3.Effect of aplysin on the proliferation and apoptosis in human gastric cancer cell SGC-7901

Aim To investigate the effects of Aplysin on the inhibition of gastric cancer cell in vitro .Methods MTT assay was used to examine the inhibition of gastric cancer cell 1ine SGC-7901 by Aplysin in different concentrations and at different times.The morphologic changes and the apoptosis of SGC-7901 was observed by inverted microscope and Hematoxylin-Eosin(HE)staining.Reverse transcriptase polymerase chain reaction(RT-PCR)assay was used to detect the changes of COX-2 mRNA expressions.Results Aplysin could decrease the proliferation significantly in a dose-dependent manner in SGC-7901 cells.When treating SGC-7901 with Aplysin in concentration of 120, 240 mg·L~(-1) for 24 h, the growth of the cell was obviously inhibited observing by inverted microscope.Aiso, when treating with the same concentration for 18 h, its chromatin became crimpled and breakdown, as well as cell shrinkage and apoptotic bodies formation when using HE staining.The apoptotic rates(%)of SGC-7901 was(15.0±2.12)%, (18.4±2.3)%, respectively, which was significantly higher than(1.4±0.55)% that in control group(P <0.01).60、120、240 mg·L~(-1) Aplysin could not effectively inhibited the mRNA expressions of COX-2(P >0.05).Conclusions Aplysin can inhibit the proliferation and induces apoptosis of SGC-7901 cells.
4.Chromosomal structural features of Yersinia pestis isolated from China

Objective To study the differences of chromosomal structure among Yersinia pestis strains isolated from China,and to investigate the reasons of chromosomal rearrangement events occurred in Yersinia pestis as well as the possibility of strain identification and phylogenetic analysis based on the chromosomal rearrangement features.Methods According to the genome sequence data downloaded from web of National Center for Biotechnology Information (http://www.ncbi.nlm.nih.gov/genome),alignment of all the coding sequences (CDSs) among five strains(American strain CO92 as reference and other four completely sequenced strains from Inner Mongolia,Jianchuan of Yunnan,Yulong of Yunnan,Naqu of Tibet in China named 91001,D182038,D106004 and Z176003 as comparison strains) was performed,and then the chromosome of Yersinia pestis was divided into several large DNA segments (named chromosomal plate in the text) according to the similarity of CDSs.Plate arrangement patterns in each strain' s chromosome and gene content of breakpoint regions were determined.Finally,genetic relationships among Yersinia pestis strains were analyzed on the basis of rearrangement diversity from paired-comparison.Results Yersinia pestis chromosomes of strains CO92,D182038,D106004,91001 were composed of 44 relatively independent plates,except strain Z176003.Gene order was very stable within each plate,while it was movable between the plates.Comparing with the reference strain CO92,13 rearrangement events occurred in the chromosomes of both strain D182038 and strain D106004,and 14 rearrangement events involved in Z176003,while 37 rearrangement events occurred in 91001.Paired-comparison data showed that only 8 plates order differences were existed between D106004 and Z176003.Forty-three breakpoint regions were identified on the chromosome of strain CO92,and 39 of them contained insertion sequences,and 25 of them were IS100.Conclusions Yersinia pestis genome represents a high degree of genetic flux,and chromosomal structures of strains are significantly different from each other.Chromosomal rearrangement events is closely related to the large number of insertion sequences in the Yersinia pestis chromosome.Rearrangement diversity among Yersinia pestis strains could reflect their genetic relationships.
5.The Diagnosis and Treatment of 64 Children with Ovarian Torsion

Objective To evaluate the clinical features and key points in treatment of ovarian torsion in children. Methods The clinical data of 64 children with ovarian torsion, hospitalized in our hospital from January 2005 to October 2012, were analyzed retrospectively. The clinical manifestations in children of all ages were summarized. The anal examina-tion, B ultrasound examination, CT examination were used for the early diagnosis of ovarian torsion. All children were per-formed laparoscopic surgical exploration. All children were found the typical lower abdominal cramps. The B ultrasound and CT examination showed non-homogeneous mass in pelvic, which suggested the early diagnosis of ovarian torsion. The detec-tion rate of anal examination reached 70.3%(45/64). Results All patients were performed laparoscopic surgical explora-tion. Five patients were treated with ovariectomy. The rest were treated conservatively. The postoperative follow-up was good. Conclusion Ovarian torsion in children was likely to be misdiagnosed. Doppler, CT, anal examination and laparo-scopic operation were important for the early diagnosis and treatment of ovarian torsion.
6.Health economic evaluation of Chinese population-based screening and opportunistic testing strategies to prevent Helicobacter pylori-related diseases

Objective:To conduct health economic evaluation of Chinese population-based screening and opportunistic testing strategies for the prevention of Helicobacter pylori ( H. pylori)-related diseases. Methods:The Markov models of H.pylori infection caused non-ulcer dyspepsia, peptic ulcer (PU) and gastric cancer were established. The cost and effects of the population-based screening, opportunistic testing, and non-intervention strategies in 100 000 population of our country were simulation calculated. Wilcoxon signed rank test was used for statistic analysis. Single factor sensitivity analysis was performed to analyze the impact of single parameter uncertainty on cost-effectiveness. Probability sensitivity was used to analyze the impact of common uncertainty of all parameters on cost-effectiveness. Results:Compared with opportunistic testing strategy and non-intervention strategy, the total cost of population-based screening strategy was reduced by 0.43 million yuan(-6.63 million yuan to 7.19 million yuan) and 4.45 million yuan(-8.60 million yuan to 27.93 million yuan), quality adjusted life years (QALY), life years and asmptomatic months were prolonged by 888.00 (479.86 to 1 574.10) and 3 032.78 (1 756.04 to 5 007.84), 651.82 (294.73 to 1 211.94) and 1 868.64 (1 045.88 to 3 148.34), 28 381.91 (19 109.54 to 43 736.72)and 102 537.13 (58 649.18 to 176 868.77) respectively, gastric cancer, deaths from gastric cancer, deaths from PU and deaths from various causes were reduced by 115 cases(52 cases to 232 cases) and 464 cases(266 cases to 803 cases), 74 cases(33 cases to 148 cases) and 260 cases(149 cases to 453 cases), 46 cases(25 cases to 72 cases) and 369 cases(210 cases to 710 cases), 80 cases(42 cases to 126 cases) and 501 cases(331 cases to 772 cases) respectively, and the differences were statistically significant ( Z=1.99, 10.54, 27.39, 27.39, 27.37, 27.39, 27.39, 27.39, 27.21, 27.28, 27.22, 27.28, 27.38, 27.39, 27.39 and 27.39, all P<0.05). Compared with the non-intervention strategy, the opportunistic testing strategy reduced the total cost by 3.89 million yuan(-2.79 million yuan to 18.18 million yuan), QALY, life years and asymptomatic months were prolonged by 1 764.77 (888.70 to 3 406.62), 1 044.16 (524.46 to 1 912.75) and 62 568.69 (30 054.07 to 129 892.76) respectively, gastric cancer, deaths from gastric cancer, deaths from PU and deaths from various causes were reduced by 311 cases(164 cases to 563 cases), 164 cases (87 cases to 301 cases), 325 cases(162 cases to 644 cases) and 399 cases(234 cases to 684 cases) respectively, and the differences were statistically significant ( Z=14.20, 27.39, 27.38, 27.39, 27.25, 27.25, 27.39 and 27.39, all P<0.01). The results of singlefactor sensitivity analysis showed that the uncertainty of any parameter did not affect the cost-effectiveness of three strategies. The results of probability sensitivity analysis indicated that the probability of cost-effectiveness advantage of the population-based screening strategy was 100% and the common uncertainty of all parameters had no effect on cost-effectiveness. Conclusions:For preventing H.pylori-related diseases, the population-based screening strategy has the lowest average cost and the best average effect. However, non-intervention strategy has the highest average cost and the worst average effect. Population-based screening strategy is worthy of promotion throughout China, and opportunistic testing strategy has a higher clinical practical value.
7.Metabolic profile and insulin resistance in patients with OSAS

8.Effects of motilin on potassium and calcium currents of rat's proximal colon smooth muscle cells

Objective To investigate the effects of motilin on the voltage dependent potassium channel and L-type calcium channel currents in rat proximal colon smooth muscle cells (PCSM) and to explore its mechanism in increasing colonic motility.Methods PCSM were isolated by collagenase.The voltage dependent potassium channel transit outward current (IKA ) and delayed rectifier current (IKdr) and L-type calcium currents (ICa(L)) were measured by whole cell patch clamp technique.Groups were analyzed by paired t-test.Results There was no significant effect of motilin on IKA and IKdr.L-type calcium channel was dose-dependently activated by motilin from 0.5 × 105 mmol/L to 10.0 ×10-5 mmol/L.At 6 × 10-5 mmol/L motilin and under - 10,0 and 10 mV stimulating voltage,maximum current density increased by 154.61%,62.69% and 21.02% respectively and activation kinetics curve obviously left shifted.Half activation voltage decreased from (2.740±1.211) mV prior administration to ( - 25.290 ± 0.614) mV (t =8.534,P =0.007 ) and there was no significant difference in slope factor. Conclusions Motilin increases colonic smooth muscle contraction by promoting calcium influx. However the frequency of colonic smooth muscle contraction could not change with frequency of equilibrium potential and action potential of colonic smooth muscle.
9.Quality Standard for Longdan Xiegan Capsule and Longdan Xiegan Soft Capsule

Objective To establish the quality standards of Longdan Xiegan Capsule and Longdan Xiegan Soft Capsule. Methods Gentianae Radix et Rhizoma, Bupleuri Radix, Scutellariae Radix, Gardeniae Fructus, Alismatis Rhizoma, Angelicae Sinensis Radix, Rehmanniae Radix and Glycyrrhizae Radix et Rhizoma were identified by TLC. Gentiopicrin in Gentianae Radix et Rhizoma, geniposide in Gardeniae Fructus, baicalin in Scutellariae Radix were determined by HPLC. Results The TLC spots developed were clear. Gentiopicrin showed good linear relationship in the range of 0.066 1-0.595 1 mg (r=1.000 0), the average recovery of Capsule was 99.34%(RSD=1.50%), and the average recovery of Soft Capsule was 96.62%(RSD=1.50%). Geniposide showed good linear relationship in the range of 0.076 1-1.369 8 mg (r=0.999 9), the average recovery of Capsule was 101.3% (RSD=1.70%), and the average recovery of Soft Capsule was 100.59%(RSD=0.79%). Baicalin showed good linear relationship in the range of 0.214 4-1.608 mg (r=1.000 0), the average recovery of Capsule was 101.12% (RSD=1.30%), and the average recovery of Soft Capsule was 98.07% (RSD=2.40%). Conclusion The method is simple and reproducible. It can be used to control the quality of Longdan Xiegan Capsule and Longdan Xiegan Soft Capsule.
10.Individualized function evaluation and monitoring by blood biochemistry and brain function monitoring in athletes

BACKGROUND:Due to the differences of sport events and individual metabolic characteristics of athletes, it is difficult to establish uniform biological indexes for training monitoring. Current individual evaluation is a longitudinal analysis relying on experience or numerical variation width, which is less objective.OBJECTIVE: To explore the individual function assessment and monitoring by monitoring blood biochemical indexes and brain function indexes in elite gymnasts, in order to accurately reflect the body changes resulting from training loads. METHODS:Thirty gymnasts from the Chinese national gymnastics team preparing for London Olympic Games were enroled in this study and monitored from the last winter training until the London Olympic Games. The blood biochemical indicators and brain function indicators were measured and assessed individually according to according to the principle of quality control (alert value=mean±SD, and controled variable=mean±2SD). RESULTS AND CONCLUSION:Under the relatively uniform test conditions, it is feasible to individualy assess the blood biochemical and brain function indexes of elite gymnasts in accordance with the principle of quality control, which is more accurate and objective to reflect the body changes under training load and the current state of athletes. In addition, the combined monitoring of blood biochemical indexes and brain function indexes is more comprehensive to evaluate the body function and status of elite gymnasts.
