The aim of this study was to explored the mechanism of inhibitory effect of chloroquine on IL-6 release induced synergistically by bacterial DNA and endotoxin. Before experiment mice were sensitized for 1 hour with D-GalN by I.P. injection. Mortality within seven days was observed after mice were challenged with CpG ODN or LPS with or without chloroquine treatment. ANA-1 cell line cells were cultivated in vitro. We investigated the influence of chloroquine on IL-6 release induced by both EC DNA and LPS. Expressions of TLR4 and TLR9 and activation of NF-?B p65 were investigated in cultured THP-1 cells pretreated with chloroquine and stimulated by EC DNA and LPS. The results showed that all mice died within 4 hours after challenged with CpG ODN and LPS. Only 4 or 5 mice pretreated with chloroquine (30mg/kg) died after challenged with CpG ODN and LPS (P

Objective To study the relationship among insulin-like growth factor 1 (IGF-1), insulin-like growth factor binding protein 3(IGFBP-3) and birth weight, length and placental weight and to discuss the endocrine factors which affected fetal growth by assaying levels of IGF-1、IGFBP-3 in umbilical cord serum. Methods The neonates were divided into appropriate for gestational age infant(AGA) and small for gestational age infant(SGA) according to birth weight and gestation. The birth weight, length and placental weight of neonates were measured and at meantime the levels of IGF-1 and IGFBP-3 were assayed by immunoradiometric assay. Results 1.There were 105 neonates, including 79 AGA and 26 SGA. There were significantly statistical differences in birth weight, length and placental weight (all, P

Objective:To detect serum levels of placenta growth factor(PIGF)throughout normal pregnancy and in patients with pregnancy-induced hypertension(PIH)as well as to explore the relationship between P1GF and pathogenesis of PIH. Methods: Serum specimens were collected from 51 healthy pregnant women as a control group and 33 women suffered from PIH as third trimester of normal pregnancy(P<0.001).There was a trend that serum levels of P1GF in PIH group decreased with the severity of PIH(P<0.001).Conclusion:PlGF play important roles in placental angiogenesis throughout pregnancy, and decreased serum levels of PlGF is associated with PIH.

Objective: To study the examination of coronary sinus (CS) and blood flow by transthoracic echocardiography(TTE) and transesophageal echocardiography(TEE).Methods: Thirty patients with supraventricular tachycardia were studied by TTE and TEE. The CS was visualized using modified 4 chamber view. The position of the probe was optimized until the coronary sinus with its ostium into the right atrium could be visualized. CS flow recordings were performed by TEE with Doppler sample volume placed in the CS within a distance of no more than 10 mm from its ostium. Results: In all patients the angle between the doppler beam and the long axis of the CS was <30°. The CS was fully displayed in 18 patients by TTE and 28 patients by TEE. The length and width of the CS were (16.53±2.57) mm and (4.51±1.30) mm by TTE, (24.11±2.46) mm and (5.06±0.97) mm by TEE.The CS flow was characterized by biphasic flow. Its flow velocity was (39±7.8), (31±6.1) and (21±4.7) cm/s respectively. The CS flow velocity-imeintegral was(43±11.6),(43±13.0),(27±8.2) cm/s. Conclusion: Echocardiography is reliable for detecting CS and its flow. TTE is more feasible for detecting CS and its flow than TEE.

A new organic-soluble co-reactant of N,N,N’,N’-tetrakis(propyl)-pentanediamine (TPPD) was synthesized and co-immobilized with tris ( 2, 2’-bipyridine ) ruthenium ditetrakis ( 4-chlorophenyl ) borate ([Ru(bpy)3][4-(Clph)4B]2) on an indium tin oxide (ITO) electrode for electrochemiluminescent (ECL) study. The hydrophobicity of TPPD prevented its leakage from the electrode surface, thus the long-term stability of the ECL electrode was enhanced. The TPPD/[Ru(bpy)3][4-(Clph)4B]2 modified-ITO electrode exhibited strong, stable and reproducible ECL signal. The ECL signal could be quenched efficiently by phenol with a quenching efficiency of 95. 4% in the presence of 0. 1 mmol/L phenol, demonstrating the potential of the modified electrode in determination of phenolic compounds. The transient-state of the electrogenerated chemiluminescence reaction was also investigated, which revealed that the TPPD/[Ru(bpy)3][4-(Clph)4B]2 modified-ITO electrode had a longer lifetime than conventional TPA-Ru(bpy)2+3 co-reactant system.

Objective:To evaluate the character of cardiac valvular calcification in uremia patients and its effect on cardiac structure and function. Methods:Thirty five cases with mitral and aortic valve calcification were selected from 326 uremia patients.Control group consisted of 33 uremia patients without valvular calcification.Their age,LVEDD,LAD,AOD,LVWT,LVEF and blood pressure matched with calcification group. The observing indexs included distribution of calcification, degree of regurgitation,LVEDD,LAD,AOD,LVWT and LVEF.The observing duration was one year. Results:The results showed that characters of valvular calcification with uremia were similar to elder valvular calcification. The degree of regurgitation was more greater in calcification group than in control group after one year. The LAD was greater in calcification group after one year. Conclusion:Cardiac valvular calcification of uremia may cause the increase of regurgitation and enlarge LAD. Objective To investigate the roles of CpG motifs in the release of cytokines induced by bacterial DNA. Methods Mice were sensitized with injection of D GalN (600 mg/kg) into the abdominal cavity 1 h before experiment. Mortality was observed after purified Escherichia coli DNA(EC DNA), calf thymus DNA(CT DNA), phosphorothioate backbone CpG oligodeoxy nucleotides (CpG ODN) and CG sequence inverted oligodeoxy nucleotides (non CpG ODN) were injected venously into mice. The injection dosage of DNA given was 30 mg/kg but ODN was 10 nmol/mouse. THP 1 cell lines were cultured in vitro . After the above reagents were added into the culture, TNF ?, IL 6, IL 1? levels were detected to observe the different abilities of these reagents to induce the release of cytokines. Results EC DNA and CpG ODN could induce the death of mice and the large amount of release of cytokines, but CT DNA and Non CpG ODN could not. Conclusion CpG ODN has the ability to induce macrophage to release cytokines largely. CpG motifs, which may play an important role in the release of cytokines induced by bacterial DNA, may probably be the structural basis of bacterial DNA.

OBJECTIVE To investigate clinical features and etiology of inhalation pneumonia.METHODS Totally 108 cases of inhalation pneumonia during from Jan 2000 to Dec 2005 were completely surveyed and analyzed. RESULTS There were underlying diseases and susceptible factors, and it was not typical in their clinical signs and symptoms.Totally 177 pathogens were isolated from sputa. There were 96 strains of Gram-negative bacilli (54.2%), 41 strains (23.2%) of Gram-positive cocci, and 40 strains (22.6%) of fungi. The 45 cases (41.7%) were with polyinfections, and 19 cases (17.6%) with double infections.CONCLUSIONS We should enhance diagnosis of inhalation pneumonia, make rational use of antibiotic, and take vigorous precautions against inhalation pneumonia.
