1.Correlation analysis between diabetic macular edema and serum E-selectin

To investigate the relationship between diabetic macular edema (DME) and serum E-selectin level.
●METHODS: Totally 90 patients with diabetic retinopathy in our hospital from October 2009 to October 2013 were observed. Retrospective analysis of the correlation between macular edema degree and serum soluble E -selectin ( sE - sel ). Ninety patients with diabetic retinopathy were divided into two groups: DME group and non - DME group. The level of sE - sel and microalbuminuria (MA) of two groups were determined and compared.
●RESULTS: There was significant difference in MA, sE-sel levels of groups ( P < 0. 05). The value of diabetic retinopathy patients with sE - sel and MA, FBG were positively correlated ( r = 0. 728, P < 0. 05; r = 0. 651, P <0. 05).
●CONCLUSlON: Elevated levels of sE-sel is a risk factor for diabetic macular edema, and sE-sel was involved in the pathogenesis of diabetic microangiopathy.
2.Multipoint Mutation and Over-expression in Pichia pastoris of Phytase Gene

According to bias in codon choice of Pichia pastoris, The phytase phyA gene from Aspergillus niger N25 was mutated without changing its amino acid sequence. The expression plasmid pPIC9k-phyAm was constructed and transformed into GS115 strain. Positive clones,of which the chromosomes were integrated with phyA gene,were identified by the phenotype and PCR. SDS-PAGE analysis suggust that the size of enzyme protein of the expression product was about 70.15kDa.Southern blotting analysis to the yeast transformants showed that phyA gene was intergrated into the chromosome genome. The phytase activity of PP-NP m-4-4 with codons optimized reached 136 000U/ml in malt wort culture medium after being induced with 36h, which was the 2.8 times of the original strain PP-NPm-8.
3.Diffusion-weighted MRI in residual foci evaluation of uterine cervical cancer after chemoradiation therapy

Objective To evaluate the usefulness of diffusion-weighted MR combined with routine T2 WI in finding the possible residual foci in uterine cervical cancers after radical chemo-radiation therapy.Methods This was a retrospective study including 25 consecutive cervical cancer patients who received hysterectomy after radical chemo-radiation therapy.All of them underwent MR examinations post-chemoradiation and just before operation.Images of T2 WI alone and those of T2 WI combining DWI were evaluated respectively by 2 senior radiologists,in order to decide whether there were residual tumors.ADC values were also measured.Taking the post-operation pathological results as the gold standard,the accuracies,sensitivities and specificities of T2 WI alone,T2 WI combining DWI,and ADC values were all calculated.Results In those 25 patients,9 were found with foci of residual cancer in operative pathology,while no cancer cells were found in the other 1 6 patients.The accuracy,sensitivity and specificity in finding the positive residual cancer using T2 WI alone were 56.0%,77.8% and 43.8%,comparing with 72.0%,66.7% and 75.0% in T2 WI combining DWI.The accuracy and specificity increased with statistical significance after combining DWI (P =0.01 6 for accuracy,P =0.031 for specificity),while the sensitivity decreased but did not reach statistically significant level (P =0.099).No difference in ADC values was found.Conclusion DWI can be used as a supplementary sequence in finding the existence of residual tumors of cervical cancer after radical chemo-radiation therapy.Routine T2 WI combing DWI increased the specificity and accuracy,but still facing the risk of decreasing sensitivity.

5.Application of Tn5 Transposon Mutagenesis Technology in Molecular and Genetic Researches of Gram-negative Bacteria

With development of wide-host-range vector systems,Tn5 transposon and its derivative vectors have been widely applied to genetic research of gram-negative bacteria.The applications of Tn5 transposon mutagenesis technology to genetic researches of bacteria were briefly discussed,including researches on biological control mechanisms of biocontrol bacteria,identification of bacterial essential genes,discovering virulence genes of bacterial pathogens,characterization of metabolism regulatory genes and genetic improvements of bacteria.
7.Correlation between personality characteristics and academic performance of vocational nursing students

Objective To investigate personality characteristics and its correlation with academic performance of vocational nursing students.Methods 153 nursing students were asked to filled out the questionnaire of Chinese College Student Personality Scale(CCSPS).The results were analyzed by 13.0 SPSS.Results Nursing students concentrated in the middle-up level in all the seven dimensions of the T-norm.The mean of directness,sensation and amiability were higer than the T-norm; The mean of tenaciousness and altruian were lower than the T-norm; There were different levels of correlation between academic performance and the four dimensions including directness,tenaciousness,rigorousness and amiability(P<0.05).Finally only the dimension of rigorousness entered into the regression equation of academic performance.Conclusion Vocational nursing students tend to have ordinary personalities.The dimension of rigorousness is the major factor influencing vocational nursing students' academic performance.So it should be targeted to promote vocational nursing students' personality perfection.
8.The Influence of Two Resins on Synthesizing HCV Polypeptides by Solid-phase Method

The solid-phase method to synthesis HCV polypetides was used.The connecting rate and the polypeptides' productivity and purity were compared when Wang resin and 2-chlorotrity choloride resin acting as vectors.By the identification of mass spectrum,the products' molecular weight was analysed.By ninhydrin method we compared to the connecting rate between two resins and amino acid.By RP-HPLC we carried out the analysis and purity of hetero peptides.The result showed the first amino acid's connecting rate along with the purity and productivity which we need when 2-chlorotrityl choloride resin acted as vector was notably higher than those when Wang resin acted as vector.So 2-chlorotrityl choloride as vector is used to synthesize HCV polypeptides,which adapts to the demand of big scale's industrialization production.
9.Clinical observation of warm needling therapy for treating knee osteoarthritis of different Chinese medical syndrome types.

OBJECTIVETo observe the difference in the therapeutic efficacy of warm needling therapy on knee osteoarthritis (KOA) patients of different Chinese medical syndrome types.
METHODSTotally 197 KOA outpatients [including wind-cold-damp retention syndrome (50 cases, 61 knees), yang deficiency cold coagulation syndrome (48 cases, 58 knees), stagnation of blood stasis syndrome (49 cases, 63 knees), and insufficiency of Shen-essence syndrome (50 cases, 66 knees)] were treated with warm needling therapy, 10 days as one therapeutic course, 3 courses in total. The symptom score and changes of clinical efficacy were assessed. The contents of interleukin-1 (IL-1), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-alpha) in the synovial fluid were determined before and after treatment.
RESULTSThe symptom scores and the clinical efficacy were improved in all syndrome groups after treatment (P < 0.05), with the best effects shown in the yang deficiency cold coagulation syndrome (P < 0.05) and the worst effects shown in the stagnation of blood stasis syndrome (P < 0.05). The contents of IL-1, IL-6, and TNF-alpha in the synovial fluid decreased after treatment in all syndrome groups (P < 0.05), with the best effects shown in the yang deficiency cold coagulation syndrome (P < 0.05).
CONCLUSIONWarm needling therapy had favorable therapeutic effects on KOA patients of wind-cold-damp retention syndrome, yang deficiency cold coagulation syndrome, and insufficiency of Shen-essence syndrome, with the best effects shown on KOA patients of yang deficiency cold coagulation syndrome.

10.Preparation and evaluation of novel mesoporous molecular sieve of baicalin surface molecularly imprinted polymers.

Taking mesoporous molecular sieve MCM-41 as a substrate, baicalin (BA) as template, acrylamide (AM) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as a cross-linking agent, ethanol as solvent, under thermal polymerization initiator of azobis isobutyronitrilo (AIBN) , a kind of selective recognition of baicalin surface molecularly imprinted polymer was synthesized. The surface morphologies and characteristics of the MIPs were characterized by infrared spectroscopy (IR) and transmission electron microscope (TEM). The adsorption properties of polymer microsphere for the template were tested by the dynamic adsorption equilibrium experiments and static adsorption equilibrium experiments. The experiment showed that the imprinting process was successfully and the well-ordered one-dimensional pore structure of MCM-41 was still preserved. Furthermore, molecularly imprinted polymers had higher selective ability for BA, then provided a new method for the efficient separation and enrichment of baicalin active ingredients from medicinal plants Scutellaria baicalensis.
