1.Expression of cyclooxygenase-1 in ovarian cancer and its significance

Objective To explore the expression of cyclooxygenase-1 in ovarian cancer and its significance. Methods Expression of COX-1 and CA125 was detected by immunohistochemistry in 37 cases of ovarian cancer and 31 cases of ovarian cyst. The serum level of COX-1 and CA125 was tested in 40 cases of ovarian cancer and 31 cases of ovarian cyst and 60 healthy volunteers by ELISA. Results The positive expression of COX-1 and CA125 was 78% and 57% in ovarian cancer and 3% and 7% in ovarian cyst,respectively. The expression of COX-1 and CA125 was 65% and 93% at the serum level. Conclusion Expression of COX-1 could act as an auxiliary diagnostic criterion in ovarian carcinoma.
2.Study on the chemoresistance of CD44+/CD24+ Siha cells to cisplatin and its mechanisms

Background and purpose:One of the reasons why cancer cells are resistant to chemotherapy is the existence of cancer stem cells. The purpose of this study was to investigate the chemoresistance of CD44+/CD24+ Siha cells to cisplatin and its mechanisms.Methods:Siha cells were cultivatedin vitro. The CD44+/CD24+ Siha cells were sorted out by fluorescence activated cell sorter (FACS) andin vitro proliferation was detected by MTT assay after treatment with the different concentrations of cisplatin. The cell apoptosis rate was detected by flow cytometry after 10 μg/mL cisplatin acted on CD44+/CD24+ Siha cells for 24, 48 and 72 h. The relative mRNA and protein expressions of Bcl-2, Oct-4 and ABCG2 were detected by quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot, respectively. Results:The survival rates of CD44+/CD24+ Siha cells treated with different concentrations of cisplatin (0.1, 1, 5, 10, 15 and 20 μg/mL) were higher than those of their parental Siha cells [(88.42±1.51)%vs (92.87±1.5)%, (79.94±1.05)%vs (84.72±1.09)%, (69.78±0.81)%vs (75.13±2.86)%, (58.97±0.70)%vs (65.79±2.71)%, (49.60±0.88)%vs (52.10±0.52)%, (45.13±0.69)%vs (48.84±1.02)%,P<0.05]. Compared with their parental Siha cells, the apoptosis rates of CD44+/CD24+ Siha cells were lower after 10 μg/mL of cisplatin acting on them for 24, 48 and 72 h, respectively [(3.05±0.16)%vs (5.17±0.27)%, (17.94±2.02)%vs (32.60±4.28)% and (40.14±3.01)%vs (56.62±5.32)%,P<0.05]. The results from both qRT-PCR and Western blot indicated that Oct-4, ABCG2 and Bcl-2 were highly expressed on CD44+/CD24+ Siha cells. A significant difference was found in Oct-4, ABCG2 and Bcl-2 expression between CD44+/CD24+Siha cells and their parental cells (P=0.015<0.05).Conclusion:CD44+/CD24+ Siha cells could be resistant to apoptosis induced by cisplatin and expressed high levels of cancer stem cell markers such as Oct-4 and ABCG2. This study lays the basis for useful isolation and further targeted therapy of cervical cancer stem cells.
3.Resistance of CD44+/CD24+ expressing cervical cancer cells on apoptosis induced by X-ray irradiation

Objective To explore whether CD44 +/CD24 + expressing cervical cancer cells are resistant to X-ray irradiation and investigate the underlying mechanism.Methods Cervical cancer cell line (Siha) was cultured in vitro and the CD44 +/CD24 + expressing cells were sorted with a flow cytometer.The cells were irradiated with 8,16 and 30 Gy of 6 MV X-rays.Colony formation test was used to evaluate the radiosensitivity of CD44 +/CD24 + expressing cervical cancer cells.Cell morphology was observed by electronmicroscopy,cell apoptosis was analyzed with a flow cytometer and also verified with a DNA ladder assay.Gene expression was determined by RT-PCR.Results After radiation,the ratio of CD44 +/CD24 + cells significantly increased.Compared to Siha cells,the radiosensitivity of CD44 +/ CD24 + cells decreased (t =93.99-400.45,P <0.05),and the expressions of bcl-2,survivin and Oct4 mRNA increased in CD44 +/CD24 + cells (t =221.35,941.65,82.27,P <0.01).Both apoptotic body and specific DNA ladder pattern were observed in cells but not in the CD44 +/CD24 + Sihacells which had no obvious morphological changes of apoptosis.Conclusions The CD44 +/CD24 + expressing cervical cancer cells are resistant to X-rays due to expression of anti-apoptosis factors.
4.Effect of Simvastatin on Severe Complications of Subarachnoid Heamorrhage

ObjectiveTo investigate the effect of simvastatin on severe complications and prognosis of subarachnoid heamorrhage (SAH).Methods98 cases with SAH were randomly divided into the treatment groups and control group (finally, there were 32 cases in treatment group, 48 cases in control group). Patients in treatment group were given simvastatin 20 mg/day, and those in control group were treated with routine therapy. The incidences of cerebral vasospasm, hydrocephalus, rebleeding and mortality between the two groups were compared.ResultsThe incidence of hydrocephalus of treatment group was 3.13%; that of control group was 18.7%, there was a significantly difference between two groups (P<0.05). There were no significant differences for incidences of cerebral vasospasm, rebleeding and mortality between two groups (P>0.05).ConclusionSimvastatin can reduce the occurrence of hydrocephalus after SAH.
5.Flavonoids from the seeds of Alpinia galanga Willd.

Ten flavonoids were isolated from the 95% ethanol extract of the seeds of Alpinia galanga Willd. with a combination of various chromatographic techniques, including silica gel, Sephadex LH-20 and preparative HPLC. On the basis of spectroscopic data analysis, they were elucidated as (2R, 3S)-pinobaksin-3-cinnamate (1), (2R, 3R)-pinobaksin-3-cinnamate (2), pinocembrin (3), pinobaksin (4), 3-O-acetylpinobaksin (5), galangin (6), galangin-3-methylether (7), kumatakenin (8), 3-methylkaempferol (9) and (2R, 3R)-3, 5-dihydroxy-7-methoxyflavanone (10). Among them, compound 1 is a new compound, compounds 2, 5 and 10 were isolated from the genus Alpinia for the first time, and others were isolated from this plant for the first time.

6.Effect of nordy on biological behaviors of malignant glioma cell line U87MG and the analysis of differential expression proteome.

OBJECTIVETo explore effects of nordy on biological behaviors of human malignant glioblastoma cell line U87MG in vitro and transplanted tumor in vivo, and to identify the differential proteome upon Nordy induced differentiation.
METHODSGlioblastoma U87MG cells were induced to differentiate by synthetic lipoxygenase inhibitor, Nordy. The drug was also given via peritoneal injection to nude mice (27 mg/kg body weight) bearing orthotopic transplanted tumors of U87MG cells in the brain. The tumor volumes and GFAP expression were measured. Total proteins of U87MG cells after Nordy treatment were analysed by two-dimensional gel electrophoresis. PDQuest 7.1 computer software was used to compare protein profiles of the treated cells with that of untreated control. Differentially expressed proteins were then selected and characterized by matrix assisted laser desorption ionization-time of flight-mass spectrometry. The functional aspects of these proteins were analyzed by bioinformatics.
RESULTSNordy suppressed both the proliferation of U87MG cells in vitro and the tumor growth of orthotopic transplanted tumors in vivo (P < 0.01). The differentially expressed proteins induced by Nordy included proliferation-associated gene A, alternative splicing factor ASF-3, eukaryotic translation initiation factor 5A, coffilin 1 (non-muscle), beta galactoside binding lectin, glyceraldehyde-3-phosphate dehydrogenase, enolase 1 and an unknown protein.
CONCLUSIONSNordy promotes the differentiation of glioblastoma cells, by which it may serve as a therapeutic agent. Various proteins identified during Nordy-induced differentiation are involved in the cell proliferation, metabolism, differentiation, apoptosis and gene transcription.

7.Analysis of liver function and myocardial enzyme level of 395 cases with infantile rotavirus enteritis

Objective To detect the damage incidence rate of liver and myocardium in infantile rotavirus (RV) enteritis.And to provide the basis for prevention and cure of liver and myocardium damage in infantile RV enteritis.Methods The liver function and myocardial enzyme detection results of 395 patients with infantile RV enteritis were collected .These results were compared with those of 40 healthy kids from health examinations . Results The levels of alanine aminotransferase ( ALT ) , aspartate amino transferase ( AST ) , serum creatine kinase (CK) and serum creatine kinase isozyme(CK-MB) from infantile patients were (34.49 ±29.13)U/L,(52.44 ± 24.10)U/L,(141.75 ±132.22)U/L and (48.69 ±32.53)U/L,respectively,which were higher than those of the healthy control [(16.00 ±3.24)U/L,(29.90 ±3.76)U/L,(101.82 ±64.56)U/L and (22.32 ±8.98)U/L,t=4.008,5.901,3.982,5.64,all P<0.05].The abnormal occurrence rates of ALT ,AST,CK and CK-MB in infantile patients were 19.49%,73.16%,12.15%,73.16%,respectively,which were higher than those in healthy controls (2.50%,20.00%,0.00%,0.00%),the differences were statistically significant (χ2 =7.128,47.397,5.464, 300.239,all P<0.05).The incidence rates of liver and myocardium damage were 19.49%and 73.16%,respectively. Conclusion There are higher incidence rates of liver and myocardium damage in infantile RV enteritis .So for the infantile RV enteritis patients ,it is necessary to detect the liver function and myocardial enzyme level .The treatment for infantile RV enteritis should be included liver and myocardial protection except antivirus and correcting dehydration .
8.A matched case-control study of papillary thyroid carcinoma.

OBJECTIVEIn order to explore the risk factors of the papillary thyroid carcinoma(PTC), a hospital-based matched case-control study was carried out in Shanghai.
METHODSMatched by gender, age (+/- 3 years old) and native place,205 pairs of cases and controls were recruited and studied. Database was established with Epi Info 6.0 software. Univariate and multivariate conditional logistic regression analysis were carried out with SPSS 11.5 software.
RESULTSData from multivariate conditional logistic regression analysis showed that often drinking tea and having better economic living standard 20 years ago were protective factors of PTC, with ORs of 0.456 and 0.221 respectively. Personal history of CT examination, familial history of cancer and the mother's age was older than 22 years when the research subject was delivered, were risk factors of PTC, with ORs of 12.935, 7.027 and 3.729 respectively. Results data from multivariate analysis on female subjects showed that the history of CT examination, mother's age was older than 22 (when the subject was delivered) and the history of gynecological disease were the risk factors of PTC,with ORs of 107.453,29.246 and 59.521 respectively. Taking bean products frequently and having higher standard of living 20 years ago were the protective factors of PTC,with ORs of 0.025 and 0. 144 respectively.
CONCLUSIONHistory of CT examination, familial history of cancer and the mother's age(older than 22 when the subject was delivered) were the risk factors of PTC. Frequent tea drinking habit and having higher standard of living 20 years ago were the protective factors of PTC. History of CT examination,mother's age (older than 22 when the subject was delivered) and the history of gynecological disease appeared the risk factors of PTC for women. Habit on frequent bean products taking and had higher standard of living 20 years ago were the protective factors of PTC to women.

9.Genetic polymorphisms in X-ray repair cross-complementing gene 1 and susceptibility to papillary thyroid carcinoma.

OBJECTIVETo study the association of polymorphisms in the X-ray repair cross-complementing gene 1 (XRCC1) and papillary thyroid carcinoma (PTC).
METHODSA hospital based, matched case-control study was carried out. The polymorphisms in XRCC1 for 105 pairs of cases with PTC and controls were identified by PCR-RFLP.
RESULTSThe frequencies of Arg/Arg, Arg/Trp and Trp/Trp genotypes at XRCC1 Arg194Trp site were 47.6%, 49.5% and 2.9% among cases compared to 45.7%, 48.6% and 5.7% among controls. There was no statistically significant difference between the two groups (chi(2) = 1.07, P = 0.59). The frequencies of Arg/Arg, Arg/Gln and Gln/Gln genotypes at XRCC1 Arg399Gln site were 46.7%, 41.9% and 11.4% among cases, while 54.2%, 42.9% and 2.9% among controls respectively. There was statistically significant difference between the two groups (chi(2) = 6.40, P = 0.04). Individuals with Gln/Gln genotype had a 3.65-fold increased risk of developing PTC compared to Arg/Arg genotype (OR = 4.65, 95% CI: 1.24 - 17.45). The multivariate conditional logistic regression analysis showed that the XRCC1 Arg399Gln polymorphism, negative life events and X-irradiation history were associated with PTC, with odds ratios of 2.71 (95% CI: 1.22 - 6.05), 5.34 (95% CI: 1.40 - 20.38) and 0.38 (95% CI: 0.12 - 0.72) respectively. However, XRCC1 Arg194Trp polymorphism, drinking tea, fruit and economic levels did not show statistically significant associations with PTC.
CONCLUSIONThe Gln/Gln genotype at XRCC1 Arg399Gln site and negative life events significantly increased while X-irradiation history decreased the risk of developing PTC.

10.Study for association between high risk human papillomavirus and cervical lesions in the samples from opportunistic screening.

OBJECTIVETo investigate the association between HPV genotypes and cervical lesion in Hybrid Capture 2 (HC2) HPV test positive samples.
METHODS602 cervical samples randomly obtained detected as HPV positive by the HC2 high-risk probe cocktail were determined by polymerase chain reaction (PCR)-reverse dot blot (RDB), among them 344 participated Thinprep Cytology test.
RESULTS569 (94.5%) samples were successfully amplified. The most common HPV genotypes were HPV16 (31.6%), 52 (16.7%), 58 (15.1%), 56 (8.1%), 39 (7.9%); HPV16 or HPV16/18 infection was significantly related to ASCUS, LSIL and HSIL; 24 samples suggested single HPV infection with the genotypes not available for HC2.
CONCLUSIONThe most common HPV genotypes in random screening were HPV16, 52 and HPV58; Our data demonstrated the pseudo-positivity of HC2 test was 4.0%; HPV16 was a index for progression of HSIL.
