1.Role of intercellular gap junction in lung injury induced by hemorrhagic shock and resuscitation in rabbits

Objective To evaluate the role of intercellular gap junction in lung injury induced by hemorrhagic shock and resuscitation in rabbits. Methods Twenty-four healthy rabbits of both sexes weighing 1.5-2.2 kg were used in this study. The animals were anesthetized with iv 3% pentobarbital 30 mg/kg,tracheostomized and mechanically ventilated. Femoral artery was cannulated for MAP monitoring, blood-letting.Right external jugular vein was cannulated for fluid infusion. Hemorrhagic shock was induced according to the method described by Wiggers. MAP was maintained at 35-40 mm Hg for 60 min. The animals were then randomly divided into 2 groups (n = 12 each):traditional treatment group (group A) and octanol group (group B). Both groups received rapid iv infusion of lactated Ringer's solution (LR solution) 1.5 ml·kg-1 ·min-1 for 30 min (T3 ).In group B 99.5% octanol (a specific gap junction inhibitor) 5 mmol/kg was injected intraperitoneally (IP) in addition to iv LR solution infusion. Thirty minutes later the animals were resuscitated with infusion of the blood withdrawn and LR solution (the volume was equal to the volume of blood loss). Then LR solution was infused iv at 2.5 ml· kg-1 · h-1 for 150 min ( T4 ). Left ventricular systolic pressure (LVSP) and HR were recorded before blood letting (T1), immediately after successful establishment of the model (T2), and at T3 and T4. The mortality during resuscitation was calculated. The animals were killed at T4. Blood samples were taken from femoral artery for determination of the serum protein concentration. The lungs were removed. The left lower lobe was lavnged. The protein concentration in broncho-alveolar lavage fluid (BALF) was measured. Lung permeability index (LPI) was calculated (LPI = protein concentration in BALF protein concentration in serum). Na+-K+-ATPase and Ca2+-ATPase activities and W/D lung weight ratio were determined and microscopic examination was performed. Results HR was significantly lower at T3 and T4 and LVSP higher at T4 in group B than in group A. The pulmonary permeability index, W/D lung weight ratio and mortality were significantly lower and the activities of Ns+-K+-ATPase and Ca2+ -ATPase were significantly higher in group B than in group A. The lung tissue danage was significantly ameliorated in group B. Conclusion Intercellular gap junction is involved in the development of lung injury induced by hemorrhagic shock and resuscitation in rabbits.
2.Analysis of the hope level and its related factors in patients with high-risk gestational trophoblastic neoplasm

Objective To study the hope level in patients with high-risk gestational trophoblastic neoplasm and to explore its related factors.Methods 83 cases of patients with high-risk gestational trophoblastic neoplasm were collected and investigated with Herth Hope Scale,Social Support Rating Scale (SSRS) and self-designed questionnaire.The results underwent analysis.Results The hope level of most patients underwent investigation was at the moderate level,among which the positive attitude for future and reality scored (10.80 ± 1.77),adopting positive action scored (10.30 ± 2.36),keeping close relationship with others scored (9.63 ± 2.13),the total scored (30.74 ± 4.82).The hope oevel was positively correlated with rigor personality (r=0.254),but negatively correlated with neuroticism (r=0.237),the difference was statistically significant.Conclusions The hope level of patients with high-risk gestational tropoblastic neoplasm is above the moderate level and is correlated with many factors.Doctors and nurses should evaluate carefully enough and take nursing measures to improve the hope level and life quality.

5.Diffusion-weighted MRI in residual foci evaluation of uterine cervical cancer after chemoradiation therapy

Objective To evaluate the usefulness of diffusion-weighted MR combined with routine T2 WI in finding the possible residual foci in uterine cervical cancers after radical chemo-radiation therapy.Methods This was a retrospective study including 25 consecutive cervical cancer patients who received hysterectomy after radical chemo-radiation therapy.All of them underwent MR examinations post-chemoradiation and just before operation.Images of T2 WI alone and those of T2 WI combining DWI were evaluated respectively by 2 senior radiologists,in order to decide whether there were residual tumors.ADC values were also measured.Taking the post-operation pathological results as the gold standard,the accuracies,sensitivities and specificities of T2 WI alone,T2 WI combining DWI,and ADC values were all calculated.Results In those 25 patients,9 were found with foci of residual cancer in operative pathology,while no cancer cells were found in the other 1 6 patients.The accuracy,sensitivity and specificity in finding the positive residual cancer using T2 WI alone were 56.0%,77.8% and 43.8%,comparing with 72.0%,66.7% and 75.0% in T2 WI combining DWI.The accuracy and specificity increased with statistical significance after combining DWI (P =0.01 6 for accuracy,P =0.031 for specificity),while the sensitivity decreased but did not reach statistically significant level (P =0.099).No difference in ADC values was found.Conclusion DWI can be used as a supplementary sequence in finding the existence of residual tumors of cervical cancer after radical chemo-radiation therapy.Routine T2 WI combing DWI increased the specificity and accuracy,but still facing the risk of decreasing sensitivity.

7.The effect of oxidative stress on obstructive sleep apnea-hypopnea syndrome combined with hypertension.

OBJECTIVE:
To detect the levels and effects of protective bioactive substances such as NO, SOD and GSH in patients with obstructive sleep apnea syndrome (OSAHS) combining hypertension.
METHOD:
To collect the data from 99 snoring patients, which were divided into control group, OSAHS group, and OSAHS combined with hypertension group. The serum levels of NO), SOD and GSH in these patients were detected perioperatively, correlation analysis were carried out between these parameters.
RESULT:
The levels of NO, SOD and GSH in control group, OSAHS group and OSAHS combined with hypertension group reduced in turn with statistically significance, and had inverse correlations with apnea hyponea index (AHDI) and lowest saturation of blood oxygen. After operation, the levels of NO), SOD and GSH were increased and the blood pressure was decreased.
CONCLUSION
The oxidative stress state is serious in patients with OSAHS. The oxidative stress process is more severe in patients with hypertension, which may play a role in the devolepment of hypertension in OSAHS patients.

8.Retrospective analysis on 222 patients with pancytopenia

Objective To investigate the patients′cell morphology characteristics in peripheral blood and bone marrow with the reduce of leukocyte,hemoglobin and platelet in peripheral blood,and analyze the common cause.Methods From June 2005 to Feb-ruary 2011,222 patients with pancytopenia treated in the hospital were enrolled in the study,whose peripheral blood and bone mar-row smears were stained by Wright,combined with histochemical staining and the clinical data of patients,the disease types were analyzed.Results In the 222 patients with pancytopenia,patients with hematopoietic system disease accounted for 84.65% (188/222),non-hematopoietic system disease accounted for 15.35%(34/222),the difference was statistically significant(P <0.05 ).In 150 patients whose peripheral blood smears were obseved,58% patients obtained positive results.Conclusion The diseases of hem-atopoietic system are the common cause of pancytopenia,but can not ignore the non-hematopoietic system diseases.Peripheral blood smear has a high value in the diagnosis of these diseases.
9.An experimental study of bio-polymer composite film as a micro-skin autograft covering

Objective To explore the promoting effects of bio-polymer composite film as a micro-skin auto-graft covering on wound healing. Methods The full thickness skin defect models were made on both sides of 30 experimental rabbits. Then, the rabbits were randomly divided into experimental group and control group. In the for-mer group, the side was covered with chitosan/glucomannan composite membrane and in the latter, the side cov-ered with acelluar porcine skin after micro-skin autograft. We obtained wound tissues at week 1 , 2, 3, 4 and 5 af-ter operation. The conditions of wound healing were observed, the rate of wound healing was calculated, HE stain-ing was made, and PCNA and CD31 were detected by immunohistochemistry. Results (1) During 2~4 weeks af-ter operation, the rate of wound healing in the experimental group was significantly higher than that of the control wound (P<0.01). (2) The amount of neutrophil in experimental group was less than that of the control after oper-ation. (3) During 1 ~ 2 weeks after operation, the expression of PCNA in the experimental group was higher than that of the control group (P < 0.01), but lower than the control wounds during 1 ~ 2 weeks after operation (P <0.01). (4) During 1 ~ 5 weeks after operation, the expression of CD31 in the experimental group was higher than that of the control group (P < 0.01). Conclusion Chitosan/glucan-mannan composite membrane as a micro-skin autograft covering may promote wound healing.
