Vibrio parahaemolyticus has been considered as one of the most important foodborne bacterial pathogens. The loop-mediated isothermal amplification(LAMP) that amplifies DNA with high specificity and rapidity under an isothermal condition was applied for rapid detection of this pathogen for the first time.A set of four primers, two outer and two inner primers, was designed specifically to recognize the thermolabile hemolysin gene(tlh) of V.parahaemolyticus.The LAMP reaction mix was optimized. The most optimal reaction temperature and time of the LAMP assay for the tlh gene were 60℃ and 60min,respectively.Genomic DNAs from 28 bacterial strains including 14 V.parahaemolyticus strains were amplified using LAMP,and no amplicon was observed in other bacterial strains. The detection limit of this LAMP assay was around 90 fg of V.parahaemolyticus genomic DNA and 24 colonies forming units for pure cultures. In addition, this method was applied to detect artificially contaminated food samples, and the detection limit was 89 cfu/g for non-cultured artificially contaminated food samples. These results suggested that detection of V.parahaemolyticus by LAMP is an effective and low-cost procedure with high specificity and sensitivity that requires no specialized equipment. This assay is expected to become a valuable tool for rapid detection and identification of V.parahaemolyticus.

Objective To analysis the health literacy status, trends and distribution characteristics of Ningbo residents during 2015-2016, and to provide basis for developing health policies and intervention strategies. Methods During 2015-2016, 9810 residents of 15-69 years old in Ningbo were investigated by face-to-face interviews using the unified national questionnaire and multistage PPS sampling method. Results Totally 9750 residents correctly filled out the questionnaire. The adjusted health literacy (AHL) of residents in 2016 was 21.73%, increased by 6.29 percentage points than 2015 (15.44%) .The monitoring results of two years both showed that there was no significant differences of AHL between men and women (P >0.05) . However, the AHL of residents with different age-groups, occupations and education levels all assumed significant differences (P<0.05) . The AHL was significantly higher in urban residents than rural residents (P<0.05) .Among the 6 health issues, the AHL of safety and first aid was the highest (2015: 56.98%, 2016: 61.32%), and the chronic disease prevention (2015:11.23%, 2016: 12.10%) and basic medical care (2015: 11.97%, 2016: 13.81%) were the lowest, also who had the smallest lifting range. Logistic regression analysis displayed that residents with higher level of education had higher AHL (OR=1.72, 95%CI: 1.61-1.83) . Comparison with civil servants, institutions, teachers and medical professionals, farmers (OR=0.74, 95%CI: 0.58-0.94), workers (OR=0.70, 95%CI: 0.57-0.86) and other occupational groups (OR=0.78, 95%CI: 0.64-0.96) had lower AHL. Conclusion The health literacy of Ningbo residents is on the rise, Knowledge of chronic disease prevention and basic medical care will be the main contents of health literacy enhancement program in the future, especially for people with less education.

Objective To study the alveolar macrophages (AM) activation in neutropenic ARDS in children with hematologic malignancy. Methods Thirteen patients of ARDS were prospectively enrolled into the study and divided into two groups, neutropenic (5 cases) and nonneutropenic (8 cases). Bronchoalveolar lavage fluid (BALF) samples were analyzed for differential and total cell counts, and alveolar activation marker expression (HLA-DR) was determined. Results In neutropenic patients, the BALF total cell count and the macrophage absolute count were significantly lower compared to those in nonneutropenic group ((62. 6 ±9. 6)/μl vs ( 124. 0 ± 6. 7)/μl,P < 0. 01 and (40. 8 ±4. 3)/μl vs (67. 6 ± 10. 7)/μl,P < 0. 01,respectively). As compared with the nonneutropenic group, the percentage of AM in neutropenic patients was significantly higher (65.9% ±9. 0% vs 54. 6% ± 8. 7% ,P <0. 05) ,the mean percentage of AM HLA-DR expression was significantly lower (35. 3% ± 5.8% vs 62. 2% ± 5. 8%, P < 0. 01 ). Conclusion These results suggest an alveolar hypocellularity and a deactivation of AM in neutropenic ARDS in children with hematologic malignancy.

Objective To investigate the levels of Th1 and Th2 cell in the peripheral and the cytokines (IFN-γ,TNF-α,IL-12,IL-10,IL-4) in the culture medium at different time in the porcine model with multiple organ dysfunction syndrome(MODS) after continuous blood purification(CBP). Methods Twenty-four young porcines were given intravenous infusion of endotoxin to induce MODS, then were randomly divided into 2 groups:CBP group(n = 12) and MODS group without CBP(n = 12). Continuous venovenous hemodiafiltration (CVVHDF) was the main mode for CBP. At baseline, onset of MODS, and 2 h,4 h,6 h after treatment of CVVHDF,we picked the blood respectively to separate the T helper cell using the beads, detected the Th1 and Th2 cell by FACS and detected the cytokines (IFN-γ,TNF-α,IL-12,IL-10,IL-4) in the culture medium by ELISA. Results The MODS group showed a obvious rise in TNF-α,the drop tendency in IL-12,IFN-γ,IL-10,Th1/Th2 ratio, and no significant change in IL-4. After the treatment, the CBP group showed the drop in TNF-α and IL-4;and increase in IL-12,IFN-γ,IL-10 and Th1/Th2 ratio. Conclusion CBP helps to dear immune function, and restore the balance of Th1/Th2 in porcine MODS model.
