2.Evaluation of Arterial Elasticity, Left Ventricular Function and Correlation by 256- slice Spiral CT in Patients with Aortic Dissection

Objective Using the function of 256-layer iCT in the determination of cardiac function and arterial elasticity index, to compare the arterial elasticity, left ventricular function and their correlation between patients with aortic dissection and non-dissected controls, and to provide a reference for the clinical treatment of patients with aortic dissection. Methods This study selected 36 cases in our emergency department,with chest, back or abdominal pain, whose clinical symptoms were highly suggestive of aortic dissection, and plain or enhanced diagnosis confirmed aortic dissection as a case group. At the same time, we randomly selected 40 cases without aortic dissection as the control group. All subjects underwent CTA one-stop scanning and measurement of elastic and cardiac function at the same level. According to the measurement results, the arterial elasticity indexes and the measured cardiac function of the case group and the control group were evaluated and compared so that to evaluate the arterial elasticity and its correlation with left ventricular function in patients with aortic dissection. Results patients's arterial expansion and compliance in patients with aortic dissection were significantly lower than the control group (P<0.05) . The mean diameter change rate and stiffness of the patients with dissection were significantly lower than those in the control group (P>0.05) . The difference was not statistically significant. The mean stiffness in patients with aortic dissection was significantly higher than non-aortic dissection patients. In patients with aortic dissection, the mean arterial diameter change rate, expandability and arterial stiffness showed a highly negative correlation. There was no significant correlation between arterial elasticity index and cardiac function in patients with Stanford type A and Stanford type B aortic dissection. There was no significant correlation between each elasticity index and cardiac function in the control group (r<0.3) . Conclusion CTA one-stop scan can be used as an important measure of arterial elasticity, cardiac function and other parameters in patients with aortic dissection. Significantly lower aortic expansion and compliance, increased stiffness, insignificant changes in cardiac function, and insignificant correlation between cardiac function and each elasticity index in patients with early aortic dissection all have a clinical significance.
3.Effects of hydrogen peroxide on intracellular free Ca2+ content in rat liver oval cells.

OBJECTIVETo study the effects and mechanism of hydrogen peroxide (H2O2) of low concentration on dynamic changes of intracellular free calcium contents ([Ca2+]i) in cultural rat liver oval cells (WB-F344 cells).
METHODSUsing Fluo-3/Am as fluorescent indicator of [Ca2+]i and it was measured by laser scanning confocal microscope system.
RESULTSThe results showed that: (1) A rapid transient spiking of [Ca2+]i occurred after the stimulation of H2O2 of low concentration (800 nmol/L). (2) The [Ca2+]i increase was abolished by pretreated with catalase (CAT) or by incubated in D-Hank's solution containing EGTA, the chelate of extracellular Ca2+. (3) The [Ca2+]i increase was not inhibited by pretreated nifedipine, Ca2+ channel blocker, but was abolished by pretreated with anthracere-9-cardoxylic acid (A9C), the Cl-channel blocker and which also blocked calcium activated non-selective cation channel (CAN).
CONCLUSIONSThese results suggest that the increase of [Ca2+]i induced by H2O2 of low concentration may be due to the influx of extracellular Ca2+ through CAN.

4.Changes of the elastic fibers and collagen fibers during the development and progression of experimentally induced tongue carcinoma in hamsters.

OBJECTIVETo investigate the relationship between the extracellular matrix (ECM) and neoplastic progression in hamster with tongue cancer.
METHODSForty-eight specimens of hamster tongue cancer were divided into control group (n=6) and experimental group (n=42). The pathological grade of the specimens was assessed (including 3 stages, namely atypical hyperplasia, carcinoma in situ and early invasive carcinoma). The sections of the tongue were stained with Masson and aldehyde-fuchsin (AF) staining for microscopic observation of the elastic fiber and collagen fiber changes.
RESULTSWithin the connective tissue cores (CTC) of the papillae in the control group was a framework of numerous and fine Gomrori's aldehyde fuchsin-positive elastic fibers. But in the stages of dysplasia and carcinoma in situ, these elastic fibers decreased and further diminished in the CTC in early invasive carcinoma. In dysplasia and carcinoma in situ stages, most of the elastic fibers collapsed with scattered elastic fibers, and the elastic fibers decreased significantly in early invasive carcinoma. The control group showed a significantly greater number of elastic fibers in the experimental group. The collagen fiber was obviously increased and irregularly arranged in dysplasia and carcinoma in situ stage; in early invasive carcinoma, the collagen fibers became thicker with deposition in the lamina propria.
CONCLUSIONAn excessive deposition of collagen fiber and reduction of the elastic fibers is an important factor contributing to the development of tongue carcinoma in hamsters.

5.Revision of infected total hip replacement:a report of 30 cases

Objective To retrospectively study revisions for infected total hip replacements in 30 cases and discuss the bacteriological characters of the infected total hip replacements,difficulties and strategies in the revision.Methods Thirty revisions of infected total hip replacements were reviewed retrospectively.There were 12 males and 18 females,with mean age of 62.5 years(31-86 years)at revi- sion surgery.Infection was presented one month to four years(mean seven months)after THA operation. The diseases for initial operation included femoral neck fractures in 12 cases,femoral head necrosis in 11,hip osteoarthritis in five and rheumatoid arthritis in two.Twelve eases were treated by one-stage revi- sion and 18 by two-stage revision.Results Before the revision operation,the hip infection were diag- nosed by bacterial culture in 18 cases including five with Staphylococcus epidermidis,four with Staphylo- coccus aureus and nine with other bacteria.Bacteria growth appeared on the specimens from 23 hip joints during the revision surgery but not on the specimens from seven hip joints.Of 12 one-stage revisions,10 cases were followed for mean 16 months,which showed infection recurrence in two eases.Of 18 two-stage revisions,13 cases were followed for mean 20 months,which showed one case with infection recurrence. The mean Harris hip score was improved from preoperative 44 to 84 at follow-up.Conclusions 1) The main bacteria in the infected hip are antibiotic resistant Staphylococcus.2)Because the revision op- eration is difficult,careful preparation before revision is important for success.The fresh surgeon should not attempt.3)The revision strategies should vary according to specific status of the cases.The infection recurrence rate is lower when using a two-stage revision strategy.4)Application of antibiotic bone cement can help improve treatment effect and facilitate functional recovery of the joints.5)The scientific rehabil- itation after operation is very important to functional recovery.
6.Application of microsurgical technique in joint replacement

Objective To report the experience of the application of microsurgery in joint replace- ment.Methods There were 22 cases,10 cases with segmental acetabular defects treated with the pedicle sartorius muscle iliac bone grafting,5 cases with vascular repair following major vascular injury of extremity during operation,6 cases with neural repair following neural injury during operation,1 case with serious injury reconstruction by elbow joint replacement and free flap.Results The operations succeeded in 22 cases without any postoperative infection.The mean follow-up was 40.1 months (3-72 months) in 22 cases,in which the joint function improved and the operative result was satisfactory with no joint pain.Conclusion Microsurgical technique can reconstruct bone and tissue defect effectively in joint replacement.
7.Progress on Detection of Metals Ions by Functional Nucleic Acids Biosensor

Functional nucleic acids are natural or artificialnucleic acid sequences with specific functions and special structures. Special metal ions are essential trace elements for human health, but excessive metal ions will be harmful to human health. The functional nucleic acids are widely used in the detection of metal ions because of their advantages such as easy modification, low price, high stability and strong specificity. This review describes the function of functional nucleic acid and metal ions, mainly including cutting type, link type, metal ion-mediated base pairing, click chemical type, conformation change type, and other types. Then, biosensor of functional nucleic acid combined with different signal output is introduced. Finally, the research significance and problems of functional nucleic acid in metal ion detection are discussed, and the future development direction and application prospect of functional nucleic acid biosensor are prospected.
8.Development and genetic polymorphism of abnormal pronuclear zygotes after intracytoplasmic sperm injection.

OBJECTIVETo compare the development of abnormal pronuclear zygotes after intracytoplasmic sperm injection (ICSI) and analyze their genetic polymorphism.
METHODSFour hundred and ninety three abnormal pronuclear zygotes after ICSI were divided into three groups based on the number of pronuclei: 347 nonpronuclear oocytes, 71 monopronuclear zygotes and 75 multipronuclear zygotes. All of them were cultured in the medium of Vitrolife G5 series(TM). Sixteen short tandem repeats (STR) of seven blastocysts were then analyzed by ABI3100.
RESULTSThe cleavage rate of nonpronuclear group (25.4%) was lower than that of the others (P<0.01), the proportion of blocked embryos in nonpronuclear group (48.9%) was significantly higher than that of the others (P<0.05), but the blastocyst rate showed no significant difference in three groups (P>0.05). The genetic polymorphism of the 16 STRs showed that the blastocysts from the nonpronuclear and multipronuclear were diploid, and one of the blastocysts from nonpronuclear oocyte was Y-bearing.
CONCLUSIONThe zygotes with abnormal pronuclei after ICSI might have development potential, and the blastocysts from nonpronuclear oocytes and multipronuclear zygotes could be diploid.

9.Effects of patient age, number and quality of embryo transferred on the outcome of in vitro fertilization-embryo transfer.

OBJECTIVETo study the effect of patient age, the number and quality of embryo transferred on pregnancy outcome of in vitro fertilization-embryo transfer (IVF-ETs).
METHODSA retrospective study was performed in infertile women who underwent a total of 1800 cycles of IVF-ET and intracytoplasmic sperm injection (ICSI). The patients were divided into three age groups, namely<30 years group, 30-35 years group, and>or=35 years group. The clinical pregnancy rate and the multiple pregnancy rate were compared when 1, 2 or 3 embryos and 0, 1, 2 or 3 good-quality embryos were transferred.
RESULTSIn patients<30 years, no significant differences was found in the clinical pregnancy rate between 1, 2 and 3 embryos transfer groups; 2 and 3 good-quality embryos transfer resulted in similar pregnancy rate, which was significantly higher than that resulted from 0 and 1 good-quality embryo transfer. Multiple pregnancy was not found in 1 embryo transfer group. In patients aged 30-35 years, the pregnancy rate showed no significant differences not only between 1 and 2 embryos transfer groups, but also between 2 and 3 good-quality embryos transfer groups; multiple embryo transfer led to significantly increased multiple pregnancy rate. In patients aged>or=35 years, the transfer of 1, 2 and 3 embryos resulted in similar pregnancy rate; transfer of 3 good-quality embryos had obviously higher pregnancy rate than 0, 1 and 2 good-quality embryos transfer groups. Increased numbers of embryos and good-quality embryos transferred were both associated with increased multiple pregnancy rate.
CONCLUSIONOne good-quality embryo transfer in patients<30 years and 2 good-quality embryos transfer in patients>or=30 years can obtain ideal pregnancy rate and reduce the incidence of multiple pregnancy. For patients aged>or=35 years, transfer of only good-quality embryo is recommended.

10.Infection of Babesia in domestic animals in Western Yunnan Province, China

The objective of this study was to investigate Babesia infection among domestic animals in Western Yunnan Province and provide scientific evidence for developing control measures.A total of 1 073 domestic animals blood samples (274cattle,395 sheep,354 dogs,33 horses and 17 donkeys) were collected in 12 counties in Western Yunnan Province.Genomic DNA was extracted and a near full-length 18S rRNA gene sequence of Babesia was amplified by using nested PCR.Babesia species was identified by Blast program and phylogenetic tree.It was indicated that 50 samples were infected with Babesia,belonging to 5 species and with the infection rate of 4.66%.Among 274 cattle blood samples,11 were infected with Babesia (4.01%).Four of them were Babesia bovis and seven of them were Babesia bigemina.Among 395 sheep blood samples,38 were infected with Babesia (9.62 %),37 of them were Babesia odocoilei-like parasites and 1 of them was Babesia capreoli-like parasites.Horses and donkeys were negative.In conclusion,domestic animals in Western Yunnan Province are infected with many kinds of Babesia,which threaten stock raising development and human health.It is necessary to strengthen prevention of babesiosis and investigate infection rate of babesiosis in human.
