1.Analysis of Clinical Features and Drug Sensitivity of 67 Newborn Infants with Klebsiella Pneumoniae

Objective To explore the clinical characteristics and drug sensitivity of newborn pneumonia of klebsiella pneumoniae,and investigate the positive rate of extended-spectrum ? lactamases(ESBLs)in klebsiella pneumoniae.Method Sixty-seven cases of sputum samples newborn pneumonia of klebsiella pneumoniae from Jan.2003 to Dec.2005 were collected.Bacteria identification and drug sensitivity test were performed.At the same time,ESBLs were detected.Results The clinical characteristics of newborn pneumonia of klebsiella pneumoniae were non special,and the positive rate of ESBLs in klebsiella pneumoniae was 17.91%.The klebsiella pneumoniae was 100%sensitivie to carbopenems,but resistant to commonly used ?-antibiotics such as penicillin and cephalosporin.ESBLs negative strains had higher drug sensitivity than ESBLs strains.Conclusions Partial klebsiella pneumoniae are ESBLs strains and insensitive to commonly used antibiotics.In order to decrease ESBLs strains,the antibiotics treatment shall be used appropriately and the selection of antibiotics shall be guided by the results of drug sensitivity tests.
2.Evaluation on the quality of cryoprecipitate prepared from fresh frozen plasma via two methods

Objective To evaluate the quality of cryoprecipitates prepared from fresh frozen plasma via deepfreeze method and siphonage method thawing at 4℃ water bath,and to select a proper preparation method for cryoprecipitates.Methods Sixty samples of cryoprecipitate prepared by deepfreeze method,and 60 samples of cryoprecipitate prepared by siphonage method were selected randomly.Factor FⅧ,fibrinogen(Fg) in cryoprecipitates were measured by coagulometer,and the qualification rates were calculated.Results The concentration of FⅧ(IU/pack) in the cryoprecipitates prepared via deepfreeze method and siphonage method were(70.30?23.68),and(161.62?39.45),respectively;the qualification rate of FⅧ prepared via the two methods were 43.33%(26/60),and 93.33%(56/60),respectively(P0.05).Conclusion Siphonage method,with a higher qualification rate of FⅧ,is simple and fast,which deserves a popular application.
3.Establishment and Application of Immunomagnetic Bead Negative Enrichment and Immunofluorescence Antibody Technique for Detection of CTCs in Peripheral Blood of Patients with Ovarian Cancer

Objective To establish a cell level-based negative enrichmen technique to detect circulating tumor cells (CTCs) in the peripheral blood of patients with epithelial ovarian cancer.Methods The colon cancer SKOV-3 cells were mixed with 2 ml whole blood from healthy donors at different ratio.Quantification of CTCs was performed using immunomagnetic bead based negative enrichment combined with immunofluorescence antibody method.The method was evaluated the recovery rate of target cells,Samples of 32 patients with ovarian cancer and 10 controls were assayed for CTCs detection by above method.Results ①The recovery rate was ranging from 64% ~80% by spiking varying numbers of SKOV-3 into 2 ml blood samples of healthy volunteers.Regression analysis of number of recovered SKOV-3 cells yielded a regression equation of Y=0.782X-1.408 and a coefficient of determination of R2 =0.998.②Did not detect CK8/18-+ circulating tumor cells in 10 controls,and CK8/18+ circulating tumor cells in 18 cases of 32 Patients with ovarian cancer.The positive rate of CK 8/18 + circulating tumor cells was significantly differences between the two groups (x2 =7.681,P<0.01).③The presence ofCTCs was significantly correlated with distant metastasis (x2 =5.776,P<0.05).Conclusion The method of immunomagnetic bead based negative enrichment combined with immunofluorescence antibody technique for CTCs detection in peripheral blood of patients with ovarian cancer has a clinical value of application and extension.
4.Obesity affects airway reactivity

Objective To explore if obesity is related to bronchial hyperresponsiveness in different sexuality.Methods A total of 1180 patients with provocation test positive were studied.We analysed the relation of body mass index(BMI)and FVC,FEV1,the relation of BMI and PC_20 FEV1 in all patients and in the men and the women,respectively.Results There was negative correlation between BMI and FVC,FEV_1 of the whole patients.There was also negative correlation between BMI and PC_20FEV_1 of the whole patients and the women but no correlation of the men.PC_20 FEV_1 of men was higher than that of women.Conclusion Obesity is related to the decrease of lung function and the increase of airway responsiveness,this affection is obviously in women.The airway responsiveness of women is higher than that of man.
5.D-siRNAs suppresse expression of COX-2 in A549 cells

Objective To cleave double-stranded RNA(dsRNA) into small interference RNAs(siRNAs) that can target multiple sites within an mRNA.Methods A549 cells were isolated to incubate with 5 g/L IL-1? for different times to detect the time-dependent expression of COX-2.To generate the long dsRNA,the COX-2 gene(728 bp) was amplified by PCR with a specific forward primer that contained a T7 promoter and a specific reverse primer that contained an SP6 promoter.Then,sense strand RNAs were generated by T7 RNA polymerase and antisense strands RNAs were generated by SP6 RNA polymerase.These single strands RNAs were annealed by the standard method.We mixed dsRNA with Dicer in reaction buffer.We recovered siRNAs using RNA Purification Column.Transfections with diced siRNAs were performed using the TransMessenger Transfection Reagent in accordance with the manufacturer's instructions.COX-2 mRNA and protein were determined by RT-PCR and Western blot respectively.PGE2 was measured by ELISA.Results IL-1? induced COX-2 protein expression in A549 cells.We recovered siRNAS that have been generated in vitro by Dicer.D-siRNAs significantly suppress the expression of COX-2 in human pulmonary epithelial.Conclusion D-siRNAs significantly suppress the expression of COX-2 in human pulmonary epithelial.
6.Efficacy and Safety of Teicoplanin on Treatment of Gram-positive cocci in Lower Respiratory Tract Infection

0.05).The clearance rate of pathogens of teicoplanin was 87.5% and vancomycin was 88.9%.There was no significant difference between them.Both drugs had side effects.The side effects of vancomycin was a little more than teicoplanin.CONCLUSIONS Both vancomycin and teicoplanin are effective drugs in the treatment of G+ cocci in lower respiratory tract infection.The side effects of teicoplanin are smaller than of vancomycin.
7.Role of nicorandil in protecting myocardium of very old patients with non-ST segment elevation myocardial infarction

Objective To study the role of nicorandil in protecting myocardium of very old patients with non-ST segment elevation myocardial infarction.Methods Ninety-six very old patients with non-ST segment elevation myocardial infarction admitted to our hospital from March 2012 to July 2016 were randomly divided into experimental group (n=48) and control group (n=48).After 4 weeks of comprehensive treatment,the patients in experimental group were given oral nicorandil (5 mg,3 times a day).The serum cTnI level was measured in 2 groups before treatment and on days 1,3,7 and 10 after treatment.The ECG of 2 groups was observed before and after treatment.Results The serum cTnI level was significantly lower in experimental group than in control group on days 3,7 and 10 after treatment (3.24±4.31 μg/L vs 10.47±t4.72 μg/L,P<0.05;0.53±0.31 μg/L vs 2.50±1.45 μg/L,P<0.01;0.09±0.07 μg/L vs O.41±0.26 μg/L,P<0.01).The ECG was significantly better in experimental group than in control group after treatment (P<0.05).Conclution Oral nicorandil can protect the myocardium of very old patients with non-ST segment elevation myocardial infarction.
8. Determination of 13 amino acids in Asini Corii Colla by RP-HPLC with precolumn derivatization

Objective: To establish a RP-HPLC method for the simultaneous determination of 13 amino acids in Asini Corii Colla. Methods: Asini Corii Colla was hydrolyzed for 3 h at 120°C using 6 mol/L hydrochloric acid, and the amino acids were determined by RP-HPLC after being derived with phenyl isothiocyanate. HPLC was performed on an Eclipse AAA column (75 mm × 4.6 mm, 3.5 μm) with acetonitrile-110 mmol/L ammonium acetate solution (1:99) as mobile phase A and acetic acid-acetonitrile-water (0.05:80:20) as mobile phase B in the gradient mode at a flow rate of 0.6 mL/min. The column temperature was 42°C, and the detection wavelength was set at 254 nm. Results: The 13 amino acids were separated within 36 min with a good linearity (r > 0.9977) in the range of the test concentration. The average recoveries (n = 3) were 95.2%-104.4% and the RSD values were less than 5.0%. Conclusion: The method is simple, accurate, and repeatable, which could be available for the quality control of Asini Corii Colla. It may also serve as a good reference for the determination of amino acids in other pharmaceuticals.

10.A preliminary study on visual cortex and optic radiation with diabetic retinopathy by 1H-MR spectroscopy

Objective To study the metabolic change of proton magnetic resonance spectroscopy (1 H-MRS)in the visual cortex and optic radiation region of patients with diabetic retinopathy (DR).Methods 1H-MRS was performed in 20 patients with DR and 20 healthy volunteers on GE 1.5 T MR system respectively.Metabolic peaks of N-acetylasparte(NAA),creatine(Cr,in 3.02 and 3.94 ppm),choline-containing compounds(Cho)and myo-inositol(mI)were observed,and the ratios were analyzed by each other.Independent-samples t test was performed between two sets of data.Results In both visual cortex and optic radiation,the ratios of ml/Cr and mI/CrSee in DR group(0.664 ± 0.052 and 1.453±0.068 in visual cortex,0.717±0.074 and 1.484±0.114 in optic radiation)were significant higher than those in normal group(0.602±0.047 and 1.249±0.044 in visual cortex,0.679 ± 0.075 and 1.334±0.089 in optic radiation,P<0.05).In DR group,the ratios of CrSec/Cr,Cho/Cr and NAA/Cr in visual cortex and optic radiation were 0.458±0.043 and 0.488±0.052,0.481±0.057 and 0.807±0.110,1.633±0.105 and 1.709±0.140 respectively.In control group.the ratios of those were 0.484±0.041 and0.502±0.056,0.471±0.065 and 0.786±0.109,1.625±0.098 and 1.716±0.135 respectively.The ratios of CrSec/Cr,Cho/Cr and NAA/Cr had no statistic difference between two groups(P<0.05).Conclusion mI/CrSec is a typical change in the visual cortex and optic radiation region,1H-MRS as a noninvasive examination could provide biochemical and metabolic informations for diabetic patients.
