4.Ultrastructure observation of biological living skin equivalent

Objective:To observe the ultrastructure of biological living skin equivalent in vitro.Methods:Keratinocytes and fibroblasts were sourced from the back skin of legally aborted human foetus.Fibroblasts were seeded into bovine type I collagen gel and cultured for 3 days.Keratinocytes were seeded on the surface of collagen gel and cultured for another 2 days,then the equivalent skin was exposed to air-liquid interface to form a protective cornified layer.Histological structure and ultrastructure of the equivalent skin were observed by light microscope and transmission electron microscope.Results:The morphogy of equivalent skin of full thickness consisted of epithlium and dermis.Epithelium was made up of stratum basale,stratum spinosum,stratum granulosum and stratum corneum.lntercellular bridge existed within the cells of different layers.Some ultrastructures in epithelium such as desmosome,Lamelle,tonofibrils and lipid droplet were found in the equivalent skin.Conclusion:Tissue-engineered skin may have the features of natural skin.
5.Anesthesia for surgery of spinal tumors

[Objective]To summarize the clinical experience on anesthesia techniques for surgery of spinal tumors.[Method]A total of 546 patients undergoing surgery of spinal tumors were retrospectively studied,and the complications of surgery were also collected.[Result]There was no perioperative death.The average degree of blood loss was 1997ml per patient who usually required a large volume of blood transfusion.Acute pumonlary edema were occurred in 3 patients who were suspected to develop larynx edema immediately after operation.A perioperative visual loss following cervical spine surgery was also founded.[Conclusion]It is important to prevent and minimize blood loss by varied techniques during operation.Peventing adverse effect of operative position on patients and protecting their airway are also crucial for spinal surgery.
6.High-performance porous beta-tricalcium phosphate bone tissue engineering scaffolds using 3D printing

BACKGROUND:Although the preparation of bone tissue engineering scaffolds can achieve satisfactory results by solvent casting/particulate leaching, in situ molding method, electrospinning, phase seperation/freeze drying, gas foaming, there are stil some deficiencies in the accuracy, pore uniformity, spatial structure complexity, personalized stents. <br> OBJECTIVE:To prepareβ-tricalcium phosphate bone tissue engineering scaffolds using 3D printing. <br> METHODS:Drug-loadedβ-tricalcium phosphate scaffolds were prepared with 3D printing, and the structure was observed to measure its porosity and mechanical strength. The scaffold was immersed in simulated body fluid for 15 weeks to observe the quality change. The scaffold was co-cultured with rat bone marrow mesenchymal stem cells for 7 days to observe celladhesion and morphological changes. Rat bone marrow mesenchymal stem cells were cultured in extracts of drug-loadedβ-tricalcium phosphate scaffold and low-glucose Dulbecco's modified Eagle’s medium containing 15%fetal bovine serum for 24, 48, and 72 hours, to determine the absorbance values and cytotoxicity grading, respectively. Meanwhile, the cells were subjected to osteogenic culture for 1 week, and <br> the alkaline phosphatase activities in two groups were detected. <br> RESULTS AND CONCLUSION:The prepared scaffold showed irregular micropores, high porosity, uniform pore distribution, high pore connectivity rate, and large compressive strength. The drug-loadedβ-tricalcium phosphate scaffold degraded completely with 15 weeks, and cancellous bone defect repair was completed in the same period. Rat bone marrow mesenchymal stem cells adhered to the surface of drug-loadedβ-tricalcium phosphate scaffold and went deep into the scaffold, showing good growth and proliferation. The activity of alkaline phosphatase was also improved. These findings indicate that the drug-loadedβ-tricalcium phosphate scaffold has good biocompatibility.
7.Program with the introduction of evidence-based medicine

We made our specific teaching program for these students with evidence-based medicine theory,according to their teaching characteristics. They studied the research methods of evidencebased medicine,and learned to search informations,write reviews,discuss clinical cases, design subjects under the guidance of teachers. Their scientific research ability was improved after these practices and training program.
8.The effect of cinobufacin combined with 5-FU on inhibiting proliferation and inducting apoptosis of human gastric carcinoma cells

Objective To study the effects of cinobufacin(cino) combined with fluorouracil(5-FU) on inhibiting proliferation and inducing apoptosis of human gastric carcinoma cells in vitro. Methods The experiment was divided into control group,cinobufacin group,5-FU group and cino+5-FU group. Cell morphological variation,cell inhibitory rate, cell cycle and ratio of apoptotic cell of human gastric carcinoma cell line BGC-823 were studied by cell culture, inverse microscopy, fluoroscopy, MTT assay and flow cytometry on different concentrations of cino and 5-FU. Results Cino could markably inhibit proliferation of human gastric carcinoma cells in time-and dose-dependent response. 9.Evaluation on the effect of marriage quality intervention by the peer education model in patients with breast cancer

Objective The aim of this study was to evaluate the effect of marriage quality intervention in peer education model among women with breast cancer.Methods Marginal quality questionnaire was used to assess in the intervention and control groups during the perioperative period.There had 120 breast cancer patients in the intervention group and 135 normal subjects for control group.The patients were followed by the peer education model and the marriage and quality education during the chemotherapy period.The marriage quality questionnaires were carried out one year after the operation.Results Marital satisfaction(38.73±7.45),couples communication(38.12±7.03)and sex life(36.77±6.96)were significantly higher than those of the patients who participated in the traditional health education group after 1 year of operation.The control subjects with traditional health education had lower satisfaction of marital life(32.59±9.29),husband and wife communication(34.41±7.39)and sexual life(32.59±6.59)in one year after surgery(P<0.001).The scores of marital satisfaction,husband and wife communication,and sex scores were significantly lower in patients with traditional health education after 1 year of operation(P<0.001).Conclusion Marriage quality interventions in the peer education model can effectively affect the marital quality of postoperative patients with breast cancer.
10.Pathogenic mechanisms of influenza virus and Staphylococcus aureus co-infection:research progress

Studies have shown that co-infection of influenza viruses with bacteria is an important cause of high mortality during the epidemic of influenza.There are at least 12 species of bacteria that have been reported to be able to co-infect with influenza.Among those species,co-infection with Staphylococcus aureus is not only the most common but also the most lethal.However,the pathogenesis of high mortality from co-infection with influenza virus/S.aureus remains elusive.In addition,co-infection of influenza virus/S.aureus can induce severe pneumonia.There is new evidence that influenza virus can reduce the host′s tolerance to pathogenic or inflammatory injury,and the two pathogens can also synergistically aggravate toxic effects on the host.Here,we review the mechanisms of severe mortality of influenza infection associated with S.aureus co-infections in order to contribute to prevention and control of influenza in the future.
