2.Exploration of Infectivity of HIV in vitro

OBJECTIVE To define the regularity of survival ability of HIV in natural environment,and prevent(infection) through contacting with positive body fluids during daily life or medical work.METHODS Having been diluted by sterile water or 10% serum RPMI 1640 medium,HIV was exposed to 4℃,room temperature(20-26℃) or 37℃ for different period of time.TCID_(50) of these samples was detected.Non-pathological samples were blind passaged for three generations.RESULTS HIV infective ability persisted more than 35 days both in(water) and medium at 4℃;whereas it persisted 7-14 days in water,14-21 days in medium at room temperature and 37℃.CONCLUSIONS HIV has higher resistance in natural environment.To prevent accidental spreading of HIV,HIV positive liquids and contaminants staffs should be treated carefully.
3.Blood hemoperfusion with resin adsorption combined continuous veno-venous hemofiltration for patients with multiple organ dysfunction syndrome

BACKGROUND: Blood hemoperfusion with resin adsorption can clean larger molecules that exceed the molecular weight cutoff of combined continuous veno-venous hemofiltration (CVVH). Hence blood hemoperfusion with resin adsorption combined CVVH (HP+CVVH) has higher ability of mediator clearance, and can improve clinical outcomes in theory. This study aimed to investigate the effect of blood hemoperfusion with resin adsorption combined continuous veno-venous hemofiltration (HP+CVVH) on plasm cytokines like TNF-α, IL-1β, IL-6, cellular immunity and prognosis in patients with multiple organ dysfunction syndrome (MODS). METHODS: This was a prospective, randomized clinical trial. A total of 30 patients who had been diagnosed with MODS were enrolled in this study. Patients were randomly allocated to routine treatment+HP+CVVH group (treatment group) and routine treatment+only CVVH group (control group). In the treatment group, patients received blood hemoperfusion with resin adsorption for 2 hours, and then received CVVH for 10 hours every day. In the control group, patients received CVVH for 12 hours only every day. The patients in the two groups received blood purification therapy for three days. The plasma of patients in the treatment group was obtained at 0, 2, 12, 24, 26, 36, 48, 50, 60 hours, 5th day, 7th day and 10th day, respectively. The plasma of patients in the control group was obtained at 0, 12, 24, 36, 48, 60 hours, 5th day, 7th day and 10th day, respectively. APACHE II score, T-lymphocytes subpopulations, blood lactate acid concentration, heart rate, breathing rate, and oxygenation index were observed. RESULTS: Plasma cytokines like TNF-α, IL-1β, IL-6 decreased markedly after HP (P<0.01);T-lymphocytes subpopulations CD3+, CD4+, CD8+, CD4+/CD8+ increased after HP+CVVH or only CVVH. The plasma concentrations of TNF-α, IL-1β and IL-6 in the two groups were not markedly different at 12, 36, and 50 hours. But on the 5th day, the plasma concentrations of TNF-α, IL-1β and IL-6 in the treatment group were lower than those in the control group (P<0.05). On the 28th day, 5 patients died in the treatment group, and 6 patients in the control group. CONCLUSIONS: Both HP+CVVH and CVVH can clean plasma cytokines like TNF-α, IL-1β, and IL-6, and improve cellular immunity and clinical symptoms and signs of patients. Compared with only CVVH, the plasma concentrations of TNF-α, IL-1β and IL-6 were lower on the 5th day, and patients have an increased survival rate on the 28 day in the HP+CVVH group.
5.Microwave radiation induces injury to GC-2spd cells.

OBJECTIVETo explore the impact of microwave radiation on GC-2spd cells.
METHODSWe exposed cultured GC-2spd cells to microwave radiation at the average power densities of 0, 10 and 30 mW/cm2 for 15 minutes and, from I to 24 hours after the exposure, we observed the changes in cell proliferation, histology and ultrastructure, cell apoptosis, and cAMP content by MTIT, light microscopy, electron microscopy, flow cytometry and ELISA.
RESULTSCompared with the control group, the GC-2spd cells showed a significant decrease in proliferation ability at 1 -24 hours after 10 and 30 mW/cm2 microwave radiation, except at 12 hours after 30 mW/cm2 radiation (P <0.05 or P <0.01), with reduced length and number of cell enation and increased intra cytoplasm vacuoles. The rate of cell apoptosis (%) was significantly increased in the 10 and 30 mW/cm2 groups at 6 hours (4.56 +/- 2.09 vs 14.59 +/- 1.09 and 8.48 +/- 1.73, P <0.05 or P <0.01) , with agglutination and margin translocation of chromatins and obvious dilation of endo cytoplasmic reticula. The cAMP content (nmol/g) in the GC-2spd cells was remarkably reduced in the 10 and 30 mW/cm2 groups at 6 and 24 hours (2.77 +/-0.24 vs 1.65+/- 0. 17 and 1.96+/-0.10, 3.02 +/-0.47 vs 2.13 +/-0.33 and 1.69 +/-0.27, P <0.05 or P <0.01).
CONCLUSIONMicrowave radiation at 10 and 30 mW/cm2 may cause injury to GC-2spd cells, which is manifested by decreased content of intracellular cAMP, reduced activity of cell proliferation, and increased rate of cell apoptosis.

6.Screening and genetic analysis of thalassemia in Sichuan District.

This paper was aimed to screen and analyze the genotypes of thalassemia of the residences in Sichuan Province. The phenotypes of thalassemia was screened by hemoglobin electrophoresis, and the genotypes of the phenotype positive cases was analyzed. There were 69 alpha-thal and 115 beta-thal phenotype positive cases in 3185 subjects. A total of 157 cases were diagnosed as thalassemia. These included 53 alpha-thal, 102 beta-thal and 2 alpha/beta-thal cases. The positive rates were 1.66%, 3.20%, 0.06%, respectively. It can be seen that Sichuan is one of the provinces with a high incidence of thalassemia. Hemoglobin electrophoresis is an important method for thalassemia screening, and GAP-PCR and PCR-RDB technology for thalassemia genotype analysis. A combination of both enables a wide range of screening and diagnosis of thalassemia.

7.Pharmacokinetics of omeprazole in Chinese Muslim and Han nationalities healthy volunteers after a single dose administration

Objective To study the pharmacokinetics of omeprazole tablets in Chinese Hui and Han healthy volunteers.Methods Twenty volunteers were involved in the study,including ten Chinese Hui volunteers and ten Han volunteers.Each healthy volunlteers were given asingle oral dose(40 mg)of omeprazole tablet.The concentration of omeprazole in plasma was determined by RP-HPLC and the pharameters were calculated by DAS 2.0 software.Results The main pharmacokinetic parameters of Chinese Hui and Han were as follows:C_(max) were (941.94±446.08) and (760.49±581.23) μg·L~(-1);t_(max) were(2.7±0.68)and(2.70±0.82)h;t_(1/2) were(2.43±2.83)and(1.60±1.28)h;AUC_(0-12) were (2.29±1.12)and(1.44±7.98)mg·h·L~(-1);AUC_(0-∞) were (2.33±1.11)and(1.47±7.69)mg·h·L~(-1),respectively.Conclusion Individual difference is significant in C_(max),AUC_(0-12),AUC_(0-∞) after an oral omeprazole,but the pharmacokinetic parameters obtained from the results of ststistical analysis show no significant difference in the main pharmacokinetic of omeprazole between Chinese Hui and Han.

9.Influencing factors and status of professional identity among undergraduate nursing students at school

Objective To explore the current situation and influencing factors of professional identity among undergraduate nursing students at school.Methods A survey was conducted with a total sample of 405 undergraduate nursing students at school using self-designed Professional Identity Questionnaire for Nursing Students.Descriptive statistics and Hierarchical multiple regressions were used to analyze the data in this study.Results For undergraduate nursing students at school,the score of professional identity of nursing students was (3.20 ± 0.43).However,internal distribution was unbalanced,with higher professional expectation and lower professional cognition.Students' gender (β =-0.243,t =-11.015 ;P < 0.05) and career choice reason (β =0.027,t =13.009 ; P < 0.05) were the influencing factors,with an overall of 50.8% of explained variance in professional identity.Whether was only child or cadre students,and what grade were no significant influencing for professional identity of students.Conclusions The results indicate that professional identity education should be implemented in each stage of nursing education.Nursing administrators should carry out the professional identity education as a systemic engineering combing macro-control with micro-treatment,analyze related reasons and adopt different measures aiming at different gender and career choice,conduct professional thought and emotional education to promote students' professional identity.
10.Application and effect of experiential services of nursing in clinical practice

Objective The purpose of this study was to explore the application effect of experiential services in clinical nursing.Methods A total of 161 nurses were selected and trained as experience member to play the role of patients' family members.And all of them were dispersed to each ward to collect the patient's or family member's feelings and their needs for nursing service,and to find out the limitations in nursing service.The Nurses Clinic Communication Comptency Scale was used to evaluate nurses' communication skills,and 172 patients were selected to survey their satisfactions.Results The total average score of clinical nurse's communication skills was (3.12 ±0.79) before implementing the experiential service,and the data was (3.29 ± 0.68) after implementing the experiential service,the difference was statistically significant (t =2.069,P < 0.05).The scores of patients' satisfaction with nurses' technology,nurses' services and hospital nursing before implementing the experiential services were all higher than those before implementing the experiential services [(3.99 ±0.78) vs (3.81 ±0.94),(4.26 ± 1.13) vs (3.85 ± 0.97),(4.13 ± 1.03) vs (3.83 ±0.95),respectively],these differences were statistically significant (t =1.933,3.611,2.808,respectively; P < 0.05).Conclusions The experiential services can improve nurse services and communication skills,which can help enhance inpatient satisfaction and promote the overall image of the hospital.
