1.Percutanenous intrapulmonary injection of IL-2 plus radiotherapy for luug cancer

Objective To evaluate the effects of percutanenous intrapulmonary injecti0n 0f IL-2 plus radiotherapy for lung cancer. Methods Twenty-two patients with inoperable non small cell lung cancer received percutanenous intrapulmonary injection of 100U ? 6 in 6 weeks; and radiation dose was 60 Gy/6wks. Results The total effective response rate (CR+PR) was 95. 4 % with the complete response rate (CR) of 40. 9 %. Couclusion Percutanenous intrapulmomary injection of IL-2 plus radiotherapy could improve the control rate of the patients with lung cancer.
2.Effect of miR-4698 mediated GALNT4 expression on proliferation and migration of hepatocellular carcinoma cells

Objective:The relative expression of miR-4698 in liver cancer tissues and cell lines was detected, and its effect on the proliferation and migration of liver cancer cells and its molecular mechanism were analyzed.Methods:Real-time fluorescence quantitative polymerase chain reaction (qRT-PCR) was used to analyze the relative expression of miR-4698 in liver cancer tissues and liver cancer cell lines. Among the lowest-expressing hepatocellular carcinoma cell lines, miR-4698 mimic and control mimic were transfected with liposome transfection method, and named as experimental group and control group. qRT-PCR was used to detect transfection efficiency. Cell counting kit-8 (CCK-8) and transwell migration experiments were used to detect the effects of overexpressing miR-4698 on the proliferation and migration ability of liver cancer cells. Bioinformatics and dual luciferase reporter gene experiments were used to predict and verify the binding of miR-4698 to target gene. qRT-PCR and Western blot were used to detect the relative expression of target gene at mRNA and protein levels, respectively.Results:Compared with the adjacent tissues, miR-4698 was significantly lower in the liver cancer ( P<0.01). Compared with normal hepatocytes, miR-4698 was significantly lower in hepatoma cell lines ( P<0.05), and the lowest in Huh7 cells ( P<0.01). After transfection, the expression of miR-4698 in Huh7 cells in experimental group was significantly increased compared with that in the control group ( P<0.01). Overexpression of the miR-4698 can inhibit the proliferation and migration of Huh7 cells ( P<0.05). Bioinformatics showed that the target gene of miR-4698 was polypeptide-n-acetylgalactosamine transferase 4 (GALNT4), and the double luciferase reporter gene confirmed that miR-4698 could bind to GALNT4 ( P<0.01). qRT-PCR showed that overexpression of miR-4698 could inhibit the expression of GALNT4 gene ( P<0.01). Western blot results showed that overexpression of miR-4698 decreased the protein expression of GALNT4, cyclin B and CDK1, and increased the protein expression of N-cadherin and ZEB-2. Conclusions:The expression of miR-4698 in liver cancer tissues and cell lines is significantly reduced. Overexpression of miR-4698 can inhibit the expression of GALNT4 gene and reduce the proliferation and migration ability of liver cancer Huh7 cells.
3.Interference of Hepatocyte Growth-Promoting Factor on Cell Apoptosis in Kidney of Rats with Renal Ischemia Reperfusion Injury

Objective To explore the effects of hepatocyte growth-promoting factor(pHGF) on renal function and cell apoptosis in kidney of rats with renal ischemia reperfusion injury(IRI).Methods Thirty-two male Sprague-Dawley rats were divided into 4 groups:sham-operated control group(groupⅠ),renal ischemia reperfusion control group(groupⅡ),one experimental group injecting pHGF(50 mg/kg,intraabdominal injection) before renal IRI(group Ⅲ),and another experimental group injecting pHGF(50 mg/kg,intraabdominal injection) after renal IRI(group Ⅳ).The animals with renal IRI exposed to 45 min bilateral renal pedicle clamping.All ischemia reperfusion rats in group Ⅰ and Ⅱ were intraabdomially injected equal volume of physiological saline(0.8 mL) at the time when the rats in experimental groups were administered 50 mg/kg pHGF.Twelve hours after IRI,samples for serum and the left renal tissue of each animal were taken.The serum sample was used to detect expression of serum creatinine(Scr),and the renal tissue sample for evaluation of apoptosis.Results Compared with the level of Scr in groupⅠ(22.775?6.508) ?mol/L,Scr was markedly higher in groupⅡ(120.850?22.237) ?mol/L(P0.05).Conclusions The laboratory investigation suggests that pHGF might be an effective pharmacological agent against renal IRI according to the findings of the evaluated parameters,and protective effect by pHGF against renal IRI might involved in the mechanisms decreasing tubular cells apoptosis.It is likely that pHGF is a potential therapentic agent in clinical renal IRI circumstances.
4.Parents′ experiences of caring for children with chronic kidney disease: a qualitative Meta-synthesis

Objective:To systematically review parents ′ experience of caring for children with chronic kidney disease (CKD) to fully understand care needs and improve the psychological state and caregiving quality of parents. Methods:The qualitative studies on parents ′ experience of caring for children with CKD were retrieved from following databases, including PubMed, Web of Science, CNKI, VIP, CBM, and WanFang Data from inception to March 2020. The quality of included researches was evaluated according to the JBI Critical Appraisal Tool for qualitative studies in Australia. The results were integrated by pooled integration methods. Results:A total of 14 studies were included. 69 results were summarized and integrated to form 10 categories. These categories extracted 4 integrated results: parents' physical and mental condition are affected, relationship between parents and their support system has changed, reconstruction of parents ′ life, unmet care needs and problems. Conclusions:Parents are crucial to the disease management of children with CKD, so clinical medical staff should not only provide medical services for children, but also pay more attention to the psychological status and needs of parents, so as to provide guidance and support to promote parents to better implement care and disease management for children.

Objective To investigate the clinical significance of retinal binding protein (RBP) and Cystatin C in blood and urine in patients with chronic kidney disease (CKD).Methods One hundred and twenty-one patients with CKD in Nephrology Laboratory of The People's Hospital of Xinjiang Uygur Autonomous Region from Aug.2012 to Jan.2013 were served as the case group.Sixty healthy check-up people were considered as control group.Enzyme linked immunosorbent assay was used to detect RBP and Cystatin C in urine,and the immune turbidimetry testing was applied to detect RBP and Cystatin C in blood.The receiveroperating characteristic (ROC) curve and area under curve (AUC) were applied to evaluate the sensibility and diagnostic value of indexes in CKD.Results The positive rate of RBP,Cystatin C in urine and blood were 89.6%,92.6%,52.5%,59.5% respectively of 12 patients in case group,higher than those in control group (all value were 0,P <0.001).ROC curve showed that the sensitivity of urine RBP and Cystatin C were higher than that in blood of case group.AUC of RBP,Cystatin C level in urine and blood were 0.915,0.974,.655,and 0.623 respectively.And 95% confidence interval were 0.877-0.954,0.956-0.992,0.575-0.736,0.543-0.702 respectively,and the differences are statistically significant (P < 0.001).Conclusion The sensitivity of urine cystatin C and RBP are significantly higher than that in blood,which might be served as a diagnostic marker of CKD and can provide important basis for clinical diagnosis.
7.Study on discrimination mode of cold and hot properties of traditional Chinese medicines based on biological effects.

OBJECTIVETo observe the effect of cold or hot properties of traditional Chinese medicines (TCM) on biological effect indexes, and analyze the contribution of variables on cold or hot properties, in order to preliminarily establish the discrimination mode for the biological effects of cold or hot properties.
METHODRats were randomly divided into the blank control group, cold TCM groups (Coptidis Rhizoma, Scutellariae Radix, Phellodendri Cortex, Gardeniae Fructus, Sophorae Flavescentis Radix and Gentianae Radix) and hot TCM groups (Aconiti Lateralis Preparata Radix, Zingiberis Rhizoma, Alpiniae Officinarum Rhizoma, Zanthoxyli Pericarpium, Cinnamomi Cortex and Evodiae Fructus), and orally administered with 10 mL x kg(-1) of corresponding TCM water decoctions for 30 d, twice a day. Altogether 53 biological effect indexes correlated to cold or hot properties of traditional Chinese medicines were founded by searching literatures. The data warehouse were established by using data-mining software Clementine12.0. Data of the blank control group, cold TCM groups (Coptidis Rhizoma, Phellodendri Cortex, Gardeniae Fructus, Sophorae Flavescentis Radix, Gentianae Radix) and hot TCM groups (Aconiti Lateralis Preparata Radix, Zingiberis Rhizoma, Alpiniae Officinarum Rhizoma, Zanthoxyli Pericarpium, Cinnamomi Cortex) were selected into a training set. C5.0 algorithm and C&R classification and regression algorithm were adopted to define the importance of variable, create the decision trees, and test hot or cold properties of Evodiae Fructus and Scutellariae Radix.
RESULTAccording to C&R classification and regression algorithm, SDH activity of livers was the most important hot or cold property, with the significance closed to 30%. It was followed by triglyceride, liver Na' -K' -ATPase enzyme, muscle glycogen and platelet distribution width, with the accuracy up to 97.39% in models. C5.0 algorithm showed that liver SDH activity was the most important hot or cold property, with the significance closed to 40%. It was followed by triglyceride, GOT, muscle glycogen and liver Na(+)-K(+)-ATPase enzyme, with the accuracy up to 98.26% in models. The possibilities that Evodiae Fructus is in hot property and Scutellariae Radix is in cold property were 100. 00% and 77.78% by using both C&R classification and regression algorithm and C5.0 algorithm.
CONCLUSIONThe SDH activity of liver is the most important biological effect index to distinguish cold and hot properties of TCMs. The discrimination pathway or mode of cold and hot properties is closely related to energy metabolism.

8.Tilting the balance of tubular cell fate toward survival contributes to ischemic tolerance in kidney

Objective To explore the role of brief ischemia pretreatment in the induction of renal ischemic tolerance,and investigate its effects on tubular cell necrosis,apoptosis and proliferation. Methods Male Sprague-Dawley rats were randomly divided into three groups,including sham-operated group (Sham),ischemia/reperfusion injured group subjected to theocclusion of both renal pedicles for 40 min followed by reperfusion(I/R),and preconditioned group with 20-min ischemia pretreatment induced 4 days before I/R(IPC).Histological changes were evaluated by PAS staining.The ultra-structure of tubular cells was observed by transmission electron microscopy(TEM).Apoptosis was confirmed by terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-biotin nick end labeling (TUNEL).The proliferation of tubular cells was evaluated with proliferating cell nuclear antigen(PCNA). Results Twenty-minites ischemia pretreatment offered both promising functional and histological protection against 40-min ischemia/reperfusion injury (P<0.01).The mortality rate wag reduced from 33%in I/R group to 0 in IPC group.The renopmtection offered by 20-min ischemia pretreatment was accompanied with reduced postischemic tubular cell apoptosis and necrosis (P<0.05), and increased cell proliferation (PCNA positive) (P< 0.01). Conclusions Brief and sublethal prior ischemia can render the kidney more tolerant to subsequent prolonged I/R injury. Its ability to tilt the balance of tubular cell fate toward survival, reducing postischemic cell death and enhancing cell proliferation, may play an important role in renal protection of ischemic preconditioning.
9.Variance analysis of contrast sensitivity for amblyopia and normal eye based on spatial frequency channels

Background Contrast sensitivity (CS) of amblyopia has been extensively studied,but its relationship with spatial frequency channels needs further research. Objective The purpose of this study was to investigate the reasons of the CS deficits in amblyopia through comparing the differences in CS between amblyopic and normal eyes from the point of view of spatial frequency channels. MethodsThe CS values of 166 normal children eyes and 143 amblyopic children abnormal eyes were measured by adopting OPTEC 6500.Then,spatial frequency channels' tuning curves were derived via principal component analysis and non-orthogonal rotation.Also,numbers and bandwidths of channels were calculated using the method of full width at half maximum ( FWHM ).All of these were used to analyze the variations of CS between amblyopia and normal eyes by comparing the numbers and the bandwidths of the channels.The reliability of spatial frequency channel was verified by a cross-validation study of 43 amblyopic children. ResultAt spatial frequency of 1.5,3.0,6.0,12.0,18.0 cpd,the mean of CS of amblyopia were 36.35±21.40,50.33 ± 33.46,46.88 ± 41.72,16.24 ± 17.26,4.67 ± 5.79,and the mean of CS of normal eyes were 49.49±24.69,87.23±40.87,93.18±51.99.36.63±24.72,15.70±13.87,with the rank-sum test results were H =27.83,66.61,68.34,78.23,89.88,P<0.05.There existed three spatial frequency channels in both amblyopia and normal eyes.At the peaks of 3.0,6.0 and 12.0 cpd,the bandwidths of normal eyes were 1.03,1.02 and 0.99 octaves,and the bandwidths of amblyopia were 1.04,1.01and 0.73 octaves.Conclusions The reduction in bandwidths of the corresponding spatial frequency channels may cause the CS deficits in amblyopia.
10.Expression of Recombinant Nematode Anticoagulant Peptide in Pichia Pastoris

Objective: To acquire recombinant nematode anticoagulant peptide ( NAP) with high anticoagulant activity. Methods: Pichia pastoris GS115 strain was transformed with recombinant yeast expression vector pPICS. 5K-rNAP. Expression of rNAP was induced with methanol after the identification of positive strains. NAP expressed in the collected yeast culture supernatant was confirmed with SDS-PAGE and Western blot. The biological activity of the products was validated with PT (prothrombin time) , INR (international normalized ratio) and APTT (activated partial thromboplastin time) , respectively. Results: The yeast strains expressing NAP were identified. The rNAP was secreted into culture supernatant with a molecular weight of about 10 kDa due to glycosylation, which is a little bigger than that predicted (8.7kDa). The anticoagulant efficiency of rNAP was confirmed with the in vitro assays. Conclusion: The recombinant nematode anticoagulant peptide with high biological activity was successfully expressed in Pichia pastoris and can be used in the future development of novel anticoagulant agent.
