1.Impacts of different stimulation cycles on outcomes of intrauterine insemination

Objective To explore the impacts of natural ovulation cycles and stimulation cycles on the outcome of intrauterine insemination (IUI) in order to improve the clinical effects of IUI. Methods 176 women received 384 stimulation cycles. According to different ovulation stimulation protocols , the women were divided into six groups including natural cycle (NC) group, clomiphene citrate (CC) group, letrozole (LE) group;human menopausal gonadotrophin (HMG ) group, CC + HMG group, and LE + HMG group. The pregnancy rate between nature cycles and ovarian hyperstimulation cycles was compared. Results The pregnancy rate was 9.33%in the nature cycle group and 13.27% in the stimulation cycle group, with a significant difference (P < 0.05);and it dif not differ significantly among the stimulation cycle groups (P > 0.05). Conclusions Use of ovulation-induction medications is one of the important factors affecting the pregnancy rate of intrauterine insemination. There are no differences in the outcome of IUI among different ovulation stimulation protocols.
2.Quantitative assessment of ischemic liver's elastic changes in rabbits using acoustic radiation force impulse (ARFI)

Objective To evaluate the change of elasticity of rabbit liver following portal inflow vessels occlusion using acoustic radiation force impulse (ARFI). Methods Twelve Japanese white big ear rabbits were used to establish total hepatic vascular exclusion animal models by occluding the portal inflow vessels. The elasticity of rabbits' right liver was detected during the stages of baseline(T0), 5 min(T1),20min(T2) ,30min (T3)after occluding the portal inflow vessels using ARFI respectively. The elasticity of rabbit liver, levels of serum's aspartate aminotransferase (AST), alanine aminotransferase (ALT) and historical results of hepatic tissue during different stage were compared. Results Shear wave velocity(Vs)of rabbits' right livers in T2 was significantly higher than that in T0 ( P <0.05) and Vs of rabbits' right livers in T3 was significantly higher than that in T2 ( P <0.05). No significant differences of serum's AST,ALT levels between T2 and T0 were found,while the Vs of rabbit live in T2 was slightly higher than that in T0 (P>0.05), serum's AST, ALT levels in T3 were significantly higher than that in T2 ( P <0.05).Historical changes of liver ischemia and injury were observed in T2 and T3 with light microscopy. Conclusions The liver ischemia and injury severity can be accurately and objectively evaluated using ARFI.
3.Idenfication of microRNAs profiles in nasopharyngeal carcinoma.

OBJECTIVE:
To filtrate and prove the different microRNAs (miRs) profiles in nasopharyngeal carcinoma.
METHOD:
Screening the different expressions of miRs between nasopharyngeal carcinoma and the inflammatory tissues by the application of expression profiling of chip high-throughput and large-scale microarray analysis. Then we used RT-QPCR technology to prove the accuracy of screening results.
RESULT:
There were significant expression differences of miRs between nasopharyngeal carcinoma and the control tissues, 144 human miRs had 2 or more fold the difference ratio. Compared with the inflammatory tissues, we have found that miRs-34b, miRs-449b and miRs-7-1 significantly low expressed in nasopharyngeal carcinoma, yet miRs-125b, miRs-184, miRs-196b, miRs-205 and miRs-24-1 expressed high. The results were consistent with the microarray analysis.
CONCLUSION
The difference expressed miRs might be closely related to the process of nasopharyngeal carcinoma, and the research on miRs profiles maybe provide a powerful target basis for early diagnosis and therapy of nasopharyngeal carcinoma.

4.Expression of SP1 in esophageal squamous cell carcinoma and its effect on proliferation and apoptosis of esophageal squamous cell carcinoma cells

Objective To investigate the expression of Specific protein1(SP1)in esophageal squamous cell carci-noma(ESCC)and adjacent normal tissues and its effect on the proliferation and apoptosis of ESCC cells.Methods The expression of SP1 protein in 121 ESCC tissues and 74 adjacent normal tissues was detected by immunohisto-chemistry.Chi-square test and Cox regression analysis were used to analyze the relationship between SP1 and clini-copathological parameters and survival prognosis of ESCC patients.SP1 siRNA(small interfering RNA)was con-structed and transfected into esophageal squamous cell carcinoma Eca1 09 and EC9706 cell lines.Western blot was used to detect the expression of SP1 after transfection.The effects of SP1 on the proliferation and apoptosis of e-sophageal squamous cell carcinoma cells were detected by cloning assay,CCK-8 cell proliferation assay and flow cytometry.Results SP1 protein was expressed in the nucleus of esophageal squamous cell carcinoma tissues,and the expression rate of SP1 protein in esophageal squamous cell carcinoma tissues was significantly higher than that in adjacent normal tissues(x2=20.568,P<0.01).Comparison between groups showed that the high expression rate of SP1 was higher in female(P=0.041),moderately or poorly differentiated(P=0.038)and T3-T4 inva-sion depth(P=0.041)ESCC(esophageal squamous cell carcinoma)patients.Log-rank test showed that the sur-vival time of patients with high expression of SP1 was shorter than that of patients with low expression of SP1(P=0.048).Multivariate Cox regression analysis showed that TNM(tumor node metastasis classification)stage(Ⅲ+Ⅳ)was a potential risk factor for shorter survival time in patients with esophageal squamous cell carcinoma(P<0.001).Cell biological experiments showed that compared with the control group,the proliferation ability of esoph-ageal squamous cell carcinoma cell lines decreased(P<0.05)and the apoptosis index increased(P<0.05)after silencing SP1.Conclusion SP1 protein is highly expressed in human esophageal squamous cell carcinoma tissues and is associated with poor prognosis in patients.Silencing SP1 can inhibit the proliferation of esophageal squamous cell carcinoma cells and promote their apoptosis.
5.Zinc finger protein-36 deficiency inhibits osteogenic differentiation of mouse bone marrow-derived mesenchymal stem cells and preosteoblasts by activating the ERK/MAPK pathway

Objective To explore the role of zinc finger protein 36 (ZFP36) in regulating osteogenic differentiation of bone marrow-derived mesenchymal stem cells (BMSCs) and preosteoblasts. Methods ZFP36 expression was observed in primary mouse BMSCs and mouse preosteoblasts (MC3T3-E1 cells) during induced osteogenic differentiation. Zfp36-deficient cell models were constructed in the two cells using RNA interference technique and the changes in differentiation capacities of the transfected cells into osteoblasts were observed. Transcriptome sequencing was used to investigate the potential mechanisms of ZFP36 for regulating osteoblast differentiation of the two cells. U0126, a ERK/MAPK signal suppressor, was used to verify the regulatory mechanism of Zfp36 in osteogenic differentiation of Zfp36-deficient cells. Results During the 14-day induction of osteogenic differentiation, both mouse BMSCs and MC3T3-E1 cells exhibited increased expression of ZFP36, and its mRNA expression reached the peak level on Day 7 (P<0.0001). The Zfp36-deficient cell models showed reduced intensity of alkaline phosphatase (ALP) staining and alizarin red staining with significantly lowered expressions of the osteogenic marker genes including Alpl, Sp7, Bglap and Ibsp (P<0.01). Transcriptome sequencing verified the reduction of bone mineralization-related gene expressions in Zfp36-deficient cells and indicated the involvement of ERK signaling in the potential regulatory mechanism of Zfp36. Immunoblotting showed that pERK protein expression increased significantly in Zfp36-deficient cells compared with the control cells. In Zfp36-deficient MC3T3-E1 cells, inhibition of activated ERK/MAPK signaling with U0126 resulted in obviously enhanced ALP staining and significantly increased expressions of osteoblast differentiation markers Runx2 and Bglap (P<0.05). Conclusions ZFP36 is involved in the regulation of osteoblast differentiation of mouse BMSCs and preosteoblasts, and ZFP36 deficiency causes inhibition of osteoblast differentiation of the cells by activating the ERK/MAPK signaling pathway.

7.Comparison of management training demands of charge nurses with different types of leadership in general tertiary hospitals

ObjectiveTo analyze the main types of the leadership in the charge nurses in ClassⅢ general hospitals by the potential category model, so as to explore the management training needs for the different types of charge nurses. MethodsBy cluster random sampling, charge nurses from 11 ClassⅢgeneral hospitals in Zhejiang province were selected from October to December of 2017 and studied by General Information Questionnaire, Leadership Questionnaire, and Training Needs Questionnaire. By Mplus 7.0 software, the potential category model of charge nurses' leadership was analyzed and all categories of charge nurses' training needs were analyzed by SPSS 19.0 software. Totally 782 questionnaires were collected with 736 valid ones left after omitting the invalid ones, yielding an effective recovery rate of 94.12%. ResultsPotential category model analysis showed that in the 736 charge nurses, 3 leadership categories of high level of leadership, medium level of leadership and low level of leadership accounted for 12.64%,55.84% and 31.52% respectively. The charge nurses had high demands on management position training, with each clauses exceeding 3.50 points, there were statistical differences in the 3 potential leadership categories of charge nurses'training demands as well as scores from each clauses(P< 0.05). There were no statistical difference between each categories in terms of the training needs of "nursing quality management","hospital infection management""occupational protection" "computer application skills" "human resource management" and "scheduling as well as basic qualify and abilities" (P>0.05). ConclusionsThere are 3 types of leadership in the charge nurses of the Class Ⅲ general hospitals. The clauses with high demands among the nurses with low level of leadership include "nursing quality management" "nursing safety management"; the clauses with high demands among the nurses with medium level of leadership include "staff stimulation and talent cultivation" "nursing safety management"; the clauses with high demands among the nurses with high level of leadership include "strategic management setup" "leadership improvement", etc.
8.The effect of staged group positive psychological intervention on resilience and professional identity of"Post-90s" nursing students during internship

Objective? To explore the effect of staged group positive psychological intervention on resilience and professional identity of "post-90s"nursing students during internship. Methods? A convenience sampling method was adopted. Nursing students of 2017 were selected as a control group (n=151) and nursing students of 2018 were selected as an intervention group (n=137). Nursing students in control group were given a conventional teaching method, while the psychological counseling team used staged group positive psychological interventions on nursing students in intervention group. Connor-Davidson Resilience Scale(CD-RISC) and Professional Identify Scale were used to evaluate the effect of intervention. Results? The CD-RISC score and Professional Identify Scale score of intervention group were (89.58±21.37)and (5.48±0.29), both higher than those of control group(t=2.253,5.725;P< 0.05). Conclusions? The staged group positive psychological intervention has positive effect on the psychological resilience and professional identity of the "post-90s" nursing students during internship, which provides a reference for further maintenance of the mental health of nursing students and stabilization of the nursing team.
9.Rational selection between internal and external drainage approaches for microinvasive/minimally invasive glaucoma surgery in intraocular pressure control

Glaucoma stands as the leading cause of irreversible blindness, with primary open-angle glaucoma being the predominant type.Although its pathogenic mechanisms are intricate, lowering intraocular pressure remains the most effective treatment.Mainstream glaucoma surgeries aim to lower intraocular pressure by increasing aqueous humor outflow.Over the past two decades, microinvasive/minimally invasive glaucoma surgery (MIGS) has emerged as a hotspot in this field, focusing on minimizing incisions.Domestic practices have followed closely, but clinical outcomes have not been ideal.Based on a comprehensive understanding of the anatomy and pathophysiology of the aqueous outflow pathway and the design principles of various MIGS procedures, ophthalmologists should recognize the advantages and limitations of each technique.It is important to rationally choose between internal and external drainage MIGS and standardize their implementation.In the future, the use of non-invasive multimodal imaging techniques for preoperative assessment of the structural and functional aspects of the eye is expected to facilitate personalized selection and planning for precise treatment.

The 8th Japan-China Hepato-Pancreato-Biliary Symposium was held in Tokyo,Japan from 22nd to 23rd November 2018.The meeting was convened coincidently with the 80th Annual Congress of Japanese Surgical Association,which attracted the participation of Chinese and Japanese hepatobiliary and pancreatic surgeons.The symposium aimed to explore the latest achievements and clinical issues of diagnosis and treatment for hepato-pancreato-biliary diseases.In this article,authors reviewed the up-to-date research information in order to share the experience,achievements and new information in the field of hepatobiliary and pancreatic diseases with colleagues.
