1.Analysis of Usage of Antibiotics and Bacterial Drug Resistance in Infected Infants of Pediatrics in Changchun

Objective To investigate the usage of antibiotics in pediatrics patients in Changchun,and detect the bacterial drug resistance of main pathogens for direction of the appropriate clinical choice of drug antibiotics.Methods A total of 900 cases of infectious patients were retrospectively investigated in antibiotic usage in the ⅢA hospital in Changchun from January 2003 to November 2005 and the data were statistics analyzed.The bacterium were isolated from all kinds of specimen of children in hospital patients from December 2004 to November 2005,according to the method of "Clinical Medicine Test Handbook".All bacterial isolated from bacterial susceptibility tes-(ting) was done with Kirby-Bauey method.Tests were performed according to the guidelines of National Committee for Clinical Laboratory Standards(NCCLS)(2004).Results Antibiotics were administered in all of the cases.Four categories and 11 kinds of antibiotics were used.Usage of the first six antibiotics were as followed: ceftezole(22.3%),cefoperazone-sulbactam(21.6%),amoxicillin clavulanic acid((19.6%),)penicilin(17.2%),azithromycin(16.40%),oxacillin(10.1%).The rate of antibiotic use was 60.6% for solo use,70.7% for bigeminy use,81.7% for intravenously guttae,18.3% for peroral administration.Eight hundred and eighteen strains of pathogens were isolated.Gram-positive coccus,gram-negative accounted for 48.0% and 52.0%,respectively.All bacterium isolated showed a high resistance against penicillins and cephalosporins.Drug resistance rate of Gram-positive bacteria to penicillin was more than 50.0%,CNS,S.aureus,E.faecalis and S.pneumonic to erythromycin and cphalosporins was over 71.4% and 35.7%.The antibiotic remaining the most active against Gram-positive bacteria was vancomycin.The drug resistance rate against imipenem and ciprofloxacin were lower than(25.0%.)The antibiotic resistance rate of Gram-negative bacteria to ampicillin,cephalosporins and aztreonam were more than 83.0%,(40.0%) and 32.0%,to piperacillin and amoxicillin-clavulanic acid were morn than 68.9%,except P.aeruginosa(33.3%).The susceptibility of Gram-negative bacteria was imipenem(
2.Effects of Recombinant Human Erythropoietin on Immune Function of Chemotherapy Mice

Objective To observe the effects of recombinant human erythropoietin(rh-EPO) on immune function of cyclophosphamide-treated mice.Methods Six-week-old mice were randomly divided into 4 groups.Cyclophosphamide plus normal saline group(CTX(+NS));cyclophosphamide plus lower and higher dosage rh-EPO group(CTX+LDrh-EPO and CTX+HD rh-EPO);normal saline control group(NS).The changes of Hb,WBC,red blood cell immune function((C_3b)-R%),T lymphocyte proliferative responsiveness optical density index A(A score),IL-2,TNF-? of 4 groups were observed.Results The CTX + NS group showed lower levels on Hb,WBC,C_(3b)-R%,A score,IL-2 and TNF-? compared with NS normal control group(P0.05).There were positive correlation between C_(3b)-R% and Ascore,C_(3b)-R% and IL-2.Conclusions 1.Cyclophosphamide treated mice have lower Hb,WBC,C_(3b)-R%,A,IL-2 and TNF-? secretion.2.rh-EPO administration have improvements with Hb,RBC immune function,T lymphocyte proliferative responsiveness and IL2 production.
3. A combination with IGF-1R inhibitior enhances the sensitivity of human non-small cell lung cancer cell line PC9/G to gefitinib

Objective: To observe the effect of gefitinib alone or in combination with AG1024 (a tyrosine kinase inhibitor of insulin-like growth factor-1 receptor) on drug resistance of human non-small cell lung cancer cell line PC9/G with acquired-resistance to gefitinib, and to discuss its possible mechanism. Methods: PC9/G cells were treated with AG1024 or gefitinib alone or in combination, the proliferative activity of PC9/G cells was detected by MTT method, and the efficacy of combination therapy was assessed by median-effects principle. Apoptosis rate of PC9/G cells was examined by flow cytometry. The expression levels of phosphorylated epidermal growth factor receptor (p-EGFR), phosphorylated-Akt (p-Akt) and the phosphorylated extracellular signal-regulated kinase (p-ERK) in PC9/G cells were detected by Western blotting. Results: PC9/G cells displayed apoptotic features after treatment with AG1024 or gefitinib alone, and the cell proliferation was inhibited to different degrees. The treatment of AG1024 combined with gefitinib had a synergistic effect on the apoptosis and the cell proliferation (P<0.05) of PC9/G cells, as well as the expression levels of p-EGFR, p-Akt and p-ERK proteins were decreased. Conclusion: AG1024 in combination with gefitinib exerts a synergistic effect, which may lead to the proliferation inhibition and the apoptosis enhancement, and eventually increases the sensitivity of human non-small cell lung cancer cell line PC9/G to gefitinib. 4. Loss of heterozygosity and abnormal expression of MTSS 1 in the progression and metastasis of advanced primary gastric carcinoma

Objective: To determine the correlation of metastasis suppressor 1 (MTSS 1) gene with the prognosis of gastric cancer by detecting the expression level of MTSS 1 in gastric cancer, and to explore its underlying molecular mechanism in the development and progression of gastric cancers. Methods: The expression levels of MTSS1 in the primary lesion, adjacent normal mucosa tissue, lymph node metastasis and other metastatic lesions from patients with gastric cancer were detected by using immunohistochemistry and RT-PCR, and the correlation of MTSS 1 expression with the clinicopathological factors was evaluated. The loss of heterozygosity (LOH) of four microsatellite loci (D8S1832, D8S2132, D8S1179 and D8S1461) in MTSS 1 gene in thirteen patients with negative-expression of MTSS1 was detected by using PCR-polyacrylamide gel electrophoresis-silver staining method. Results: The positive expression rates of MTSS 1 were 33.9% (42/124) in the primary lesions and 16.3% (8/49) in the lymph node metastasis and other metastatic lesions. Loss expression of MTSS 1 was significantly associated with the advanced T stage, lymph node metastasis, advanced disease stage, and the poor histological differentiation. Loss or down-regulation of MTSS 1 expression was significantly correlated with the poor survival rate in both univariate and multivariate analysis. A higher frequency of allelic loss was observed in the primary (46.2%) or metastatic (69.2%) gastic cancer lesion with negative expression of MTSS 1. Conclusion: The loss and decreased expression of MTSS 1 may play an important role in the development and progression of gastric cancers, and MTSS 1 gene may become a potential predictor for clinical outcome of patients with gastric cancer. OBJECTIVE:To provide reference for improving licensed pharmacist qualification system and the legislation of Li-censed Pharmacist Law in China. METHODS:The implementation of licensed pharmacist system in China was introduced;based on the experience of licensed assistant physician system in China and pharmacy technician qualification system in USA,the necessi-ty and feasibility of setting up licensed assistant pharmacist were interpreted. RESULTS&CONCLUSIONS:At present,it is neces-sary to draw lessons from licensed assistant physician system in China and pharmacy technician qualification system in USA and set up licensed assistant pharmacist because of the shortage of licensed pharmacist,low threshold of qualification admittance,non-stan-dard licensed qualification of primary(assistant)pharmacy technician,absence of Licensed Pharmacist Law. It is recommended to promote the legislation of Licensed Pharmacist Law through clearing the definition and responsibility of licensed assistant pharma-cist,confirming examination scope and the threshold of qualification admittance,etc.
6.Therapeutic effect of the co-administrated metformin and rosiglitazone in treatment of type 2 diabetes mellitus

Objective To study the therapeutic effect of co-administrated metformin and rosiglitazone in the treatment of type 2 diabetes mellitus(T2DM).Methods 53 patients with T2DM were divided into 2 groups in random:co-administrated glipizide and metformin(group A)and co-administrated metformin and rosiglitazone(group B).All cases were treated for 6 months.Blood pressure(BP),blood glucose(BG),body mass index(BMI),hemoglobin A1c(HbA1c),insulin,and blood lipid were determined and the HOMA-insulin resistance index(HOMA-IR)was calculated.Results Both treatments were effective on hyperglucose,hypertension,hyperlipidemia and high BMI(P<0.05-0.01),while group B could lower the level of insulin(P<0.05)and HOMA-IR(P<0.01).Conclusions Treatment of co-administrated metformin and rosiglitazone can improve the abnormal carbohydrate and lipid metabolism,higher levels of BP and BMI,and ameliorate insulin resistance.
7.The inf1uences of QRS comp1ex duration in patients with chronic 1eft heart fai1ure

Objective To study the effects of QRS-complex duration of patients with chronic left heart failure on their in-hospital prognosis. Methods Total 174 patients admitted for chronic left heart failure ( New York Heart Association class 3 and 4 ) from January 2014 to June 2015 were enrolled the study. They were divided into two groups according to the QRS duration at admission:normal QRS duration group (QRS ≤120 ms, n=145) and prolonged QRS group (QRS ﹥120 ms, n=29). The differences of clinical characteristics and incidences of exacerbated left heart failure, fatal arrhythmias and cardiac death during hospitalization were compared between the two groups. The influences of QRS duration on in-hospital adverse cardiovascular events was analyzed by logistic regression. Resu1ts The proportion of males (75. 9% vs. 24. 1%, P=0. 001), plasma B-type natriuretic peptide (BNP) (7. 1 ± 0. 8 vs. 6. 6 ± 1. 0, P=0. 02), left ventricular end diastolic diameter (LVEDd) [(60. 7 ± 9. 9)mm vs. (53. 5 ± 10. 8)mm, P=0.001], left ventricular end systolic diameter (LVESd) [(49.1 ±13.3)mm vs. (39.7 ±13.0)mm, P﹤0. 001], and the incidence of exacerbated left heart failure (20. 7% vs. 4. 8%, P = 0. 003), fatal arrhythmias (55. 2% vs. 21. 4%, P ﹤0. 001) and cardiac death (6. 9% vs. 0. 7%, P =0. 019) during hospitalization were significantly higher in the prolonged QRS group than in the normal QRS group. The incidence of left heart failure deterioration among female patients in the prolonged QRS group was higher than that in the normal QRS group ( 28. 6%vs. 6. 1%, P=0. 034). QRS complex duration was positively related to LVEDd ( r=0. 4019, P﹤0. 001) and LVESd ( r =0. 3289, P ﹤0. 001 ) . LVEF in male patients was significantly lower than in female patients (40. 0% ± 16. 7% vs. 53. 2% ± 17. 6%, P﹤0. 001). On the contrary, LVEDd [(59. 4 ± 9. 4) mmvs. (50.3±10.6)mm,P﹤0.001],LVESd[(46.6±12.5)mmvs. (36.2±12.4)mm,P﹤0.001] were greater in male patients than in female patients. After adjusting for gender , age, cigarette smoking, history of high blood pressure, serum creatinine, low-density lipoprotein cholesterol, LVEF, LVEDd, LVESd, use of angiotensin converting enzyme inhibitors ( ACEI) or angiotensin receptor blockers ( ARB) and aldosterone receptor blockers, multiple logistic regression analysis showed that prolonged QRS complex duration is an independent risk factor of adverse prognosis for the patients with HF during hospitalization (OR=4. 21,95%CI:1. 59-11. 12,P=0. 004), and female gender is a protective factor for them ( OR=0. 304,95%CI:0. 116-0. 793,P=0. 015). Conc1usions The incidences of left heart failure deterioration, fatal arrhythmias and cardiac death in the chronic left heart failure patients with prolonged QRS duration were higher than in those with normal duration. Female gender is a protective factor for chronic left heart failure.

9.A preliminary study on species differences among Papaver somniferum L,Papaver rhoeas L and Cannabis sativa L by AFLP technique

Objective To disclose the inter-species differences among Papaver somniferum L, Papaver rhoeas L and Cannabis sativa L using AFLP analysis. MethodsThe root, stem, foliage, flower, seed of Papaver somniferum L, foliage of Papaver rhoeas L and Cannabis sativa L were collected respectively. DNA was isolated with AxyPrep Kit, and double-digested by EcoR I and Mse I, then oligonucleotide adapters were ligated. After Pre-amplification, selective amplification was performed using 6 pairs of fluorescent primer. The DNA fragments were analyzed using a CEQ 8000 DNA Fragment Analyzer. Results27 to 46, 5 to 20 and 4 to 31 pieces of amplified products were detected from Papaver somniferum L, Cannabis sativa L and Papaver rhoeas L respectively, and significant difference was observed among these three groups of products. The identical species-specific peaks were identified from root, stem, leaf, flower and seed DNA from the same Papaver somniferum L. ConclusionThe diacritical results from Papaver somniferum L, Papaver rhoeas L and Cannabis sativa L, as well as the identical results of different parts from the same plant demonstrated that AFLP analysis could be used possibly to determine the species origin of unknown plants samples.
