1.Aorta Imaging with Multi-slice Spiral CT

Objective To investigate the optimum scanning parameters for multi-slice spiral CT aorta imaging so as to emphasize the contrast delay time and the method of image reconstruction.Methods 21 cases of patients were scanned with contrast enhancement by the method of bolus tracking.The contrast medicine was injected through elbow vein.Flow speed was 3.0ml/s,and contrast usage was 1.5 to 2ml/ kg.Scanning was triggered when the density of the enhancement at ROI arose to a prescribed value.After data acquisition and reconstruction,the aorta was displayed.Results The average time for the influx of contrast in aorta was 20 seconds.Data was reformatted with the thickness of 2.5mm and 50% overlapping.VR,CPR,MIP and SSD images were reconstructed.Conclusion With constant tracking of density by Smartprep software,the optimized scanning parameters and the proper image post-processing techniques,aorta can be clearly displayed.[Chinese Medical Equipment Journal,2008,29(2):88-89]
2.The evaluation of risk factors predicting perioperative mortality in patients with colorectal cancer

Objective To investigate the potential risk factors for the death of patients with colorectal cancer during perioperative periods.Methods The clinical data of 545 patients with colorectal cancer were analyzed retrospectively.17 factors,which may influence mortality of patients with colorectal cancer during perioperative periods were evaluated.These enumeration data were analyzed with x2 test,then those factors with obvious difference in statistics were further analyzed by multi-factor Logistic regression analysis.The independent risk factors affecting the mortality of patients with colorectal cancer during perioperative periods were obtained from the analysis.Results There were 6 independent risk factors derived that had significant impacts on perioperative death that were postoperative complications,operation method,non-planed reoperation,preoperative electrolyte disturbance,preoperative hypoalbuminemia,and age(P < 0.05).Conclusions Many potential factors could affect perioperative mortality of patients with colorectal cancer undergoing surgical procedure.
3.Effects of different concentrations of propofol on apoptosis in hippocampal neurons in neonatal rats in vitro

Objective To investigate the effects of different concentrations of propofol on the apoptosis in hippocampal neurons in neonatal rats in vitro.Methods Primary hippocampal neurons were prepared from newborn Sprague-Dawley rats (aged 7 days) and cultured for 7 days.The neurons were randomly divided into 7 groups (n =8 each):control group (group C),propofol 4,8 and 12 μg/ml groups (groups P1-3),and fat emulsion 4,8and 12 μg/ml groups (groups F1-3).The cells were cultured for 24 h in the culture medium containing propofol 4,8 and 12μg/ml in groups P1-3,respectively.The cells were cultured for 24 h in the culture medium containing fat emulsion 4,8 and 12 μg/ml in groups F1 3,respectively.The cell morphology was examined by microscopy after 24 h culture.The expression of caspase-3 (by immuno-histochemistry) and neuronal apoptosis were detected.The neuronal apoptosis rate was calculated.Results Compared with group C,the neuronal apoptosis rate and caspase3 expression were significantly increased in groups P1-3 (P < 0.05 or 0.01).The neuronal apoptosis rat and caspase-3 expression were increased in a concentration-dependent manner in groups P1-3 (P < 0.05).The neuronal apoptosis rate and caspase-3 expression were significantly lower in groups F1-3 than in groups P1-3 (P < 0.05).There was no significant difference in the neuronal apoptosis rate and caspase-3 expression between groups F1-3 (P > 0.05).The damage to neurons was induced in groups P1-3 and most severe in group P3.Conclusion Propofol can promote the apoptosis in hippocampal neurons in neonatal rats in vitro in a concentration-dependent manner.
5.Immunogenicity of Different Genetic Type Rotavirus NSP4 in Mice

NSP4, as the diarrhea-related protein of rotavirus, is becoming an attractive candidate for vaccine development. To compare the immunogenicity of NSP4 from different genetic groups, we constructed eukaryotic expression plasmids comprising the NSP4 genes from four different genetic types using the pCI vector. The recombinant vectors were designated as pCI-97B6, pCI-97S36, pCI-97S34 and pCI-97SZ8, respectively. Following the conformation of the transient expression of the constructs in 293 cells, the plasmids were respectively subjected to the 5 round i. m. inoculation of BALB/c mice. The specific antibodies against NSP4 as well as the IgG1/IgG2a subclasses of immunoglobulin in mice sera were examined with indirect ELJSA after each immunization. The results showed that the immunization of plasmids expression NSP4s could elicit not only humoral but also cellular immunity, but the humoral immune response is dominant. There is a difference of immunogenecity among the NSP4 of different genetic type. Further studies were needed to focus on the relationship between the immunogenicity and protection effect.
7.Influence of iridosides of cornus officinalis on glomerular mesangial cells cultured with AGEs

Aim To investigate the influence of iridosides of cornus officinalis(ICO) on morphology, cell cycle and oxidative stress of glomerular mesangial cells(GMC) cultured with advanced glycation end products(AGEs). Methods GMC were incubated in culture medium containing AGEs in the presence of ICO and aminoguanidin for 48 hours. At the same time, the control and model groups were added. Then the cultured GMC were stained by mixed fluorescence liquid and observed under fluorescence microscope. Cell cycle of GMC were analyzed using flowcytometry. The content of MDA, activity of SOD and GSH-Px in GMC supernatant were measured. The level of ROS was detected using flowcytometry. Result Morphology analysis showed that the morphology and structure of normal GMC were normal. The structure of most cells in AGEs were unclear, cell counts increased largely and they grow intensively. Cell cycle analysis showed that cell percentage of S phase increased and G_0/G_1 reduced. The level of ROS, MDA remarkably increased, and SOD, GSH-Px activity reduced. When the ICO were added, cell morphology tended to be basically normal and cell counts decreased, the percentale of S phase also decreased. The level of ROS, MDA and the SOD, GSH-Px activity restored in comparison with the model groups. Conclusion ICO can prevent the cultured GMC from lesion caused by AGEs. ICO may protect GMC from AGES to retard the progression of diabetic nepropathy by partially inhibiting the occurrence of oxidative stress.
8.Clonality analysis for differentiating multicentric origin and intrahepatic metastasis in multiple hepatocellular carcinomas

Objective To explore the differential diagnostic significance of clone analysis for multicentric occurrence (MO) and intrahepatic metastasis (IM) in hepatocellular carcinomas (HCCs).Methods Loss of heterozygosity (LOH) and microsatellite instability (MSI) were analyzed by microsatellite polymorphism test and the integration sites of HBV were assessed by Southern blot in each nodule of the HCCs. The clonalities were then compared between each nodule of the same patient and the diagnosis of MO or IM was concluded. Finally, the results based on clonality analysis were compared with those according to clinicopathological and imaging data. Results According to the results of LOH and MSI in 79 nodules and nontumorous tissue from 35 cases of mutiple HCCs, 5 (14.3%)and 29 cases (82.9 %) were divided into MO and IM, respectively. Both MO and IM presented simultaneously in 1 patient (2.9%). The integration sites of HBV could be analyzed in 77 nodules from 34 multiple HCCs. Among them, 6 (17. 6%) and 27 cases (79.4%) were divided into MO and IM, respectively. Both MO and IM presented simultaneously in 1 patient (2.9%). The classification results of microsatellite polymorphism test and HBV integration sites analysis demonstrated a significant positive correlation (rs = 0.909, P<0.001). Nevertheless, neither the classification of microsatellite polymorphism test nor that of HBV integrate sites analysis was correlated with the discrimination according to clinicopathologic and imaging data (rs=0. 133, P=0. 468, rs =0. 262, P=0. 155, respectively). Recurrence in patients in the MO group occurred significantly later than that in IM cases who were diagnosed by clonality analyses (P=0. 001). Conclusion The clonality analysis based on the results of LOH and MSI or assessments of HBV integrate sites is useful for the differential diagnosis of MO and IM and their treatment and prognosis.
9.Application and clinical significance of 24-hour multichannel intraluminal impedance-pH monitoring in gastroesophageal reflux disease

Objective To investigate the application and clinical significance of 24-hour multichannel intraaluminal impedance-pH (MII-pH) monitoring in gastroesophageal reflux disease (GERD).Methods Fiftythree patients with GERD were enrolled in this study according to the Montreal consensus (consulting for twice reflux a week or above) from July 2011 to June 2012.Patients were divided into erosive esophagitis (EE,n =25)group and non-erosive reflux disease (NERD,n =28) group after endoscopy and MII-pH monitoring.Fifteen healthy volunteers were recruited as the normal controls.The change of pH and MII-pH parameters were compared among the three groups and the significance of the change was investigated.Results Twenty-four-hour pH monitoring showed that all the acid reflux events in the EE and the NERD groups were significantly higher than those in the control group (P < 0.05).MII-pH monitoring showed that the frequencies of total reflux,acid reflux,weakly acidic reflux and percentage of acid reflux in the GERD group were higher than in the control group (total reflux:83 (54,118) vs.62 (44,111) vs.42 (20,70),P =0.003 ; acid reflux:45 (25,79) vs.22 (11,45) vs.3(1,10),P =0.000 ;weakly acidic reflux:36(18,47) vs.43(21,82) vs.23(11,43),P =0.001 ;percentage of acid reflux:53% (37%,81%) vs.32% (13%,48%) vs.11% (1%,23%),P =0.002).The frequency and percentage of acid reflux in the EE group were higher than those in the NERD group (P =0.000)The percentage of weakly acidic reflux in the NERD group was higher than in the EE group (66% (43%,79%) vs.46% (21%,57%),P <0.01).The frequencies of liquid reflux and mixed reflux in the GERD groups were higher than those in the control group (22 (12,40) vs.18 (12,26) vs.9 (4,18) ; 54 (39,79) vs.42 (25,77) vs.29(14,48) ;P <0.01).The frequency of gas reflux in the control group was higher than in the GERD group (86(56,207) vs.31 (14,62) vs.34 (15,119),P < 0.01).The frequency and percentage of proximal reflux in the GERD group were significantly higher than in the control group (28(18,41) vs.16(12,34) vs.3 (2,9) ; 33% (22%,49%) vs.29% (22%,35%) vs.11% (6%,22%),P < 0.001).The percentage of symptom positive index in patients with acid reflux,non-acidic reflux and total reflux in the EE group were 36.0% (9/25),20.0% (5/25) and 56.0% (14/25) respectively which were higher than in the NERD group (21.4% (6/28),14.3% (4/28) and 35.7% (10/28)).Conclusion MII-pH monitoring can detect more reflux events.Acid reflux plays an important role in GERD.The detective rate of GERD will be elevated when combined with MII-pH monitoring in the diagnosis.MII-pH monitoring has a distinct advantage in diagnosing GERD.
10.Myocardial ischemia analysis based on electrocardiogram QRS complex in time domain

Objective ST-T complex change, which represents the ventricle repolarization phase, is the main clinical indicator in detecting myocardial ischemia (MI) based on electrocardiogram (ECG) signals.However, its feature point location is not accurate due to interferences. In this paper, a new approach about myocardial ischemia analysis was proposed based on QRS complex. Methods QRS complex, representing the ventricle depolarization process, was used to analyze myocardial ischemia, and some parameters were extracted synthetically in time domain. Then they were used for statistical analysis of myocardial ischemia states and non-myocardial ischemia states. Results Five parameters had significant differences after verification of Non-MI signals in MIT-BIH database and MI signals in long-term ST database (LTST) and they were: QRS upward and downward slopes, transient heart rate, R angle and Q angle in a triangle QRS. Conclusion Five parameters extracted from QRS complex had significant differences. The proposed method provides an important basis for myocardial ischemia detection.
