1. Effect of Zuojin Formula on proliferation and apoptosis of human GES-1 cells infected by Helicobacter pylori

Objective: To study the effect of Zuojin Formula on proliferation and apoptosis of human normal gastric epithelial cells (GES-1) cells infected by Helicobacter pylori. Methods: GES-1 cells were infected by H. pylori at different multiplicity of infection (1∶1, 50∶1, 100∶1, 200∶1, 300∶1) for 12, 24, 48 h; GES-1 cells infected by H. pylori were treated with different concentrations (0.5, 1.0, 2.0, 4.0 μg/mL) of Zuojin Formula, and the cells were harvested after 12, 24, and 48 h. The proliferation activity of cells was detected by CCK-8, and the apoptosis rate of GES-1 cells was measured by Anexin V-FITC apoptosis detecting kit and flow cytometry. Moreover, the expression of Caspase-3 was detected by Western blot and the morphological changes of apoptotic cells were detected by Hochest staining. Results: After infected by H. pylori, the cell viability showed a descending trend while the apoptotic rate showed tendency to ascend, with the increasing of multiplicity of infection and infection time. GES-1 cells were treated with multiplicity of infection of H. pylori at 100∶1 for 12, 24, 48 h, and the cell viability decreased to (80.57 ± 1.21)%, (70.04 ± 3.21)%, and (67.74 ± 2.91)%, while the apoptotic rate increased to (23.74 ± 1.71)%, (53.60 ± 1.87)%, and (70.67 ± 2.87)%. After co-cultured with 1.0 μg/mL Zuojin Formula for 24 h, the cell viability increased to (97.67 ± 1.04)%, and the apoptotic rate decreased to (31.04 ± 1.02)%, and the difference was statistically significant compared with the corresponding model group (P < 0.01). The results of Western blotting showed that the protein expression of Caspase-3 of all treatment groups was obviously decreased. Based on the morphological point of view, this result was further verified. Conclusion: GES-1 infected by H. pylori NCTC11637 can inhibit cell proliferation and induce apoptosis, while ZuoJinFang could protect GES-1 cells from cell damage.
2.Effect of sensorimotor training on standing balance of the stroke patients at recovery stage

Objective To study the effect of modified sensorimotor training (SMT) method on standing ba- lance of the stroke patients during their recovery stage. Methods Sixty stroke patients at recovery stage were ran- domly divided into an intervention group and a control group. The intervention group was trained by modified SMT method which combined Thera-band with partial body weight support (PBWS) system, while the control group was trained only with their standing balance in the parallel bars based on the neurodevelopment therapy (NDT) method. Both groups were given the same medications as well as physical therapy, acupuncture and OT. The patients in the two groups practiced standing balance in front of a mirror daily, 40 minutes every day and 6 days every week for 4 weeks. The balance abilities of patients were evaluated by Berg balance scale (BBS) , and their lower extremity func- tions were assessed by simplification Fugl-Meyer assessment (FMA). Objective To explore an effective mode for the prevention and control of hypertension in agricultural and pastoral areas in Xinjiang by comparing the effect of mode A with that of B in blood pressure-lowering treatment. Methods 1445 patients with hypertension in agricultural and pastoral areas were included in this study. They voluntarily received mode A or B blood pressure-lowering treatment. The changes in heart rate , blood pressure, lipid, and glucose, and the incidences of cardiovascular and cerebrovascular events and adverse events were noted;and the effect of modes A and B was compared. Results 87.07%of the patients chose mode A group, while 12.93%chose mode B. The rate of pressure control was 70.71%in mode A group and 68.75%in mode B group , with no significant statistical difference (P>0.05). In 12th month of treatment, the decreased level of systolic pressure was (19.09 ± 20.33)mmHg in the group with mode A and (14.14 ± 17.85) mmHg in the group with mode B, showing a significant statistical difference between the two groups (P=0.047); and the declined level of diastolic pressure was (11.17 ± 13.23)mmHg and (8.17 ± 11.17) mmHg, respectively, with no significant statistical difference. Conclusion Mode A blood pressure-lowering treatment can effectively control blood pressure in hypertensive patients living in agricultural and pastoral areas in Xinjiang.
4.Risk factors of neonatal tetanus in Wenzhou, China: a case-control study

Introduction:Neonatal tetanus is a major cause of neonatal mortality in many developing countries and remains a major public health problem. This study aimed to determine risk factors associated with neonatal tetanus in Wenzhou, China.Methodology:Medical records of neonatal tetanus cases from 17 hospitals over a 13-year period (2000–2012) were reviewed for potential risk factors. Controls were selected from neonates with diseases other than tetanus who were admitted to the same facility during the same period. The potential risk factors of the neonatal tetanus group were compared with the control group using univariate analysis and an unconditional logistic regression model.Results:A total of 246 neonates with tetanus and 257 controls were included in this study. 5.Blockage of U251 cells in G0/G1 through MAPK signaling pathway by LRRC4.

OBJECTIVE:
To explore the effect of LRRC4, a glioma suppressive gene, on blocking U251 cells in G0/G1 by MAPK signaling pathway.
METHODS:
LRRC4 was transfected into U251 cells, and at 24 hour of post-transfection, cells were split at a 1:3 dilution, challenged with 500 microg /mL G418 and formed a stable transfected clone pool. RT-PCR, Northern blot and Western blot were used to identify the stable transfectants. ERK, JNK and P38 expression changes were analyzed by Western blot. FACS analysis, Luciferase reporter gene assay and Western blot were used to detect the cell cycle and cyclin D1.
RESULTS:
LRRC4 down-regulated the expression of phosphorylated ERK2 and up-regulated the expression of total protein JNK2 (a key molecule of MAPK signaling pathway) and phosphorylated c-Jun. LRRC4 decreased the expression of mutation P53, cyclin D1 activation and its expression. U251 cells were blocked in G0/G1 by LRRC4.
CONCLUSION
LRRC4 can decrease JNK2, up-regulate the phosphoralated c-Jun, down-regulate mutant P53 and cyclin D1, and therefore block U251 cells in G0/G1.

6.Biomechanical research on upper femur with vertical and bending stress.

OBJECTIVETo study the bone state with ultimate stress by examining biomechanical distribution of upper femur in Chinese, in order to accumulate more experiences for clinical work.
METHODSTotally 60 Chinese femurs from fresh cadavers were randomly divided into two groups. All the femurs were cleaned, and the body age ranged from 36 to 72 years old, averaged 56.4 years, including 41 from males, and 19 from females. These two groups underwent mechanical stress and bending stress tests. Special mechanical laboratory and machines were used to get the information. Results about the loading value at each testing point under stress were collected.
RESULTSThe four faces of the upper femur suffered different stress under external forces. The bone on upper femur can tolerate more mechanical stress than bending stress. Medial and lateral region of the femur neck and the rear side of the small tuberosity section were themain position enduring the vertical stress. The rear position of the base femur neck and the small tuberosity section were the main regions enduring the bending stress. Those main positions had strong cancellous bones. The intertrochanteric fracture fixation and artificial femoral stems were designed depending on this biomechanical basis.
CONCLUSIONAccording to our experiment result, doctors need to chose more effective fixations for upper femur fracture, and femoral stems for the patients. More information should be collected by further researches.

7.The expression and role of Cathepsin B in intracranial aneurysm wall.

OBJECTIVESTo detect the expression of Cathepsin B (CatB) in the intracranial aneurysm wall and its effect to the apoptosis of vascular smooth muscle cells, aimed at clarifying the pathological formation mechanism of intracranial aneurysm.
METHODSFrom November 2006 to February 2009, 20 intracranial aneurysm samples were collected as the experimental group, and 6 cases of normal pallium artery samples were collected as the control group. Immunohistochemical technique was used to evaluate the expressions of CatB, alpha-smooth muscle actin (alpha-SMA) and Caspase-3. The expression of CatB mRNA was evaluated by real-time PCR. The ultrastructure of intracranial aneurysms were observed by using the transmission electronic microscope.
RESULTSCompared with the normal pallium artery specimens, the expression of CatB and Caspase-3 both significantly increased in the intracranial aneurysm walls where alpha-SMA decreased (P < 0.05). The mean expression of CatB mRNA in intracranial aneurysm samples was about 3.8-folds than that in control group (P < 0.01). There were excessive apoptotic vascular smooth muscle cells (VSMCs) in the tunica median, and typical apoptotic body were observed in some aneurysm walls.
CONCLUSIONCathepsin B may be involved in the formation and the progression of intracranial aneurysm.

8.Study on the effect of vibsane-type diterpenoids of Viburnum odoratissimum on human HepG2 cell growth and its underlying mechanism.

OBJECTIVETo study the antiproliferation effect on HepG2 cells and its underlying mechanism of the active chemical composition of the Viburnum Odoratissimum.
METHODS3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) reduction assay and trypan blue dye exclusion assay were used to assess the effect of vibsane-type diterpenoids on the proliferation of various tumor cells. Alterations in cell cycle and apoptosis were determined by flowcytometry. The enzymatic activity of caspase-3/7 was measured by Apo-ONE homogeneous Caspase-3/7 Assay kit.
RESULTSCompound 1 #, a vibsane-type diterpenoid, was found to significantly inhibit the growth of HepG2 cells by anticancer proliferation activity screening. It was demonstrated that the modified groups on side chain coupled to C11 site affected the cell growth-inhibition activity of compounds by structure-activity analysis. In addition, HepG2 cell line was most sensitive to compound 1 #, which induced growth arrest of HepG2 cells in a dose- and time-dependent manner. Study on the mechanisms underlying these effects indicated that compound 1 # induced significant G0/G1 phase arrest of HepG2 cells in a time- and concentration-dependent manner. Meanwhile, It was found that higher concentrations of compound (5-10 micromol/L) caused evident increase in the unmber of apoptotic cells and dose-dependent activation of caspase-3/7.
CONCLUSIONVibsane-type diterpenoids could significantly inhibit the growth of HCC HepG2 cells. Induction of cell cycle arrest and apoptosis may play important roles in their anticancer effects.

9.A light field-based fast computation of digitally reconstructed radiographs.

Generation of digitally reconstructed radiographs (DDRs) is key to image-guided radiation therapy and widely used for 2D-3D image registration to verify the patient's position. The classic algorithms to generate DRRs with ray casting is time consuming to hamper the registration process. The authors describe the attempt to extend the light field rendering techniques from computer graphics, using the light field computed from ray casting to interpolate a DRRs image from a novel fixed viewpoint. Because most of the computation is performed in a preprocessing step, the speed is greatly accelerated and satisfactory DRRs can be obtained.

10.Urinary S100B protein and lactate/creatinine ratio measurements: a tool for the early identification of neonatal hypoxic-ischemic encephalopathy.

OBJECTIVETo investigate the value of urinary S100B protein and lactate/creatinine ratio determination in early identification of neonatal hypoxic-ischemic encephalopathy (HIE).
METHODSThe levels of urinary S100B protein and urinary lactate/creatinine ratio were detected in 58 full-term newborn infants with HIE on the first, second and third day after birth. The severity of clinical manifestations, including the degree of encephalopathy, was assessed within 7 days after birth. Twenty five normal neonates were enrolled into the control groups.
RESULTS(1) The urinary S100B level of HIE neonates was significantly higher in samples collected throughout the monitoring period than those of the normal control groups (all P < 0.001). The urinary lactate/creatinine ratio of the HIE neonates was also significantly higher than that of normal control groups within the first day (P < 0.001). (2) A significantly positive correlation was found between the level of urinary S100B protein within three days and the urinary lactate/creatinine ratio within the first day and between the level of urinary S100B protein within three days and clinical degree (P < 0.05). (3) When S100B concentration was 0.47 microg/L and urinary lactate/creatinine ratio was 0.55, the sensitivity and specificity of detecting the third day urinary S100B alone, were respectively 90.4%, 91.9%. Detecting it associated with the first day urinary lactate/creatinine ratio could increase the sensitivity and specificity (respectively 98.8% and 97.4%) for predicting development of HIE.
CONCLUSIONOn the basis of clinical manifestations of asphyxic neonatals, detecting the level of urinary S100B within three days and the first day urinary lactate/creatinine ratio may be of important value in early diagnosis and grading of HIE.
