2.Effect of all-trans retinoic acid on expression ofα-SMA in rat model with pulmonary hypertension

Objectives To explore the possible mechanism of all-trans-retinoic acid (ATRA) therapy for pulmonary ar-terial hypertension by observing the effect of ATRA on the expression ofα-smooth muscle actin (α-SMA) in monocrotaline (MCT)-induced rats with pulmonary hypertension. Methods Thirty SD rats were randomly divided into control group, MCT-induced pulmonary hypertension group (model group) and ATRA treatment group (therapeutic group). MCT (60mg/kg) was injected to the rats in the model group and the therapeutic group to induce pulmonary arterial hypertension. And at the same day ATRA [30mg/(kg?d)] was given to rats in the therapeutic group and continually given for 28 days. The mean pulmonary arte-rial pressure and right ventricular hypertrophy index (RVHI), percentage of wall thickness (WT%) and percentage of wall area (WA%) of pulmonary arterioles were evaluated at day 28. The expression ofα-SMA mRNA and protein in the lung tissue were detected by Real-time PCR and Western blot. Results Mean pulmonary arterial pressure, RVHI, WT%and WA%were signiif-cantly higher in the model group than those in the therapeutic group (P<0.01) but these data in the therapeutic group were higher than that in the control group (P<0.01). The expression ofα-SMA mRNA and protein were higher in the model group than those in the control group and the therapeutic group (P<0.01), while there was no difference was found between the therapeutic group and the control group (P>0.05). Conclusions ATRA has therapeutic effect on pulmonary hypertension through down-regulation ofα-SMA expression.
3.Immune hyporesponsiveness induced by immature dendritic cells prolonged rat kidney allograft survival

Objective To study the effect of immature dendritic cells (imDC) transfusion in combination with bone marrow transplantation (BMT) on rat kidney allograft survival and its possible mechanism. Methods Renal allograft from DA rat was transplanted to Lewis rat. Forty recipient rats were randomized into 5 groups: (1) Negative control group; (2) imDC group: Lewis rats accepted imDC transfusion in the amount of 2? 107 only; (3) BMT group: Lewis rats accepted bone marrow transplantation in the amount of 2?108 only; (4) imDC + BMT group: Lewis rats accepted both imDC and BMC; (5)Third party donor group: Lewis rats accepted renal allograft from Wistar rats. One-way MLR was performed to assay splenic cell proliferation to allogeneic T cells. Exogenous IL-2 was added at the outset of another group as the former one-way MLR. Normal Lewis rat accepted splenic cells from tolerant rat in the amount of 1?108. DTH was assayed in the trans-tolerance model. Cells from spleen and thymus were harvested from recipient rats for detecting chimerism by flow cytometry. Results The median survival time (MST) of the renal allografts was (7. 12 ? 1. 25) days in negative control group, (24. 36 ? 3. 20)days in imDCs group and (7. 87 ? 2. 10)days in BMT group, respectively. Objective To establish a cell level-based negative enrichmen technique to detect circulating tumor cells (CTCs) in the peripheral blood of patients with epithelial ovarian cancer.Methods The colon cancer SKOV-3 cells were mixed with 2 ml whole blood from healthy donors at different ratio.Quantification of CTCs was performed using immunomagnetic bead based negative enrichment combined with immunofluorescence antibody method.The method was evaluated the recovery rate of target cells,Samples of 32 patients with ovarian cancer and 10 controls were assayed for CTCs detection by above method.Results ①The recovery rate was ranging from 64% ~80% by spiking varying numbers of SKOV-3 into 2 ml blood samples of healthy volunteers.Regression analysis of number of recovered SKOV-3 cells yielded a regression equation of Y=0.782X-1.408 and a coefficient of determination of R2 =0.998.②Did not detect CK8/18-+ circulating tumor cells in 10 controls,and CK8/18+ circulating tumor cells in 18 cases of 32 Patients with ovarian cancer.The positive rate of CK 8/18 + circulating tumor cells was significantly differences between the two groups (x2 =7.681,P<0.01).③The presence ofCTCs was significantly correlated with distant metastasis (x2 =5.776,P<0.05).Conclusion The method of immunomagnetic bead based negative enrichment combined with immunofluorescence antibody technique for CTCs detection in peripheral blood of patients with ovarian cancer has a clinical value of application and extension.

9.A controlled study of cortisol, thyroid hormone and autonomic function in anxiety disorder

ObjectiveTo investigate the features of the cortisol ( COR),thyroid hormone and autonomic function of anxiety disorder,and to observe the relationships between clinical subtypes,insomnia,gender factors and the neuroendocrine and autonomic function.Methods60 anxiety disorder patients and 60 healthy controls were randomly selected from a three-grade class A hospital over the same period.Blood at 8:00am to test the plasma COR,T3,T4,TSH level and examined the heart rate variability (HRV) on the same day,the anxiety disorder patients also finish the self-rating anxiety scale (SAS).ResultsThe plasma COR,TSH level and LF/HF index were significantly higher( (531.58 ± 218.59 ) nmol/L,P< 0.0l ; (3.50 ± 1.35 ) MIU/L,P< 0.05 ;3.87 ± 3.37,P<0.0l ),while the SDNN,LF,HF index were significantly lower in anxiety disorder than that in the control group (27.34±13.13,P<0.01;338.72 ±396.34,P<0.01;135.77 ±149.95,P<0.01).The plasma COR,TSH level and LF/HF index were significantly higher(P < 0.05,P<0.01,P<0.01 ),while the SDNN and HF index were significantly lower in panic disorder than that in the generalized anxiety disorder (P < 0.01 ).The plasma COR,TSH level and LF/HF index were significantly higher,while the SDNN,LF,HF index were significantly lower in panic disorder than that in the control group (P<0.01 ).The plasma COR was significantly higher,while the SDNN and HF index were significantly lower in generalized anxiety disorder than that of the control group (P<0.01 ).The plasma COR level and LF/HF index were significantly higher (P<0.01,P<0.05),while the SDNN,LF,HF index were significantly lower in anxiety disorder with or without insomnia than that in the control group (P< 0.01 ).The plasma TSH level was significantly higher in anxiety disorder with insomnia than that in the control group (P< 0.05 ).There were no significant difference of the neuroendocrine and autonomic function between anxiety disorder with or without insomnia (P > 0.05 ).The plasma COR,TSH level were significantly higher,while the SDNN,LF,HF index were significantly lower in the female group than that in the male group (P<0.01).The total SAS scores were significantly positively correhted with plasma COR levels,and was significantly negatively correlated with SDNN,HF index in the anxiety group.ConclusionThere are neuroendocrine and autonomic nervous system dysfunctions in anxiety disorder,and these dysfunctions are related with clinical subtypes,insomnia,and gender.
10.Effects of electric stimulation on Nogo-A expression in hippocampus of rats

Objective To explore the effects of chronic electrical stimulation (ES) on Nogo-A expression in the hippoeampus of rats.Methods Thirty male Wistar rats were randomly divided into a control group and 4 exper- imental groups.The rats in the control group were given sham ES,while those in the experimental groups received 1, 3,6,or 9 days of ES before being sacrificed for the detection of Nogo-A expresson in the hippoeampus by immunohis- tochemistry and Western plotting.Results There was a positive correlation between the level of Nogo-A expression and the duration of ES,as shown by the immunohistochemistry technique.Western blotting showed the same result. Conclusion In general,seizure occurred 8 days after electrical stimulation began.Elevated Nogo-A expression in the hippocampus began earlier than seizures in the epilepsy model groups.
