Objective:To establish an experimental model of residual ridge resorption after mandibular incisor extraction and to study the occurrence mechanisms.Methods:Thirty male Wistar rats were used, the edge of the right mandibular incisor was cut every three days(a total of three times) and the incisor was extracted at three days after the final cut. The animals were sacrificed 0,1,2,4,8,12 weeks respectively after the extraction and the mandibles were dissected out. The length of the alveolar bone and the ratio of extracted side length to unextracted side length were measured on soft X-ray photographs and the mandibles were processed for histomorphometry.Eighteen male Wistar rats were used to observe the morphologic alteration of periodontal tissues by hematoxylin and eosin when the incisor crown was cut at different time intervals.Results:As compared with the unextracted side, the length of alveolar ridge of the extracted side was significantly decreased at 4,8,12 weeks;the ratio of extracted side length to unextracted side length significantly decreased at 4 week after tooth extraction(P

Objective: To evaluate the properties of PLA-PEG as a biodegradable scaffold material combined with rhBMP-2 for bone construction.Methods:PLA-PEG/rhBMP-2 multipore compound was prepared,then compression intensity and compression module were tested.The mandibular defect model was made in 20 rabbits,then PLA-PEG/rhBMP-2 was implanted into the defects on one side,and PLA-PEG without rhBMP-2 was implanted into the defect on another side as the control.The bone specimens were retrieved to make density analysis with X-ray photogram following sacrifice of the rabbits 2,4,8 and 16 weeks respectively after operation,then the specimens were decalcified to make histopathological observation by means of HE stain.Results:Compression intensity(MPa) of the PLA-PEG/(rhBMP)-2 compound was 30.05?3.12,and compression module(MPa) was 371.67?12.37.At 2,4,8 and 16 weeks,more new bone was observed,and higher intensity(P
4.Efficacy andβcell function improvement of Linagliptin in combination with Metformin in newly diagnosed T2DM patients

Objective To explore the efficacy and safety of Linagliptin in combination with Metformin in newly diagnosed T2DM patients. Methods A total of 140 newly diagnosed T2DM patients were enrolled in this study. All the patients were admitted in the Endocrinology and Metabolism department of our hospital from January 2013 to October 2013. Random number table method was used in patients’ selection. All the patients were divided into two groups :Linagliptin plus Metformin group (Linagliptin group ,n=70) and Glipizide plus Metformin group (Glipizide group ,n=70) ,and followed up for 12 weeks. Efficacy and safety of treatment were compared between the two groups. Results The difference of FPG ,2 hPG and HbA1 c did not reach the statistical significance between the two groups at baseline treatment (P>0.05). FPG ,2 hPG and HbA1 c were significantly lower after treatment compared with baseline in both groups (P<0.05). FC-P ,2 hC-P ,HOMA-β,HOMA-IR ,IAI and adiponectin level at baseline were similar between the two groups ( P> 0.05 ). All these indicators were improved after treatment in both groups (P< 0.05) ,and were significant better in Linagliptin group than in Glipizide group. Adverse events rate were significantly higher in Glipizide group than in Linagliptin group (18.57%vs 5.71% ,χ2 = 5.423 ,P= 0.020 ). Conclusion The efficacy of Linagliptin in combination with Metformin in newly diagnosed T2DM patients was similar with Glipizide plus Metformin. The β cell function improvement was better and APN level was higher after Linagliptin treatment.
5.Association of TGF? and TGF?3 polymorphisms with nonsyndromic cleft lip and cleft palate

Objective:To study TGF? and TGF?3 polymorphisms in patients with nonsyndromic cleft lip with or without cleft palate(NSCLP,CLP or CL)and cleft palate only(CPO).Methods:TGF? and TGF?3 DNA was extracted and amplified from peripheral leukocytes of 56 cases of NSCLP(40 of CLP and 16 of CL),26 of CPO and 28 of unrelated controls.The primers were designed according to the 3'untranslated region of TGF? and 5th exon of TGF?3 published in the Internet.The PCR products were analyzed by single-stranded conformation polymorphism(SSCP).The aim fragments were further conformed by DNA sequencing after being cloned into pGEM-T vector.Results:The 345 bp fragment of TGF? and 193 bp of TGF?3 were amplified from NSCLP,CPO and control samples.In SSCP analysis,three alleles of TGF? and two of TGF?3 were found.Sequencing results showed three DNA polymorphic sites in TGF? and one in TGF?3 which were all base shifts.There was no difference of each genome between patients and controls.Conclusion:There are no direct association between 3'UTR of TGF? or 5th exon of TGF?3 and NSCLP or CPO in Chinese.
6.Experimental study of cleft palate repair by rhBMP-2/Co/PLGA composite membrane

Objective:To study whether the guided bone regeneration (GBR) technique can be used to achieve good bone formation in the defects of palate. Methods:Bone defect in the shape of triangle with the height of 4 cm and base line of 1.5 cm was created in the middle of palate in each of 16 adult dogs. The dogs were equally divided into experimental group(A) and control group(B). rhBMP-2/Co/ PLGA membrane was implanted in the defects in group A and the Co/ PLGA membrane in group B. The maxillary bone were observed by CT scan at 4, 8, 12 and 24 weeks after operation. The defect repair was observed by histological examination. Results:CT and histological examinations showed that new bone formation in group A was better than that in group B. 24 weeks after operation the defects in group A were completely repaired by new bone, those in group B partly repaired. Conclusion:The rhBMP-2/Co/PLGA composite membrane can be used to repair the defects in palate.
7.The influence of fusing conditions on the surface structure and morphology of SP1 SiO_2 in dental resin composites

Objective: To evaluate the influence of fusing conditions on the surface structure and morphology of the SP1 SiO2. Methods:The SP1 SiO2 samples were sintered under 500, 650, 800, 950 and 1 100 ℃ at a rate of 250 ℃/h and maintained for 10, 30 min, 3 h respectively. The surface structure was characterized by FT-IR. Results:The characteristic peak of Si-OH bond at 958 cm-1 took off gradually with the rising temperature and prolonged time. Under the conditions of 950 ℃ maintained for 10 min and 30 min, and 650 ℃ maintained for 3 h, it disappeared completely. The particles of the SP1 SiO2 became larger while the size distribution became narrow. Conclusion: Fusing conditions can affect the Si-OH content and the size of the SP1 SiO2, as well as the size of distribution.[
8.Stability and time of miniscrew used as orthodontic anchorage in dogs

Objective To evaluate the effects of different loading occasions of orthodontic force on the stability of miniscrew as an anchorage.Methods The titanium miniscrews were respectively implanted into canine and posterior region of both upper and lower jaws of 3 crossbred dogs.The implants were immediately loaded with 200 g orthodontic force or loaded after 2 weeks of implantation.The dogs were sacrificed after 8 weeks.The samples including the microscrew implant and its around bone tissue were extracted,fixed and X-ray photographed.Then the specimens were decalcified,embedded,sectioned and stained with HE and GOMORI.The sections were examined under light microscope.Results The stability of miniscrew in posterior region of jaws was higher than that in canine region in X-ray photograph.There was more bone trabecular formation between miniscrew and jaw bone in non-immediately loaded group than those in immediately loaded group with HE and GOMORI.Conclusion The non-immediately loaded implant as an anchorage can afford more stability than the immediately loaded implant.

10.Correlation between pathogen-associated molecular patterns and periodontitis.

Pathogen-associated molecular patterns (PAMPs) are conservative molecules associated with groups of pathogens or their products. These molecules are recognized by relevant receptors. PAMPs induce the expression of inflammatory cytokines through the signal cascade. The role of PAMPs in the initiation and development of periodontitis is recently attracting attention. PAMPs induce the expression of inflammatory mediators after they are recognized in the periodontium. This process damages the periodontal soft tissue and osseous tissue, thus resulting in periodontitis. The results of this study will provide an excellent resolution for the treatment of periodontitis by blocking the pathogenic pathway of PAMPs.
