1.Biological characteristics of tumor infiltrating regulatory T cells and the application of regulatory T cells in tumor immunotherapy

Regulatory T cells (Treg cells) have a critical role in the maintenance of immune homeostasis. However, tumor infiltrating Treg (TITR) contributes to weaken anti-tumor specific immune response through enhancing immunosuppressive effect in tumor microenvironment (TME), leading to tumor escape from immune surveillance. Recently, Treg cells-based immunotherapy has achieved good anti-tumor effects with the appearance of immune checkpoint inhibitors, chemokines and their receptors antagonists, specific Treg cells selective target genes knockout and novel drugs. This paper reviews the biological characteristics of TITR and discusses the prospect and application of Treg cells-based immunotherapy.
2.Study on HPLC Fingerprint of Guizhou Zheng'an Standardized Cultivation of Chaenomeles Speciosa

This study was aimed to establish HPLC fingerprint of Chaenomeles speciosa for the place of production and quality control. HPLC was used to comparatively analyze 7 groups of C. speciosa crude drugs from Guizhou Zheng'an and 8 groups of C. speciosa crude drugs from other different production places. The results showed that after the mutual mode of HPLC fingerprint was set up, and the 11 common peaks were pinpointed, comparison of similar degree to crude drugs of other producing areas had been carried on. The main kinds of ingredient in C. speciosa from different places are nearly the same, but the content of each component was different. It was concluded that the method can be used as the identification and quality control of Guizhou Zheng'an C. speciosa, as well as the evaluation of herb quality and standardized planting of GAP, establishment of standard operating procedures of planting and the quality control of C. speciosa.
3.Thinking and suggestion on the teaching of hospital pharmacists standardized training

To cultivate more excellent pharmacists, ensuring patients rational drug use, standardiza-tion training of hospital pharmacists in Beijing has been carried out since 2000, and the wonderful effect has been achieved. The teaching methods mainly include centralized training model, small lectures, practice teaching and self-study. The training teaching content mainly includes prescription audit, drug dispensing and management as well as the pharmaceutical care. In this paper, based on the analysis of the status quo, it is recommended that detailed training formulate, teaching methods and content of the second stage of the training about the training management policy, and the clinicians teaching should be added to the contents of teaching. The thought moral qualities, laws and regula tions, as well as academic and research training should also be added to the teaching content.
4.Retrospective analysis of new clinical characteristics in adult patients of mycoplasma pneumoniae pneumonia

0.05)in sex.Most patients had no basic diseases.5% patients were above 65 yrs old.98.75% patients had 0~2 CURB-65 scores.Consistent fever and cough were the main symptoms in these patients.There were multiple changes in CT images.Except ground glass attenuation,other signs such as consolidation,global or tumor-like shadow,scattered multiple patchy infiltration shadow,pleural effusions and enlarged mediastinal lymph nodes or migrated lesions were commonly seen.Macrolides antibiotics had poor effect.Conclusion New changing tendency of adult mycoplasma pneumoniae pneumonia has occurred,clinical symptoms are severer with multiple changes in radioactivity and the treatment has become more difficult.
5.Directional differentiation of bone marrow mesenchymal stem cells into oligodendrocytes induced by the combination of various neurotrophic factors

BACKGROUND: Myelination following axonal regeneration is a key factor affecting the recovery of spinal cord injury. Oligodendrocyte survival directly affects the myelination following axonal regeneration. OBJECTIVE: To investigate the feasibility of differentiation of rat bone marrow mesenchymal stem cells (BMSCs) into oligodendrocytes induced by neurotrophic factors. DESIGN, TIME AND SETTING: The cell molecular biology in vitro study was performed at the Laboratory of Department of Orthopaedics, Tongji Hospital from September 2006 to June 2007. MATERIALS: A total of 5 Sprague Dawley rats aged 2-4 weeks, of both gender were selected. Bilateral femur and tibia bone marrow was obtained to harvest BMSCs. METHODS: At passage 4, BMSCs were incubated in serum-free medium, supplemented with N2, 20 ng/mL basic fibroblast growth factor, 20 ng/mL epidermal growth factor for 48 hours, and incubated in medium containing 500 ng/mL insulin-like growth factor I and N2 for 3 days. MAIN OUTCOME MEASURES: Morphological changes were observed using an phase contrast microscope. Semiquantitative RT-PCR was utilized to detect specific marker mRNA expression of oligodendrocytes. Using neuron marker anti-microtubule-associated protein, astrocyte marker anti-glial fibrillary acidic protein, oligodendrocyte marker anti-galactocerebroside, anti-myelin basic protein antibody, immunocytochemical staining was performed to detect the positive rate of the differentiation of BMSCs into oligodendrocytes. RESULTS: Morphological changes in BMSCs during the differentiation into oligodendrocytes: After the induction, a majority of BMSCs presented the morphological characteristics of oligodendrocytes. Cytoplasm retraction towards nucleus, cell process extension towards outwards, and strong refraction were found. With the prolongation of time, several cell processes connected and formed a typical net-shape structure. Specific marker mRNA expression of oligodendrocytes: Following induction, specific strap of myelin basic protein mRNA and galactocerebroside mRNA could be detected. Positive rate of oligodendrocytes: During induction, the positive rates of galactocarebroside, myelin basic protein and microtubule-associated protein were 65%, 45% and 10%, respectively. CONCLUSION: The combination of epidermal growth factor, basic fibroblast growth factor and insulin-like growth factor can effectively promote the directional differentiation of BMSCs into oligodendrocytes.
6.Proliferation and differentiation of precartilaginous stem cells in response to stable expression of hTGF-β3

Objective To study the effect of stable expression of reconstructed human transforming growth factor-β3 ( hTGF-β3) on proliferation of precartilaginous stem cells ( PSCs) and their differentiation into cartilage chondrocytes.Methods After isolated and purified by immunological microbeads,PSCs of rats were transfected with pcDNA3.1 ( + )-hTGF-β3.MTT reduction assay ( MTT) and flow cytometry were performed to investigate the effect of transduction on proliferation and DNA synthesis.Biosynthesis of hTGF-β3 and expressions of cartilage associated genes and proteins were examined by qRT-PCR,immunohistology and Western blot.Results hTGF-β3 was expressed in PSCs stably.Compared with non-transfection group,PSCs' DNA synthesis level and proliferation rate were significantly increased after transfection.Quantitative real-time PCR and immunological investigation suggested up-regulated expression of specific genes and proteins of chondrocyte and increase of deposition of chondrocyte typical extracellular matrices proteoglycan and collagen type Ⅱ .Conclusions Gene enhanced PSCs can stably express hTGF-β3 protein to promote proliferation of PSCs and induce differentiation of PSCs in to chondrocytes,which provides a fresh approach to cartilage tissue engineering.
7.Integrative treatment of multiple trauma patients with severe craniocerebral injury in emergency center (with 148 cases report)

Objective To explore the significance and important measure of integrative treatment of multiple trauma patients with severe craniocerebral injury in emergency center. Methods One hundred and forty-eight multiple trauma patients with severe craniocerebral injury in emergency center from November 2002 to February 2006 were analysed retrospectively. Result In total 148 cases, 72 were cured and 26 dead, 7 were in status of plant man, 17 experienced severe deformity, and 26 did mild deformity. Conclusion Multiple trauma patients with severe craniocerebral injury usually experience a serious situation and rapid development. Therefore emergency doctors are required for organizing salvage of such patients, at the same time treatment and diagnosis are implemented. Firstly the most important key to successful salvage is appropriate disposal of fatal injury and early elimination of shock. Proper surgical choice, especially at the first time, inspection and protection of visceral functions, and attention to nutritional support are other vital methods to gain more successful salvage. ICU is also emphasized for its essentiality.
8.Living-related liver transplantation for the treatment of Wilson's disease

Objective To evaluate living-related liver transplantation (LRLT) for the treatment of Wilson′s disease. MethodsBetween Jan. 2001 and Oct. 2003,LRLT was performed in 20 patients (mean age 11.1 years) of late staged Wilson′s disease. Among them emergency transplantation was performed in 3 patients. Donor livers were all from patient′s parents. ResultsSurgery was successful in all donors and recipients, liver function test and serum ceruloplasmin reached normal level one month after transplantation. One patient died of severe rejection. Discharged patients were followed up from 2~33 months (mean 18.9 months). [WT5”HZ]ConclusionsLiving related liver transplantation is an effective treatment for Wilson′s disease complicated with hepatic dysfunction.
9.Electromagnetic fields and the proliferation and matrix synthesis of annulus fibrosus cells

Objective To study the biological effects of sinusoidal electromagnetic fields(EMFs)on proliferation and extracellular matrix(ECM)formation by annulus fibrosus(AF)cells in rats.Methods AF cells isolated from rats were randomly divided into a control group and an experimental group.The cells in the experimental group were stimulated with an EMF,while those in the control group were held under the same culture conditions but with no EMF.Flow cytometry and MTT were performed to observe the effects on the ceU cycle and proliferation.Collagen and aggrecan expression were examined after amplification with a reverse transcriptase polymerase chain reaction(RTPCR).Sulfated glycosaminoglycan(sGAG)content wag detected by applying the Alcian blue method. Results AF cell proliferation was not significant until after 4 days of stimulation.Compared with the control group,the expression of type Ⅰ and Ⅱ collagen and Aggrecan were up-regulated,and sGAG content Was increased in the experimental group.Conclusion AF cell proliferation was enhanced by EMF.Gene expression of collagen type Ⅰ and Ⅱ and Aggrecan increased.a8 well as sGAG levels.The results suggest an approach for treating of intervertebral disc degeneration.
10.Application of cerebral glucose metabolism in Parkinson's disease patients with cognitive impairment

Objective To investigate the change of the cerebral glucose metabolism in Parkinson's disease(PD) with cognitive impairment.Methods Fifty patients with PD and twenty normal controls were enrolled in this study.Based on MMSE score,the PD patients were divided into PD-NC(n=25) and the PDD(n=25) group.Twenty age-and sex-matched physical health examinationer were colleted as control group.Using the SPM to analyze the characteristics of the metabolism in different cognitive level io PD patients.Results (1) Compared with control group,P D-NC group showed significant hypermetabolism in occipital (-24,-92,10),parietal (-48,12,-16) and temporal lobe(52,8,-32).(2)Compared with control group,the glucose metabolism of PDD group decreased in most cerebral cortex and subcortex.(3)Compared with PD-NC group,the metabolism of parietal lobe (-42,-48,40),occipital lobe (-22,-96,-6) and tetnporal lobe (-42,-58,16) reduced in PDD group.Conclusion Cortical brain metabolism decreases in PD-NC group.The degree and scope of glucose metabolism of PDD group will increase with the progress of cognitive dysfunction.These features could help with the diagnosis and differential diagnosis of cognitive dysfunction in Parkinson's disease.
