1.Method evaluation of plasmodium DNA template extraction from dried blood on filter paper

Objective To evaluate the method of plasmodium DNA template extraction from dried blood on filter paper . Methods Collectedand tested 47 blood samples of suspected imported malaria patients ,the DNA was extracted and tested by using conventional procedures and PCR ,then count and analyze test results of matched design .Results The Kappa value was 0 .492 when compared filter paper dried blood spots with thick and thin blood smear test results ,and the Kappa value was 0 .686 when com‐pared with whole blood sample .Using the results of thick and thin blood smear and whole blood samples as the gold standard to e‐valuate the sensitivity and specificity of filter paper dried blood spot method respectively ,the sensitivity was 75 .0% ,71 .4% and specificity was 78 .9% ,100 .0% .Conclusion Collected blood samples using filter paper can′t get good results in PCR test ,still needs further research .

Objective To investigate the expression and its clinical significance of serum urotensin Ⅱ(UⅡ)and nitric oxide(NO)in chronic cor pulmonale patients in acute exacerbation.Methods Totally 55 cases of chronic cor pulmonale in acute exacerbation(chronic cor pulmonale group)from Respiratory Department of First Affiliated Hospital of Jiamusi University during Nov.2006 to Mar.2007 and 40 healthy volunteers(normal control group)were collected.The levels of serum UⅡ and NO were detected in 55 cases of chronic cor pulmonale in acute exacerbation and 40 normal controls respectively.The condition of everyone's smoking history was collected.Arterial blood gas analysis was made in 37 cases of chronic cor pulmonale in acute exacerbation.The relationship among the levels of serum UⅡ,NO and PaO2,PaCO2,smoking history was observed.Results The levels of serum UⅡ and NO in chronic cor pulmonale in acute exacerbation were significantly higher than those of normal control group(all P

Purpose To study the effect of China cobra venom active factor(CCVAF) from China cobra venom on endothelial cells and its mechanism.Methods MTT experiment was adopted to evaluate the effect of CCVAF on bovine arteria pulmonalis vascular endothelial cells(BAVEC).The Eosin-Coomassie brillient blue and rhodamine-phalloidin method was used for actin cytoskeleton.Flow cytometry for [Ca~(2+)]_i and spectrophotometry were used for lactate dehydrogenase(LDH) and nitrogen oxide(NO) levels in cell culture supernatant.Results CCVAF(0.625-20 μg/mL) inhibited the proliferation of BAVEC in dose-dependent manner,and IC50 of CCVAF on BAVEC was 2.45 μg/mL. After CCVAF and BAVEC coincubation, it was showed that regression of intercellular conjunctions and disorder of F-actin distribution occurred. The content of [Ca~(2+)]_i, [LDH] and [NO] increased respectively.Conclusion CCVAF can inhibit BAVEC proliferation and it maybe associated with the change of cytoskeleton and increasing of [Ca~(2+)]_i,[LDH] aod [NO].
