AIM: To study the chemical components from the bioactive extract of Chuanminshen violaceum Sheh et Shan in Sichuan province. METHODS: The isolation and purification of this extract were conducted by means of silica gels column chromatography. The structures of the compounds were elucidated based on their physical, chemical features and spectrum data. RESULTS: 8 compounds were isolated from the extract. They were elucidated as hexadecanoic Acid(A),2,6-dimethoxybenzoic acid(B),succinic acid (C),combination of sigmasterol and sitosterol(1(∶)1)(D),chrysophanol(E),8-hydroxy-5-methoxypsoralen(F),4-hydroxy-3-methoxybenzoic acid(G) and stigmasteryl 3-O-?-D-glucopyranoside(H). CONCLUSION: Compounds B,C,E,G were isolated from Chuanminshen violaceum for the first time.

Objective To construct a tyrosine kinase B(TrkB) targeted RNA interference (RNAi) lentiviral vector. Methods Four oligonucleotides targeting rat TrkB gene were synthesized and cloned into lentiviral vector pXZRNAi 1.0 to construct recombinant lentiviral vectors pXZRNAi-shTrkB-1,2,3,4.Neural stem cells prepared from rat hippocampus were infected with these high-titer viruses.Real-time PCR was employed to detect the TrkB mRNA expression and western blot was used to assess the gene silencing efficacy of these recombinants. Results Enzyme digestion and DNA sequencing results demonstrated that these shRNAs were correctly inserted into lentiviral vectors and the four recombinants were constructed successfully with the titer of 8.6 × 105cfu/ml.The infection efficiency of the letivirus on neural stem cells reached 80%.Compared with the uninfection group, the expression levels of TrkB mRNA in neural stem cells decreased significantly after transfected with pXZRNAi-shTrkB-3 and 4((66.7 ± 5.5) % and(76.8 ± 4.9) % respectively,P < 0.05) ; and the protein expression levels were also significantly decreased ((68.5 ± 4.3)% and (78.2 ± 5.1)% respectively,P < 0.05).Conclusion The lentiviral vectors for TrkB have been successfully constructed with high yield of lentivirus, which provides versatile method for assessing gene function in neural stem cells.

From November 2014 to July 2017,637 patients with acute coronary syndrome (ACS) were included in the analysis, among whom there were 48 cases with prior ischemic stroke (7.5%).The risk factors, history, severity of coronary artery disease, medication status, and incidence of adverse cardiovascular and cerebrovascular events (cardiac death,re-infarction,heart failure, stroke) were analyzed.Compare with patients without prior ischemic stroke (control group) patients with prior ischemic stroke (study group) had lower rates in administration of beta blockers [50.00%(24/28) vs.69.78%(411/589),x2=8.02,P<0.05]and interventional therapy[56.67%(17/30) vs.81.86%(334/408),x2=11.15,P<0.05].However,there were no significant differences in medication of dual antiplatelet, statins and angiotensin converting enzyme inhibitors or angiotensin receptor blocker between two groups (P>0.05);and there was no significant difference in major adverse cardiovascular events between two groups (P>0.05).In the future, more studies are needed for clinical management of this group of patients.
