1.Effects of curcumin on the expressions of NR2A,NR2B and apoptotic neurons in hippocampus after ischemia-reperfusion in rats

Aim To investigate the effect of curcumin on the expressions of NR2A, NR2B and apoptotic neurons in hippocampus after global brain ischemia-reperfusion in SD rats. Methods Global ischemic model induced by 4-vessel occlusion was adopted. Rats were randomly divided into 5 groups: sham group (SH), ischemia-reperfusion group (I/R), curcumin 30(Cur 30), 100 (Cur 100) and 300 mg?kg-1 group (Cur 300). At different time points after ischemia, animals were designated as subgroups 2 h, 6 h, 1 d, 3 d, 7 d. Cell morphology was observed on HE stain slides. The apoptotic neurons were detected in CA1 region by TUNEL method. Expressions of NR2A and NR2B in hippocampal CA1 and CA3/DG regions were detected by Immunohistochemical technique. Results At each time point the number of apoptotic neurons was much more in I/R group than that in SH group and Cur 100 group,and in Cur 300 group than that in Cur 100 group. The expression of NR2A was higher in the hippocampal CA1 and CA3/DG regions in Cur 100,Cur 300, Cur 30 and SH groups than that in I/R group at each time point(P
2.Expression of apoiipoprotein H in childhood primary nephrotic syndrome

Objective To study the expression of apolipoprotein H (ApoH) in childhood primary nephrotic syndrome (PNS) and to discuss its role in PNS. Methods Immunohistochemistry staining and real-time quantitative polymerase chain reaction(RT-PCR) were performed to evaluate the expression of ApoH in renal tissues of 78 patients with PNS and 14 cases of normal controls. Serum albumin, serum lipid, proteinuria and urinary retinol binding protein (RBP) were tested before renal biopsy in all patients. Tubulointerstitial lesions were investigated. Results (l)There was positive expression of ApoH in renal tissues of PNS patients and normal controls,mainly in the proximal tubules by immunohistochemistry staining. ApoH mRNA was also shown in all renal tissues by RT-PCR. ApoH protein expression was positively correlaed with its mRNA expression(r=0.264, P 0.05) whereas these data displayed no significant difference between two groups. 3.Changes and clinical significance of serum 25-hydroxy-vitamin D levels in rheumatoid arthritis

Objective To determine the serum level of 25-hydroxyvitamin D[25(OH)D] in rheumatoid arthritis (RA) patients and to assess the association of 25(OH)D with clinical presentations.Methods Serum 25(OH)D levels were detected by enzyme-linked immunosorbent assay (ELISA) in 130 cases with RA and 80 healthy controls.The detailed clinical data of the RA patients were recorded and bone mineral density (BMD) were measured by dual-energy X-ray absor-ptiometry (DXA).Sharp score of both hands were measured for evaluating the effects of 25(OH)D on bone erosion.T-test and one-way ANOVA test were used for data analysis,and x2 test was used to compare the differences between groups.Pearson's test was adsopted for correlation analysis.Muhi-variate analysis and Logistic analysis were carried out for risk factors identification.Results ① The serum levels of 25 (OH)D were markedly lower in the RA group than the control group [(17±6) ng/ml vs (23±6) ng/ml,t=-6.624,P<0.01],while cases of 25(OH)D insufficiency/deficiency in the RA group were more than the control group (98.5% vs 81.5%,x2=26.291,P<0.01); ② Negative correlation was detected between 25(OH) D levels and the following:duration of morning stiffness,tender joint count (TJC),swollen joint count (SJC),erythrocyte sedimentation rate (ESR),C-reactive protein (CRP),health assessment questionnaire (HAQ) of the patients with RA (r=-0370,-0.307,-0.243,-0.369,-0.175,-0.381,both P<0.05),respectively; ③ 25 (OH)D levels were signific-antly lower in group with moderate and severe disease activity than in group with stable or low disease activity (both P<0.05); Insufficiency/deficiency of 25 (OH)D was the risk factor for disease activity by multiple regression analysis (b=-0.46,P=0.029); ④ No statistically significant association was detected between 25(OH)D and degree of bone erosion in RA (P>0.05); ⑤ BMD was classified into three groups:normal,osteopenia and osteoporosis,and significant differences of serum 25(OH)D levels were found by compared with each group (P<0.01).Normal serum 25 (OH)D level was a protective factor for RA-induced osteoporosis by Logistic rcgression analysis (OR=0.898,95%CI 0.830-0.972,P=0.008).Conclusion Significantly low 25 (OH)D level could be found in patients with RA.Negative correlation is detected between 25 (OH)D level and disease activity and osteoporosis respectively in patients with RA.Insufficiency/deficiency of 25 (OH)D is the risk factor for disease activity and RA-induced osteoporosis.

5.Effect of nano-SiO2 exposure on spatial learning and memory and LTP of hippocampal dentate gyrus in rats.

OBJECTIVETo study the effect of nano-SiO2 on spatial learning and memory.
METHODSTwenty-four male rats were randomly divided into 3 groups: control group (C group), low dose group (L group) and high dose group (H group). The rats were intragastrically administrated with nanometer particles at 25 and 100 mg/kg body weight every day for 4 weeks. After exposure, the ability of learning and memory of rats was tested by Morris water maze, and electrophysiological brain stereotactic method was used to test long-tear potentiation (LTP) in dentate gyrus (DG) of the rats.
RESULTSThe increase rate of body weight in H group was reduced significantly compared with C group ( P < 0.05). In the space exploration experiment of Morris water maze test, the escape latency of H group was longer than that of C group (P < 0.05). The rats of H group spent less time in finding the target quadrant (P < 0.05) . The rate of LP induction of H group was significantly lower than that of C group (P < 0.05). After high fre quency stimulation (HFS), The changes of amplitude of population spike (PS) of L group and H group were lower than those of C group significantly (P < 0.05, P < 0.01).
CONCLUSIONNano-SiO₂may result in impairment of spatial learning and memory ability by reducing the rate of LTP induction and the increase of PS in hippocampus.

Objective To prepare the rabbit abdominal aorta atheromatous plaque model ,and to monitor its forming process by ultrasound .Methods Totally 60 Japanese male white rabbits(mdel group ,dead 6 rabbits) fed by high fat diet and the abdominal a‐orta atheromatous plaque formation process was monitored by ultrasound ,20 normal rabbits were taken as control .The abdominal aorta atheromatous plaque was finally confirmed by pathology .Results 52 rabbits in the model group were successful in preparing the abdominal aortic plaque model .The thickness of intima‐media complex was obviously higher than that of the control group .Con‐clusion High fat diet is an effective method for preparing the rabbit atherosclerosis model .The arterial atheromatous plaque forma‐tion is the typical characteristic of atherosclerosis .The high frequency ultrasound can better evaluate the formation process and con‐dition of rabbit abdominal aorta atheromatous plaque .
7.Study on quality standard for Humulus scandens.

To control the quality of Humulus scandens, the quality standard was established in this study. According to the method recorded in the Appendix of Chinese Pharmacopoeia (2010 Edition) , the water and ash inspections were carried out. The component luteoloside and cosmosiin in Humulus scandens were identified and assayed by TLC and HPLC. The results showed a strong characteristics microscopic of Humulus scandens, and trichoromethane-methanol-formic acid (10: 3: 0. 3) as the mobile phase of TLC, the spots at 365 nm with a UV lamp was clear. The 16 batches of samples were analyzed by HPLC with a gradient elution of acetonitrile and phosphate solution (0.2%) at a flow rate of 1.0 mL · min(-1) and detected at 350 nm. The content of luteoloside was 0.015%- 0.651% (average 0.148%); the content of cosmosiin was 0.003%-0.118% (average 0.036%). The linear calibration curve of luteoloside and cosmosiin was acquired in the ranges of 0.011-0.364 g · L(-1) (r = 1.000 0) and 0.003-0.096 g · L(-1) (r = 1.000 0), respectively. The average recovery was 100.5% and 98.5%, respectively. The methods are convenient and reliable, which can be ap- plied for quality assessment of Humulus scandens.

8.Population pharmacokinetic modeling and evaluation of propofol from multiple centers.

In order to successfully develop the effective population pharmacokinetic model to predict the concentration of propofol administrated intravenously, the data including the concentrations across both distribution and elimination phases from five hospitals were analyzed using nonlinear mixed effect model (NONMEM). Three-compartment pharmacokinetic model was applied while the exponential model was used to describe the inter-individual variability and constant coefficient model to the intra-individual variability, accordingly. Covariate effect including the body weight on the parameter CL, V1, Q2, V2, Q3 and V3 were investigated. The performance of final model was assessed by Bootstrapping, goodness-of-fit and visual predictive checking (VPC). The context-sensitive half-times and the infusion rates necessary to maintain the concentration of 1 microg x mL(-1) were simulated to six subpopulations. The results were as follows: the typical value of CL, V1, Q2, V2, Q3 and V3 were 0.965 x (1 + 0.401 x VESS) x (BW/59)(0.578) L x min(-1), 13.4 x (AGE/45)(-0.317) L, 0.659 x (1 + GENDER x 0.385) L x min(-1), 28.8 L, 0.575 x (1 + GENDER x 0.367) x (1 - 0.369 x VESS) L x min(-1) and 196 L respectively. Coefficients of the inter-individual variability of CL, V1, Q2, V2, Q3 and V3 were 29.2%, 46.9%, 35.2%, 40.4%, 67.0% and 49.9% respectively, and the coefficients of residual variability were 24.7%, 16.1% and 22.5%, the final model indicated a positive influence of a body weight on CL, and also that a negative correlation of age with V1. Q2 and Q3 in males were higher than those in females at 38.5% and 36.7%. The CL and Q3 were 40.1% increased and 36.9% decreased in arterial samples compared to those in venous samples. The determination coefficient of observations (DV)-individual predicted value (IPRED) by the final model was 0.91 which could predict the propofol concentration fairly well. The stability and the predictive performance were accepted by Bootstrapping, the goodness-of-fit and VPC. The context-sensitive half-times and infusion rates necessary to maintain the concentration of 1 microg x mL(-1) were different obviously among the 6 sub-populations obviously. The three-compartment model with first-order elimination could describe the pharmacokinetics of propofol fairly well. The involved fixed effects are age, body weight, gender and sampling site. The simulations in 6 subpopulations were available in clinical anesthesia. The propofol anesthesia monitor care could be improved by individualization of pharmacokinetic parameter estimated from the final model.

10.The Synchronous Detection and Significance of β-Glucuronidase Both in Serum and Tumorous Tissue in Non-Hodgkin Malignant Lymphoma Patient

Objective: Our purpose was to explore the change regularity of β-glucuronidase (β-G) in body of patients with Non-Hodgkin malignant lymphoma. Methods: β-G was examined synchronously both in the serum and in the tumor tissue of 13 cases patient with Non-Hodgkin malignant lymphoma by using the method of enzymlinked immunsorbent assay (ELISA) and immunohistochemistry separately. Among them, 3 cases were studied by using the immuno electron microscopic technique. Results: β-G was highly expressed both in the serum and tumorous tissue in patients with non-Hodgkin malignant lymphoma and there was obviously difference as compared with the control group (P<0.01). Conclusion: The combined detection with functional and morphological methods to β-G, it may be assistant target to early discovery and early diagnosis of Non-Hodskin malignant lymphoma.
