2.Study on influential factors associated with the long-term effect of allergic rhinitis specific immunotherapy.

OBJECTIVE:
To investigate the influential factors associated with the long-term effect of allergic rhinitis (AR) specific immunotherapy (SIT).
METHOD:
Retrospective analyzed the following visual analog scale(VAS) before and after the specific immunotherapy of 219 patients with allergic rhinitis. Logistic regression and pair t-test were used to explore the key factors influencing the prognosis.
RESULT:
(1) The gender and the VAS before specific immunotherapy (P < 0.05) were associated with the long-term effect of allergic rhinitis specific immunotherapy, while family history, the age of specific immunotherapy, number of allergens and stitches and with other allergic diseases (P > 0.05) were not associated with the long-term effect of specific immunotherapy; (2) Compared VAS of different endpoints with VAS before the treatment,there were significant differences. The primary endpoints were the time following no more than half a year (total effective rate: 50.0%), half a year to 1 year (total effective rate: 51.7%), 1 to 2 year (total effective rate: 47.1%), 2 to 3 year (total effective rate: 54.5%) and over 3 year (total effective rate: 40.7%).
CONCLUSION
Apart from the gender and the VAS before specific immunotherapy,genetic and other factors can not significant influence the long-term effect of specific immunotherapy. The effect of specific im- munotherapy seems to last for 3 years at least. Increasing stitches can not improve long-term effect of SIT,but it can consolidate the effect. The long-term effect of specific immunotherapy tends to drop off with fluctuations over time and the higher of VAS before SIT,the better effect of long-term SIT. Further more, the long-term effect of SIT of male is worse than female. In consequences, we speculate epigenetics related to the interaction between environmental factors and genetic factors plays an important role in the long-term effect of allergic rhinitis specific immunotherapy.

4.Growth state and adenovirus infection efficiency of human umbilical cord-derived mesenchymal stem cells in 3 different culture systems

BACKGROUND: In vitro culture condition and culture efficiency are different in reported umbilical cord-derived mesenchymal stem cells, and lacked of unified standards. Different derived mesenchymal stem cells have different biological properties. Therefore, it is very necessary to establish a simple and high-performance culture system for umbilical cord-derived mesenchymal stem cells. OBJECTIVE: To observe the growth state of human umbilical cord-derived mesenchymal stem cells in different culture systems in vitro and adenovirus infection efficiency. METHODS: Mesenchymal stem cells were separated from healthy full-termed delivery fetus using collagenase digestion method and purified by adherent culture. These cells were cultured and amplified in DMEM (low glucose), MesenPRO RS~(TM) Medium and STEMPRO~(R) MSC SFM in vitro. The 3-5 passage mesenchymal stem cells were infected by the Ad5-EGFP, Ad5/11-EGFP, Ad5/35-EGFP as multiplicity of infection (MOI)=1, 10, 100. Viral infection and green fluorescence expression were observed at post-infection 24, 56 and 72 hours using inverted fluorescence microscope. RESULTS AND CONCLUSION: The cell morphology in STEMPR~(R) MSC SFM was different from other two culture system and these cells were not easy to adherent after trypsin digestion. Cell doubling time in the MesenPRO RS~(TM) Medium was shorter than other two groups. Mesenchymal stem cells were infected by Ad5/35-EGFP with higher efficiency than other two kinds of adenovirus, but part of cells appeared apoptosis. The infection efficiency of Ad5/11-EGFP was highest. The fluorescence intensity was gradually increased with increased MOI.
5.Effect of sodium captopropane sulfonic on expressions of matrix metalloproteinase-9 mRNA and tissue-inhibitor of metalloproteinase-1 mRNA in lungs of paraquat poisoning rats

OBJECTIVE To study the changes in matrix metalloproteinase-9 (MMP-9) mRNA, tissue-inhibitor of metalloproteinase-1 (TIMP-1) mRNA and the ratio of MMP-9 mRNA/TIMP-1 mRNA in the lungs of paraquate poisoning rats, and to investigate the protective effects of sodium dimercaptopropane sulfonate (DMPS, Unithiol). METHODS One hundred and twenty male SD rats were divided into 12 groups (n=10) randomly: normal control, DMPS control, PQ poisoning 1, 3, 7, 14 and 28 d model groups, (DMPS+PQ) 1, 3, 7, 14 and 28 d groups. PQ poisoning model was established by intraperitoneal injecting paraquate. The expression of MMP-9 mRNA and TIMP-1 mRNA in lung tissues was detected by RT-PCR. RESULTS ①By observing the changes of action and observing the lung tissues sections, the rats' PQ poisoning models was successfully established. ②The histopathology of lung showed infiltration of inflammatory cell in acute phase(within 2 weeks), the inflammation decreased gradually after 2 weeks, hyperplasia of collagen and pulmonary fibrosis were instead. Howerer, the pathological changes were alleviated obviously in the (DMPS+PQ) groups. ③Compared with the PQ groups, the expression of MMP-9 mRNA and TIMP-1 mRNA in lungs diminished greatly in the DMPS+PQ groups after rats injected DMPS(P<0.05); the ratio of MMP-9/TIMP-1 mRNA was bigger at 7, 14 and 28 d in the DMPS+PQ groups after rats injected DMPS. CONCLUSION Down-regulation of the expression of MMP-9 mRNA and TIMP-1 mRNA and up-regulation ratio of MMP-9 mRNA/TIMP-1 mRNA by DMPS may be one of mechanisms by which pulmonary injury and pulmonary fibrosis are prevented in the acute paraquate poisoning.
6.Safety of rush allergen-specific immunotherapy in allergic rhinitis patients.

OBJECTIVE:
The safety of rush immunotherapy (RIT) in Chinese allergic rhinitis (AR)patients is unknown. The purpose of this prospective was to assess the safety differences between RIT and conventional immunotherapy in Chinese AR patients, and then discuss the clinical application feasibility of RIT.
METHOD:
A one-year study period was set for this study. The enrolled patients were divided into 2 groups according to their preference of therapy: RIT or conventional immunotherapy using standardized house dust mite allergen vaccine. For safety evaluation, the local and systemic adverse reactions were recorded throughout the both groups initial phase. Week 0 (W0), Week 2 (W2), Week 5 (W5), Week 17 (W17) were set as observation time points for leukotriene (LT-B4) and so on. The Generalized Mixed Linear Model with SPSS13. O and the chi-square test with SAS 9. 1.3 were used for Statistics.
RESULT:
Fifty-two cases were enrolled into the RIT group, of which 49 patients have completed the established treatment study, and 3 cases were lost to follow-up. In the conventional immunotherapy group, 35 cases were enrolled, of which 32 have completed established treatment study, and 3 cases were lost to follow-up. The local and systemic adverse events of AR RIT appeared to be similar to those of conventional therapy and LT-B4 was descended steadily in the two groups.
CONCLUSION
Processed in advance Chinesear with drugs, RIT is similar to the safety of conventional immunotherapy.

7.Effect of huaier granule combined with TEC neoadjuvant chemotherapy on disease control rate and serum HER2-ECD, TAP levels in patients with advanced breast cancer

Objective:To investigate the effect of huaier granule combined with docetaxel + epirubicin + cyclophosphamide (TEC) neoadjuvant chemotherapy on the disease control rate and the levels of extracellular fragment of human epidermal growth factor receptor 2 (HER2-ECD) and tumor abnormal protein (TAP) in patients with advanced breast cancer.Methods:From April 2013 to June 2016, 94 patients with advanced breast cancer in Shengjing Hospital Affiliated to China Medical University were selected and randomly divided into observation group ( n=47) and control group ( n=47). The control group was treated with TEC neoadjuvant chemotherapy, and the observation group was treated with TEC neoadjuvant chemotherapy combined with huaier granules for 12 weeks. The curative effect, incidence of adverse reactions, cellular immune function indexes (CD8 + , CD4 + , CD4 + /CD8 + ), serum HER2-ECD and TAP levels were compared before and 12 weeks after treatment. The patients were followed up for 3 years after treatment, and the 1-year, 2-year and 3-year survival rates were compared between the two groups. Results:After 12 weeks of treatment, the disease control rate of the observation group was higher than that of the control group, with statistically significant difference ( P<0.05); during the treatment, the incidence of gastrointestinal reaction, neutropenia, thrombocytopenia and abnormal liver function of the observation group was lower than that of the control group, with statistically significant difference ( P<0.05); after 12 weeks of treatment, the serum CD8 + level of the observation group was lower than that of the control group, and the serum CD4 + , CD4 + /CD8 + level was higher than that of the control group, with statistically significant difference ( P<0.05); after 12 weeks of treatment, the serum levels of HER2-ECD and TAP in the observation group were lower than those in the control group, with statistically significant difference ( P<0.05); the 3-year survival rate in the observation group was higher than that in the control group, with statistically significant difference ( P<0.05). Conclusions:Huaier granules combined with TEC neoadjuvant chemotherapy in patients with advanced breast cancer can reduce serum HER2-ECD, TAP levels, which can enhance the immune function of the body, reduce the toxic and side effects, improve the long-term survival rate, and have a significant effect.
9.Rat intestine absorption kinetics study on cucurbitacin B-sodium deoxycholate/phospholipid mixed nanomicelles with in vitro everted gut sacs model.

To investigate the absorption kinetics of Cu B-SDC/PLC-MMs in rat different intestinal segments and compared with the absorption of Cu B suspension. The in vitro everted gut sacs model was established to study the absorption characteristics of Cu B-SDC/ PLC-MMs in rat duodenum, jejunum, ileum and colon, and the content of cucurbitacin B was detected by HPLC method, and the effects of concentrations on intestinal absorption were evaluated as well. The results showed that the absorption of Cu B-SDC/PLC-MMs was linearity at different intestine segment and different concentrations (R2 > 0.9), which was consistent with zero order rate process. The Ka of different intestine segments showed a concentration-dependent increasing along with the raised concentration of Cu B-SDC/ PLC-MMs, indicating that it was likely to be a mechanism of passive absorption. The best absorption site of Cu B-SDC/PLC-MMs was ileum, and its absorptions in different intestinal segments were superior to cucurbitacin B suspension. SDC/PLC-MMs could significantly enhance the intestinal absorption of cucurbitacin B, and the study of intestinal absorption kinetics of Cu B-SDC/PLC-MMs had gave a support to its further reasonable solidfication.

10.Effect of different treatments of dentin surface on sheer bond strength between different bonding agents and dentin.

OBJECTIVETo evaluate the effect of Zinc oxide-eugenol cement and Gluma desensitizer on the sheer bond strength of three kinds of dentin bonding agents. The three dentin bonding agents were Zinc phosphate cement, Glass ionomer cement and Super-Bond C&B. To find the theory depending for the using of different protective methods and the selecting of different kinds of dentin bonding agents in prepared abutment teeth.
METHODSThe buccal surfaces of ninety freshly extracted human premolars were flattened to expose an adequate area of lower dentin. Followed by wet-grinding on a series of silicon carbide paper from number 320, 400, 600 grit to produce the dentin bonding surface. The teeth roots were embedded in self-curing resin with the crown out of the resin. The embedded ninety teeth were divided randomly into three groups. The group A was control and the dentin surfaces were not treated. The group B was covered with a paste of Zinc oxide-eugenol cement. The group C was covered with Gluma desensitizer. Calculating the sheer strength between three bonding agents and dentin after the two treatments of dentin surface. The results were statistically assessed with SPSS software. Dentin surfaces were observed with scanning electron microscope (SEM).
RESULTSThe sheer bond strengths of Zinc phosphate cement had significant decrease (P<0.05), especially the C1 group. The sheer bond strengths of Glass ionomer cement and Super-Bond C&B had no significant difference.
CONCLUSIONZinc oxide-eugenol cement and Gluma desensitizer could reduce the sheer bond strength of Zinc phosphate cement with dentin surface. Zinc oxide-eugenol cement and the Gluma desensitizer could not effect Glass ionomer cement and the Super-Bond C&B with dentin.
