Objectives To observe the pre and post-operational changes of the expressions of survivin, caspase-3 and CD44V6 in patients with colorectal cancer after intra-abdominal implantation of sustained releasing fluorouracil. Meth-ods Sixty-four patients with colorectal cancer (Dukes’stage of B and C) were divided into treatment group and control group, 32 patients in each group. The standard radical surgery was performed in two groups of patients. The fluorouracil im-plants were implanted intra-abdominally in treatment group. The peripheral blood levels of surviving and caspase-3 were de-tected by RT-PCR. The level of CD44V6 was detected by flow cytometry in two groups of patients. Results There were no significant differences in levels of survivin, caspase-3 and CD44V6 before surgery between two groups (P>0.05). The level of survivin (0.362 ± 0.183) was significantly lower at 14 days after operation in treatment group than that of control group (0.585±0.207), but the level of caspase-3 (2.001±0.146) was significantly higher than that of control group (1.654±0.111). The levels of CD44V6 were significantly lower in treatment group (1.857±0.535) and control group (3.471±0.496) after opera-tion than those before operation (9.557±1.170 and 9.729±0.943, P<0.05), and the level of CD44V6 was significantly lower in treatment group than that of control group (P<0.05). Conclusion The implant for the sustained release of fluorouracil showed a positive impact on micrometastases and prognosis of colorectal cancer, while improved the long-term efficacy of postoperative colorectal cancer.

ObjectiveTo evaluate the influence on left ventricular(LV) twist, and the relationship between LV twist and function in the rabbits with ventricular aneurysm formed after myocardial infarction by two-dimensional ultrasound speckle tracking imaging.MethodsTwenty-six healthy rabbits were divided into two groups, six as control group, twenty as experimental group. The model of myocardial infarction and ventricular aneurysm were established by ligaturing left anterior descending coronary artery and left circumflex artery. After four weeks, according to the pathological examination of whether ventricular aneurysm formed or not,the experimental group was divided into myocardial infarct group and ventricular aneurysm group.Echocardiography was performed to measure rotations (at the levels of mitral annulus and apex in LV short-axis view),LV twist, left ventricular cavity size and function.ResultsCompared with the myocardial infarct group,LV end-diastolic diameter(LVEDD) and LV end-diastolic volume(LVEDV) were larger in the ventricular aneurysm group, but LV ejection fraction (LVEF),LV fractional shortening (LVFS),apical rotation and LV twist were lower in the ventricular aneurysm group than the myocardial infarct group. Both apical rotation and LV twist had shown a great correlation with LVEF.The conclusion was also shown in the ROC curve that sensitivity and specificity of diagnosis cardiac dysfunction were 76.9% and 80.0% respectively when the apical rotation was 1.21°.Conclusions There was a closely relationship between the change of apical rotation and the LV systolic function. When the apical rotation dropped to 1.21°,an apparent decline of LV systolic function was implied.

Objective To evaluate whether three chronic kidney disease epidemiology collaboration (CKD-EPI) equations (CKD-EPI2009Scr,CKD-EPI2012SCysC and CKD-EPI2012Scr-SCysC) are applicable in the prediction of glomerular filtration rate (GFR) in Chinese patients with diabetic nephropathy (DN).Methods One hundred and eight patients with DN who were hospitalized in the First Affiliated Hospital of Guangzhou Medical University with GFR being measured by dynamic renal imaging with 99mTc-DTPA from June 2012 to April 2014 were enrolled in this study.GFR measured by dynamic renal imaging with 99mTe-DTPA was used as the reference value (rGFR).GFR was estimated by the CKD-EPI2009Scr equation, the CKD-EPI2012SCySC equation, and the CKD-EPI2012Scr-SCysC equation (labeled as eGFR1,eGFR2,eGFR3).The correlation,30% accuracy, staging consistency, deviation and diagnostic accuracy were compared among the three CKD-EPI equations. Results The rGFR in 108 DN patients was (61.78±26.51) ml· min-1· (1.73 m2)-1.The correlation between three eGFRs and rGFR was significant (all P < 0.01),the correlation coefficients were 0.738,0.708,0.782.The 30% accuracy were 74.07%,52.78%,67.59%,The 30% accuracy of eGFR1 and eGFR3 were higher than eGFR2 (all P < 0.05),but there was no significant difference between eGFR3 and eGFR1 (x2=0.874,P=0.436).The staging consistency was not ideal,Kappa values were 0.391,0.180 and 0.422.For the deviations between three eGFRs and rGFR,there was no significant difference between eGFR3 and rGFR (P > 0.05),eGFR1 underestimated rGFR,eGFR2 overestimated rGFR (all P < 0.01).The results of the Bland-Altman chart showed that consistencies between three eGFRs and rGFR were poor, the degree of deviation of eGFR3 was the smallest. The area under the ROC curve (AUC) of three eGFRs was 0.878,0.883 and 0.915.The AUC,sensitivity, specificity, overall compliance rate and Youden index of eGFR3 were the highest. Conclusions The eGFRs predicted by the three CKD-EPI equations showed good relevance, accuracy and diagnostic accuracy with the rGFR,but poor in consistencies.Comparatively,CKD-EPI2012Scr-SCysC may be better than others, but its consistency limits exceeds the acceptable limits. Therefore, the applicability of using the three CKD-EPI equations to predict the GFR in Chinese DN patients requires a larger sample and multiple verifications as well as further improvement.

Objective To investigate the effect of somatostatin on the electrophysiological changes of early diabetic rats, and to clarify its therapeutic effects on retinopathy in early diabetic rats. Methods 20 Wistar male rats, which were induced into diabetic rat models by intraperitoneal injection of streptozotocin (STZ),were randomly divided into somatostatin therapy group and model control group, 10 in each group. The rats in somatostatin therapy group were injected with somatostatin 10μg·kg-1 ·d-1,for 8 weeks;the rats in model control group received the same volume of saline.Another 10 male Wistar rats were used as normal control group. The changes of blood glucose and electroretinogram were measured, and the indicators were used for statistical analysis. Results Compared with model control group, the blood glucose was declined in somatostatin therapy group at the 8th week (P<0.05).Compared with normal control group, the latent periods of a-wave,b-wave of rod-response and max-response were significantly extended in somatostatin therapy group and model control group at the 1 st, 2nd,4th,6th,and 8th week (P<0.05).The latent periods of a-wave,b-wave of rod-response and max-response in somatostatin therapy group were shorter than those in model control group at the 8th week (P<0.05 ). Compared with model control group, the amplitude had no changes at each observation time point in somatostatin therapy group (P>0.05).Conclusion Somatostatin has a therapeutic effect on early diabetic retinopathy in rats.
