2.Full length genome sequencing and phylogenetic analysis of two strains of Getah virus

Abstract: Objective The main aim of the study is to sequence the complete genome of two Getah virus strains (GS11-155 and HNDZ1712-1) isolated in Gansu Province and Hainan Province in 2011 and 2017 respectively and analyze the molecular and genetic evolution of the two strains compared with M1, which was first isolated in 1964 in Hainan Province, China. Methods Genome of two newly isolated Getah viruses were sequenced by virus gene amplification technique, and the genomic database of Getah viruses was established. The molecular characteristics and genetic evolution of the viruses were analyzed by bioinformatics software. Results The genome length of two new isolated Getah virus strains (GS11-155 and HNDZ1712-1) was 11 690 nt and 11 621 nt, respectively. Both strains had the structural characteristics of Alphavirus genome. Although the nucleotide sequence lengths of structural genes, non-structural genes and non-coding junction regions of the two strains were identical, the nucleotide sequence lengths of the 5' and 3' non-coding regions of the viral genomes were a few different. The 3'UTR repeats elements in the genomes of the two virus strains did not change. It was 97.7% and 98.1% different of nucleotide and amino acid homology between both strains of Getah virus, HNDZ1712-1 isolated in 2017 and M1 isolated in 1964 in Hainan Province. Interesting, Gansu 2011 cluster and Hainan 2017 cluster were emerged leading by both strains GS11-155 and HNDZ1712-1 respectively, those two clusters totally independent with M1 virus isolated from Hainan in 1964 in whole genome phylogenetic analysis first. Conclusions Although the HNDZ1712-1 was also isolated from mosquito samples in Hainan Province, it was in a completely different evolutionary branch from the M1 isolated from Hainan Island in 1964, and was closely related to the strain isolated from Gansu Province (GS11-155) thousands of kilometers away. It is suggested that the two new strains of Getah virus are different from the Getah virus isolated in 1964.
3.Evaluation of a health education project on brucellosis and behavioral intervention in Mehlis District, Qiqihar City, Heilongjiang Province

Objective To evaluate the effects of a health education and behavior intervention project,and to provide.a scientific basis for brucellosis control.Methods By stratified cluster sampling method,target population from 6 township in Meilisi District were randomly selected to carry out baseline survey.Of these 6 townships 4 were selected as intervention townships,and two as control townships.A health education and behavior intervention program was carried out in intervention townships,and after intervention target population from 6 township were participated in the questionnaire survey.Results Awareness rate of common sense knowledge of farmers in intervention townships was 84.33% (5237/6210) after intervention which was significantly higher than 53.56% (2003/3740),before the intervention (x2 =112.49,P < 0.01),and also higher than 54.15 % (1787/3300) of the control after the intervention (x2 =101.53,P < 0.01).Average awareness rate(82.13%,2550/3105) of common sense on prevention after intervention was significantly higher than 58.77%(1099/1870) before the intervention (x2 =57.19,P < 0.01).After intervention,average awareness rate of common sense on prevention was significantly higher than that of the average of control townships (70.36%,1161/1650,x2 =25.49,P < 0.01).The awareness rate of treatment before intervention was 36.80% (116/299),which increased to 76.36% (436/571)after intervention (x2 =119.38,P < 0.01).After the intervention the awareness rate of intervention townships was significantly higher than that of the controls (33.72%,88/261,x2 =139.69,P < 0.01).In survey of these high-risk behavior in intervention townships,in addition to cattle and sheep which were regularly immunized in the intervention townships(35.91%,121/337),and the control townships(32.01%,97/303,x2 =1.08,P > 0.05) and their difference was not significant,other processing such asapoblema,killing cattle,sheep,cattle to deliver,cutting the wool,regular disinfection,and accuracy of high-risk behavior protection were significantly higher than those of controls[89.83%(106/l18),91.07%(51/56),84.75%(150/177),66.67%(32/48),73.78%(242/328),51.22%(42/82),75.56%(34/45),33.69%(63/187),27.78%(15/54),21.63%(61/282),x2 value were 38.00,5.82,103.84,15.84 and 173.67,all P < 0.05].Conclusion After implementation of health education and behavioral intervention,farmers brucellosis prevention knowledge and awareness of risk behaviors have significantly improved,which have achieved the desired effect.
4.Value of ultrasound elastography in detecting small breast tumors.

BACKGROUNDDetecting small breast tumors is difficult for conventional ultrasound. The goal of this study was to assess the value of ultrasound elastography in characterizing small breast tumors and to compare its sensitivity, specificity and accuracy with conventional ultrasound.
METHODSA total of 308 breast tumors less than 2 cm in size from 283 in-hospital patients examined with both conventional ultrasound and ultrasound elastography were retrospectively analyzed. The results were compared to surgical pathology.
RESULTSThere were 104 malignant and 204 benign lesions. The sensitivities of sonography and sonoelastography were similar (P < 0.05), and the sensitivity of the two modalities combined improved remarkably to 97.1%. The mean elastic score of malignant and benign tumors less than 2 cm were 3.76 ± 1.01 and 1.73 ± 0.99, respectively (P < 0.05), and the mean elastic score of the false-negative lesions on conventional ultrasound was 3.61 ± 1.14.
CONCLUSIONSUltrasound elastography in combination with conventional ultrasound can improve the sensitivity for detecting small breast tumors. It is also valuable in detecting small malignant tumors which are difficult to diagnose with conventional ultrasound. Ultrasound elastography can be a useful adjunct to conventional ultrasound in diagnosing small breast tumors.

5.Preliminary study of XELOX regimen as the first-line chemotherapy in advanced or recurrent gastric cancer.

OBJECTIVETo evaluate the efficacy and toxicity of the combined therapy with oxaliplatin and capecitabine (XELOX) in patients with advanced or recurrent gastric cancer.
METHODSForty-one patients with previously untreated advanced or recurrent gastric cancer received intravenous infusion of oxaliplatin at the dose of 130 mg/m(2) on day 1 and oral administration of capecitabine at 1000 mg/m(2) twice a day on days 1-14. The chemotherapy was repeated every 2 weeks for a median of 4 cycles.
RESULTSTwo of 41 patients achieved a complete response, and 15 had partial responses, with an overall response rate of 41.5%. Stable disease was observed in 11 patients and progressive disease in 9. The median time to progression and overall survival was 6.2 months and 11.8 months. All the 41 patients were evaluated for toxicity according to NCI criteria, 4 showed grade 3-4 neural toxicity, 4 had hematological toxicity and 3 had hand-foot syndrome.
CONCLUSIONThe XELOX regimen shows good efficacy with an acceptable toxicity profile in advanced or recurrent gastric cancer patient.

7.Application of Quality Control Circle in Health Education in Stroke Rehabilitation Nursing

Objective To explore the effect of quality control circle (QCC) on health education in stroke rehabilitation nursing. Methods Such activities were conducted in the VIP ward of neurological rehabilitation in our hospital as: theme selecting, activity planning, status mastering, cause analyzing, countermeasures formulating and implementing, etc., in accordance with QCC theory from January to April, 2014. Results The satisfaction of stroke patients to health education rose from 80.1% to 91.1% (P<0.001), and the dissatisfaction to guidance of activities of daily living and prevention of complications decreased significantly (P<0.05) after the implementation of QCC program. All the circle members benefited from the improvement of QCC technique, team work, professional knowledge, communication and coordination, activity confidence, duty and honor. Conclusion Carrying out the QCC activities can play a positive role on health education in stroke rehabilitation nursing.
8.Human immunodeficiency virus/sexually transmitted infection,risk behavior and sexual networks among men who have sex with men in Taizhou city,Zhejiang province

Objective To study the prevalence of human immunodeficiency virus(Hiv),sexually transmitted infection(Sti),risk behavior and the sexual neworks among men who have sex with men (MSM)in Taizhou city,Zhejiang province.Methods cross-sectional Astudy was applied with venue-based sampling in 2 MSM gathering sites in Taizhou.'Informed Consent'principle was applied and MSM were studied through a structured questionnaire.Blood samples were collected from thee who accepted free and confidential HIV/STI counseling and then tested for HIV,syphilis,HCV and HsV-2 antibodies with ELISA.HIV positive sera were certified with western blot.Results 106 MSM were investigated and 97 qualified questionnaires were collected.25.0%(23/92)of these MSM have ever had l female sex partner while 47.8%(44/92)had 2 or more.14.3%(13/91)of them reported having had 1 male partner who had engaged in anal sex and 80.2%(73/91)had 2 or more.22.1%(19/86)of therh had partieipated in group sex but 62.5%(55/88)of them did not always use condom when having analintercourse.15.1%(14/93) of them had l orfll sex partner while 75.3%(70/93)having 2 or more.38.9%(37/95)of them had sex with female sex worker,and 35.5%(33/93)had sex with male-to-male sex worker.15.3%(13/85)0f them had once been male-to-male sex worker themselves.3.9%(3/77)of them were found HIV positive in blood tests,with 24.7%(18/73)positive of syphilis,15.1%(11/73)positive ofHSV-2 but HCV appeared to be negative.46 csses reported their egocentric recognition networks,with mean degree of 5.91(ranging0-10),and mesa density of 0.548(ranging 0.000-1.000).43 sexual networks were identified,with mean degree of 2.70(ranging 0-10),and mean density of 0.246(ranging 0.000-1.000).Conclusion Risk behaviors,such as multiple sex partners,IOW proportion of condom use and commercial sex engagement both with heterosexuals and homosexuals,were extensively existed among MSM in Taizhou,and the prevalence of HIV/STI was relatively high.Their sexual networks seemed complicated but there might be in place of some sukstantiaUy isolated MSM groups with high risk of Hlv/sTl infection.More study should be applied to identify the relationship between scxual networks and HIV/STI transmigsion.
9.Application of microfluidic chip analytical systems in ABO genotyping.

Limitations of polyacrylamide gel or agarose gel electrophoretic methods in genotyping research affect the interpreting of detection results. In order to develop a simple and reliable method for appraising results of ABO genotyping detection, the microfluidic chip analysis system was established by using microfluidic chip to replace the gel electrophoresis and combining with multiplex-PCR-RFLP technique. 150 blood samples were tested by this microfluidic chip analysis system with multiplex-PCR-RFLP technique to evaluate its stability and accuracy. The results showed that all the testing results were consistent with serologic ABO genotyping results and 1 blood sample with decrease of B antigen caused by CML was identified. In conclusion, the established microfluidic chip analysis system is stable and reliable technique. Application of this technique enables the ABO genotyping results to be more objective and accurate.
