Objective To evaluate the effect of intrathecal ketamine on nitric oxide synthase (NOS)activity in the spinal dorsal horn via a rat model of sciatic constriction injury(SCI). Methods Thirty-six male SDrats weighing 160-180g were randomly divided into six groups(n = 6 in each group): group Ⅰ sham operation;group Ⅱ SCI; group Ⅲ-Ⅵ intrathecal ketamine + SCI. In group Ⅰ right sciatic nerve was exposed but noligature was placed around sciatic nerve. In group Ⅱ-Ⅵ four ligatures were placed around the right sciatic nerveand hed without obstructing the blood supply of the nerve. In group Ⅲ -Ⅵ ketamine 12. 5?g (group Ⅲ ),50?g(group Ⅳ ), 0?g (group Ⅴ ) or 300?g (group Ⅵ ) was given intrathecally 30 min before and 1,2 and 3 daysafer surgery. On the 7th and 14th day after operation thermal and mechanical hyperalgesia were measured with ice-cold water and von-Frey filaments. The animals were decapitated 14 days after SCI. The I_(4-6) lumbar spinal cordwas immediately removed and the spinal dorsal horn was dissected on ice and homogenized. The homogenate wascentrifuged at 3 500 r/min for 10 min and the protein content was determined. NOS achvity in the spinal dorsal hornwas measured using ultraviolet spectrophotometer. Objective To investigate the effect of intrathecal ketamine on the intracellular calcium ion concentration [Ca2+ ]1 in the spinal dorsal root ganglion (DRG) neurons in a rat model of chronic neuropathic pain produced by chronic constrictive injury (CCI) .Methods Thirty-six male SD rats weighing 160-180 g were randomly divided into 3 groups ( n = 12 each) : Ⅰ sham-operated group; Ⅱ CCI group and Ⅲ ketamine + CCI group. The animals were anesthetized with intraperitoneal pentobarbital 40 mg?kg-1 . The right sciatic nerve was exposed and 4 loose ligatures were placed on the trunk of the nerve at 1-2 mm interval. In sham-operated group (Ⅰ) the sciatic nerve was exposed but not ligated. Intrathecal catheter was implanted at L4,5 and correct placement was confirmed by aspiration of cerebro-spinal fluid. In group Ⅲ ketamine 1 mg ? kg-1 was administered intrathecally. 30 min before and on the 1st, 2nd, 3rd, 5th, 7th, 9th and 11th day after operation. In group Ⅰ and Ⅱ normal saline (NS) was given intrathecally instead of ketamine. Thermal and mechanical hyperalgesia were measured with ice-cold water and von-Frey filaments on the 7th and 14th day after operation. The animals were killed by cervical dislocation on the 7th (n = 6) and 14th ( n = 6) day. Bilateral DRG of L4-6 spinal nerve were removed and homogenized and centrifuged at 5 000 r/min. Purpose To assess the effect of pelvicfloor electrical stimulation treatment by transperineal sonography, so as to provide guidance for postpartum pelvic floor rehabilitation.Materials and Methods Thirty-one delivery women who received postpartum pelvicfloor rehabilitation were selected as research group and 30 non-pregnant young women as control group in the study. Pelvicfloor electromyography treatment was taken in research group, and transperineal sonography was undergone in quiet state and pressure state (Valsalva operation) before treatment, after the fifth treatment and after the tenth treatment respectively to measure the bladder neck to the public symphysis edge distance (Dr, Ds), bladder neck angle (θr,θs) and bladder rotation angle (θ). A comparison was made between the research group and control group.Results①Dr and Ds in research group were both lower than those in control group (P<0.05), butθr,θs andθ were all higher than those in control group (P<0.05).②Dr and Ds showed a gradual increase (P<0.05) whilstθr presented a gradual decrease before the treatment, after the fifth treatment and after the tenth treatment (P<0.05).Conclusion Natural childbirth may damage the function of pelvicfloor; electrical stimulation of pelvic therapy is effective on postpartum pelvicfloor rehabilitation; and transperineal sonography can objectively assess the effect of this therapy.

Objective To investigate the roles of sodium hyaluronate (SH) in the preservation of corneas. Methods Referred to M-K prescription Preserving liquid MMK was employed as the control. Different concentrations of SH added into the preserving liquid MMK were employed as the experiment groups: group Ⅰ (0.05% SH), group Ⅱ (0.03% SH), and group Ⅲ (0.01%). At 3, 5, 7, and 10 d after preservation in the preserving liquids, the ultrastuctures of the rabbit corneas were observed by scan electron microscopy and light microscopy, the corneas were stained by typan blue alizarin red staining, and the activities of SDH and G-6-P were detected by enzymohistochemistry. Results There was difference in macrostructure of the corneas in each group. Morphological changes in group Ⅰ were not obvious, but those in the control group were remarkable. Trypan blue-alizarin red staining indicated the highest vitality of corneal endothelial cells (CECs) in group Ⅰ, but the lowest in the control group. In addition, the activities of SDH and G-6-P in each group were significantly different. Conclusion The addition of SH in reformed M-K liquid can prolong the duration of the mid-term preserved corneas and the protective effect is in a dose-dependent manner.

Objective: To find out the attitudes of university students and postgraduates to donating corneas after their death in Chongqing. Method: In order to find out the attitudes to the donation of corneas and remains after their death, the understanding degree of the knowledge on donation and the actual participation, cue formulated 10 questions including 3-4 selective answers each one to have a questionnaire investigation among university students and postgraduates of three universities in Chongqing. Result: In 311 questionnaires, 288 of them are valid. 53.82 % of students investigated are willing to donate their corneas and 77.08% of them support the donation, but the majority don't want to go through the formalities of donation and don't know how to deal with this; nearly 2/3 of them don't know an eye bank has been set up in Chongqing; 77% of them agrees to legislate or make the local regulations as soon as possible. Conclusion: There are quite a number of volunteers in the university students, but the number of the actual participants is small. Therefore we need to strengthen the propaganda of the eye bank to impel more students to comprehend the knowledge of donation and join the volunteers.

Objective To investigate the effects of ultrashortwave (USW) diathermy and electrical stimulation (ES) used in combination with nerve growth factor (NGF) in the treatment of experimental sciatic nerve injury.Methods Sixty adult Wistar rats were randomly divided into a normal control group, a model group, an NGF group,a physiotherapy group and a combined treatment group. A model of sciatic nerve injury was established in the latter four groups. Beginning on the 2nd day after the operation, no treatment was given in the model group, NGF was injected in the NGF group, diathermy and ES were administrated to rats in the physiotherapy group, and the combined treatment group was treated with USW diathermy, EW and NGF. Function, electrophysiology and morphology were evaluated at the 2nd, 7th, 14th and 30th days after the operation Results The average sciatic nerve function index (SFI), nerve conduction velocity (NCV) and nerve regeneration in the NGF, physiotherapy and combined treatment groups were significantly better than in the model group, with those in the combined treatment group improved to the greatest extent. At the 30th day there was no significant difference between the combined treatment group and the normal control group in terms of SFI, NCV, axon regeneration or myelin sheath thickness. The number of myelinated nerve fibers and the average axon diameter in the combined treatment group and normal control group were significantly higher than those in the model, NGF or physiotherapy group. Conclusions With NGF injection, additional application of USW diathermy and ES may significantly enhance the regeneration of the sciatic nerve and aid functional recovery after injury.

In order to investigate the genetic basis of morphological variation of tetraploid plantlets of Atractylodes macrocephala, diploid plantlets were taken as experimental material, sterile filtration colchicine was used to soak 0.5-1.0 cm long buds. The difference between morphology and stomatal of diploid and tetraploid of A. macrocephala was compared, and genome polymorphism was explored by AFLP. The results showed that the buds dipped in 0.1% colchicine solution for 36 h was optimal conditions to induce tetraploid of A. macrocephala with induction rate of 32.0%. Morphological indexes such as leaf area index, leaf length and width, the density of stomas and the number of chloroplast of tetraploid were distinctly different from diploid. Four hundred and fifty-one bands ranging with 80-500 bp were amplified with 24 pairs of primers, the rate of polymorphism was 32.59%. These amplification sites of diploid were different from tetraploid of A. macrocephala, and the differences in morphology of them were reflected in the DNA polymorphism.
