Objective To study the antimicrobial resistance of isolated pathogens from surgical infections in Hubei area. Methods The diameters of the inhibition zones of surgical isolates around antibiotic susceptibility test discs in 15 hospitals were computerfiled and analysed by the software of “WHONET-4” according to NCCLS published in 1999.Results A total of 1314 surgical isolates were collected between October 1998 and September 1999.S.aureus,P.aeruginosa,E.coli,Enterobacter sp.and CNS were the main microorganisms.44.6% of staphylococcus was resistant to oxacillin.Imipenem, amikacin, ceftazidime and ciprofloxacin were most active against Gram-negative bacilli with the sensitities (in descending order) of 84.4% to 57.2%.Conclusion Antimicrobial resistance should be seriously considered during surgical therapy and prohylaxis with antimicrobial agents. The tendency to antimicrobial resistance of clinical isolates should be followed up continuously.

Objective To study the correlation between chronic infection of three microorganism and coronary artery lesion by detecting their antibodies and high-sensitivity C-reactive protein(hs-CRP) .Methods According to their severity of coronary artery lesion ,patients enrolled in the study were divided into 4 groups ,including normal control group(n=37) ,single-branch lesion group (n=36) ,double-branch lesion group(n= 30) and multi-branch lesion group(n= 29) .Serum Chlamydia pneumonia IgG(CPN-IgG) ,Helicobacter pylori IgG(HP-IgG) ,human cytomegalo virus IgG (HCMV-IgG) and hs-CRP tests were performed .Results CPN-IgG ,HP-IgG ,HCMV-IgG positive rates and the mixed infection positive rates increased sequentially from normal group to multi-branch group .There were significant differences of CPN-IgG ,HP-IgG ,HCMV-IgG and hs-CRP contents among the 4 groups (P<0 .05) .Logistic analysis showed that CPN-IgG ,HP-IgG and HCMV-IgG were dangerous factors to coronary artery lesion . Conclusion Chronic infections of three microorganism might affect the development of coronary artery lesion .

Chinese Journal of Experimental Ophthalmology 2015;33(3):250-254
Background Application of available technology and objective indexes are very important for the early diagnosis, monitoring and therapeutic evaluation of primary open angle glaucoma (POAG).Many studies have determined retinal damage in structure and function in POAG.However, the study on the association of structural damage and functional abnormality in early POAG is still lack. Objective This study was to evaluate the relationship between structural and functional changes of retina in early stage of POAG.Methods A prospectively pilot study was performed under the approval of Ethic Committee of Shenzhen Eye Hospital from January 2011 to June 2013.Based on Helsinki Declaration, written informed consent was obtained from subject prior to entering the cohort.Ninety-five eyes of 95 POAG patients were included as the study group, and 41 eyes of 41 non-glaucoma subjects were enrolled at the same period as controls. The structural parameters of retinas were measured using RTVue-100 OCT and Cirrus HD-OCT respectively, including macular ganglion cell complex (GCC)-Avg thickness and peripapillary retinal neural fibril layer (RNFL)-Avg thickness;and the functional parameters of retinas were obtained by Humphrey visual filed analyzer and RETI scan 3.15 system respectively, including MD of visual field and PhNR of flash electroretinogram (F-ERG).The associations between the GCC or RNFL thickness and M D or amplitude of PhNR were evaluated by linear and curvilinear regression models. Results The MD,GCC-Avg,RNFL-Avg and PhNR amplitude were (-0.68±1.72)dB,(97.17± 4.82)μm,(102.51±8.74) μm and (49.61±11.01)μV respectively in the control subjects, and those in the POAG patients were (-10.82±9.87) dB,(75.07±12.29) μm,(69.09±12.96) μm and (28.38± 11.52) μV,showing significant differences between them (t =6.549,11.118,-15.061,9.956,all at P=0.001).The curvilinear regression model appeared to better describe the relationship between GCC thickness and MD (R2 =0.595,F=97.089,P<0.001) ;while a linear regression model seemed to be better fit for the relationship between GCC thickness and amplitude of PhNR (R2=0.437,F=103.413,P<0.001).RNFL thickness analysis showed the similar regression models with MD and amplitude of PhNR as GCC thickness, but R2 values were higher between the RNFL thickness and MD (R2 =0.606,F =101.666,P<0.001) or amplitude of PhNR (R2 =0.454,F=54.983,P<0.001).Conclusions Both GCC thickness and RNFL thickness show a curvilinear relationship with MD and a linear relationship with amplitude of PhNR.Goodness-of-fit of RNFL thickness is superior to GCC thickness.

BACKGROUND: Cryopreservation can decrease tissue and organ immunogenicity. The effects of cryopreservation on cell immunogenicity are disputed.OBJECTIVE: To investigate the effects of liquid nitrogen cryopreservation on osteoblast immunogenicity. DESIGN: Randomized, controlled ,paired-sample experiment. SETTING: This study was performed in the Laboratory Center, Qilu Hospital Affiliated to Shandong University between July 2003 and March 2004. MATERIALS: Four New Zealand rabbits of either gender were included for this study. 3H-TdR was provided by Nuclear Medicine Institute of Shandong University. METHODS: Osteoblasts were cultured from the tibial periosteum of New Zealand rabbits and then cryopreservated in the liquid nitrogen for 3 months and defrosted. Cryopreservated and thawn osteoblasts were set as cryopreserved group and freshly cultured osteoblasts were set as non-cyropreserved group. Major histocompatibility complex (MHC)-I positive rate was examined by flow cytometry assay prior to and after cryopreservation. Simultaneously, mixed lymphocyte-osteoblast cultures were established. Lymphocyte stimulation index was calculated after counting the flares using β liquid scintilloscope. MAIN OUTCOME MEASURES: MHI-I antigen positive rate and lymphocyte stimulation index prior to and after cryopreservation of osteoblasts. RESULTS: MHI-I antigen positive rate and lymphocyte stimulation index of osteoblasts was significantly higher in the non-cryopreserved group than in the cryopreserved group (P<0.01). CONCLUSION: The immunogenicity of cryopreserved osteoblasts was significantly decreased. Liquid nitrogen cryopreservation is an ideal method to decrease the immunogenicity of osteoblasts.

Objective:To predict the basic physicochemical properties ,structure and function ,and linear B-cell epitopes of the capsid protein VP1 of coxsackievirus A6(CVA6).Methods: The amino acid sequence of the CVA6 VP1 was analyzed using Bioedit software and various online tools including SubLoc ,TargetP and the others from ExPASy Bioinformatics Resource Portal. Results: The CVA6 VP1 protein was a hydrophilic protein with a relative molecular weight of 33.6 kD and an isoelectric point of 7.92.This protein containsed 24 phosphorylation sites , but no signal peptide , transmembrane domains and possible fatty acylation sites. Its secondary structure was characterized by the richest random coils , and 48.52 percent of its amino acid residues exposed at the solution inter-face.Epitope prediction by Bepipred showed a number of potential B cell epitopes in the protein ,the highest antigenicity index among them located in the region of amino acids residue 155-165.Conclusion:The basic physicochemical properties ,structure and function characteristics ,and potential linear B-cell epitopes of CVA 6 VP1 were successfully predicted , which laid foundations for the further study on the protein and the preparation of vaccines and immunological diagnostic reagents for CVA 6 infection.
