Objective To explore the treatment effect and mechanism of A-NK cells on the subcutaneous trans?planted tumor of oral tongue squamous cell carcinoma in nude mice through observing the external growth and hyperplasia of A-NK cells and to provide theoretical evidence for squamous cell carcinoma treatment. Methods A-NK cells and NA-NK cells were both derived from healthy human peripheral blood and cultured in vitro. Cell growth was observed under micro?scope. The squamous cell carcinoma model in nude mice was established through subcutaneous implanting of Tca8113 cells. Then they were randomly assigned into three groups who were injected with either saline solution, or A-NK cells or NA-NK cells paraneoplastically. All animals were sacrificed after 33 days when tumor were isolated then weight and change in tumor size were assessed. Finally curve of tumor growth was drawn. Results Under the microscope, the proliferation of A-NK cells peak in 15 days and NA-NK cells peak in 12 days. After 3 weeks, the number of A-NK cells increased by 39.33 times while the number of NA-NK cells increased by 16.33 times. The Volume of tongue squamous cell carcinoma in saline solu?tion group was larger than that in A-NK cells and NA-NK cells groups, and volume in the NA-NK cells group was larger than that in A-NK cells group. The volume of tongue neoplasms in different groups, time, and interaction effects are statisti?cally significant (P<0.01). The tongue neoplasms weight in the saline solution group was greater than that in the A-NK cells and NA-NK cells group, and the weight in NA-NK cells group was greater than that in A-NK cells group, and the difference are statistically significant (P<0.05). Conclusion A-NK cells and NA-NK cells can significantly inhibit the subcutane?ous transplanted tumors in nude mice and the anti tumor effect of A-NK group is stronger than NA-NK.

Objective To establish a multi-drug resistant model of temporal lobe epilepsy and observe the effect of hippocampal stimulation on pharmacoresistant epileptic rats and its possible mechanism with the use of indexes including after discharges (AD) of the amygdalae, the stimulus-induced seizures and the extracellular levels of γ-aminobutyrate (GABA).Methods Totally,120 Wistar rats were used for the amygdaloid kindled model of epilepsy by chronic stimulation of amygaloid basal lateral nucleus.Based on the successful kindled model of epilepsy,we selected the pharmacoresistant and pharmacosensitive epileptic rats according to their response to phenobabital and phenytoin. We then divided the pharmacoresistant epileptic rats into the hippocampal stimulation group and control group, with 8 rats in each group.Low-frequency stimulus was conducted for two weeks in the hippocampal stimulation group. The hippocampus extracellular fluid collected by microdialysis was then used to determine the levels of GABA by a high performance liquid chromatography method after hippocampal stimulation. Results The stimulus-induced seizures were inhibited significantly, and the frequency of the AD was decreased,as well as the amplitude, compared with the control group. The extracellular level of GABA in the 8:00-9:00 am and the 8:00-9:00 pm was (32.69 ± 7.80) and (35.76 ± 6.27) μg/ml,respectively,both significantly increased as compared with the control ((26.58 ± 6.87) μg/ml,t =-21.45,P =0.000 ; (31.50 ± 4.87) μg/ml,t =-15.74,P =0.000).Conclusions The low-frequency hippocampal stimulation inhibits the seizures of the kindled model of epilepsy and decreases the frequency, the amplitude,as well as the duration of the amygdale AD in the pharmacoresistant epileptic rats, which may be contributed to the increased GABA content in extracellular fluid of the brain after stimulation.

In the present study, a new compound named 17-(6-cinnamamido-hexylamino-)-17-demethoxygeldanamycin (CDG) was obtained by introducing the cinnamic acid (CA) group into the 17-site of geldanamycin (GDM). The anti-cancer effects of CDG in vitro and in vivo were evaluated. MTT assay was used to examine the inhibitory effect of CDG on the proliferation of MCF-7, HepG2, H460 and SW1990 cells. Immunofluorescent staining flow cytometry combined with Annexin V-FITC/PI staining were used to detect apoptotic cells. Transwell assay was used to analyze the effect of CDG on cell invasion and migration ability. Western blotting was used to detect the expression levels of RAF-1, EGFR, AKT, CDK4 and HER-2 of MCF-7, HepG2 and H460 cells. The toxicities of CDG and GDM were evaluated in mice. Using the subcutaneously transplanted MCF-7 xenograft in nude mice, inhibitory effect was evaluated in vivo. The results showed that CDG inhibited the proliferation of cancer cells (IC50: 13.6-67.4 microg.mL-1). After exposure to CDG for 48 h, most cells presented typical morphologic changes of apoptosis such as chromatin condensation or shrunken nucleus. The rates of apoptosis of MCF-7, HepG2, H460 and SW1990 cells incubated with 10 microg.mL-1 CDG were 23.16%, 27.55%, 22.21%, 20.47%, respectively. A dose-dependent reduction of migration of four cell lines was found after exposure to CDG. The decreased levels of RAF-1, EGFR, AKT, CDK4 and HER-2 showed that CDG possessed HSP90 inhibitory effect. The result of animal toxicity test on the mice suggested that CDG had lower toxicity than GDM. Meanwhile, CDG inhibited the growth of MCF-7 xenografts of athymic mice.
