1.Effect of Doxycycline on Gastric Cancer Cell Proliferation and Notch1, MMP-9 Expression

Objective To investigate the effect of Doxycycline on gastric cancer cell line BGC-823 proliferation and Notch1, Matrix metalloproteinase-9(MMP-9)expression in order to shine a light on new anticancer drugs. Methods Final concentration of 0, 5, 10, 20, 40 mg/L doxycycline were added into human gastric cancer cell line BGC-823. Cell pro-liferation was detected by MTT;Cell cycle distribution was assessed by flow cytometry;Notch1, MMP-9 protein expression was revealed by Immunoblot. Results Doxycycline can inhibit proliferation of human gastric cancer cells line BGC-823, and its effect is dose and time-dependent(P<0.01). Doxycycline alters distribution of gastric cancer cell line BGC-823. With increasing drug concentration, the proportion of cells in S phase dropped(P<0.01). Notch1 expression rose and MMP-9 expression decreased(P<0.01). Conclusion Doxycyclinecan inhibited gastric cancer cell line BGC-823 prolif-eration and up-regulating Notch1 might be one mechanisms.

6.Research on the Consciousness of Cornea Donation among University Students

Objective: To find out the attitudes of university students and postgraduates to donating corneas after their death in Chongqing. Method: In order to find out the attitudes to the donation of corneas and remains after their death, the understanding degree of the knowledge on donation and the actual participation, cue formulated 10 questions including 3-4 selective answers each one to have a questionnaire investigation among university students and postgraduates of three universities in Chongqing. Result: In 311 questionnaires, 288 of them are valid. 53.82 % of students investigated are willing to donate their corneas and 77.08% of them support the donation, but the majority don't want to go through the formalities of donation and don't know how to deal with this; nearly 2/3 of them don't know an eye bank has been set up in Chongqing; 77% of them agrees to legislate or make the local regulations as soon as possible. Conclusion: There are quite a number of volunteers in the university students, but the number of the actual participants is small. Therefore we need to strengthen the propaganda of the eye bank to impel more students to comprehend the knowledge of donation and join the volunteers.
8.Effects of sodium hyaluronate in reformed M-K liquid on prolonging duration of the preserved corneas

Objective To investigate the roles of sodium hyaluronate (SH) in the preservation of corneas. Methods Referred to M-K prescription Preserving liquid MMK was employed as the control. Different concentrations of SH added into the preserving liquid MMK were employed as the experiment groups: group Ⅰ (0.05% SH), group Ⅱ (0.03% SH), and group Ⅲ (0.01%). At 3, 5, 7, and 10 d after preservation in the preserving liquids, the ultrastuctures of the rabbit corneas were observed by scan electron microscopy and light microscopy, the corneas were stained by typan blue alizarin red staining, and the activities of SDH and G-6-P were detected by enzymohistochemistry. Results There was difference in macrostructure of the corneas in each group. Morphological changes in group Ⅰ were not obvious, but those in the control group were remarkable. Trypan blue-alizarin red staining indicated the highest vitality of corneal endothelial cells (CECs) in group Ⅰ, but the lowest in the control group. In addition, the activities of SDH and G-6-P in each group were significantly different. Conclusion The addition of SH in reformed M-K liquid can prolong the duration of the mid-term preserved corneas and the protective effect is in a dose-dependent manner.
9.Influence of Astragalus Injection on glomerular mesangial cells cultured with AGEs

AIM: To investigate the influence of Astragalus Injection on morphology,cell cycle and oxidative stress of rat's glomerular mesangial cells(GMC) cultured with advanced glycation end products(AGEs). METHODS: GMC were incubated in culture medium containing AGEs in the presence of Astragalus Injection and aminoguanidin for 48 h.At the same time,the normal and control groups were established.Then the cultured GMC was stained by mixed fluorescence liquid and observed under fluorescence microscope.Cell cycle of GMC was analyzed using flowcytometry.The activity of SOD、MDA and GSH-PX in GMC supernatant were measured by test kit.The level of ROS was detected by flowcytometry. RESULTS: Morphology analysis showed that the morphology and structure of normal GMC were normal.The structure of most cells in AGEs was unclear,cell counts increased markedly and they grew intensively.Cell cycle analysis showed that cell percentage of S phase increased and G_0/G_1 reduced.The level of ROS,MDA remarkably increased,and SOD,GSHPX activity reduced.Whereas Astragalus Injection was added,cell morphology tended to be basically normal and cell counts decreased,the percentage of S phase also decreased.The level of ROS,MDA and the SOD,GSH-PX activity restored in comparison with the control groups. CONCLUSION: Astragalus Injection can prevent GMC from lesion caused by AGEs.Astragalus Injection may protect GMC from retarding the progression of diabetic nepropathy by partially inhibiting the occurrence of oxidative stress.
