1.Recombinant adenovirus vector-delivered report gene expression in rats kidney through different ways

Objective To investigate expression of recombinant adenovirus(rAV) vector-mediated report gene in kidney, and to seek for the best passway of rAV vector-mediated gene transfer in vivo.Methods Thirty Sprague-Dawley(SD) rats were divided into 5 groups randomly. rAV-GFP(1013viral particales?L-1) was injected through different ways. 2, 7 and 14 d after injection, GFP expression in left kidney, right kidney, liver and lung was evaluated by fluorescence microscope and computer software. Results GFP gene expressed in kidey and reached peak 7 d after injection expression. Conclusion rAV vector-mediated report gene can be transducted in kidney. The rAV-mediated transgene expression in kidney is a potential strategy in the treatment of renal diseases.
2.Determination of β-Galactosidase Activity by Convolution Spectrum Method

The optimum convolution spectra of β-galactosidase and the linear equation of Qj=1.906×10-2+4.946×10-3 C+1.909×10-4 C2,r=1.0000 under the best condition were established by the convolution spectrum method.Based on the chromogenic substrate o-nitrophenyl-β-galactoside,a method for the determination ofβ-galactosidase activity in the COS-1 extraction sample which had been transfected transiently expression vector of β-galactosidase was determined by using the convolution curve transformation technology.The convolution spectrum method was compared with the absorption method and the result was satisfactory.The method was simple,convenient,fast and valuable to the biochemical analysis.
3.Identification of chlorin metal complexes by the convolution spectrum method

Objective: To qualitatively identify 5 chlorin metal complexes which had similar structures. Methods: The compounds were identified by the convolution curve transformation technology and computer information process technology. Results: The results were demonstrated by match comparison and three dimensional differential diagram of convolution spectra. Five chlorin metal complexes were identified satisfactorily and 10 nonidentity identification results were acquired. Conclusion: The convolution spectrum method is simple and feasible for the identification of the compounds which have similar structures. [

Objective To investigate the effects of remote preconditioning on inflammatory cytokines and respiratory index of rabbit lung injured by ischemia/reperfusion.Methods Eighteen rabbits were randomly divided into three groups(6 each):control group(C),ischemia-reperfusion group(I/R)and remote preconditioning group(R).The plasma concentrations of interleukin-6(IL-6),tumor necrosis factor-?(TNF-?)and interleukin-10(IL-10)were measured before ischemia and 60,120 and 180 min after reperfusion.Respiratory index(A-aO2/PaO2)was calculated before ischemia and 15,30,60,120 and 180 min after reperfusion.The animals were sacrificed after reperfusion,and the left lung was removed for calculation of wet/dry(W/D)ratio and lung permeability index,histological examination was done with light microscope,and diffuse alveolar damage(DAD)scores was estimated.Results The plasma concentrations of IL-6 and TNF-? were significantly higher in I/R group than in C group(P

6.Examination of coronary sinus and blood flow by transthoracic and transesophageal echocardiography

Objective: To study the examination of coronary sinus (CS) and blood flow by transthoracic echocardiography(TTE) and transesophageal echocardiography(TEE).Methods: Thirty patients with supraventricular tachycardia were studied by TTE and TEE. The CS was visualized using modified 4 chamber view. The position of the probe was optimized until the coronary sinus with its ostium into the right atrium could be visualized. CS flow recordings were performed by TEE with Doppler sample volume placed in the CS within a distance of no more than 10 mm from its ostium. Results: In all patients the angle between the doppler beam and the long axis of the CS was <30°. The CS was fully displayed in 18 patients by TTE and 28 patients by TEE. The length and width of the CS were (16.53±2.57) mm and (4.51±1.30) mm by TTE, (24.11±2.46) mm and (5.06±0.97) mm by TEE.The CS flow was characterized by biphasic flow.Its flow velocity was (39±7.8), (31±6.1) and (21±4.7) cm/s respectively. The CS flow velocity-imeintegral was(43±11.6),(43±13.0),(27±8.2) cm/s. Conclusion: Echocardiography is reliable for detecting CS and its flow. TTE is more feasible for detecting CS and its flow than TEE.
7.Oxidative stress in human skin fibroblasts induced by UVB irradiation

Objective To observe the aging,apoptosis,cell cycle arrest and oxidative stress in human skin fibroblast(HSF)induced by UVB,and to detect the expression profiles of p66Shc,a determinant of oxidative stress response and life span,in this process.Methods HSF cells were exposed to UVB at a subcytotoxic dosage twice a day for three days.The cells without exposure served as control.After another 24-hour culture,SA-β-Gal staining was performed to evaluate the senescence state of the cells,flow cytometry to observe cell apoptosis;cell cycle arrest was detected by serum starvation and flow cytometry:ELISA was applied to detect intracellular levels of superoxide dismutase(SOD)and malondialdehvde(MDA),and Western blotting to analyze the expression of p66Shc protein.Results The percentage of cells positive for SA-β-Gal staining increased from 0 to 98.3% after UVB radiation,which strongly suggested an aging state of HSF cells.The percentage of apoptotic cells increased from 0.96% to 37%.and 80.07% of the HSF cells were arrested in G0/G1 phase following the irradiation.Intracellular SOD activity decreased from(52.35±4.97)ng/g to(7.81±0.68)ng/g(P<0.01).while intracellular MDA was found to increase from(3.52±0.34)ng/g to(33.91±3.20)ng/g(P<0.05).The p66Shc protein was found to be weakly expressed in HSF in 24 hours following the exposure to UVB,and a stronger expression was noted 48 hours later.Conclusions HSF cells are induced into a state of senescence associated with oxidative stress after UVB irradiation,which may be applied as an in vitro model in aging research.The expression of p66Shc is increased in HSF during this process,and further studies are needed to explore the relation between p66Shc and oxidative stress as well as cellular aging.
8.Biological characteristics and induced differentiation of bone marrow-derived mesenchymal stem cells from sensitized mice in vitro

AIM: To evaluate the biological characteristics and differentiating potentials of bone marrow-derived mesenchymal stem cells(MSCs) from sensitized mice by allogeneic splenocyte transfusion in vitro.METHODS: Adherent culture method was applied for culturing the bone marrow-derived MSCs from sensitized mice.The cell morphology was examined and the surface marker profiles were analyzed by flow cytometry.The differentiating potentials of the MSCs into osteogenic,adipogenic and myogenic lineages were explored.The bone marrow-derived MSCs from the normal mice were collected and served as controls.RESULTS: Both the bone marrow-derived MSCs from sensitized and normal mice were exhibited a homogeneous distinctive morphology and were positive for the surface markers CD29,CD105,CD44 and Sca-1,negative in CD 34 and CD11b.The abilities of both MSCs to differentiate into osteogenic,adipogenic and myogenic pathways in the same condition were also observed.CONCLUSION: There is no difference in the biological characteristics and induced differentiating potentials between the sensitized mouse bone marrow-derived MSCs by allogeneic splenocytes transfusion and the MSCs from normal mice.

10.Plasma brain natriuretic peptide levels in elderly male patients with type 2 diabetes and primary osteoporosis

Objective To evaluate plasma natriuretic peptide brain (BNP) levels in elderly male patients with type 2 diabetes and primary osteoporosis.Methods A total of 122 elderly male patients with type 2 diabetes were divided into 3 groups according to bone mineral density(BMD):normal group (41 cases),osteopenia group (40 cases) and osteoporosis group (41 cases),and another 33 age matched healthy subjects as control group.Plasma BNP levels were determined by ELISA.Results Plasma BNP levels in osteoporosis group [(1.95 ± 0.49) pmol/L] and osteopenia group [(1.64±0.48) pmol/L] were significantly elevated compared with that in normal group [(1.32±0.38) pmol/L] and control group [(1.26±0.39) pmol/L] (all P<0.01).There was a statistical difference between osteoporosis group and osteopenia group (t=3.539,P<0.05),and also between normal group and control group (t=2.726,P<0.05).Plasma BNP levels had negative correlation with BMD of 2na-4th lumbar vertebra (r=-0.366) and femoral neck (r=-0.375),body mass index (r=-0.288) and estrodiol (E2) (r=-0.352) (all P<0.05); while had a positive correlation with parathyroid hormone (PTH) (r=0.353,P<0.05).Conclusions With BMD declining,plasma BNP levels are elevated in elderly male type 2 diabetes,which may be related to the compensatory increase in PTH and the decrease in estradiol.
