Objective To study the effects of Ginkgo biloba extract(EGb) on the volume of cerebral infarction and expression of caspase-3 in ischemic-reperfusion rats.Methods Setting up the brain ischemic and reperfusion model in rats by Nagasawa way.Both groups rats treated with small dose of EGb and large dose of EGb were injected intraperitoneally with EGb 50 mg/kg and 100 mg/kg at 30 min before the trial and 1 h after cerebral ischemic respectively.The infarction volume and caspase-3 expression were detected by TTC staining and immunohisto-chemistry.Then the results were compared with ischemic-reperfusion group.Results In groups treated with small dose of EGb and large dose of EGb,infarction volumes [(83.4?2.9)mm3,(37.0?2.5)mm3] were smaller obviously than that in the ischemic-reperfusion group [(138.8?6.1)mm3](all P

3.Clinical Study of Blood Glucose and Severity in Patients with SIRS

Objective To study the value of blood glucose changes and the severity of the disease in patients with the systemic inflammatory response syndrome(SIRS).Methods In 328 SIRS patients without diabetes,intravenous blood glucose was monitored before open intravenou channel and subsequently 3~8h after intervals.The blood glucose was reviewed if glucose was abnormal 24h later.Results The blood glucose was higher than normal in 287(87.5%)patients with SIRS,of which 44 cases were death(13.4%).The higher the blood glucose level was,the higher the mortality rate was.Conclusion Blood glucose of patients with SIRS was elevated.If the blood glucose was 3 times higher than the normal,patients with SIRS had a higher mortality rate.
4.Effects of early pregnancy exposure to hazardous indoor air pollutants due to interior decoration and hyperthermia on neurobehavioral development of postnatal rats

Objective To investigate the effects of early pregnancy exposure to hazardous indoor air pollutants due to interior decoration and hyperthermia on neurobehavioral development of postnatal rats.Methods Twenty-two pregnant rats were randomly divided into 4 groups.Each group was placed in pollutants cabinet,hyperthermia cabinet,hyperthermia+pollutants cabinet and empty cabinet respectively for 2 h per day for 10 d since their pregnancy.The weight and neurobehavioral development of postnatal rats were observed at different time points.The number of apoptotic and dead neurons in the CA1 region of hippocampus was observed in postnatal rats.The expression of the NR1 subunit of NMDA receptor of hippocampal neurons was also detected.Results Indoor pollutants and hyperthermia could accelerate apoptosis and necrosis of hippocampal neurons in CA1 region of postnatal rats and increase the expression of NR1 subunit of NMDA receptor.Furthermore,hazardous indoor pollutants and hyperthermia could induce remarkable influences on postnatal rats’ neurobehavioral development,learning and memory capability.Indoor pollutants combined with hyperthermia aggravated these effects.Conclusion Exposure to environment of the hazardous indoor pollutants especially combined with hyperthermia during early pregnancy has remarkable influences on the nervous system of postnatal rats.
5.THE PATHOLOGICAL CHANGES OF ULTRASTRUCTURE IN BRONCHIAL MUCOSA OF TYPE 2 DIABETIC PATIENTS

Objective To investigate the pathological changes of ultrastructure of mucosa in various bronchial segments from type 2 diabetic patients. Methods Sixteen cases of type 2 diabetic patients were selected,2-3 pieces of bronchial mucosa and submucosal tissue of the lesion were taken from various bronchi during bronchoscopy and these samples were observed under a transmission electron microscope. Results The basal lamina of bronchial capillary were diffusely thickened and mostly showed onion-skin like change,protein deposited around and mixed with basal menbrance;irregular highly electron dense materials were found to deposite around capillary,capillary lumen became narrow or even collapsed,neutrophilic leucocyte marginated in lumen and adhered with endothelium;protein deposited in the interstitial;endothelial cells and pericytes had dark cell changes.The cistern of rough endoplasmic reticulum dilated and vesicle formed.Conclusion Bronchial mucosa and its surrounding tissues show characteristic pathological changes of diabetes,bronchial is also the target organ of chronic diabetic damage.

Objective To investigate health behavior level and self-efficacy in patients underwent coronary artery stent implantation,and to discuss the relationship between health behavior and self-efficacy and to offer proof to the intervention strategy of health behavior.Methods HPLP Ⅱ,self-efficacy questionnaire were used as main tools.106 patients underwent coronary artery stent implantation were investigated.Statistics analyses were performed using the Statistical Package for Social Science(SPSS)13.0.Results (1)The score of total HPLPⅡwas 130.08±21.25,the range of that was 84~180.The health behavior level of47.2%patients was general or worse,and that of 52.8%patients was good or better.(2)The score of self-efficacy was 71.37±16.32,the range of thatwas 38~110.(3)There were significant positive relationship in the scores of total HPLP Ⅱ and the six elements,and self-efficacy.The correlation coefficient was 0.389~0.916(P<0.01).Conclusion The health behavior level of patients underwent coronary artery stent implantation was not in an ideal state.Self-efficacy has a positive effect on health behavior.The importance motivating factor should be attached to nursing practice and education by medical staff,and medical staff should establish and supervise to carry out corresponding intervention measure.

BACKGROUND:Self-assembling short-peptide has been shown to be a kind of hovel nanometer biomaterial.Following culture combined with chondrocytes,self-assembling short-peptide can promote eell marex secretion,cell division and proliferation.and eell phenotype maintenance.For this reason,it has become a candidate for all ideal cell scaffold used for cartilage tissue engineering.OBJECTIVE:To observe the biological characteristics of rabbit articular chondrocytes cultured in three-dimensional nanoscale self-assembling short.peptide KLD-12 hydrogel seaffold.DESIGN.TIME AND SETTING:An observational experiment was performed at the laboratory of Institute for Nanobiomedical Technology and Membrane Biology,Sichuan University between November 2007 and April 2008.MATERIALS:Nanoscale short-peptide KLD-12 with a sequence of Ac-KLDLKLDLKLDL-CONH2 was synthesized by Shanghai Biotech Company,China.METHODS:Chondrocytes were harvested from New Zealand rabbit articular cartilage by enzyme digestion.Following in vitro culture.chondrocytes of passage 3 were inoculated into 4 g/L self-assembling short-peptide KLD-12 solution at a density of 5x 109/L.Peptide gel formation was induced by phosphate buffered saline for three-dimensional culture.MAIN OUTCOME MEASURES:①Observation of chondrocyte morphology in the short-peptide hydrogel with inverted phase contrast microscope.②Detection of intracellular matrix secretion by toluidine blue staining and immunohistochemistry.③Measurement of mRNA expression levels of aggrccan,type Ⅱ collagen and type Ⅰ collagen in chondrocyte-seeded hydrogels by reverse transcription-polymerase chain reaction.RESULTS:①The viability of chondrocytes digested and isolated from rabbit articular cartilage was over 95%.④Chondrocytes cultured in three-dimensional short-peptide hydrogel maintained their round shape,proliferated actively,and aggregated in clusters.In addition.cartilage lacuna could be seen surrounding the clusters.③Both toluidine blue staining and type Ⅱ collagen immunohistochemistry results were positive.The mRNA expression levels of aggrecan and type Ⅱ collagen were increased with time.④Reverse transcription-polymerase chain reaction results revealed that after 3 weeks of in vitro culture in the short-peptide hydrogel,chondrocytes maintained the capability to secrete aggrecan and to express type Ⅱ collagen.CONCLUSION:Self-assembling short-peptide KLD-12 three-dimensional hydrogel Call well inainrain the normal cellular morphology,function and proliferative capability of chondrocytes
9.Immune hyporesponsiveness induced by immature dendritic cells prolonged rat kidney allograft survival

Objective To study the effect of immature dendritic cells (imDC) transfusion in combination with bone marrow transplantation (BMT) on rat kidney allograft survival and its possible mechanism. Methods Renal allograft from DA rat was transplanted to Lewis rat. Forty recipient rats were randomized into 5 groups: (1) Negative control group; (2) imDC group: Lewis rats accepted imDC transfusion in the amount of 2? 107 only; (3) BMT group: Lewis rats accepted bone marrow transplantation in the amount of 2?108 only; (4) imDC + BMT group: Lewis rats accepted both imDC and BMC; (5)Third party donor group: Lewis rats accepted renal allograft from Wistar rats. One-way MLR was performed to assay splenic cell proliferation to allogeneic T cells. Exogenous IL-2 was added at the outset of another group as the former one-way MLR. Normal Lewis rat accepted splenic cells from tolerant rat in the amount of 1?108. DTH was assayed in the trans-tolerance model. Cells from spleen and thymus were harvested from recipient rats for detecting chimerism by flow cytometry. 10.Application of Modified Pie-crusting Technique in Releasing Medial Tightness During Primary Total Knee Arthroplasty

Objective To analyze and evaluate the safety and efficacy of modified Pie-crusting ( PC) technique for releasing medial tightness during primary total knee arthroplasty ( TKA) . Methods We completed primary TKA by the same performer with modified PC technique in 30 patients (34 knees) with genu varus from March 2014 to June 2016.By using a special curved scalpel with width limit of 3 mm and depth limit of 5 mm to poke the tension parts , we released the anterion bondle of superficial medial collateral ligament ( sMCL) and posteromedial corner structures ( PMCS) during tension happened in extension , and we released the posterior bondle of sMCL during tension happened in flexion .According to the gap value measured intraoperatively , we divided these cases into three groups: extension with flexion tension group ( 10 knees ) , extension tension group ( 13 knees ) , and flexion tension group (11 knees).The difference between medial and lateral gap value no more than 1 mm was defined as gap balance.We calculated the gap balance rate of each group .Series of weighted frontal X-ray were conducted at fixed period to evaluate the varus angle of the knee postoperatively.The range of motion (ROM), HSS scores and WOMAC scores were also recorded at the same time . Results Among the 34 knees , 31 knees reached the medial and lateral gap balance at both extension and flexion .There was a difference of 2 mm in medial and lateral gap value at extension in 1 knee and the same difference at flexion in the other 2 knees.The total postoperative gap balance rate was 91.2%( 31/34 ).The constrained inserts were implanted in 3 cases.No technical-related complications happened after the surgery.After the releasing procedure, the flexion gap value had an increase of 1 mm (range, 1-3mm) in the extension tension group , and the extension gap value had an increase of 1 mm ( range, 1-2 mm) in the flexion tension group, without significant difference (Z=-1.118, P=0.264).The ROM was 83.3°±14.7°preoperatively and 100.7°±14.2° postoperatively (t=-7.714, P=0.000).The median alignment of the knee was 11.5°(range, 7°-32°) preoperatively and 1° (range, 0°-4°) postoperatively (Z=-5.092,P=0.000).The HSS scores were (42.7 ±16.3) points preoperatively and (88.1 ± 9.9) points postoperatively (t=-21.868, P=0.000).The WOMAC scores were (76.2 ±8.2) points preoperatively and (11.4 ± 9.7) points postoperatively (t=31.726, P=0.000).All of them were significantly improved in comparison with those before the surgery. Conclusions Using modified PC technique is safe and effective in medial releasing during primary TKA .Both extension and flexion gap value will be affected by releasing tensed fiber at extension or flexion position .
