2.Study on the relation between TCM constitution and familial hereditary in patients with type 2 diabetes

Objective This clinical research will compare the constitution of patients with positive and negative family history in order to study on the relation between TCM constitution and the familial hereditary in type 2 diabetes patients.Methods The data of family history,general conditions,Chinese medical constitutional types of 231 type 2 DM patients in Dongzhimen Hospital were collected and analyzed.Results Among patients with family history,the number of patients' mothers with diabetes was significantly more than that of fathers'(x2=22.319,P<0.05).There was a positive correlation between phlegm-dampness constitution r=0.215,blood stagnant constitution r=0.268 and female patients' family history(P<0.01).And there was a positive correlation between the familial hereditary of DM and the familial hereditary of hypertension r=0.328,hyperlipidemia r=0.223,obesity r=0.151 and coronary artery diseases r=0.215.Conclusion Among type 2 DM,female had stronger familial hereditary.It indicated that diabetic mothers had a stronger influence on following generations than diabetic fathers,especially those female patients of phlegm-dampness constitution or blood stagnant constitution.There was inner correlation between the familial hereditary of DM and the familial hereditary of hypertension,hyperlipidemia,obesity and coronary artery disease.
3.Electro-acupuncture decreases hydrogen sulfide concentration in brain cortex of baby rats with hypoxic-ischemic brain damage

Objective To establish hypoxic-ischemic brain damaged (HIBD) rat model,investigate whether H2S and cystathionine-β-synthase (CBS), the key enzyme for its generation, may be a mediator of electro-acupuncture(EA) stimulation treatment for HIBD. Methods Thirty-two healthy Sprague-Dawley neonatal rats were divided into four groups randomly: sham control group ( n = 8 ); sham + EA group ( n =8); HIBD control group ( n = 8); and HIBD + EA group ( n = 8 ). HIBD rat models were established on their 7-day-old. From the next day ,rats of sham + EA group and HIBD + EA group were electric stimulated 30 min daily for 14 d,BAIHUI and DAZHUI as the acupoints. Control ones were just fixed at the same time,without acupuncture. The rats were sacrificed on the 22 nd day, one day after the treatment course. Cortical H2S concentration was measured by sensitive sulphur electrode assay. The CBS protein expression was measured by western blot analysis and immunohistochemistry for localization. Results The concentration of cortical H2S in HIBD control group was (26. 83 ± 4. 31 ) nmol/mg protein, which was significantly higher than that of sham control group[(22. 78 ± 1.54) nmol/mg protein]( P < 0. 01 ). The H2 S levels in HIBD + EA group and sham + EA group were ( 18.08 ± 2.71 ) nmol/mg protein and ( 18.91 ± 2. 78 ) nmol/mg protein, respectively. Compared with corresponding control group, they were much lower( P < 0. 01 ). The expression of CBS protein in rats with EA stimulation decreased in cortex compared to corresponding control group( P <0. 05 ).Conclusion EA can down-resulate H2S/CBS pathway. This may be one of the mechanisms of how EA contributes to the recovery of brain damage.
4.Study on the distribution characteristics of Hyperthyroidism's syndrome

Objective To Analyze the distribution characteristics of Hyperthyroidism's syndrome and the relationship between symptoms and thyroid hormones. Methods The TCM syndromes of a total of 78 patients with hyperthyroidism were differentiated. The relationship between syndrome and general condition of patients (gender and age, etc.) were analyzed;correlation between symptoms and thyroid hormones were also studied. Results Of all 78 patients, patients with yin deficiency and hyperactive yang accounted for 43.6%; patients with internal disturbance of phlegm-fire accounted for 30.8 %; and patients with hypteracitivity of heart-fire and liver-fire accounted for 25.6%. The severity of all the symptoms showed positive correlation with hyroid hormones (the value of r is positive, P<0.05); and no linear relationship with TSH (P>0.05).Conclusion The syndrome of yin deficiency and hyperactive yangI was the most common syndrome type. Thyroid hormones had direct proportion with the severity of syndrome.

With the high incidence of diabetic nephropathy, the clinical trials of traditional Chinese medicine (TCM) in diabetic nephropathy area play an important role in the development of this subject. Therefore, follow-ing the principles of medical ethics, issues of fully understanding professional requirements of ethics review for clinical research of diabetic nephropathy and the protection of patients' rights are questions which should be solved. This article was based on the investigations on ethical needs of experts and patients in diabetic nephropathy area of TCM clinical trials . Discussions and considerations were made on results . A the more per-fect and regulated medical informed consent formwork for diabetic nephropathy of TCM clinical trials was made based on these consultations .

Objective To examine serum lipid levels and to investigate the risk factors of dyslipidemia in children and to study the indices of screening test for dyslipidemia of children in Beijing.Methods Serum total cholesterol(TC),triglyceride(TG),high density lipoprotein-cholesterol(HDL-c) and low density lipoprotein-cholesterol(LDL-c) were detected by HITACHI 7060 automatic biochemical analyzer in Beijing students.Body height, weight,upper body height,waist circumference,upper arm circumference and blood pressure were measured.Body mass index(BMI) was calculated according to the measurement of body height and weight.Waist-to-height ratio(WHR) was also calculated.Then a Logistic regression analysis and ROC study were conducted to examine the relationship between the above indices and dyslipidemia.Results Of 962 students,107 were found dyslipidemia.The prevalence rate was 11.12%.The upper body height,upper arm circumference,BMI,WHR,systolic pressure and diastolic pressure in children with dyslipidemia were higher than those of normal subjects(P

9.Paclitaxel and cisplatin inhibit the proliferation of nasopharyngeal cancer stem cells and promote apoptosis via the Wnt/beta-catenin pathway

BACKGROUND:Cancer stem cel s have self-renewal ability and can differentiate into new tumors. Cancer stem cel s are the source of tumor formation and recurrence, and they can make tumors insensitive to radiotherapy and chemotherapy.
OBJECTIVE:To explore the effect of paclitaxel plus cisplatin on the proliferation and apoptosis of nasopharyngeal cancer stem cel s (NPCSCs) and involved signal pathways.
METHODS:NPCSCs were sorted by immunomagneticbeads and were treated with paclitaxel, cisplatin or their combination. The expression of caspase-3, activated caspase-3 and Bcl-2, which are related to apoptosis, was determined by western blot. The expression ofβ-catenin and its downstream proto-oncogene, c-myc, was also determined by western blot. The activity of the Wnt/β-catenin pathway was inhibited by knocking downβ-catenin expression orβ-catenin inhibitor XAV939. Proliferation and apoptosis of NPCSCs were detected by MTT and flow cytometry, respectively.
RESULTS AND CONCLUSION:Either paclitaxel or cisplatin could inhibit proliferation and induce apoptosis of NPCSCs. The expression of apoptosis marker, activated caspase-3, was increased and the expression of the inhibitor of apoptosis, Bcl-2, was declined. Combined use of paclitaxel and cisplatin had synergistic effect when used together. Either paclitaxel or cisplatin could inhibit the expression ofβ-catenin and c-myc, suppressed the proliferation and induced the apoptosis of NPCSCs by inhibiting the activity of Wnt/β-catenin pathway. These results indicate that the combined use of paclitaxel and cisplatin may inhibit the proliferation of NPCSCs and promote apoptosis via the Wnt/β-catenin pathway.
10.An analysis of risk factors for brucellosis in employees working in the sheep market in Tongxiang of Zhejiang Province, 2010-2015

Objective To understand the epidemiological characteristics of brucellosis among risk population and analysis risk factors, and to provide a reference basis for prevention and treatment of brucellosis. Methods Retrospective survey method was used to collect information on brucellosis and epidemiologically confirmed cases in Tongxiang City from 2010 to 2015. The employees gender, age, occupation, job seniority, contact area, contact history to abortion cub and with or without protection measures were analyzed by single factor and multiple factors analysis, calculate odds ratio (OR) and 95%confidence interval (CI). Results Totally 1 450 people were investigated and 43 were confirmed positive (2.97%),new cases were 37 with joint pain 86.49% (32/37), low temperature 81.08%(30/37), and weak 62.16%(23/37) as the main body symptoms. Most people were infected in the summer season, accounted for 8.07%(18/223); female cases were relatively higher, accounted for 5.38% (12/223);people younger than 30 years old had the highest positive rate 24.14%(7/29);people working in the processing of animal products had the highest positive rate 6.42%(12/187). The employees gender, age, the types of work, the job seniority, the district distribution and the season were all related to the prevalence of the disease (all P< 0.05) analyzed by Chi-square test and Fisher's exact probability method. By single factor analysis, gender (male, OR =0.397, 95%CI:0.178-0.884), age (31-60 years old, OR = 0.254, 95%CI:0.081-0.796), and types of work (processing personnel, OR =3.211, 95%CI: 1.424 - 7.238) and contacting young animal (OR = 3.544, 95%CI: 1.729 - 7.264), practitioners operating tool (OR=0.231, 95%CI:0.070-0.766) were related to the prevalence of the disease;by multiple factors analysis, contacting young animal (OR=5.198, 95%CI:2.078-13.000) was a risk factor for brucellosis abortion, and practitioners operating tools (OR = 0.161, 95%CI: 0.045 - 0.577) were brucellosis protective factors such as wearing gloves . Conclusion We suggest to strengthen health education and health promotion in professional key populations, do a good job in the epidemic monitoring and promote occupational protection awareness, and decrease the infection of brucellosis.
