1. Inhibitory effect of recombinant adenovirus expressing TEM8 gene against angiogenesis in mouse model of glioma

Objective: To evaluate the inhibitory effects of recombinant adenovirus carrying TEM8 (Ad-TEM8) against angiogenesis and growth of mouse glioma G422 cells in vitro and in vivo. Methods: The Ad-TEM8 was constructed and subcutaneously injected to immunize Kunming mice, and then the splenic lymphocytes were collected for co-culture with G422 cells in vitro. The growth inhibition of G422 cells was observed and IFN-γ secretion by the activated lymphocytes were examined by ELISOT method. In addition, the anti-tumor and anti-angiogenesis effects ofAd-TEM8 were observed in Kunming mice carrying G422 xeno grafts. Results: Ad-TEM8 effectively stimulated the generation of cytotoxic Tlymphocytes (CTLs)in Kunming mice and the secretion of IFN-γ. Subcutaneous injection of Ad-TEM8-induced cytotoxicity effectively inhibited in vivo tumor growth and angiogenesis. Conclusion: Ad-TEM8 can induce CTLs in mice, inhibit tumor growth in vitro and in vivo, and disrupt tumor vasculature. The anti-angiogenesis effect might be a mechanism for its in vivo anti-tumor effect; and TEM8 may serve as a novel target for anti-angiogenesis therapy of glioma.
2. Effect of recombinant adenovirus livin siRNA on proliferation and apoptosis of U251 cells

Objective To investigate the silencing effect of siRNA targeting livin gene on the proliferation and apoptosis of U251 cells. Methods Adenoviral Vector of siRNA targeting livin was constructed and transfected into U251 cells. The gene silencing efficiency was monitored by RT-PCR and Western blotting analylis; the proliferation and apoptosis of U251 cells were detected by MTT assays and flow cytometry, respeclwey Mean while, the therapeijlic effect of the recombrnarrt adenoVirus was also observed en tumor-beaeing mice. Resutts The constructed recombinant adenoVirus effeclwely lnhlbited the expreseion of livn at both mRNA and proton leVlls; it also lnhlbited the proeferalion and promoted the apoptds of U251 cells. In addilton, the recombinant adenoVirus effetively inhibited the humor growth in mice and improved the mean survival period of tumor-beaeing mice. Conclusion Sileneing livin gene expression with recombination adenovirus carrying siRNA targeting liven can inhibit the proliferation and promote the apoptosis of U251 cells, which casts new lights on the treatment of glioma.

4.Cow's milk challenges in gastrointestinal cow's milk allergic diseases.

OBJECTIVETo understand the clinical features of gastrointestinal cow's milk allergy in children, and to assess the importance of cow's milk challenge.
METHODAn analysis was performed on the clinical manifestations and the challenge results of 50 children who received cow's milk challenges after admission to the department of gastroenterology, Children' s Hospital of Zhejiang University School of Medicine from January,2009 to December, 2012. The value of immunoglobulin E antibody was also analyzed among the 50 children, 25 cases were male and the other 25 were female. The youngest subject was 1. 6 months old, and the oldest was 20 months, most of the cases were younger than 6 months (36 cases).
RESULT(1) Diarrhea (27 cases, 54%) and hematochezia (25 cases, 50%) were the most common clinical features, vomiting, hematemesis and abdominal distention were rare. (2) Cow's milk challenges failed in 58% of the cases, 90% of whom showed delayed allergy. Diarrhea (19 cases, 73%) was the major later presentation, whereas the immediate hypersensitivity showed angio-edema, gastrointestinal symptom and rash. (3) The neutrophil count ((3.8 ± 2.8) x 10(9)/L vs. (2.5 ± 1.3) x 10(9)/L) was higher after challenge among children who failed the challenge. The change in the count of blood cell, neutrophil and platelet was studied, however, there were no statistical differences between the challenge-failed children and the passed ones. (4) Forty-seven cases had milk specific immunoglobulin E antibody test, and 5 showed positive results, 4 of whom were seen among the challenge-failed children.
CONCLUSIONDiarrhea and hematochezia was the most common clinical manifestation, and cow's milk protein induced proctocolitis was the most common disease in practice. It is important and necessary to perform cow's milk challenge.

5.Effects of Tibetan medicineManu-Xitang granules on the expression of prostaglandin E2 and TNF-? on adjuvant arthritis rats

Objective To study the effect of Tibetan MedicineManu-Xitanggranules on the expression of prostaglandin E2 and TNF-a on adjuvant arthritis (AA) rats.Methods A total of 60 rats were randomly divided into the control group, the model group, the Aspirin group, the low-, medium- and high- dose Manu-Xitang granules groups (each group with 10). Complete Freund's adjuvant method was used for the adjuvant arthritis model except the control group. All the groups started treatment at 8th day, and the treatment last 20 days. The low-, middle- high-dosage groups were treated with 2.0, 1.0, 0.5g/(kg body weight) Manu-Xitang granules, while the the Aspirin group with ASP (0.27 g/kg), control and medol groups with the equal Volume saline. The body weight, the swelling of primary side of arthritis index were observed. The levels of PGE2 and TNF-α were measured by ELISA.Results At 6th, 12th, 18th day, compared with the model group, the body weights in Aspirin group, the low-, middle-, high-dosage groups significantly increased (P<0.05 or P<0.01); at the 6th day, the foot swelling (0.40% ± 0.18%, 0.50% ± 0.25%, 0.55% ± 0.35%vs. 0.85% ± 0.15%) in the Aspirin group, middle-, and high-dosage groups significantly decreased (P<0.05 or P<0.01). And the PGE2 (0.66 ± 0.31vs. 0.39 ± 0.11) and TNF-a (0.72 ± 0.24vs. 0.50 ± 0.15) in the high- dose group were significantly lower than the model group (P<0.05).Conclusions The Tibetan medicineManu-Xitanggranules could treat AA through the changes of PGE2 and TNF-a.
6.Effect of serotonin on subdiaphragmatic vagus nerve afferent discharge and its mechanism in rats

Objective To investigate the effect and mechanism of serotonin (5-HT) and secretin(SEC) on afferent discharge of vagus in rats. Methods Spontaneous afferent discharge of subdiaphragmatic vagus nerve was recorded in urethane anesthetized rats. The effects of 5-HT, 5-HT3 receptor antagonist and 5-HT+SEC on the discharge of subdiaphragmatic vagus nerve were investigated by intravenous injection of different dosage of 5-HT(3, 10, 30 ?g/kg), Granisetron (1 mg/kg) and 5-HT+SEC. Results Intravenous 5-HT caused obvious exciting effect on spontaneous afferent discharge of subdiaphragmatic vagus nerve. The 5-HT3 receptor antagonist significantly inhibited the effects of 5-HT. 5-HT+SEC could augment the effect of 5-HT on spontaneous discharge. Conclusion The exciting effect of 5-HT on afferent discharge of subdiaphragmatic vagus nerve is possibly mediated by 5-HT3 receptor related to vagus nerve afference. Secretin can augment the exciting effect of 5-HT on spontaneous discharge of vagus afferent nerve.

10.Clinical Efficacy of Topiramate, Carbamazepine and Sodium Valproate in the Treatment of Epilepsy Secondary to Encephalitis

Objective:To observe and compare the clinical curative effect and safety of topiramate,carbamazepine and sodium valproate on the epilepsy secondary to encephalitis.Methods:80 cases of patients with epilepsy secondary to encephalitis who were treated in our hospital from January 2010 to September 2015 were selected and divided into the topiramate group,carbamazepine group and sodium valproate group,which were treated with topiramate,carbamazepine and sodium valproate respectively.The treatment effect,congnitive scores (including executive function and visual space,name,language,abstract,attention,delayed memory,directional),adverse reaction rate of three groups were compared.Results:The effective rate oftopiramate group was the highest(80.65%,25/31),carbamazepine was the lowest (70.00%,21/30),but there was no significant difference between the three groups (P>0.05).After treatment,the scores of executive ability and visual spatial,naming,abstraction,attention,orientation,language in topiramate group were significantly higher than those of carbamazepine group and sodium valproate group (P<0.05);the incidence of adverse reactions of topiramate group was 12.90%,which was significantly lower than that of carbamazepine group (36.67%) and sodium valproate group (29.62%)(P < 0.05).Conclusions:Topiramate,carbamazepine and sodium valproate have equal therapeutic effect on epilepsy secondary to encephalitis,but topiramate had less adverse reactions and best safety.
