Objective To explore effects of shuanghuanglian combined with levofloxacin on antibiotic resistance of Staphylococcus Aureus ATCC29213 to levofloxacin. Methods Tissue cage infection model with Staphylococcus aureus was established in rabbits, and the infected animals were given with levofloxacin alone ( group A ) or in combination with shuanghuanglian ( group B) for 5 days respectively. Steady-state concentration of levofloxacin in tissue cage, bacteria recovery and bacterial resistance in tissue cage infection model were studied. Results Steady-state concentration of levofloxacin in tissue cage was not significantly different between group A and group B. The recovery rate of bacteria was significantly lower in group B than in group A (20. 0% vs. 100. 0%). The minimum inhibitory concentration (MIC) was lower in group B than in group A. Conclusion Shuanghuanglian combined with levofloxacin is helpful to reduce antibiotic resistance of Staphylococcus aureus to levofloxacin, indicating that some Chinese traditional medicine combined with antibiotics can reduce antibiotic resistance.

Objective To investigate relationship between levels of follicle-sitimulating horomone receptor(FSHR) and ovarian response induced by gonadotropin hormone, and whether the FSHR expression is correlated with in vitro fertilization(IVF) outcome. Methods Granulose cells were collected from 43 women receiving IVF-embryo transplantation(IVF-ET).According to the number of oocyte, the women were divided into three groups: low response(15).The expression intensity of FSHR was measured by immunohistochemistry technique. The expression intensity of FSHR on the granular cell, the embryological and clinical outcomes were compared and analyzed. Results The expression of FSHR was significantly different in three groups with the highest in high response group(P0.05).The FSHR level was positively correlated either with the number of oocyte (r=0.719) or with the serum E2 levels(r=0.516,P0.05). Conclusion Ovarian response to gonadotropin hormone stimulation is correlated with the level of FSHR in the granulose cells. The development of follicles may be influenced by it.

Objective To develop a rapid and efficient single cell PCR method for the non-invasive prenatal hemophilia A genetic diagnosis. Methods Six STR markers (DXS15,DXS9901,G6PD, DXS1073,DXS1108 and F8Civsl3) closely linked to F Ⅷ gene were examined in 118 healthy individuals and 12 hemophilia A families by muhi-plex fluorescent PCR.Three markers with the higher heterozygote rate and diagnostic rate were chosen and combined with amelogenin gene to establish the four-plex nested fluorescent PCR on single lymphocyte level. Results The single lymphocyte amplification rates of Amelogenin,DXS15,FSCivsl3 and DXSI073 were 94.3%,91.4%,100% and 100% in the normal male, and 100% ,97.1% ,97.1% and 97.1% in the normal female's respectively. The female was heterozygous in DXS1073 and DXS15.However,no allele dropout was found in both markers.Neither the false negative nor the false positive amplification was observed. Conclusion The single cell four-plex nested fluorescent PCR method established in this study is convenient and efficient, and will be hopefully employed in the clinical non-invasive prenatal genetic diagnosis for hemophilia A.

OBJECTIVETo study the anti-asthmatic effect of sodium ferulate (SF) and its mechanism in guinea pig asthmatic model.
METHODSGuinea pigs were sensitized with ovalbumin as animal asthmatic model and treated with 3 different concentration of SF for 8 days. Levels of endothelin (ET) and nitric oxide (NO) in blood and lung tissue, and eosinophil (EOS) in blood and bronchoalveolar lavage fluid (BLAF) was counted at the end of trial.
RESULTSSF could significantly lower the ET content and increase the NO concentration in serum and lung tissue, reduce the number of EOS in blood and BLAF (P < 0.05, P < 0.01). Stronger effect was showed in the high dose group.
CONCLUSIONMechanism of anti-asthmatic action of SF might be to increase NO concentration, lower ET content, alleviate EOS infiltration.

Objective To investigate the relationship between urinary iodine level and thyroid disease. Methods The study used a case-control design. One hundred and nine patients with thyroid disease from the Affiliated Hospital of Shanxi Institute for Endemic Disease Control were selected as case group from 2011 to 2012,and these patients were divided into three groups:Graves's disease (GD) group (n =48),chronic lymphocytic thyroiditis (HT) group(n =34) and thyroid nodules group(n =27).Sixty-two healthy people from the same region were selected as a control group.Urinary iodine was determined using arsenic cerium catalytic spectrophotometry, thyroid autoantibody (TRAb) and thyroid peroxidase antibody (TPOAb) was detected using electrochemiluminescence, while iodine absorption rate was measured using thyroid function analyzer, and thyroid volume was measured using type-B ultrasonic method. The relationship between urinary iodine level and patients with thyroid disease was compared with that of control group. Results Urinary iodine levels of patients with GD,HT,thyroid nodules and control groups were 313.95,375.20,220.20 and 196.50 μg/L,respectively.Urinary iodine levels of patients with GD and HT groups were higher than that of control group(Z =3.238,4.275,all P < 0.0125).Urinary iodine level of patients with HT was higher than that of thyroid nodules(Z =3.762,P < 0.0125).Iodine uptakes of GD,HT,thyroid nodules and control groups were (84.20 ± 16.90)%,(23.51 ± 6.72)%,(28.34 ± 8.02)% and (29.31 ± 8.41)%; TRAbs of patients with GD,HT,thyroid nodules and control groups were (58.57 ± 20.31)%,(2.54± 1.00)%,(2.98 ± 0.83)% and (3.01 ± 1.21)%; TPOAbs of patients with GD,HT,thyroid nodules and control groups were (117.03 ± 57.21)%,(251.00 ± 98.20)%,(16.81 ± 9.87)% and (15.00 ± 7.23)%.Iodine uptake,TRAb and TPOAb of GD group were higher than those of control group(P < 0.05).TPOAb of HT group was higher than that of control group(P < 0.05).Urinary iodine levels of GD group and HT group were positively correlated with TPOAb(correlation coefficient were 0.462,0.478 all P < 0.05).Conclusions Excessive iodine intake is found in patients with GD and HT.Determination of urinary iodine is helpful for individualized iodine supplementation.

A HPLC method has been developed in the current investigation for simultaneous determination of three chemical markers of by akangelicin, imperatorin and isoimperatorin in Radix Angelicae Dahuricae. Separation was performed at 30 degrees C. on achromatographic column of Platisil ODS C18 (4.6 mm x 250 mm, 5 μm). The mobile phase was acetonitrile-water. The flow rate was 1.0 mL x min(-1) and the detection wavelength was 254 nm. The results showed that the three chemical markers could be well resolved and that in the selected linear range, all calibration curves of the three chemical markers showed good linearity (R2 ≥ 0.999 8). The recoveries of byakangelicin, imperatorin and isoimperatorin were 100.83%, 100.10% and 103.52%, respectively, and RSD were 1.7%, 0.77% and 0.41% (n = 6), respectively. The data suggested that the developed HPLC method had good reproducibility, robustness, and accuracy, which was suitable for the quality control of Radix Angelicae Dahuricae. Applications of this method showed that the three chemical markers had higher contents in the Bozhou Anhui and Changge Henan than others.
