antiretroviral drugs, and drug resistance test.
