1.Protective effects of cannabinoid receptor agonist WIN55,212-2 preconditioning on spinal cord ischemia reperfusion injury in rats

Objective To explore the effects of cannabinoid receptor agonist WIN55,212-2 preconditioning on spinal cord ischemia reperfusion injury in rats.Methods A total of 32 male Sprague-Dawley rats was randomly divided into four groups (n =8):sham group,control group,dimethyl sulfoxide(DMSO) group which was given intraperitoneally DMSO 0.3 ml 30 min before ischemia reperfusion,and WIN group which was given intraperitoneally WIN55,212-2 1 mg/kg 30 min before ischemia reperfusion.Each rat was neurologically assessed at 24 h and 48 h after reperfusion by Tarlov scale,and the number of normal motor neurons at anterior horn of the spinal cord was recorded.Res uits The Tarlov scale of WIN group was significantly higher than that control and DMSO groups (P < 0.05).There were more normal motor neurons at anterior horn of the spinal cord in WIN group than those in control and DMSO groups (P < 0.05).Conclusions Cannabinoid receptor agonist WIN55,212-2 preconditioning might attenuate spinal cord ischemia reperfusion injury.
2.The clinical observation of flurbiprofen as pre-emptive analgesic used in patients undergoing craniotomy

Objectives To observe the effect of flurbiprofen as preemptive analgesic and it's relative adverse reactions in pa-tients undergoing selective craniotomy .Methods Sixty patients undergoing selective craniotomy were divided into two groups randomly by age,sex,location of craniotomy:observation group (Group-flurbiprofen) and control group (Group-saline).Monitored sonoclot signal and estimate bleeding amount during the operation .Evaluated the quality of emergence after operation .Recorded the VAS pain scores 2 hours,6 hours,12 hours and 24 hours after operation ,and the occurrence of adverse reactions as nausea , vomiting and respiratory de-pression.Results The concentration of prostaglandin E2(PGE2) in the serum of the observation group was lower than that of control group( P <0.05).There were no significant differences in ACT (activated coagulation time) and CR (clot rate) of the two groups( P>0.05),but the PF (platelet function) of observation group was lower than that of control group ( P <0.05).There were no signifi-cant differences in awakening time and extubation time of the two groups ( P >0.05).The 2 hours'pain scores of the two groups was of no significant difference( P >0.05),but the pain scores of observation group were lower than that of control group in 6 hours,12 hours and 24 hours after operation( P <0.05).The occurrence of nausea , vomoting and respiratory depression had no significant differences ( P >0.05) .Conclusions Flurbiprofen as preemptive analgesic can reduce the postoperative pain in patients undergoing cranioto -my,without increasing the occurrence of nausea , vomit and respiratory depression .It can effect the platelet function ,but could not in-crease the amount of bleeding .

4.Strategy of antibody-drug conjugates in preclinical safety evaluation

Recently,increasing cancer researches focus on antibody-drug conjugates(ADCs) which can improve the anti-tumor potency with less adverse effect while benefiting patients in the future. However,safety evaluation of ADCs is a big challenge because of complex components as well as in experience in preclinical studies. In this review,the authors reviewed the mode of action,hazard risks,and toxicity observed in preclinical/clinical studies of ADCs,summarized the preclinical studies of Adcetris(brentuximab vedotin)and Kadcyla(ado-trastuzumab emtansine),and suggested a better strategy of ADCs in preclinical safety evaluation.
5.Urine biomarkers after acute kidney injury in rats induced by gentamycin

OBJECTIVE To investigate the time and dose relation of new urine bio markers in rat model of acute kidney injury induced by genta mycin (GM)to search for more sensitive,noninvasive and specific markers than traditional approaches to monitor nephrotoxicity.METHODS SD Rats were im treated with GM5,20,80 mg·kg -1 or saline once daily.Rats were randomly divided into 20 subgroups:treated for 1 ,3,7,14 d and 14 d followed with 28 d recovery period.Ten rats per group (5 rats per sex)were scarified at 24 h after the last dosing or the end of recovery period.Blood sa mples were col-lected for blood urea nitrogen (BUN)and creatinine(CRE)analysis.Urine was collected at each nec-ropsy for urine protein by dry che mistry method,for kidney injury molecule-1 (KIM-1 )analysis by ELISA, and for β2-microglobulin (β2-MG)analysis by ELISA.Kidneys were obtained for histological exa mination after HE stains.RESULTS Positive protein(3 +)was noted for several fe male animals treated for 7 or 14 d at 80 mg·kg -1 and the tendency of recovery were noted at the end of recovery period.Co mpared with those in saline control group treated for 7 d,the seru m BUN and CRE levels for fe males and the CRE level for males were significantly increased at 80 mg·kg -1 (P <0.05),and the BUN level showed the tendency of increase for males at 80 mg·kg -1 (P >0.05).When treated for 14 d,the seru m BUN and CRE levels for fe males and males at 80 mg·kg -1 and the seru m CRE level for fe males at 20 mg·kg -1 were significantly increased when compared with those in saline control group(P <0.05). The seru m BUN and CRE recovered to base line for all animals treated for 14 d followed by a 28 d recov-ery period.Histopathological observation of kidney tissues indicated that focal tubule dilatation was noted for animals treated for 3 d at 20 and 80 mg·kg -1 ,infla mmatory cell infiltration and focal tubule dilatation were noted at 20 mg·kg -1 and focal renal tubular epithelial cell degeneration,infla mmatory cell infiltra-tion,focal casts (lightly)were noted at 80 mg·kg -1 for animals treated for 7 or 14 d.For animals treated for 14 d followed by a 28 d recovery period,only basophilic tubules and renal casts were noted at 80 mg·kg -1 .New urine bio markers determination indicated that KIM-1 level was significantly increased at 20 and 80 mg·kg -1 for animals treated for 3,7 or 14 d when compared with that in saline control group (P<0.05).For animals treated for 14 d followed by a 28 d recovery period,the KIM-1 level was still significantly higher than saline control group for males and fe males at 80 mg·kg -1 and males at 20 mg·kg -1 (P <0.05 ),but there was evidence for reversal.The β2-MG level was significantly increased at 80 mg·kg -1 for animals treated for 3 d(P<0.05),at 20 or 80 mg·kg -1 for animals treated for 7 or 14 d(P<0.05 or P<0.01 ),when compared with that in the saline control group.For animals treated for 14 d followed by a 28 d recovery period,the β2-MG level was still significantly higher than saline control group for males and fe males at 80 mg·kg -1 and fe males at 20 mg·kg -1 (P <0.05),but there was also evidence for reversal.CONCLUSION Urine KIM-1 and β2-GM are more sensitive and specific markers for early diagnosis of kidney injury induced by GM when compared with the traditional approaches to monitor nephrotoxicity.
6.Changes of the femur biomechanics in fluorosis rats

Objective To observe the changes of femur biomechanical properties in fluorosis rats. Methods Fifty Wistar rats of thirty-day old, weighing 90-100 g, were randomly divided according to body mass into fluorosis and control group of 25 each. Fluorosis group drank tap water containing 100 mg/L of fluoride, the control group drank tap water. The rats were observed of dental growing status and killed after feeding 6 months. Their femurs underwent tensile strength, impact, shear, bending experiments. Results The deteetable rate of dental fluorosis was significantly higher in fluorosis group[92%(23/25)] than control group[0(0/25),X2=38.97, P<0.01]. Biomeehanical data in fluorosis group(225.67±11.81,1.94±0.15,76.62±6.10,39.96.3±3.90) were lower than those of the control group(244.70±13.38,2.39±0.19,87.72±7.05,45.75±3.75) in experiments of tensile strength (MPa), impact toughness (J/mm2), shear and bending strength (MPa). The difference was statistically significant(t=3.372,5.879,3.756 and 3383, respectively, P<0.01) between the two groups. Conclusion Fluorides affected bone metabolism in rats, femur biomechanieal properties ehanged in fluorosis rats.
7.Clinical analysis and CT features of microscopic polyangiitis with lung involvement

Objective To investigate the pathologic foundation,clinical characteristics and CT features of microscopic polyangiitis(MPA)with lung involvement.Methods Clinical data of 6 patients of MPA with lung involvement in 2003-2006 were retrospectively analyzed.Results The types of lung imaging manifestations as the followings:(1)Mutiple flaky shadow in both lung in 3 cases.(2)Ground-glass shadow in 1 case.(3)Disseminated interstitial lung involvement in 2 cases.The main respiratory symptom was haemoptysis except cough and expectoration.Renal damaging often occurred besides the lung.Six cases with positive antineutrophil cytoplasmic antibody(ANCA)and 5 cases with positive P-ANCA/MPO-ANCA were found in laboratory examination.Conclusions The CT manifestations of MPA with lung involvement have no characteristic features.Clinical characteristics and laboratory examination can help to understand and diagnose MPA with lung involvement early.
9.DJ-1 gene and Parkinson disease

Recently,more and more attention have been focused on the role of genetics in the pathogenesis of Parkinson disease.DJ1 gene is a newly discovered gene related to Parkinson disease.Here,the structure,tissue location and pathological properties of the DJ-1 Protein are reviewed.The DJ1 mutant protein and its association with early onset and sporadic Parkinson disease are also discussed in the review.
10.Establishment of Abcb1 knock out rat and Abcb1 humanized rat models

Objective To knock out the Abcb1 gene of rat,and establish the Abcb1 humanized rat model based on the Abcb1 knock out rat.Methods The animal model was established using BAC and CRISPR/Cas9 technology,and was analyzed by PCR, RT-PCR and real-time PCR.Results Establishing a rat model expressing human Abcb1 stably by transfer the 153 kb BAC containing human Abcb1 promoter and cDNA into rat genome, and establishing the Abcb1 knock out rat at the same time.Establishing the Abcb1 humanized model by crossing these two strains together.The expression pattern of Abcb1 in Abcb1 humanized rat is different from the wild type rat.The Abcb1 humanized model express not only the human Abcb1 gene but also has similar expression pattern as human.Conclusions The Abcb1 knock out rat and the Abcb1 humanized rat were successfully established, and this model is close to human concerning about the drug metabolism related to Abcb1.
