BackgroundAdenoid cystic carcinoma is a malignant tumor originating from lacrimal gland epithelium with high recurrence rate and mortality because of its invasiveness. Although surgery, radiotherapy and chemotherapy are used clinically, the curative effect is not enough satisfied. ObjectiveThis study was to provide an experimental basis for clinical application of 125 Ⅰ seeds interstitial brachytherapy. MethodsHuman ACC-2 cell links were transplanted subcutaneously in the back of 30 SPF BALB/C nude mice to establish the ACC model, and 25 of these mice with suitable sizes of tumor were selected and randomly divided into the G1 (0. 4 mCi 125 Ⅰ ), G2 (0. 6 mCi 125 Ⅰ ) ,G3(0. 8mCi 125 Ⅰ ) ,and G4( 1.0mCi 125 Ⅰ ) groups according to the different therapeutic radioactivity treatments, with 5 nude mice for each group, and a 125 Ⅰ seed without radioactivity was used in 5 mice as the control group. The dimensions of the tumors were measured at 2-day intervals and the inhibition rates of tumors were calculated. Nude mice were killed 14 days later by the broken neck method, and the amount of apoptosis and necrosis as well as the maximum effective radius of tumor to 125Ⅰseed were detected under the transmission electron microscope and routine pathological examination. ResultsFourteen day after operation, the dimension of tumors was (3713.19±243.23)mm3 in the G0 group;while in the G1 ,G2 ,G3 and G4 groups, the dimensions of tumors were (3113.35±316.54) mm3,(2635.85±261.21) mm3, (2538.37±312.16) mm3 and(1686.28±231.65) mm3,respectively, showing a significant decrease in comparison with the Go group( P<0. 05 ). The tumor inhibitory rates in the G1 ,G2,G3 and G4 group were(20. 11±3.09)%, (36. 18±2.54)% ,(40. 83±4. 17)% ,and(66. 63±5.34)% ,with an obvious elevation with the increase in the dose of 125 Ⅰ ( F=120. 240,P=0. 000). Correlative analysis showed that the intensity of radioactivity from 125Ⅰhad a positive correlation with tumor inhibitory rate (r =0. 653,P =0. 008 ). The maximum effective radius were ( 5.2 ±0.5 ) mm, ( 6.4 ±0. 7 ) mm, ( 7.4 ±0.4 ) mm, and ( 8.2 ±0. 5 ) mm in the G1 ,G2, G3 and G4 groups, with the considerable differences among them (F=29. 22, P=0. 000). Radioactivity of 125 Ⅰ exhibited positive correlation with the maximum effective radius ( r =0. 609, P =0. 004). Conclusions125 Ⅰseed implantation brachytherapy can inhibit the growth of the transplanted ACC in BALB/C nude mice by suppressing the proliferation of tumor cells. It is a safe ,feasible and effective method to treat adenoid cystic carcinoma.

Chinese Journal of Experimental Ophthalmology 2013;(1):34-38
Background Diabetes mellitus (DM) can provoke the apoptosis of retinal cells and downregulate the expression of calcitonin gene related peptide (CGRP) in the retina.Capsaicin promotes the release of CGRP and elicits protective effects on human organs. However, whether CGRP protects retinal cells in diabetic retinopathy (DR) is still unclear. Objective The study was designed to examine the effect of capsaicin on the apoptosis of retinal cells in diabetic rats and its relationship with CGRP.Methods Forty clean healthy adult male Sprague-Dawey rats were randomly divided into the diabetes group, capsaicin pretreated group, streptozocin (STZ)control group, capsaicin control group and plain control group, with 8 rats per group. The diabetic model was established by the intraperitoneal injection of 60 mg/kg in all rats except those of the plain control group.0.4 mL of a 1% capsaicin injected at 20 mg/kg was subcutaneously injected for 3 consecutive days prior to model establishment in the capsaicin pretreated group, after which 1.2 mL of STZ was intraperitoneally injected on the fourth day.Rats from the STZ control group were administered intraperitoneally 1.2 mL of 0.1 mol/L,pH 4.5,citrate buffer. The capsaicin control group received subcutaneous injections of 0.4 mL of 1% capsaicin at 20 mg/kg for 3 consecutive days, after which 1.2 mL of 0.1 mol/L,pH 4.5,citrate buffer was administered intraperitoneally. The rats were sacrificed at the tenth week after model establishment and retinal specimens were prepared for the apoptosis assay by TUNEL staining and the quantitative analysis of caspase-3 activity.Expression of CGRP in the retina and serum was detected using ELISA.The use of experimental animals followed the Regulations for the Administration of Affairs Concerning Experimental Animals by State Science and Technology Commission. Results Retinal cell apoptosis was mainly localized to the retinal ganglion cell (RGC) layer. The apoptosis rate of RGCs was (43.4±5.0)% in the DR model group and (30.0±5.1)% in the capsaicin pretreated group, showing a significant difference (t =5.930,P<0.01).Compared with the DR model group and capsaicin pretreated group, the apoptosis rates of the DR control group (12.4±9.9) % and the capsaicin control group (17.6-±6.1) % were significantly lower (t =8.800,t =4.925,P<0.01).The apoptosis rate of the plain control group was (16.2±6.9)%,exhibiting significant differences in comparison with the DR control group and capsaicin control group (t =-0.989,t =0.951,P>0.05).The specific activity of caspase-3 was (2.19±0.86) in the DR model group and (1.96±0.56) in the capsaicin pretreated group, presenting a significant difference (t =-0.515,P<0.05).Those of the DR control group and capsaicin control group were (1.47±0.14) and (0.74±0.27),respectively,with considerable decline in comparison with the DR model group and capsaicin pretreated group (t=2.142,t=2.797,P<0.05).The retinal and serum CGRP levels were (424.4±44.2)and (148.8±39.1) ng/L,respectively,displaying significantly lower levels than (543.2±74.4) and (237.5±78.7) ng/L (t =3.070,2.359,P<0.05) from the capsaicin pretreated group. Conclusions Apoptosis of retinal ganglion cells occurs in the STZ-induced diabetic rats.Pretreatment of capsaicin reduces retinal cell apoptosis, which may be associated with an increase of CGRP in the retina.

Objective To explore the clinical manifestation, imaging examination, treatment and prognosis of Alagille syndrome in a child combined with hepatocellular carcinoma. Method The clinical manifestation, assistant examination and diagnosis of Alagille syndrome combined with hepatocellular carcinoma were analyzed in the child, and the pertinent literature were reviewed. Results The 6-year-old girl was admitted to hospital for repeated jaundice, and had a history of cardiac surgery. After admission, the patient was found to have a typical face look such as frontal bossing, sunken eyes, pointed chin and hypertrophy of nasal tip. Blood biochemistry showed intrahepatic cholestasis and increased alpha-fetoprotein. Abdominal ultrasonography revealed diffuse multiple solid lesions in the liver. And magnetic resonance imaging of the liver indicated that the liver was enlarged and multiple solid space occupying masses. Jagged 1 gene detection showed heterozygosis mutation of c.1205delC. Conclusion Alagllie syndrome complicated with hepatocellular carcinoma in childhood is extremely rare, and early diagnosis and long-term follow-up are of positive significance for its treatment and prognosis.

Objective To investigate the therapy effects of bone marrow mesenchymal stem cells (MSCs) on renal recovery after ischemia-reperfusion injured. Methods Seventy-two Spargue-Dawley rats were randomly divided into 4 groups as normal control, sham operated control, I/R group(n=30) and the MSCs group. Rats were subjected to 45 min bilateral renal ischemial-reperfusion injury with microvascular clips, after 60 min of reperfusion they were injected in BrdU positive MSCs(1?106/mL)intravenously. The animals were sacrificed at 12 h, 3 d, 7 d, 14 d, and 42 d after reperfusion, and the bilateral kidney and blood samples were harvested. The blood urea nitrogen(BUN) and serum creatinine(Scr) were measured on automatic biochemistry analyzer. Renal morphologic changes were scored with Paller’s criterion on hematoxylin and eosin(H&E) stained sections. The apoptosis of tubular cells were examined by terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL). PCNA positive tubular cells were detected immunohistochemically as proliferation index. And confocal microscopy were used to identified the distribution of BrdU positive MSCs. Results After 12 h, 3 d of reperfusion, the Scr value of MSCs group were signifcantly lower than I/R group(P

Objective: To evaluate the clinical efficacy of sinew-regulating bone-setting manipulations plus exercise therapy in treating chronic non-specific low back pain (CNLBP). Methods: A total of 65 CNLBP patients were divided into two groups by the random number table method. Thirty-three cases in the treatment group were intervened by sinew-regulating bone-setting manipulations plus exercise therapy; 32 cases in the control group were intervened by medium-frequency electrotherapy plus exercise therapy. Before and after treatment, visual analog scale (VAS), dynamic and static muscle endurance of low back, median frequency (MF) of surface electromyography (sEMG) and Oswestry disability index (ODI) were used to evaluate the low back function. The therapeutic efficacy was estimated after treatment. Results: The two groups each had 2 dropouts during the study. The total effective rate was 90.3% in the treatment group versus 66.7% in the control group, and the between-group difference was statistically significant (P<0.05). After treatment, the VAS score, dynamic and static muscle endurance of low back, MF of sEMG and ODI score all changed significantly in both groups (all P<0.05); all the items in the treatment group were significantly different from those in the control group (all P<0.05). Conclusion: Sinew-regulating bone-setting manipulations plus exercise therapy can effectively release pain in CNLBP patients, increase muscle endurance of the low back and improve the quality of life, and its therapeutic efficacy is more significant than that of medium-frequency electrotherapy plus exercise therapy.

Measurement of cervical lordosis is the basic method for evaluating cervical function, and important reference for determine treatment decision. However, how to choose appropriate measurement in accordance with different situation, as well as the relationship among these methods is not clear. An increasing number of studies suggested that different measurements could directly affect the judgment of cervical lordosis. Therefore, comparative study of cervical vertebrae plays an important role in clinical treatment for cervical spondylosis under different cervical curvature conditions.
