1.Effect of Acupuncture on Postpartum Stress Urinary Incontinence

Objective To observe the effect of acupuncture on postpartum stress urinary incontinence. Methods 100 natural birth mothersmet the selection conditions were divided into control group (n=50) and treatment group (n=50). The control group accepted pelvic floormuscle training and the treatment group accepted acupuncture and moxibustion in addition. They were assessed 4 weeks after treatment. Results84% in the treatment group improved and it was 56% in the control group ( χ2 =10.54, P<0.01). Conclusion Acupuncture may improvethe efficacy of pelvic floor muscle training on postpartum stress urinary incontinence.
2.Optimization the methodology of isolating human ubilical cord mesenchymal stromal cells from Wharton's jelly and examination of their passage effect on immune phenotype using flow cytometry

Objective To observe the effects of different collagenase digestions on isolating human umbilical cord mesenchymal stromal cells (MSC) from Wharton’s jelly, to exam their differentiation ability and to investigate their passage effect on the immune phenotype. Methods Human umbilical cord samples were digested by collagenaseⅠorⅡorⅣfor 4-18 hours then were passed through sieves . Cells were collected by centrifugation then inoculated in DMEM/F12 medium at concentration within range of 4.8×103-1×104/cm2 to compare the effect of different digestions on MSC. Von kossa staining and tetracycline fluorescence was used to label the osteogenic differentiation capacity of MSC. Also RT-PCR was employed to identify the differentiate capacity of MSC into myocardial-like cells. The immunophenotype of MSCs were detected by flow cytometry after subculture. Results Using collagenaseⅠdigestion, the number of MSCs isolated from human umbilical cord in Wharton’s jelly and their vitality were much higher while the period to show cell extension and primary culture time were shorter than those using collagenaseⅡorⅣdigestions. The analysis of surface marker revealed that the expression of positive markers include CD29, CD44, CD73, CD90 and CD105 did not change with passages while the negative markers such as CD31, CD34 and HLA-DR increased significantly with passages;Differential experiments induced in vitro show that human umbilical cord MSC in wharton’s jelly had the ability to differentiate into osteoblasts and myocardial-like cells. Con?clusion The human umbilical cord MSC in Wharton’s jelly was successfully isolated by collagenaseⅠdigestion. This meth?od was simple with a high success rate while cell loss and damage were minimum. This makes large-scale cultivation possi? ble. Negative markers increased with cell passages. This phenomenon revealed that MSC showed directional differentiation.
3.Surveillance on multidrug-resistant organism infection in patients with malignant tumor

Objective To understand multidrug-resistant organism (MDRO) healthcare-associated infection(HAI) in patients with malignant tumor, so as to provide basis for making HAI prevention and control measures.Methods Targeted surveillance method was used to survey MDRO HAI in patients with malignant tumor in a hospital from January 1 to December 31, 2014, WHONET 5.3 software was used to analyze the data.Results A total of 54 056 patients with malignant tumor were surveyed, HAI occurred in 3 542 (6.55%) patients, 847(23.91%)of which were MDRO HAI.Most patients(54.55%) with MDRO HAI were >60 years old.A total of 2 606 bacterial strains were isolated from patients with HAI, 847 (32.50%) of which were MDROs, and most were extended-spectrum beta-lactamases (ESBLs)-producing Escherichia coli (n=459, 54.19%).The major site of MDRO HAI was lower respiratory tract (47.34%).The top 3 departments of detection of MDROs were departments of thoracic surgery(17.12%), colorectal and anal surgery(9.92%), and general surgery (8.26%).Conclusion Incidence of MDRO HAI is higher in patients with malignant tumor, surveillance of high risk population and monitoring of antimicrobial resistance of pathogens should be strengthened, so as to reduce the occurrence of HAI.
4.Ningxin Granule for Pediatric Viral Myocarditis:A Clinical Observation of 30 Cases

Objective To observe the therapeutic effect of Ningxin Granule Heart-Quieting Granule for pediatric viral myocarditis.Methods Totally 60 cases of pediatric viral myocarditis were randomly divided into treatment group (n=30) and control group (n=30).There was no significant difference between two groups in sexuality,age and pathogenetic condition.Patients in treatment group took Ningxin Granule,while ATP,CoA,Vit C,Inosine and Ribavirin were given to control group by intravenous drip.Results The treatment group was superior to control group in relieving symptoms,reducing the bearing premature,CK-MB,cTnI and LVEDD,and increasing LVEF.Conclusion Ningxin Granule can obviously relieve symptoms and signs,reduce the bearing premature,decrease the level of myocardial enzyme and improve the cardiac function.
5.Role of endotoxemia in the occurrence and development of cirrhosis

AIM: To study the roles of endotoxemia in the occurence and development of cirrhosis. METHODS: The cirrhosis of rats were made by oral intake of thioacetamide (TAA-treated rats). A separate group of rats were subjected to a injection of small dosage of lipopolisaccharride (LPS) in the period of thiocetamide treatment in order to observe the possible effect of LPS on the forming of cirrhosis. RESULTS:Both plasma endotoxin levels and hepatic collagen contents were increased in TAA-treated rats,and there was a linear correlation between them ;Plasma and hepatic tumor necrosis factor (TNF?)、endothelin-1(ET-1)、nitric oxide(NO)、and MDA levels were all increased,and plasma endotoxin was correlated with all of their plasma contents;Only TNF? levels and hepatic collagen contents of TAA+LPS group were increased significantly compared with TAA group. CONCLUSION: These results suggest that endotoxemia (ETM) could play an important role in the development of cirrhosis mainly through activating Kupffer cells, which secrete TNF?, ET-1, NO and free radicals could be involved in the process.
6.Role of endotoxemia in the development of hepatocarcinoma in rats

AIM: To explore the effect of endotoxemia on hepatocarcinoma course induced by TAA. METHODS: The rat cirrhosis was induced by intaking TAA (0.03%) for four months, hepatocarcinom for six months. Lipoplysaccharide (100 ?g/kg) was administered subcutaneously every two days from the fifth month to the end of sixth month in TAA-treated rats. The plasma endotoxin content, ?-glutamyl transpeptidase(?-GT) activity, DNA index and aneuploidy index were determined, the point mutation of N-ras, p53 gene was detected in hepatocarcinoma rats. RESULTS: Endotoxin increased protein overexpression of p53, bcl-2 and N-ras , p53 gene mutation point and free radical production, reduced antioxidase activity and aggravated DNA damage in hepatocarcinoma rats. CONCLUSION: Endotoxin could accelerate hepatocarcinogenesis promoted by TAA.

Objective To investigate the relationship of nitric oxide with nitric oxide synthase system in fetal rat kidney and the mechanism of renal injury after acute intrauterine ischemia(hypoxia)/reperfusion ischemia(hypoxia)/reperfusion. 8.Comparison of cranial base morphology between high-angle and low-angle malocclusion patients

Objective:To compare the differences of cranial base morphology between high-angle and low-angle malocclusion patients with geometric morphometrics.Methods:Pretreatment lateral cephalograps of 80 skeletal Class I patients(40 hypodivergent,40 hyperdivergent)were selected.All cephalographs were scanned,and 13 homologous landmarks of cranial base were identified and digitized.Average geometries of cranial bas e were created by means of Procrustes analysis.Thin-plate spline analysis was applied to cranial base configurations to determine local morphology differences between the two vertical facial types.Results:1.high-angle patients had the cranial base characteristics as follow:(1)Horizontal and vertical expansion occured in the sphenoida region,(2)The anterial and posterior cranial bases circled S point opposite direction revolving,which caused cranial base angle to reduce significantly.2.The low-angle the cranial base characteristics was just opposite with high-angle′s.Conclusion:The shape of cranial base was different between high-angle and low-angle malocclusion patients.
9.Analysis on antimicrobial susceptibility of bacteria on Poloxamer 407 gel

Objective To evaluate the antimicrobial susceptibility of bacteria cultured on a Poloxamer 407(P407) thermosensi-tive in-situ gel .Methods The modified Kirby Bauer(K-B) disc diffusion method was adopted to determine the drug susceptibility of 9 kinds of bacterium isolated on the P407 and the Mueller Hinton(MH)agar culture medium, at the same time the diffusion rates of different antibacterial drugs on the P407 and the MH agar culture medium were compared;the sodium dodecylsulfate polyacrylam-ide gel electrophoresis(SDS-PAGE) was adopted to analyze the outer membrane proteins (OMPs) expression of Pseudomonas aeruginosa on these two kinds of culture medium .Results In the antimicrobial susceptibility tests, the inhibition zones on the P407 gel were found to be generally smaller than those on the MH agar.The difference in the diffusion rates of the antibacterial drugs on these two kinds of culture medium had no statistically significant difference (P> 0.05);the OMPs expression of Pseudomonas aeruginosa was similar to its growth expression on the P407 culture medium ,but different from its expression on the MH agar .Con-clusion P407 as an excipient of bacteria culture medium can be used in the screening test of the biofilm antimicrobial susceptibility .
10.Agrobacterium-mediated genetic transformation of Poncirus trifoliata and PCR analysis

Object To establish an effective system for Agrobacterium mediated genetic transformation of Poncirus trifoliata Raf Methods The explants used for transformation were the epicotyls from P trifoliata; the Agrobacterium tumefaciens strain was EHA101, provided with the vector plasmid pGA482GG; the coat protein gene (CTV cp gene) was introduced into the transformation plasmid The transformation was proved by PCR and histochemical GUS assay Results In trans for mation of P tri foliata, 20 days epicotyls were suitable for transformation Shoot regeneration frequency was high when cocultivation time was 2-3 days The presence of acetosyringone during cocultivation could enhance the efficiency of transformation Histochemical GUS assay showed that 70 0% of the resistant plants were GUS positive Extra gene was proved to be transformed into P trifoliata plant by PCR analysis Conclusion An effective genetic transformation system has been established for P trifoliata This system will be useful for resistant breeding of P trifoliata
