Observe and study the morphological changes and effects on yoleperoxidase (MPO) of traditional Chinese medical complex Intestine-clearing Suppository (ICS) in preventing and treating ulcerative colitis (UC).Method.. Divide the experimental rats randomly into 6 groups, i.e., blank, salicylazosulfapyridine (SASP), model, high, middle and low dosage of ICS, except the blank group, makeother rats experimental UC model with albumen immunologic method of rabbit colon mucosa, further divide them randomly into 2 experimental groups, i. e. , prevention and treatment, afterwards at 2nd~21st day, give prevention group the medicine, 22nd~35th day, treatment group, then decapitate them and take the colon tissue, and observe it morphologically under optical microscope, measure active MPO, analyse statastic results. Result: Through general morphological observation ,we found there were serious congestion ,edema, mucosa erosion, many large ulcers in the rats of model group; less serious mucosa erosion and quantity of ulcers in SASP and low-dosage ICS groups: small ulcers, not serious mucosa erosion, low degree of congestion and edema in high-and middle-dosage ICS groups; the scores of inflammatory injury of ICS groups were markedly lower than that of model (P<0. 01), meanwhile, the preventive and reating effectiveness of high-and middle-dosage groups was better than that of SASP (P<0.05). Under the optical microscope, there were large area of mucosa coloboma in the rats of model group, the ulcer penetrated to muscular layer and serous coat; in SASP group and low-dosage ICS group, there were erosive mucosa and inflammatory cell infiltration, granulation on the base; in middle-and high-dosage groups, large part of colon mucosa recovered, granulation formed on the base, ulcers were obviously less than that in model group (P <<0.01); there had marked difference compared with that in SASP group (P<0.05). The MPO active values of high-and middle-dosage groups were markedly lower than that of model group (P < 0.01). Conclusion: Traditional Chinese complex ICS had good prevention and treatment to rats UC, itseffectiveness was positively correlated with dosage and better than that of SASP suppository.

DNA microarray generally refers to gene chips. Its basic principle is that a large number of oligonucleotide molecules were fixed on the support, and then hybridized with the labeled samples, and then the chip hybridization signal strength was scanned to determine the number of target in the samples. Gene chips can trace the nucleotide sequence in the samples for testing and analyzing. Its characteristics of high-throughput,rapid and parallel acquisition of biological information are better than that of other traditional gene detection technology. It has been widely used in various fields of medical research. This article reviews the application of DNA microarray technology in the study of orthopedics including biological characteristics, formation and development, injury and repair, degeneration and regeneration of articular cartilage.

[Objective] To observe the curative effects and the influence of activity of Superoxide dismutase(SOD) and contents of Maleic dialdehyde(MDA) of Qingchangyukui Gel in Rats with Experimental Ulcerative Colitis. [Method]Ulcerative Colitis model was established in Wistar rats by injecting trinitro-benzene-sulfonic acid (TNBS)into anus, and then treated with Qingchangyukui Gel and salisy-lazosulfa-pyridine(SASP) respectively. Isolating the colon and measuring the SOD and MDA after treating for two weeks. [Results]Colon mucous membrane in the model group had serious erosion, ulcer, damaging of glandular organ, high inflammation grade; activity of SOD in model group was significantly lower than in normal group and contents of MDA increased. The other groups, especially in Qingchangyukui Gel group, had different levels of inflammation repairing, ulcer healing, increasing of SOD and reducing of MDA. [Conclusion]TNBS can cause Colon mucous membrane inflammation and ulcer, reducing of SOD and increasing of MDA , which are the typical pathological change of Ulcerative Colitis; the Qingchangyukui Gel has the effects of increasing the activity of SOD and reducing contents of MDA, therefore it can effectively treat the UC rat.

BACKGROUND: Based on previous studies, the combination of basic fibroblast growth factor (bFGF) with graft may accelerate the procedure of vascular invasion of anterior cruciate ligament (ACL) graft. The antigenicity of graft could be inhibited by the destruction of major histolocompatibility complex (MHC) through the treatment of allogenous tendon by freeze. The freeze. dried tendon showed advantages including prolonged storage time. availability for transport and possibility of commercial application. There is no experimental and clinical study on the graft substance of bFGF combined tendon in ACL reconstruction in animal model so far. OBJECTIVE: To observe histologically the effect of exogenous application or bFGF combined to freeze-dried tendon on angiogenic enhancement in early ACL reconstruction. DESIGN, TIME AND SETTING: Controlled animal study, which was performed in the Department of Orthopeadics, Xijing Hospital. Fourth Military Medical University of Chinese PLA between June 2006 and June 2007.MATERIALS: Fourteen dogs were used in the experiment. METHODS: Extensor digitorum longus tendon was harvested from the rest 2 dogs and treated by freeze-dry as graft for other experimental dogs. bFGF(100 u g/L)was combined to freeze-dried tendon and then transplanted into one side knee to substitute the original ACL. While only freeze-dried tendon was used in the transplantation at the other side as control. MAIN OUTCOME MEASURES: Twelve of them were randomly divided into 6 groups according to the 6 time points,i,e.,1,2,3,4,5,6 weeks after surgery(2 dogs in each group). The histological observation with HE staining was done under microscope to mainly observe the angiogenesis in the transplanted ACL. RESULTS: Neovascularization occurred at the 2nd to 3rd weeks and reached the peak at the 4th to 5th weeks postoperatively at the experimental sides. By contrast. The neovascularization occurred at the 4th to 5th weeks postoperatively at the control sides. Neovascularization in the combined group was longer and deeper than that in the control group. CONCLUSION: The time of neovascular formation and the depth of vascular penetration into the tendon in the group of bFGF combined to freeze-dried tendon are superior to those in the control group.

ObjectiveTo investigate the molecular response of adult and neonate ovine articular cartilage to acute mechanical injury.MethodsAn established in vitro model was used to compare gene expression difference of ovine articular cartilage explants at different developmental stages 24 hours after mechanical injury and the gene expression was compared between these models and that of the uninjured controls by microarray analysis.Total RNA was isolated from the tissue samples, linearly amplified, and applied to a 15 208 Ovine probes cDNA microarray(Agilent).Validation for selected genes(PPARγ,GRO TNC and LDHA) was obtained by real-time polymerase chain reaction.Comparisons between groups were performed by variance analysis.ResultsThere was significant difference in gene expression in adult and neonatal ovine articular cartilage after mechanical injury.Eighty-six genes were significantly manipulated at least 2-fold following mechanical injury for neonate sheep and 83 genes for adult sheep (P<0.05).Conclusion Our findings indicate that mechanical injury to adult and neonatal ovine articular cartilage results in the activation of a series of signaling responses. We could identify four significant genes that are up or downregulated in response to acute mechanical injury.Significant functional clusters including genes associated with wound healing, articular protection, repair integration, and energy metabolism.Of these,PPARG could be specifically identified as novel target molecules and potential chondroprotective agent involved in traumatic cartilage injury and cartilage integrated repair.
