1.Efficacy of Controlled- Release Glipizide in Treatment of Type 2 Diabetes

ObjectiveTo compare the results of controlled- release glipizide and immediate-re- lease glipizide in treating type 2 diabetes mellitus.Methods22 patients took controlled-release glipi- zide as treating group, 20 took immediate-release glipizide as compared. The glucose tolerance, insulin, C- peptide and HbA1c were measured before and after treatment. ResultsBoth of controlled-release glipizide and immediate-release glipizide had good results for controlling blood sugar, but controlled-re- lease glipizide did not increase insulin and C-peptide level. Conclusion The controlled-release glipizide had safe and reliable results for controlling blood sugar and decreasing insulin resistence.
2.Relationship between homocysteine,C-reactive protein and P-selectin in coronary artery disease

Objective To evaluate the concentration of serum homocysteine,C-reactive protein and the expressions of P-selectin and their relationship in patients with coronary artery disease(CAD).Methods Our study included 44 patients with CAD and 22 negative controls based on coronary angiography(CAG).The concentration of serum Hcy,CRP and the expressions of PS were tested before CAG in 66 patients.Results The concentration of serum Hcy and CRP were markedly increased and the expressions of PS on platelets were significantly increased in patients with CAD compared to control group(P
3.Study on Quality Standard of Hovenia dulcis Thunb Extract

Objective To establish a quality standard of Hovenia dulics Thunb. Method The presence of quercetin was identified and assayed by TLC and HPLC, respectively. Result Linearity of marker was obtained over the range of 1.32 ~ 5.28 ?g (r =0.9999). The average recovery rate was 98.78% (RSD=1.08%). Conclusion TLC is specific. The method of quality is accurate, reappearance, simple, rapid, and suitable for the quality control of Hovenia dulcis Thunb.
4.Health-Related Quality of Life of Children with Subclinical Epileptiform Electro encephalographic Discharges

Objectives To study health-related quality of life(HRQOL) of children with subclinical epileptiform electroencephalographic discharges.Methods Subjects were 30 children with subclinical epileptiform electroencephalographic discharges,50 children with clinical seizures,28 children without clinical seizures and 30 controls.Guardians completed a valid epilepsy-specific HRQOL questionnaire for children,the quality of life in children epilepsy questionnaire(QOLCE).Results Children with subclinical epileptiform electroencephalographic discharges have 8 subscales scores lower than control group,2 subscales scores higher than group with clinical seizures.Conclusions Quality of life of children with subclinical epileptiform electroencephalographic discharges is better than that of children with clinical seizures.However cognitive,behavior,daily life and social activity are impaired by subclinical epileptiform electroencephalographic discharges and lowered the HRQOL of children.
5.Application of copy number variation analysis based on next-generation sequencing in the genetic analysis of missed abortion chorionic villi

Objective To investigate the value of copy number variation analysis based on next-generation sequencing (NGS-CNVA) in the genetic analysis of missed abortion chorionic villi. Methods From August 2012 to May 2014, chorionic villi from 74 cases of missed abortion at 6-13 gestational weeks in Haidian Maternal and Child Health Hospital were collected and analyzed by karyotype analysis and NGS-CNVA. The results of the two methods were compared. Results (1) Karyotype analysis was carried out for the villi from the 74 missed abortion patients. Thirty cases were euploid, 26 cases were aneuploid, while 18 cases had structural abnormalities. The resolution of the karyotyping was 320 bands and the average report time was 22 days. (2) All of the 74 samples obtained NGS-CNVA results and the report time was 7-10 days. (3) The NGS-CNVA results of 56 cases were consistent with karyotype. Among them, 28 cases (28/56, 50%) had no copy number variants (CNV), and 19 cases (19/56, 34%)had CNV between 1 Mb and 10 Mb. 9 cases (9/56,16%) had CNV≥10 Mb found by NGS-CNVA, but not found by karyotyping. (4) According to the results of NGS-CNVA, karyotype were reviewed. The reviewed results found 7 cases with CNV<10 Mb and 3 cases with CNV≥10 Mb in 30 cases which got normal karyotype results at the first analysis. (5) Among the 18 cases of structural abnormalities, 6 cases were Robertsonian translocation. Sequencing technology could confirm the specific area of chromosome deletion/duplication in 8 cases, but could not locate them. Conclusions NGS-CNVA has lower failure rate, higher resolution, lower specimen requirement and shorter report time than karyotype analysis when used for the genetic analysis of missed chorionic villi . NGS-CNVA could be a useful genetic analysis method for the missed abortion villi.
6.Analysis of the Rate of Common Genetic Mutations of Deaf in Pregnant Women

Objective To carry out a molecular screening of Chinese common deafness gene mutations in Chinese pregnant women group,so as to expatiate on the content,provide molecular epidemiological data,reduce the birth rate and provide a theoretical basis to the deaf children. Methods The molecular detection was done to the pregnant women underwent normal antenatal care in our hospital,using gene chips to screen the four com?mon deaf genes(GJB2,GJB3,SLC26A4 and mitochondrial 12S rRNA)in China;then,the newborn infants carrying mutations were treated with the hearing screening,using the methods of Otoacoustic Emissions(OAE)and Brainstem Auditory Evoked Potentials(BAEP),and the husbands of mutation carrying pregnant women were adopted molecular testing of the deaf susceptibility genes in order to investigate the correlation of the rate of pregnant women carrying the mutant genes and newborn infants deafness. Results Totally 2 067 cases of pregnant women were accepted to do the molecular screening,there were 110 cases of deafness mutations detected(5.320%),in which GJB2 gene(67 cases),GJB3 gene(6 cases), SLC26A4gene(33 cases),mitochondrial 12SrRNAgene(4 cases)mutation detection rates were 3.240%,0.290%,1.600%and 0.190%,respec?tively;especially:GJB2gene 235 del C,GJB2gene 299 del AT double mutant 1 case;GJB2gene 299 del AT,GJB3gene 538 C>T double mutant 1 case;GJB2 gene 235 del C,SLC26A4 gene IVS7?2 A>G double mutant 1 case. About 108 cases children newborn accepted to do the hearing screening,in which 3 cases had problems with the left ear,3 cases with the right ear,and 4 cases with the double ears. Conclusion The use of ge?netic deafness gene chip to do the molecular diagnostics in pregnant women can be convenient,fast and efficient for prenatal diagnosis of deafness, which provides a theoretical basis and good method for reducing the birth rate of deaf children and should be popularized more widely.
8.A novel dipeptidyl peptidase IV inhibitors developed through scaffold hopping and drug splicing strategy.

Though all the marketed drugs of dipeptidyl peptidase IV inhibitors are structurally different, their inherent correlation is worthy of further investigation. Herein we rapidly discovered a novel DPP-IV inhibitor 8g (IC50 = 4.9 nmol.L-1) which exhibits as good activity and selectivity as the market drugs through scaffold hopping and drug splicing strategies based on alogliptin and linagliptin. This study demonstrated that the employment of classic medicinal chemistry strategy to the marketed drugs with specific target is an efficient approach to discover novel bioactive molecules.

9.Study of HOXA5 gene expression in breast carcinoma.

OBJECTIVETo study mRNA and protein expression of HOXA5 gene in breast carcinoma, to correlate the expression of HOXA5 gene with clinicopathologic parameters and to explore the possible role of HOXA5 gene in carcinogenesis, progression and metastasis of breast carcinoma.
METHODSTaqMan real-time reverse transcriptase-polymerase chain reaction (RT-PCR) was applied on 60 cases of primary breast carcinoma and 24 cases of benign mammary lesions in order to detect mRNA expression of HOXA5 gene. Immunohistochemical study using polyclonal antibody against HOXA5 was also performed. Statistical analysis was carried out to analyze the correlation between HOXA5 gene expression and various clinical parameters in these breast cancer patients.
RESULTS(1) The relative expression level of HOXA5 mRNA ranged from 0.73 to 193.07 (average = 20.85) in primary breast carcinoma, in contrast to 5.42 to 81.91 (average = 30.94) in benign mammary lesions. Compared with benign mammary lesions, a significant reduction in expression of HOXA5 mRNA was noted in primary breast carcinoma (P < 0.01). (2) There was a decreased or completely diminished HOXA5 protein expression in breast carcinoma. (3) HOXA5 mRNA expression was significantly lower in lymph node-positive cases, when compared with that in lymph node-negative cases (P < 0.05). A significant difference of HOXA5 protein expression was also observed in both groups (P < 0.01). Immunohistochemical staining of HOXA5 was either negative or weakly positive in lymph node-positive cases. On the other hand, moderately or strongly positive HOXA5 staining was noted in lymph node-negative cases. (4) Neither mRNA nor protein expression of HOXA5 gene correlated with clinicopathologic parameters such as age of patients, size of tumor, clinical stage, pathologic subtype or histologic grade (P > 0.05).
CONCLUSIONSDisordered expression of HOXA5 gene may play a role in the carcinogenesis of breast cancer. Reduced expression of HOXA5 gene may be related to the metastatic potential of breast carcinoma cells.

10.Observation on therapeutic effect of turtle probing the cave needling method on diabetic gastroparesis.

OBJECTIVETo compare therapeutic effects of acupuncture therapy and Motilium on diabetic gastroparesis.
METHODSSixty cases were randomly divided into a needling method group and a medication group, 30 cases in each group. The needling method group were treated with turtle probing the cave (a kind of needling manipulation) as main, with pair-acupoints, Zhongwan (CV 12) and Zusanli (ST 36), Neiguan (PC 6) and Sanyinjiao (SP 6) selected, once a day, two weeks constituting one course. The medication group were treated with oral administration of 10 mg Motilium, thrice each day, 30 minutes before meals, two weeks constituting one course. The therapeutic effects were observed after 2 courses.
RESULTSThe total effective rate was 93.3% in the needling method group and 73.3% in the medication group, with a significant difference between the two groups (P<0.05). The needling method group in improvement of epigastric distention and pain, eructation, nausea and vomiting was better than the medication group.
CONCLUSIONThe therapeutic effect of turtle probing the cave needling method on diabetic gastroparesis is significantly better than that of Mutilium.
