Object To develop a method for the determination of polysaccharide in TIANCI CAPSULE, which is a new drug dosage form containing the bouillon of fresh water mussels with immunopotentiating activity Methods Total polysaccharide was determined by phenol sulfuric acid method with glucose as the standard, and spectrophotometer assay absorbency at 490 nm Results The linear range of polysaccharide was 10～80 ?g/mL, r=0 999 7 The average recovery rate was 98 67% (n=5), RSD=2 58% Through sample assay, the content of polysaccharide was 267 7～300 6 mg in each TIANCI CAPSULE Conclusion The method is simple, rapid and reliable, and may be used for the quality control of this preparation

ATP-sensitive potassium channel(KATP) consists of a 4.4 complex of an inwardly rectifying Kir6.x pore plus a sulfonylurea receptor,which is an ATP-binding cassette transporter.KATP has been indentified in a variety of tissues and recognized as an important drug target.It connects cell metabolism with cell electric activity.KATP has been proposed to play protective roles during heart failure,arrhythmia,myocardial infarction,stress,myocardial ischemia and hypertension.In this review,a summary of KATP is presented with molecular structure,localization,regulation,cardiovascular protective effect and its mechanisms.

Objective:To discuss the variation of parameters in ambulatory blood pressure for neurally mediated syncope(NMS) in children,and to analyze the diagnostic value of ambulatory blood pressure pattern in children with NMS.Methods: Forty-seven children with NMS [20 males and 27 females,mean age(11.7?2.8) years] who came from Peking University First Hospital from July,2007 to March,2008 were included in the study.Twenty-three healthy children [12 males and 11 females,mean age(11.0?3.2) years] were recruited as control group.And the hemodynamic patterns of NMS were detected.Based on the hemodynamic patterns,they were divided into vasovagal syncope(VVS) group [16 children in total,7 males and 9 females,mean age(11.5?2.8) years] and postural tachycardia syndrome(POTS) group [31 children in total,13 males and 18 females,mean age(11.7?2.9) years].Parameters of children with different hemodynamic patterns in ambulatory blood pressure and diagnostic value of ambulatory blood pressure pattern were analyzed.SPSS 10.0 software was used for the statistical analysis of these data.Results: Mean diastolic pressure of the whole day,mean diastolic pressure in the day-time and mean systolic pressure at night in POTS group were higher than those of the control group,reapectively(P0.05).The systolic pressure differences in the day-time and at night in VVS group and POTS group were lower than those of the control group,respectively(P0.05).The non-spoon pattern rate of fluctuation curve of ambulatory blood pressure in VVS group and POTS group was higher than that of the control group(68.8% vs 17.4%,64.5% vs 17.4%,P0.05).The sensitivity,specificity and diagnostic coincidence of ambulatory blood pressure pattern to NMS was 66.0%,82.6% and 71.4%,respectively.Conclusion: There was autonomic nerve adjustment imbalance in children with NMS.And the diagnostic value of ambulatory blood pressure pattern to children with NMS was high.

Object To develop a capillary GC-MS analytical method for identification and deter- mination of ginkgolide A, B, C and bilobalide (GA, GB, GC and BB) in Ginkgo biloba L. leaves. Methods The leave samples were extracted in ultrasonic bath with ethanol-water (20∶80). The extract was purified by liquid-liquid extraction with ethyl acetate followed by solid-phase extraction on a column mixed with acid Al 2O 3, active carbon and celite. The terpenes were trimethylsilylated by BSTFA (with 1% TMCS) for 60 min at 100 ℃ and determined by GC-MS with HP-5 MS capillary column in the selected-ion monitoring mode. The intense fragment ions were chosen as monitoring ions for quantitative analysis. Cholesterol was used as an internal standard. Column temperature gradient: initial temperature 180 ℃, maintained 1 min, and then increased at 20 ℃/min to 260 ℃, and finally at 2 ℃/min up to 300 ℃, maintained 2 min. Results The retention times of GA, GB, GC and BB were 13.7,14.3,15.3 and 6.8 min, the major fragmentation ions (monitoring) were at m/z 537, 625, 713 and 455 (299), the average recoveries of GA, GB, GC and BB were 102.0%, 99.4%, 96.0%, 96.3%, RSD were 0.54%, 2.40%, 1.98% and 2.43%, respectively. Conclusion This method is repeatable, specific, accurate and easy to operate. It is adoptable for quality and quantity analysis of terpene lactones from G. biloba leaves.

Objective To explore possible impact of endogenous hydrogen sulfide(H2S) on connective tissue growth factor(CTGF) expression in rat pulmonary artery with high pulmonary blood flow.Methods Thirty-two male SD rats,weighing 120～140 g,were randomly divided into 4 groups:shunt group,shunt+PPG group,sham group and sham+PPG group.After 4 weeks of experiment,Rat lung tissue H2S content was determined by a modified sulfide electrode method.Plasma ET-1 concentration was detected by radioimmunoactivity,and lung tissue ET-1 mRNA expression of rat was determined by quantitative competitive reverse transcriptional polymerase chain reaction(RT-PCR).Pulmonary artery connective tissue growth factor(CTGF) protein expression of rats was investigated by immunohistochemistry.Results After 4 weeks of experiment,lung tissue H2S content plasma ET-1,lung tissue ET-1 mRNA and CTGF expression increased significantly in rats of shunt group as compared with that of sham group(P

Aim To investigate the effect of sulfur dioxide (SO2) on blood pressure and aortic structure of the spontaneous hypertensive rat(SHR).Methods Sixteen male SHRs at the age of 4 weeks were randomly divided into SHR control group and SHR+Na2SO3/NaHSO3 group,respectively.Another eight male WKY rats served as WKY control group.Five weeks later, the blood pressure was determined.The rat aortas were dyed with Hart's Method. The morphometric parameters including outer radius, lumen radius and the wall thickness were calculated by Leica workstation. Objective To study the change of the renal endogenous hydrogen sulfide (H2 S) pathway in the development of salt-sensitive hypertension in Dah1 rats.Methods Sixteen male Dah1 rats,in accordance with the random number table,were randomly divided into control group and high salt group fed with diet containing 80 g/kg NaCl.After 8 weeks,24 h urine sodium,24 h urinary protein,serum creatinine and serum urea were measured.The microstructural and ultrastructural changes in kidney were observed with light microscope and electronic microscope.The serum and kidney H2S contents were determined by using sulphur-sensitive electrode method.The mRNA levels of cystathionine β-synthase(CBS),cystathionine γ-lyase(CSE) and mercaptopyruvate sulfurtransferase(MPST) in renal tissue were determined by means of real-time polymerase chain reaction(RT-PCR).The protein expressions of CBS,CSE and MPST in renal tissue were detected by using Western blot.Results Compared with control group,high salt group rats had a significant rise in blood pressure,declined renal function,damaged renal structure,segmental glomerular sclero sis,small artery wall thickening,and occlusion of the lumen.Moreover,the endogenous H2S pathway in kidney of Dah1rats with high salt diet was downregulated markedly,demonstrated by the decreased serum and kidney H2S content,the reduced renal CBS,CSE and MPST mRNA expressions and CBS protein expression of kidney tissue.Conclusion The endogenous H2S/CBS pathway is downregulated during the development of salt-sensitive hypertension in Dah1 rats.
