1.Protective effect of imatinib on myocardial fibrosis in uremic rats

Objective To evaluate the effect of imatinib in improving myocardial fibrosis in uremic rats through regulating the expression of PDGFRα.Methods Seventy two rats were divided into three groups ,which were Sham group ,5/6 group and 5/6+I group .All The rats in 5/6 group underwent the 5/6 nephrectomy and the rats in 5/6+I group were given imatinib by ga‐vage after the operation of 5/6 nephrectomy .Hearts were harvested for HE and Sirius red staining at 8 weeks post surgery .The ex‐pression of PDGFRαwas assessed with immunohistological staining .The real‐time PCR was employed to detect the PDGFRαmR‐NA level in hear samples .Results The urine protein ,Scr ,BUN of the 5/6 group and 5/6+I group were higher than that of Sham group(P<0 .01) .The myocardial pathological score in Sham group was significantly lower than that of 5/6 group (P<0 .01) ,and the score in 5/6+I group was significantly lower than that of 5/6 group (P<0 .01) .The collagen volume fraction (CVF) in 5/6 group was significantly higher than that in Sham group (P<0 .01) .And the CVF in 5/6+ I group was higher than that in Sham group (P<0 .05) ,but lower than that of 5/6 group (P<0 .05) .The expression ratio of PDGFRαmRNA and staining rate in 5/6 group and 5/6+I group were both much higher than that in Sham group (P<0 .01) ,and the expression in 5/6+I group was signif‐icantly lower than that in 5/6 group (P<0 .05) .Conclusion These data suggest that the tyrosine kinase inhibitor imatinib reduces heart injury and attenuates myocardial fibrosis in uremic rat by mechanisms associated with the inhibition of the expression of PDGFRα.
2.Levels of volatile sulfur compounds and the analysis of related factors in oral cavities of 384 health subjects in Chengdu.

OBJECTIVEThe purpose of the study was to investigate the prevalence of oral malodor in a group of health cohorts in Chengdu, China by using different diagnostic methods.
METHODSVolatile sulfur compounds (VSCs) level was evaluated by using Halimeter. The organoleptic score and tongue coating index were also evaluated. A questionnaire interview was conducted at the same time.
RESULTSThere were 21.61% subjects whose oral VSCs level were more than 300 ppb. No significant difference was found between male and female for the VSCs values. The subjects with malodor (score > or = 6) evaluated by organoleptic score were 28.91%, and the difference between the male and female was significant (P < 0.05). Further, a significant correlation could be detected between the VSCs level, organoleptic score and tongue coating index (P < 0.05).
CONCLUSIONThe study indicates that nearly one of fourth people suffer from oral malodor. Malodor has significant correlation with tongue coating, so cleaning of tongue dorsum is very important to reduce oral malodor.

3.The effect of ART in the prevention of dental caries in Chinese school children

Objectives: To assess the effects of ART in the preventio n of dental caries in school children.Methods: Standard instrument s and procedures of ART were used in 191 fissure seal in 140 children aged 12~1 3 years. The material used was a high-strength glass-ionomer material (Ketac-M olar, ESPE). The treatments were evaluated annually after ART by clinical examin ation.Results: The cumulative 1-,2- and 3-year retention rat es of the sealant were 89.6%,78.8% and 71.9% respectively. The incidence of recu rrent caries 2 years and 3 years after ATR was 1.6% and 2.8% respectively. Conclusion: ART approach for preventing tooth decay in school childre n is effective.
4.Effect of minocycline on glutamate uptake in periventricular zone of neo-natal rats after hypoxia

AIM:To investigate the role of minocycline on glutamate uptake in the periventricular zone and its putative mechanism after hypoxic exposure in neonatal rats.METHODS: A model of hypoxic-ischemic brain damage (HIBD) was developed by putting postnatal 1 d rat pups in 5%O2 for 3.5 h.The glutamate level in periventricular zone was measured by liquid chromatography coupled with tandem mass spectrometry assay ( LC-MS/MS) after hypoxic exposure for 4 h and 1 d.The dynamic changes of glutamate transporters EAAT1, and EAAT2 during developmental period in periventricular zone were determined by Western blot.Moreover, the expression of EAAT1, EAAT2, Iba-1, IL-1β, TNF-αand TGF-β1 was also detected by Western blot after hypoxic exposure for 4 h and 1 d in that region.The effects of mino-cycline on all parameters mentioned above were tested after minocycline treatment at the same time points and in the same region.RESULTS:After hypoxic exposure, glutamate level was increased, but it was decreased after minocycline treat-ment.EAAT1 and EAAT2 kept a low expression level at the first postnatal week, but a predominant elevation was found at the end of the second postnatal week.The expression of EAAT1, EAAT2, Iba-1, IL-1βand TGF-β1 was increased at 1 d after hypoxic exposure.EAAT1 and TNF-αexpression was significantly up-regulated, while EAAT2 was down-regulated af-ter minocycline treatment.CONCLUSION: Minocycline inhibits the increase in the glutamate level after hypoxia in periventricular region of the neonatal rats.The mechanism may relate to the selective regulation of glutamate transporters, rather than the inhibition of neuroinflammation in periventricular zone.
5.Research on radix notoginseng for improving CKD rat renal fibrosis

Objective To investigate the effects of notoginseng and guiding medicinals mediated notoginseng for improving the renal inter stitial fibrosis in rats with chronic kidney-disease(CKD)by regulating TGF-β signaling pathway.Methods A total of 100 male SD rats were randomly divided into five groups:normal group(NOR,n =20),model group(CKD,n =20),radix notogin-seng group(RN,n =20),radix notoginseng plus platycodi group (RNP) and radix notoginseng plus cinnamon group (RNC,n =20).Except for the NOR group,the CKD rat model in other groups was established by adenine gavage.After modeling,the NOR group and CKD group were given the same volume of normal saline by gavage,while the group RN,RNP and RNC were given corresponding drugs by gavage,for 4 weeks.After 4 weeks,the rats in each group were sacrificed for collecting serum and detecting the renal function(serum Scr,BUN),the renal tissues were taken for conducting HE and Masson staining.Then the renal tissue pathological damage severity was observed.The expressions of FN and LN in kidney tissue were detected by immunohistochemistry and the expressions of TGF-[β,α-SMA were detected by Western blot method.Results Compared with the NOR group,the model group exhibited the renal dysfunction(P<0.01),renal interstitial severe fibrosis manifestation and increased collagen deposition(P<0.05),and the expression of kidney tissues α-SMA(P<0.01),TGF-β(P<0.01),FN and LN were significantly increased.Compared with the model group,the renal function in various treatment groups was improved,Scr(P<0.01)and BUN(P<0.01)were significantly decreased,the renal interstitial fibrosis degree was reduced,collagen desposition was decreased(P<0.05),renal tissue α-SMA(P< 0.05),TGF-β(P<0.05),FN and LN expression were reduced to some extent,in which the effect of RNC group was stronger than that of the RN group and RNP group.Conclusion Notoginseng and guiding medicinals mediated notoginseng can retard the progression of renal interstitial fibrosis caused by adenine in CKD rat in varying degrees,its mechanism maybe reduce the expression of TGF-β protein.
6.Application of failure mode and effects analysis in reducing inpatients' fall injury

Objective To formulate effective safety management method for falls and reduce inpatients' fall occurrence rate and fall injury rate. Methods Setting up a multi-sectoral fall management team and the failure mode and effect analysis (FMEA) was used to form a fall assessment, education, prevention and disposal management system. Taking the inpatients of Changshu Hospital Affiliated to Soochow University from January 2014 to December 2017 as the research object, the incidence and injury rate of falls of inpatients before and after FEMA implementation were compared. Results After implementation, fall occurrence rate reduced from 0.091‰ in 2014 and 2015 to 0.076‰ in 2016 and 2017, however there was no statistical significance (P>0.05). The rate of injury above Grade 2 caused by falls decreased from 42.86% to 21.25%. The difference was statistically significant (P< 0.05). Conclusions Using FEMA quality improvement tools to form a fall management system can effectively reduce the fall injury rate of hospitalized patients.
7.Scutellarin inhibits caspase-11 activation and pyroptosis in macrophages via regulating PKA signaling

Inflammatory caspase-11 senses and is activated by intracellular lipopolysaccharide (LPS) leading to pyroptosis that has critical role in defensing against bacterial infection, whereas its excess activation under pathogenic circumstances may cause various inflammatory diseases. However, there are few known drugs that can control caspase-11 activation. We report here that scutellarin, a flavonoid from Erigeron breviscapus, acted as an inhibitor for caspase-11 activation in macrophages. Scutellarin dose-dependently inhibited intracellular LPS-induced release of caspase-11p26 (indicative of caspase-11 activation) and generation of N-terminal fragment of gasdermin D (GSDMD-NT), leading to reduced pyroptosis. It also suppressed the activation of non-canonical nucleotide-binding oligomerization domain-like receptor family pyrin domain containing 3 (NLRP3) inflammasome as evidenced by reduced apoptosis-associated speck-like protein containing a CARD (ASC) speck formation and decreased interleukin-1 beta (IL-1β) and caspase-1p10 secretion, whereas the NLRP3-specific inhibitor MCC950 only inhibited IL-1β and caspase-1p10 release and ASC speck formation but not pyroptosis. Scutellarin also suppressed LPS-induced caspase-11 activation and pyroptosis in RAW 264.7 cells lacking ASC expression. Moreover, scutellarin treatment increased Ser/Thr phosphorylation of caspase-11 at protein kinase A (PKA)-specific sites, and its inhibitory action on caspase-11 activation was largely abrogated by PKA inhibitor H89 or by adenylyl cyclase inhibitor MDL12330A. Collectively, our data indicate that scutellarin inhibited caspase-11 activation and pyroptosis in macrophages at least partly via regulating the PKA signaling pathway.
